Improving the Nutritive Value of Full-Fat Rice Bran for Broiler Chickens Using a Lipase-Based Enzyme Preparation

  • Two experiments were conducted to test the hypothesis that a lipase-based enzyme preparation would increase the AME content of full-fat rice bran (FFRB) by increasing fat digestibility when fed to broiler chickens. Experiment 1 used FFRB from Australia and lasted for 35 days, while Experiment 2 used FFRB from Thailand and lasted for 14 days. Rice bran was substituted in a maize-soybean diet at levels of 90 g/kg (Experiment 1) and at 90 and 180 g/kg in Experiment 2. Total collections of excreta were used for determination of AME content and fat digestibility. In Experiment 1, the enzyme increased the AME content of FFRB between days 4-7, 18-21 and 32-35 by 6.1-16.1% (p>0.05), however this was not associated with improved fat digestibility. In Experiment 2, the enzyme enhanced the AME content of FFRB between days 4-7 (10.42 vs. 9.06, p=0.107) and 11-14 (11.94 vs. 9.93, p=0.041), but again, this was not caused by increased fat digestibility. Inclusion of 180 g/kg depressed the AME content of FFRB by 7.4-11.5% (p>0.10) in conjunction with decreased (p<0.05) fat digestibility between 0-14 days of age. Improvements in bird growth with the enzyme were seen in Experiment 2 but not in Experiment 1. Increases in AME content of FFRB per se were not caused by enhanced fat digestibility, suggesting that the side activities associated with the preparation must have acted singularly or in combination to improve AME content and bird performance. These data show that the response of FFRB to the lipase-based enzyme preparation was dependent upon the geographical origin of the rice bran and the level of FFRB substituted in the basal diet.

Absence of DNA Polymorphisms in Myzus persicae (Homoptera: Aphididae) in Relation to their Host Plants (기주식물 종류에 따른 복숭아혹진딧물(Myzus persicae)의 DNA Polymorphism 비교)

  • DNA polymorphisms were analyzed for 8 clones of the green peach aphid, Myzus persicae Sulzer, by random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR). The insect has different host preferences and was even classified into two different species, M. persicae Sulzer and Myzus nicotinae Blackman by their morphological characters, but this point is still in arguement. To identify the differences between two types of the green peach aphid by RAPD-PCR, the template DNA was extracted from 4 clones each of tobacco-feeding and non-tobacco-feeding forms and one hundred primers of 10-nucleotideslong were tested in PCR. The amplified DNAs were analyzed by agarose gel electrophoresis. Eighty-three primers gave amplified DNA fragments with 1 to 22 in number and 500 to 20,000 base pairs in length, but no amplification was observed in the other 17 primers. The average number of fragment per each amplification was about 13. In the case of 82 out of 83 random primers, band patterns of amplified DNA were identical among 8 clones, even though some differences were noticed in the intensity of specific bands. Polymorphism was detected by only one primer within the tobacco-feeding forms, but not between the two host types. The results did not detect any relationship between RAPD polymorphism and their host preference.

Floristics of bryophytes in Dongbaek-dongsan at Seonheul Gotjawal (선흘 곶자왈 동백동산의 선태식물상 연구)

  • This study presents a survey on the bryophytes in Dongbaek-dongsan located at Seonheul Gotjawal on the north-eastern face of Mt. Halla in Jeju Island, Korea. A total of 85 taxa belonging to the Bryophyta (22 families 44 genera 62 species), Marchantiophyta (12 families 16 genera 23 species) were found. Among them, 3 moss species, Diphyscium perminutum Takaki, Racomitrium japonicum Dozy & Molk., and Isopterygium minutirameum (M$\ddot{u}$. Hal.) A. Jaeger. were new additions to the bryophyte flora of Korea. The liverwort index was 27.0%. Predominant life-form was weft. The rates for the bryophytes dominating in mesic to hygric sites were higher than for the bryophytes mainly observed in xeric habitats. We considered that the composition and distribution of species are affected by the different environmental factors according to the evergreen broad-leaved forests, wetlands and exposed spaces. Moreover, the rocks were the substrate type that play a major role in providing the micro-habitats for bryophytes in this area. Particularly, the bryophytes of swamps in this area were very unique and various for its limited space and showed plant communities with mosaic spatial pattern, along with vascular plants. We suggest more detailed and comprehensive studies on the swamps where endemic fern Mankyua chejuense B. -Y. Sun, M. H. Kim & C. H. Kim inhabits to provide valuable information for the conservation and restoration of the species and habitats.

