Genomic changes of c-myc, c-H-ras in benzo(a)pyrene and dimethylbenz(a)anthracene treated human lymphoblast NC-37 cells (Benzo(a)pyrene과 dimethylbenz(a)anthracene에 의한 사람 림프아세포(NC-37)의 c-myc, c-H-ras 유전자 변화)

  • To investigate genomic changes in c-myc gene by a chemical carcinogen, human lymphoblast NC-37 cells were exposed to benzo(a)pyrene(BP) and dimethylbenzanthracene(DMBA), and the c-myc gene expression was evaluated by Northern and Southern blot hybridization techniques. The results are as follows: When the genomic DNA of NC-37 cells exposed to several concentrations(1.25, 2.5 and 5ug/ml) of BP concentration. However, the c-myc gene was most significantly enhanced with 2.5ug/ml of BP. The expressions of c-myc gene in NC-37 cells was stimulated by BP and DMBA. Addition of TPA reduced the gene expression BP-treated cells, whereas it enhanced the gene expression in DMBA-treated cells. The expression of c-H-ras gene was slightly increased by treatment with BP and DMBA alone and in combination with TPA, however the magnitude of increase was not significantly different between each other. The expressions of c-myc c-H-ras genes in Burkitt's lymphoma cells were greater than those in NC-37 cells. When the DNA extracted from NC-37 cells exposed to various concentrations of BP were amplified by polymerase chain reaction using a primer set containing c-myc exon I, the amplified products were of the same size in all groups. To evaluate the BP toxicity in E.coli to which human c-myc gene-cloned pBR322 vector was inserted, Southern blot hybridization was conducted on c-myc genes digested with EcoRI/HindIII and Smal/Xbal restriction enzymes, and observing that in 2 ug/ml BP-treated cells a 3.5kb fragment was generated in addition to 1.3kb fragment which can be observed in normal cells. Direct nucleotide sequence analysis of polymerase chain reaction products showed a mutation of G$\longrightarrow$A transition at the Smal recognition site.

Gibbs Ensemble Monte Carlo Simulation for Vapor-Liquid Equilibrium of Binary Mixtures $CO_2/C_3H_8$, $CO_2/CH_3OCH_3$, and $CO_2/CH_3COCH_3$

  • Gibbs ensemble Monte Carlo simulations were performed to calculate the vapor- liquid coexistence properties for the binary mixtures $CO_2/C_3H8$, $CO_2/CH_3OCH_3$, and $CO_2/CH_3COCH_3.$ For all the molecules the potential between sites in different molecules was simply calculated by the Lennard-Jones potential. Density of the mixture, composition of the mixture, the pressure-composition diagram, the chemical potential of component, and the radial distribution function were calculated at vapor- liquid equilibrium. The composition and the density of both vapor and liquid from simulation agreed considerably well with the experimental values over a wide range of pressures. The radial distribution functions in the liquid mixtures showed that $CO_2$ molecules tended to form cluster with each other and $C_3H8$ molecules also aggregated each other due to the weak interaction between $CO_3$ and $C_3H8$ molecule. However the interaction potentials between the same components were similar to those between the different components in the liquid mixtures $CO_2/CH_3OCH_3$ and $CO_2/CH_3COCH_3$.

  • This study was undertaken to find out the effect of heating time and temperature, pH and NaCl concentration on heat sensitivity of egg albumen during heat treatment. Sharp increase of the turbidity and rapid decrease of the foaming power were observed when egg albumen was heated at above $60^{\circ}C$. Egg albumen became opaque when it was heated at $60^{\circ}C$ for above 13 minutes or at $65^{\circ}C$ for above 5 minutes. The turbidity was markedly increased at below pH 7 and the foaming power was largely decreased at around pH 4.0 by the heat treatment ($60^{\circ}C$, 5 min). The foam stability was slightly decreased at alkaline pH range by the heat treatment (60t, 5 min). The addition of NaCl up to 0.3M decreased the turbidity. There was no effect of NaCl addition on the foaming power, but the foam stability was decreased by the addition of NaCl at above 0.2M before and after the heat treatment ($60^{\circ}C$, 5 min).

  • A k-hypertournament H on n vertices, where $2{\leq}k{\leq}n$, is a pair H = (V,A), where V is the vertex set of H and A is a set of k-tuples of vertices, called arcs, such that for all subsets $S{\subseteq}V$ with |S| = k, A contains exactly one permutation of S as an arc. Recently, Li et al. showed that any strong k-hypertournament H on n vertices, where $3{\leq}k{\leq}n-2$, is vertex-pancyclic, an extension of Moon's theorem for tournaments. In this paper, we prove the following generalization of another of Moon's theorems: If H is a strong k-hypertournament on n vertices, where $3{\leq}k{\leq}n-2$, and C is a Hamiltonian cycle in H, then C contains at least three pancyclic arcs.

  • Srivastava noticed the existence of three additional complete triple hypergeometric functions $H_A$, $H_B$ and $H_C$ of the second order in the course of an extensive investigation of Lauricella's fourteen hypergeometric functions of three variables. In 2004, Rathie and Kim obtained four summation formulas containing a large number of very interesting reducible cases of Srivastava's triple hypergeometric series $H_A$ and $H_C$. Here we are also aiming at presenting two unified summation formulas (actually, including 62 ones) for some reducible cases of Srivastava's $H_C$ with the help of generalized Dixon's theorem and generalized Whipple's theorem on the sum of a $_3F_2$ obtained earlier by Lavoie et al.. Some special cases of our results are also considered.

