Effect of Increased Intracellular pH by HMA on the Cytotoxicity of Combined Treatment of Hyperthermia and Chemotherapy (HMA의 세포내 산도증가가 온열과 항암화학 치료의 세포독성에 미치는 효과)

  • Purpose: The enhanced cytotoxic effect of combined treatment of hyper-thermia and chemotherapy by increasing intracellular acidity with HMA was investigated. Materials and Methods: FSall tumor cells were injected on the hindlegs of female $C_3H$ mice. When the tumor volume reached about 200mm3, experiments were performed on the groups classified as follows: Group I :Control, Group II : Melphalan alone (2.5mg/kg, 5mg/kg, 10mg/kg, 15mg/kg), Group III : Heat alone $(42.5^{\cdot}C$ for 1 hour) Group IV : Melphalan + Heat $(42.5^{\cdot}C$ for 1 hour), Group V : HMA(10mg/kg) + Melphalan(5.0mg/kg) + Heat$(42.5^{\cdot}C$ for 1hour). Each group included 8-12 mice on each experiment HMA (3-amino-6-chloro-5-(1-homopiperidyl )-N-(diaminomethylene) -c-pyrazinecarboxamide), an analog of amiloride which increases intracellular pH(pHi) was dissolved in dimethyl sulfoxide (DMS) and injected into the tumor-bearing mice through the tail vein. 10mg/kg of HMA and each dose of melphalan were injected into peritoneum of the tumor-bearing mice 30 minutes before heating. Tumor growth delay was calculated when the tumor volme reached at $1500mm^3$ Excision assay was performed on each group and repeated 2-4 times. Results : Tumor growth delay of each experimental groups at $1500mm^3$ were 9, 10, 13 and 19 days respectively. In vivo-in vitro excision assay using FSall tumor cells, the cytotoxicity of each experimental groups was $1.2{\times}10^7,\;1{\times}10^7,\;6{\times}10^6,\;1.7{\times}10^6\;and\;1{\times}10^5$ clonogenic cells/gm respectively When HMA was added to the combined treatment of heat and .chemotherapy, the tumor growth was delayed more than combined treatment without HMA i.e., 6 days tumor growth delay at $1500mm^3$ of tumor volume. Conclusion: The combined effect of cytotoxicity by heat and chemotherapy can be much more enhanced by HMA.

The effect of Puerariae Radix on ovariectomy-induced bone loss in C3H/HeN mice

  • Osteoporosis is a major worldwide public health problem that poses a great economic burden to society. Puerariae Radix, the dried root of Pueraria lobata (Wild.) Ohwi, has been widely used in Asia. This study investigated the effects of Puerariae Radix (PR) on bone loss in ovariectomized (OVX) mice. C3H/HeN mice (10 weeks old) were divided into sham and OVX groups. The OVX mice were treated with vehicle, $17{\beta}-estradiol$ ($E_2$), PR (oral administration, 250 mg/kg/day) or PR (intraperitoneal administration, 50 mg/kg/every other day) for 6 weeks. Grip strength, uterus weight, serum alkaline phosphatase (ALP), estradiol concentration and osteoclast surface levels were measured. Tibiae were analyzed using microcomputed tomography. There were no significant differences in the degree of grip strength, body weight and uterine weight between OVX group and PR-treated group. As compared with the OVX group, the serum estradiol levels were significantly increased in the PR-treated group. PR (i.p.) significantly preserved trabecular bone volume, trabecular bone number, structure model index and bone mineral density of proximal tibiae metaphysic. The administration of PR lowered serum ALP and osteoclast surface levels in OVX mice, suggesting that PR can reduce the bone turnover rate in mice. The results indicate that the supply of PR can prevent OVX-induced bone loss in mice.

  • Gymnophclloides seoi is a human intestinal trematode prevalent on southwestern islands in Korea. In the present study, we investigated whether G. seoi metacercariae can grow and develop into adults by in vitro cultivation. The metacercariae were obtained from naturally infected oysters, and cultured in uitro for 5 days under three conditions; $37^{\circ}C/5%{\;}CO_2,{\;}41^{\circ}C/8%{\;}CO_2,{\;}or{\;}41^{\circ}C/15%{\;}CO_2$, in NCTC 109 complete media containing 20% FBS and 1% antibiotics-antimycotics. The degree of worm growth and development was compared with that grown in uiuo of C3H mice. The length of the worms cultivated in uitro was $200-300{\;}\mu\textrm{m}$, not significantly different from metacercariae, whereas the length of the worms recovered from C3H mice was significantly larger, $300-400{\;}\mu\textrm{m}$. The worms produced eggs when grown in C3H mice or cultured in vitro for 2 days under $41^{\circ}C/8%{\;}CO_2{\;}or{\;}41^{\circ}C/5%{\;}CO_2$, but not when cultured under 37$^{\circ}C/5%{\;}CO_2$. Among the in vitro conditions, $41^{\circ}C/15%{\;}CO_2$ was best for egg Production, although the number of eggs was about half of worms obtained from C3H mice. In conclusion, in vitro cultivation of G. semi metacercariae into egg-pioducing adults was partially successful under culture conditions of $41^{\circ}C/5%{\;}CO_2{\;}or{\;}41^{\circ}C/8%{\;}CO_2$.