Effects of several factors on pollen germination in Platycodon grandiflorum

  • Pollen germination and its' viability in bellflower hybrid system are of great importance. The present study was conducted to investigate the several factors underlying the pollen germination of Platycodon grandiflorum and obtain the basic data for effective artificial pollination for the production of sound specifies. The pollens of Platycodon grandiflorum started germination from one hour after planting, and the germination was actively progressed as time dependent manner. For lighting conditions, the germination of pollens under the light was faster by more than twice than that without the light. Furthermore, the germination was better in the high temperature rather than in the low temperature. The germination rate was higher in the $30^{\circ}C$. For the carbon source, the germination rate was better at the concentration of 15% regardless of the kinds. In particular, the highest value was observed with glucose. The germination rate was decreased substantially as the increasing with the higher pH. The dynamic germination of pollens was observed at the pH 5. With respect to the growth regulator, the higher concentration of NAA induced the higher the germination rate. $GA_3$ showed a good germination rate in $0.05mg{\cdot}L^{-1}$. Meanwhile, for kinetin, lower concentration increased the germination rate, unlike NAA. The higher concentrations of boric acid degraded the germination rate, and the addition of boric acid of $10mg{\cdot}L^{-1}$ demonstrated higher germination rate than the addition of other growth regulators. Notably, the addition of asparaginic acid exhibited the similar results in all test sectors regardless of concentration, whereas a little higher result was observed in the high concentration sector. Taken together, the results concluded that the boric acid was considered as one of the essential minerals that played an important role on the germination of pollens.

Quality Characteristics of Pumpkin Jam when Sucrose was Replaced with Oligosaccharides during Storage (올리고당을 첨가한 호박잼 저장 중 품질 특성 변화)

  • The physicochemical and sensory qualities of pumpkin jams, in which sucrose was replaced with oligosaccharides, were investigated during storage at 20$^{\circ}C$. pumpkin jam was prepared with steamed grind pumpkin, mixed with either sucrose only(50%), corn syrup(COS, sucrose 30%+COS 20%), fructooligosaccharide (FTO, sucrose 30%+FTO 20%), isomaltooligo saccharide (IMO, sucrose 30%+IMO 20%), or galactooligo-saccharide(GTO, sucrose 30%+GTO 20%). The final sweetness of each pumpkin jam was 64$^{\circ}$ Brix. During 60days of storage there were no differences in acidity and pH among the treatments. Reducing sugar content was higher in the pumpkin jam containing COS compared to 次e others. During storage Lightness(Hunter L), redness(a value) and yellowness(b value) increased, of which L and b values were the highest in COS, and the a value were higher in sucrose compared to the other sugars. Adhesiveness and hardness of textural properties were the highest in sucrose. Sensory evaluation results showed that the mean scores of over-all acceptability during storage did not significantly decrease until the 30th day of storage, compared to the freshly made jams. Physicochemical and sensory characteristics of pumpkin jams during storage in a PCA plot were comprised of the first principal component (99.44%) and the second principal component (0.54%).

A Study on the Quality and Sensory Characteristics of Ginseng Pickles(JJangachi) (인삼 절임식품의 품질 및 관능특성에 관한 연구)

  • The purpose of this study is to investigate the quality and sensory characteristics of ginseng Jeolim, or ginseng pickles. The moisture contents of ginseng pickles decreased significantly with increase in soy sauce and in storage period. The L-value of pickle solution decreased significantly with increase in soy sauce and in storage period while its a-value and b-value increased significantly with increase in soy sauce and in storage period. The salinity and sugar contents of pickle solution increased significantly with increase in soy sauce; however, they decreased with increase in storage period. The pH of pickle solution decreased significantly with increase in soy sauce and in storage period. As the results of acceptance test, apparence, flavor, taste, texture, and overall acceptance were higher in C3. In conclusion, ginseng Jeolim showed great preference with adding soy sauce, and the preference has increased with 20% of soy sauce added and 14 day storage period.

Effect of Liquid Fertilizer Application using Fish-meal, Bone-meal and Sesame oil-cake on Seed Germination and Growth of Tomato (어분, 골분 및 참깨박을 이용한 발효액비 제조에 따른 무 발아 및 토마토 생육에 미치는 영향)

  • This study aimed to investigate the physicochemical characteristics of fish meal, bone meal, and sesame oil cake, which are readily available by-products from agriculture and fisheries, during the process of liquid fertilizer fermentation, and to examine the effects of liquid fertilizer application on seed germination and growth of tomatoes. During processing the fermentation for liquid fertilizers by using fish meal, bone meal, and sesame oil cake liquid fertilizers, the pH of the fertilizer increased in the order of bone meal > fish meal > sesame oil cake, and the concentration increased rapidly up to 30 days in all types of liquid fertilizer. The nitrogen content of the liquid fertilizers increased as fermentation progressed in the order of fish meal > bone meal > sesame oil cake. The phosphorus content increased as fermentation progressed and the highest was 1.0 % in the liquid fertilizer of sesame oil cake. The germination rate and its index of radish seeds were compared for different dilutions of each of the liquid fertilizers. Excluding the 10-fold dilution of the fish meal and oil cake liquid fertilizer, all the treatment groups showed a germination rate ≥ 95 % and the germination index tended to increase with dilution rate of liquid fertilizers. For responses of tomato growth, there were no significant differences among the liquid fertilizer treatment groups; however, the organic content, microbial density, and microbial biomass C in the soil were higher than chemical fertilizer treatment. These results demonstrated that there were differences in the characteristics of liquid fertilizers depending on the materials used, and that liquid fertilizer can be used for nutrition management for the organic crop cultivation.