The Isolation of Bacillus sphaericus 366M-9 Producing New Cephalosporin-C Deacetylase (CAH) and its Enzymatic Characterization (신규 Cephalosporin-C Deacetylase(CAH) 생산 균주인 Bacillus sphaericus 366M-9의 선발 및 그 효소학적 특성)

  • Several microorganisms (esterase-producing group) were isolated by the solid selective media containing-naphtylacetate. Among them, strain 366M-9 having a high activity of cephalosporin-C deacetylase (CAH; EC 3.1.1.41) was selected. The strain 366M-9 was identified as Bacillus sphaericus on the basis of morphological, physiological, and biochemical characteristics. The production of CAH reached at maximum value after 32 hrs, when cultivated in the optimal medium containing dextrin 2.5%, peptone 2.5%, sodium chloride 0.5%, dipotassium phosphate 0.25%, ferrous sulfate 0.02%, and 7-ACA 0.1% at $30^{\circ}C$ with initial pH 6.0. The CAH was purified by 3 steps with ammonium sulfate precipitation, adsorption chromatography on hydroxyapatite column, and Sephadex G-200 gel chromatography. The final enzyme preparation was homogeneous as judged by the analysis of SDS-PAGE and HPLC. Optimum temperature and pH for CAH activity were $50{\circ}C$ and around 7.0, respectively. And the enzyme was stable at pH 6.0~8.0, up to $50^{\circ}C$. The Michaelis-Menten constants ($K_{m}$ ), $V_{max}$ were 0.87 mM and 1.22 unit/ml, respectively.

Synthesis of Vaterite Powders with a Spherical Shape by the Precipitation Method (침전법에 의한 구형 Vaterite분말의 합성)

  • CaCO$_3$ powders were synthesized by aqueous solution reaction of CaC1$_2$ㆍ2$H_2O$-(NH$_4$)$_2$CO$_3$ system with NH$_4$OH at 45$^{\circ}C$ and pHs 8, 9, 10, and 11 and in the concentration range of 0.1∼5 M and its polymorphism, morphology and size were investigated. In order to investigate the influence of pH on nucleation, pH was adjusted before and after reaction respectively. When pH was adjusted after reaction a formation ratio of vaterite was increased with increasing pH and concentration but vaterite was formed with calcite. But, when pH was adjusted before reaction, the formation rate of vaterite was increased with increasing pH and concentration. resulting in a phase-pure vaterite with a spherical shape and 2∼5 $\mu\textrm{m}$ in size. It was found that solubility of alkaline vaterite was decreased with increasing OH- ions in the high pH solution. When pH was adjusted before nucleation in the high concentration range, in particular, decreasing of solubility disturbed transformation of initially formed numerous vaterite to calcite.

Stability of a Silica Membrane in the HI-$H_2O$ Gaseous Mixture (HI-$H_2O$ 기상 혼합물에서 Silica 막의 안정성)

  • The stability of the prepared silica membrane by chemical vapor deposition (CVD) method in the HI-$H_2O$ gaseous mixture was evaluated aiming at the application for hydrogen iodide decomposition in the thermochemical IS process. Porous $\alpha$-alumina having pore size of 100 nm was modified by the different CVD temperature using tetraethoxysilane as the Si source. The CVD temperature was $700^{\circ}C$, $650^{\circ}C$, and $600^{\circ}C$. The $H_2$/H$_2$ selectivities of the modified membranes which were measured by single-component permeation experiment showed 43.2, 12.6, and 8.7 at $600^{\circ}C$ for the M1 (CVD temperature was $700^{\circ}C$), M2 (CVD temperature was $650^{\circ}C$) and M3 membranes (CVD temperature was $600^{\circ}C$), respectively. Stability experiment in the HI-$H_2O$ gaseous mixture was carried out at $450^{\circ}C$. The prepared silica membrane at $600^{\circ}C$ of CVD temperature was more stable than that at the other CVD temperature.

  • has been synthesized using the layered organic-inorganic hybrids, Cu2(OH)3(CH3COO)·H2O as a precursor. Cubic Cu2O is synthesized by reducing Cu2(OH)3(CH3COO)·H2O with glucose in water at 75oC. The effects of precursor and glucose are investigated. The structure of Cu2(OH)3(CH3COO)·H2O plays an important role in preparing the uniform size of Cu2O.

Determination of Natural Gas Components by Gas Chromatographic Multicolumn System (기체 크로마토그래피 복합컬럼 시스템에 의한 천연가스 성분의 정량)

  • A multicolumn system consisted of two valve(10-port and 6-port valve)-three column (28% DC 200, SP 1700 and Chromosorb 102 column) was developed. Nine natural gas components composed of $N_2$, $CH_4$, $CO_2$, $C_2H_6$, $C_3H_8$, $i-C_4H_{10}$, $n-C_4H_{10}$, $i-C_5H_{12}$$n-C_5H_{12}$ completed all the baseline separation within 18 minutes. The accuracy and the precision of this system was tested. The retention times and the peak areas were determined with a repeatability between 0.02 and 0.16%, and less than 1%, respectively. Calibration curves for natural-gas components were plotted by the partial pressure injection method of pure gases, and good linear relationships for each component were presented. By using these calibration curves the accuracy of the multicolumn system compaired with that of the single column system for a certified standard gas of natural gas. As a result, relative error in the single and the multicolumn system was less than 0.5% and 0.04%, respectively. The result of application of this system in the analysis of importing LNG composition showed that the heating values calculated by the multicolum system were estimated lower compared with those calculated by the single column system and consequently, the importing price of LNG was able to be cut down.