Multiphasic Analysis of Growth Curve of Body Weight in Mice

  • The present study describes the analysis of the multiphasic growth function (MGF) to body weight in laboratory and wild mice. Three genetic groups of laboratory mice (Mus musculus domesticus) designated $CF_{{\sharp}1}$, C3H/HeNCrj and C57BL/6NCrj, and a genetic group of Yonakuni wild mice (Mus musculus molossinus yonakuni, Yk) were used. Mean body weights of each genetic group-sex subclass from birth to 69 days of age taken at 3-day intervals were analyzed by a monophasic, diphasic and triphasic functions for describing growth patterns. A comparison among the three functions of the MGF was based on the goodness-of-fit criteria: residual standard deviation (RSD), adjusted R-square (Adj $R^2$) and Akaike's information criterion (AIC). Result of this study indicated that body weight averaged heavier for males than for females. Among the four genetic groups within both sexes, $CF_{{\sharp}1}$ showed the highest, subsequent followed by C3H/HeNCrj, C57BL/6NCrj and Yk. Comparison among the three functions revealed that the triphasic function was the best fit to growth data, with the lowest RSD, the highest Adj $R^2$ and the lowest AIC, for the four genetic groups. For the triphasic function, RSD within each genetic group-sex subclass was similar for males and females. Adj $R^2$ was 0.999 for all genetic group-sex subclasses. AIC for laboratory mice males and females ranged from -70.48 to 66.50 and from -92.81 to -68.64, respectively; whereas for Yk wild mice males was -74.29 and females -78.42.

Effect of Korean Red Ginseng on radiation-induced bone loss in C3H/HeN mice

  • This study investigated the effects of Korean Red Ginseng (KRG) on radiation-induced bone loss in C3H/HeN mice. C3H/HeN mice were divided into sham and irradiation (3 Gy, gamma-ray) groups. The irradiated mice were treated for 12 wk with vehicle, KRG (per os, p.o.) or KRG (intraperitoneal). Serum alkaline phosphatase (ALP), tartrate-resistant acid phosphatase, estradiol level, and biomechanical properties were measured. Tibiae were analyzed using micro-computed tomography. Treatment of KRG (p.o., 250 mg/kg of body weight/d) significantly preserved trabecular bone volume, trabecular number, structure model index, and bone mineral density of proximal tibia metaphysic, but did not alter the uterus weight of the mice. Serum ALP level was slightly reduced by KRG treatment. However, grip strength, mechanical property, and cortical bone architecture did not differ among the experimental groups. The results indicate that KRG can prevent radiation-induced bone loss in mice.

Efficient Expression of hG-CSF cDNA from an IRES-Dependent Bicistronic Vector Targeted to Mammary Gland of Transgenic Mice

  • Previously, we observed high level expression of goat β-casein/genomic hGH fusion gene in mammary gland of transgenic mice. To develop an expression vector to make a human granulocyte-colony stimulating factor (hG-CSF) protein efficiently produced in milk of transgenic animals, we designed a new bicistronic vector using the goat β-casein/genomic hGH fusion gene as regulation sequences for expression and internal ribosome entry site (IRES) as a mediator for second gene expression. This vector was constructed by insertion of encephalomyocarditis virus (EMCV) IRES-dependent second gene region coupled with hG-CSF cDNA into 3' untranslated region of an intact hGH gene. By microinjcetion, four transgenic mice were generated and three of them transmitted the bicistronic vector to their progeny. (omitted)

Effect of Hwangryunhaedok-tang Extracts on DNCB-induced Allergic Contact Dermatitis (DNCB로 유발된 알레르기성 접촉피부염에 황련해독탕이 미치는 영향)

  • Objectives : HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-inflammatory effect of HRHDT, we treated HRHDT-skin in Balb/c mice model induced contact hypersensitivity. Methods : Contact hypersensitivity, a local inflammatory respinse of skin, was induced by spreading the back skin of Balb/c mice with 1% DNCB. HRHDT-skin was prepared by dissolving 3% 1,3-butylene glycol extract of HRHDT in solution and treated 2 weeks on the back skin. Results: HRHDT-skin significantly reduced TEWL and erythema by 0.4-1% of DNCB treatment compared with control group. HRHDT-skin reduced IgE on serum obtained from blood of DNCB-treated Balb/c mice. Histopathological examination showed that thickening of the epidermis, hyperkeratosis and the infiltration of inflammatory cells were found in Balb/c mice under conventional circumstances. Conclusions : These results showed that HRHDT-skin could be used as a pharmaceutical material with antiinflammatory effects by reducing IgE in contact hypersensitivity dermatitis Balb/c mice by DNCB.

