Effect of NO Treatment during Shelf Life of 'Hayward' Kiwifruit after Storage at Cold Temperature (Nitric Oxide 처리가 저온 저장된 키위과실의 상온 유통 중 품질에 미치는 영향)

  • The effect of nitric oxide (NO) treatment on the quality of kiwifruit, cv. Hayward, was studied at room temperature after cold storage for one or three months at $0^{\circ}C$. Kiwifruits cold-stored for one month were treated with $200{\mu}L{\cdot}L^{-1}$ NO and subsequently transferred to room temperature to monitor quality changes over the course of their shelf life. Weight loss was high in fruits not treated with NO. Ethylene production was delayed for two days by NO treatment, and respiration rate was reduced to less half than that of the control. The kiwifruits stored for three months were treated with $N_2$ and 100, 200, or $500{\mu}L{\cdot}L^{-1}$ NO, or air alone. The highest weight loss was observed in kiwifruit treated with $100{\mu}L{\cdot}L^{-1}$ NO. While ethylene production was high in fruits treated with $100{\mu}L{\cdot}L^{-1}$ NO and without the treatment, it was relatively low in the kiwifruit treated with 200 and $500{\mu}L{\cdot}L^{-1}$ NO. Firmness was abruptly decreased in fruits not treated with NO, while the kiwifruit exposed to $200{\mu}L{\cdot}L^{-1}$ NO maintained the s ame level of f irmness for 9 days a t room t emp erature. In addition, growth o f Botrytis cinerea was inhibited by NO as compared with the air and $N_2$ treatments. Our findings indicate that NO can be used effectively for prolonging shelf life and maintaining fruit quality during distribution after cold storage. The optimum NO concentration for cold-stored kiwifruits was found to be $200{\mu}L{\cdot}L^{-1}$.

A New Whole Crop Barley Cultivar 'Younghan' with Good Quality, Resistance to Lodging and Viral Disease and High-Yielding (양질 내병 내도복 다수성 청보리 품종 '영한')

  • 'Younghan' (Hordeum vulgare L.), a winter barley for forage use, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, National Institute of Crop Science, Rural Development Administration in 2008. It was derived from a cross between $F_1$['YB3433-3B-5'/'YB3135-3B-2-3'] and 'YB3135-3B-2-3'. 'SB971305-B-B-B-4-4' line was selected for its earliness, resistance to disease and good agronomic characteristics. The promising line showed both high yield and lodging resistance in the yield trials in Iksan in 2004 to 2005, and designated as 'Iksan420'. The line was subsequently evaluated for winter hardiness, earliness, and yield at eight locations throughout Korea for two years from 2007 to 2008 and finally named as 'Younghan'. It has the growth habit of IV, erect plant type, green leaf and thick culm in diameter. Its heading date was April 24, and the maturing date was May 25 in paddy field conditions, which were one day earlier than those of the check cultivar 'Youngyang'. The cultivar 'Younghan' had better winter hardiness, and resistance to lodging and BaYMV than those of the check cultivar did. The average forage dry matter yield of 'Younghan' was approximately 12.0 MT/ha in adapted region. 'Younghan' also showed 8.5% of crude protein content, 27.2% of ADF, and 67.1% of TDN, including higher grade of silage quality for whole crop barley. This cultivar would be suitable for (area with daily minimum temperature in January) above $-8^{\circ}C$ in Korean peninsula.