  • The effects of CaO, $CaO+MgSO_4$, and $CaO+MgSO_4$+compound fertilizers(NPK) on soil chemical properties of acidified forest soils were studied in Quercus spp. and Pinus rigida stands in Mt. Namsan and Mt. Surak in urban areas, and Kwangnung in a mountain area. The soil samples were collected in November 1995 after every year fertilization from November 1991 through April 1995. The fertilizations affected soil chemical properties. Soil pH increased after fertilizations compared with control. However, the effect was different between the stand types and the areas. Organic matter and total nitrogen content were not changed, while exchangable cations such as calcium and magnesium increased after fertilizations. However, these ration concentrations after fertilizer treatments were lower in P. rigida than in Quercus spp. stands. These rations also showed increased leaching characteristics more in the urban area than in the mountain area.

Supplementing Rhodobacter sphaeroides in the diet of lactating Holstein cows may naturally produce coenzyme Q10-enriched milk

  • Objective: To examine the effects of Rhodobacter sphaeroides (R. sphaeroides) supplementation as a direct-fed microbial (DFM) on rumen fermentation in dairy cows and on coenzyme Q10 (CoQ10) transition into milk, an in vitro rumen simulation batch culture and an in vivo dairy cow experiment were conducted. Methods: The characteristics of in vitro ruminal fermentation were investigated using rumen fluids from six cannulated Holstein dairy cows at 2 h post-afternoon feeding. A control treatment was included in the experiments based on a typified total mixed ration (TMR) for lactating dairy cows, which was identical to the one used in the in vivo study, plus R. sphaeroides at 0.1%, 0.3%, and 0.5% TMR dry matter. The in vivo study employed six ruminally cannulated lactating Holstein cows randomly allotted to either the control TMR (C-TMR) treatment or to a diet supplemented with a 0.5% R. sphaeroides culture (S-TMR, dry matter basis) ad libitum. The presence of R. sphaeroides was verified using denaturing gradient gel electrophoresis (DGGE) applied to the bacterial samples obtained from the in vivo study. The concentration of CoQ10 in milk and in the supernatant from the in vitro study was determined using high performance liquid chromatography. Results: The results of the in vitro batch culture and DGGE showed that the concentration of CoQ10 significantly increased after 2 h of R. sphaeroides supplementation above 0.1%. When supplemented to the diet of lactating cows at the level of 0.5%, R. sphaeroides did not present any adverse effect on dry matter intake and milk yield. However, the concentration of CoQ10 in milk dramatically increased, with treated cows producing 70.9% more CoQ10 than control cows. Conclusion: The CoQ10 concentration in milk increased via the use of a novel DFM, and R. sphaeroides might be used for producing value-added milk and dairy products in the future.

Changes in Serum Vitamin E and Trace Mineral Levels and Other Blood parameters in Growing Thoroughbred Horses During the Period of Pasture Grazing and stable Feeding (Thoroughbred 육성마의 방목과 사사기간 중 혈청 비타민 E, 미량광물질 및 기타 화학치의 변화)

  • Nutritional adequacy of growing Thoroughbred horses raised in an alternate feeding system - grazing during late spring through late fall and stable feeding for the rest of seasons - was assessed by determining vitamin E and trace mineral levels in the serum and blood chemistry related to nutrition and health. During the stable feeding in winter and early spring, 50 growing female horses were fed concentrates (1.4% of their body weight), grass hay (0.62%) and alfalfa hay (0.37%). For the grazing period, the same horses were fed supplementary concentrates (1.1%) during late spring through early summer, and concentrates (1.1%) and alfalfa hay (0.5%) during late summer through late fall. Blood samples were collected before grazing in early spring, and during grazing in early summer through late fall. Serum vitamin E, BUN, GTP, total bilirubin and direct bilirubin levels were increased (P<0.01) by grazing compared to those measured before the initiation of grazing. Horses had lower (P<0.01) serum Fe contents in early summer than in late fall or in time of stable feeding. Stable feeding increased (P<0.01) serum Cu content compared to grazing in both early summer and late fall. In late fall, serum Zn level increased (P<0.01) compared to that found in the other seasons. Blood glucose and creatinine levels decreased (P<0.01) after grazing. Results indicate that supplementations of some minerals and vitamin E are not always necessary in diets for growing horses and should be done after careful evaluation of diets with regard to concentrations and biological availability of minerals.