Potentiality of Anti-idiotypic Antibodies Mimicking GD2 to Induce Cellular Immunity (GD2 유사 항이디오타입 항체의 세포면역 유발 잠재성)

  • Background: Disialoganglioside GD2 is a tumor-associated antigen that is overexpressed on tumor cells of neuroectodermal origin, such as melanoma, small cell lung carcinoma and neuroblastoma. Immunity against GD2 has anti-tumor activities, but GD2 is poorly immunogenic. Anti-idiotypic antibodies that mimic GD2 may induce more effective immune responses than GD2 antigen itself, because they are protein antigens and are known to be able to break immune tolerance. In our previous study, we produced anti-idiotypic antibodies mimicking GD2 (3A4 and 3H9), which induced humoral immunity. However, cellular immunity is essential to eradicate tumor cells in vivo as well as humoral immunity. In the present study, we investigated whether these anti-idiotypic antibodies 3A4 and 3H9 could induce cellular immunes responses. Methods: BALB/C mice were immunized with anti-idiotypic antibody 3A4 or 3H9, or normal mouse IgG as a negative control. Lymphoproliferative responses, cytokine production responses, and delayed-type hypersensitivity reactions were measured in mice immunized with the anti-idiotypic antibodies. Results: Both the anti-idiotypic antibody 3A4 and 3H9 induced GD2-specific lymphoproliferative responses and $IFN-{\gamma}$ production of lymph node lymphocytes in BALB/C mice. Only anti-idiotypic antibody 3H9 induced significant GD2-specific delayed-type hypersensitivity in the mice. Conclusion: These results show that anti-idiotypic antibodies 3A4 and 3H9 have the potentiality of inducing GD2-specific cellular immune responses that cannot be induced by the native antigen GD2 itself.

Effects of Anti-Helicobacter pylori IgY Powder to Protect Mice from Helicobacter pylori (Helicobacter pylori 감염생쥐에서 항-Helicobacter pylori 난황항체 분말의 효과)

  • Effects of anti-Helicobacter pylori IgY powder on H. pylori infection were evaluated 3 and 7 weeks after powder feeding by urease, PCR, and histological tests, and specific IgG assay of murine gastric tissue using mouse model. To produce anti-H. pylori IgY powder, laying hens were immunized with H. pylori prior to egg yolk harvest. C57BL/6 mice showing high response to H. pylori were infected with H. pylori and fed with the anti-H. pylori IgY powder. In urease and PCR tests, urease activity and gene count of anti-H. pylori IgY powder-fed group significantly decreased in comparison with control. Histological results indicated anti-H. pylori IgY powder effectively protected mice from H. pylori.

Evaluation of Local Lymph Node Assay as an Alternative Method for Skin Sensitization Potential in Baltic Mice (Balb/c 마우스에서 Local Lymph Node Assay(LLNA)를 01용한 피부 감작성 시험 대체시험법 연구)

  • Allergic contact dermatitis (skin sensitization) may be caused by a wide variety of chemicals. A murine local lymph node assay (LLNA) has been developed as an alternative to guinea pig models for assessing the contact sensitization potential of chemical. This study was carried out to evaluate the skin sensitization potential for chemicals in Balb/c mice by LLNA. Contact allergen, dinitrochlorobenzene (DNCB), respiratory allergen, toluene diisocyanate (TDI) and a weak allergen, $\alpha$-hexlycinnamaldehyde (HCA) were wed as positive chemicals and irritant, sodium lauryl sulfate(SLS) also wed as a reference chemical in this study. The weights of lymph node in the mice treated with DNCB, TDI, and HCA were increased compared to vehicle control. There was a significant increase in lymph node weight of mice treated with high concentration of SLS compared to vehicle control. The stimulation index (SI) of Lymph node cell in the mice treated with DNCB, TDI, and HCA revealed over three-fold increase compared to vehicle control by $3^H$-thymidine uptake. All allergens correctly identified in this LLNA study wing Balb/c mice. These results suggest that LLNA wing Balb/c mice could be a useful method for screening the allergenic potential of chemicals. The expression of IL-2 mRNA was slightly increased in draining auricular lymph node cell of the mice treated with TDI and HCA by RT-PCR. However the IL-2 levels in DNCB and SLS of treated animals were not significantly changed.