Synecological Study of the Naturalized Plant Communities in Old-Andong City (구 안동시역에 분포하는 귀화식물군락의 생태학적 연구)

  • The present study was undertaken to classify and describe the spring naturalized plant communities in old-Andong city by the methodology of the ZM school of phytosociology. As a result, the vegetation was classified into the six communities and two subcommunities: A. Bromus tectorum community A-a. Bromus japonicus subcommunity, A-b. Melica onoei subcommunity; B. Poa pratensis community; C. Rumex crispus community; D. Oenothera lamarckiana community; E. Ambrosia artemisiifolia var. eiatior community; F. Rumex conglomeratus community. The total taxa of the naturalized plant communities consisted of 31 families, 86 genera, 114 species and 13 varieties. Of which the taxa of the naturalized plant species consisted of 8 families, 21 genera, 25 species and 1 variety. The result of Bray-Curtis ordination revealed that the plots suveyed were arranged according largely to the vegetation units of' the communities. Also the interspecific affinity was examined by an analysis of interspecific association and the main component species in the communities were divided into two groups. The naturalized rate was higher in the communitise affected by strong human impacts, while was lower in the communities affected relatively less by human impacts. On the other hand the seasonal changes of the communities and the naturalized rate between the spring and the autumn were investigated. Many naturalized communities present in Spring were replaced by the other native ruderal communitues in the Autumn. The naturalized rate based on the dominance was largely decreased over from the spring to the Autumn.

Experimental and epidemiological studies on the life cycle of Echinostoma hortense Asada, 1926(Trematoda: Echinostomatidae) (남한강류역(南漢江流域)의 호르텐스극구흡충(棘口吸蟲) 감염실태(感染實態)와 생활사(生活史)에 관(關)한 연구(硏究))

  • Recently there have been some reports on human infections of Echinostoma hortense in Korea. It was found that a few species of freshwater fishes were playing the role of the second intermediate host of E. hortense. However, molluscan intermediate host has not been identified yet in Korea. The present study aimed to establish the life cycle of E. hortense in laboratory. Experimental studies such as egg production from the rat, development of the eggs in vitro, exposure of miracidia to freshwater snails, shedding pattern of cercariae from infected snails, morphology of cercariae, cercarial infection to the second intermediate host and infection of metacercariae to the definitive hosts were done. In addition, epidemiological surveys on the infection status in inhabitants and house rats, and on the natural infection of larval echinostomes in the snails and fishes were carried out along the South Hangang-river. The results obtained were as follows: 1. The eggs deposited from adults in physiological saline were cultivated at room temperature($20{\sim}24^{\circ}C$). The miracidia were firstly observed on 8 days after cultivation, and 85.5% of the eggs contained the mature miracidia on 11 days after cultivation. More than 90% formed the miracidia when cultivated at temperature $22{\sim}27^{\circ}C$. Hatching of the miracidia began on 12 days after cultivation and continued for a week. The size of the miracidia was $103.0{{\times}}51.4{\mu}m$ in average. The motility of the miracidia were active up to 8 hours after shedding, but they were all dead within 10 hours after shedding. 2. A freshwater snail, Radix auricularia coreana was cultivated in aquaria. A hatched $F_1$ snails from the egg masses were exposed to 20 miracidia respectively. Escape of cercariae started on 15 days after infecton. Radix auricularia coreana was experimentally identified as the first intermediate host of E. hortense in Korea. 3. Cercarial shedding started on $15{\sim}20$ days after infection by snail, continued for about 10 days (8.8 days in average). Infected snails were dead within 32 days after the miracidial infection. About 1,335 cercariae($328{\sim}1,994$) per snail were shed in its life, and 119 cercariae in average per snail per day were shed. The cercariae were motile for more than 24 hours, and then squirming at the bottom until death. The body and tail sizes of cercariae were $356{\times}186{\mu}m$ and $510{\times}68{\mu}m$ in average, respectively. The rediae parasitized in the snail hosts were found mainly around the pericardial regions, and their size was $1,575{\times}258{\mu}m$ in average. The numbers of developing cercariae in a mature redia were 14 in average ($7{\sim}20$ in range). The numbers of rediae in a snail were 102 in average on 15 days after miracidial infection and 221 in average on 28 days. 4. Three uninfected Misgurnus anguillicaudatus, less than 6.5cm long were used in for the cercarial infection. They were all exposed with 755 cercariae, and examined at 5-day intervals starting from 10 days after infection. All the fishes were infected with metacercariae of E. hortense and a total of 275 was found infected(36.4%). The metacercariae were fed to rats and the adult worms were obtained on 15 days after infection. 5. The infected rats began to deposit the eggs on 11 days after infection. The number of eggs deposited per day per worm (EPD/worm) was $400{\sim}500$ on 3 weeks after infection and was increased to $1,000{\sim}1,500$ on 4 to 17 weeks, then decreased to 800 on 21 weeks after infection. 6. A total of 745 stool specimens collected from 576 male and 169 female residents of 8 different villages along South Hangang basin was examined. Out of 745 specimens, the eggs of Echinostoma sp. were found in 2 cases (0.3%). Of 34 house rats one showed egg-positive (2.9%). 7. Total 971 Radix auricularia coreana collected from 7 sampling stations were examined for shedding of cercariae. Three snails (0.3%) shed the cercariae of E. hortense. A total of 119 out of 542 freshwater fishes (22.0%) had the metacercariae of E. hortense. The fishes parasitized with the metacercariae were 4 out of 14 examined species. The infection rates of 4 species were 34.1% (106 out of 311) in Misgurnus anguillicaudatus, 30.4% (7 out of 23) in Misgurnus mizolepis, 4.3% (2 out of 46) in Moroco oxycephalus and 22.2% (4 out of 18) in Odontobutis obscura interrupta. In summarizing the above results, the first intermediate host of E. hortense was found as Radix auricularia coreana in Korea. Also, it took about 46 days for the shortest completion of a life cycle of E. hortense in summer; that is, 10 days for miracidial development in eggs, 15 days for cercarial development in the snail, about 10 days for metacercarial development in the second intermediate hosts, and 11 days for the maturation as the adults in the definitive hosts. The natural infection rates of E. hortense in the intermediate hosts were relatively high but those in the definitive hosts were low in the middle areas of South Hangang basin.

Chemical Components and Antimicrobial Activity of Garlics from Different Cultivated Area (산지별 마늘의 화학성분 및 항균활성)

  • The chemical components and antimicrobial activities of garlic from different area were investigated and analyzed to provide basic data for functional food materialization and processing. Hunter's values of garlic from different area were L 53.41~57.15, a -3.49~-4.38 and b 11.47~17.55. The moisture, crude protein, crude fat, nitrogen free extract, crude fiber and ash were 65.24~71.96, 6.24~9.35, 0.21~0.49, 19.01~22.72, 0.58~0.95 and 1.01~2.01%, respectively. The major minerals of garlic from different area were Na(27.22~112.03), Mg(18.17~32.56), K(242.16~569.28), Ca(28.60~63.93), P(117.72~265.21 mg%) and major free sugars were sucrose, glucose and fructose. The major amino acids of garlic from different area were proline, arglmne, glutamic acid and aspartic acid and content of total amino acid was 2,709.33~4,561.04 mg%. The ascorbic acid content of garlic from different area was 2.966~8.673 mg%. Composition of fatty acids of garlic from different area were linoleic acid, oleic acid and palmitic acid, unsaturated fatty acid and saturated fatty acid contents were 72.18~74.35 and 25.65~27.82%, respectively. Antimicrobial activities of garlic extracts as different area increased depends on concentration and showed the high antimicrobial activities against Gram(+) and Gram(-).

Genetic Environments of Hydrothermal Vein Deposits in the Pacitan District, East Java, Indonesia (인도네시아 동부자바 빠찌딴(Pacitan) 광화대 열수 맥상 광상의 성인 연구)

  • The hydrothermal vein type deposits which comprise the Kasihan, Jompong and Gempol mineralized areas are primarily copper and zinc deposits, but they are also associated with lead and/or gold mineralization. The deposits occur within the Tertiary sedimentary and volcanic rocks in the Southern Mountain zone of the eastern Java island, Indonesia. Mineralization can be separated into two or three distinct stages (pre-and/or post- ore mineralization stages and main ore mineralization stage) which took place mainly along pre-existing fault breccia zones. The main phase of mineralization (the main ore stage) can be usually classified into three substages (early, middle and late) according to ore mineral assemblages, paragenesis, textures and their chemical compositions. Ore mineralogy and paragenesis of the three areas in the district are different from each other. Pyrite, pyrrhotite (/arsenopyrite), iron-rich (up to 20.5 mole % FeS) sphalerite and (Cu-)Pb-Bi sulfosalts are characteristic of the deposits in the Kasihan (/Jompong) area. On the other hand, pyrite + hematite + magnetite + iron-poor (2.7 to 3.6 mole % FeS) sphalerite assemblage is restricted to the Gempol area. Fluid inclusion data suggest that fluids of the main ore stage evolved from initial high temperatures (near $350^{\circ}C$) to later lower temperatures (near $200^{\circ}C$) with salinities ranging from 0.8 to 10.1 equiv. wt. percent NaCl. Each area represents a separate hydrothermal system: the mineralization at Kasihan and Jompong were largely due to early fluid boiling coupled with later cooling and dilution, whereas the mineralization at Gempol was mainly resulted from cooling and dilution by an influx of cooler meteoric waters. Fluid inclusion evidence of boiling indicates that pressures of ${\geq}95$ to 255 bars (${\geq}95$ bars for the Gempol area: $\approx$ 120 to 170 bars for the Jompong area: $\approx$ 140 to 255 bars for the Kasihan area) during portions of main ore stage mineralization. Equilibrium thermodynamic interpretation indicates that the evolution trends of the temperature versus fS2 variation of ore stage fluids in the Pacitan district follow two fashions: ore fluids at Kasihan and Jompong changed from the pyrite-pyrrhotite sulfidation stage towards pyritehematite- magnetite state, whereas those at Gempol evolved nearly along pyrite-hematite-magnetite reaction curve with decreasing temperature. The sulfur isotope compositions of sulfide minerals are consistent with an igneous source of sulfur with a ${\delta}^{34}S_{{\Sigma}s}$ value of about 3.3 per mil. The oxygen and hydrogen isotopic compositions of the fluids in each area indicate a progressive shift from the dominance of highly exchanged meteoric water at early hydrothermal systems towards an un- or less-exchanged meteoric water at later hydrothermal systems.

Reduction of Microbial Populations on the Surface of Fresh Ginseng by Various Washing Treatments (세척처리에 따른 수삼 표면의 미생물 제어효과)

  • Surface cleaning is both essential and troublesome when a consumer seeks to eliminate soil attached to the surface of fresh ginseng because all ginseng purchased in the market is covered with soil, reflecting the post-harvest situation. To facilitate ginseng use at home, a fresh-cut type of ginseng is required. As a first step toward production of such ginseng, several washing and dipping treatments were investigated with respect to surface cleaning and reduction of microbial populations on fresh ginseng. In terms of microbial distribution on the surface of fresh ginseng, higher levels of viable bacteria (6.63 log CFU/each) and fungi (5.12 log CFU/each) were present on the rhizome head than on other regions of the root. Of the washing treatments tested, hand-brushing was effective for surface cleaning and to reduce microorganism levels on fresh ginseng, but use of a high-pressure water spray followed by hand-brushing was optimally effective. To further reduce the levelsof microorganisms on the surface of fresh ginseng after washing, additional dipping treatments in 70% (v/v) ethanol and electrolyzed acidic water (at pH 2.3) were somewhat effective but showed no significant differences compared with other dipping treatments tested, including a 3 ppm ozone solution, a 200 ppm sodium hypochlorite solution, or hot water at $50^{\circ}C$.

Association of Interleukin-27 rs 153109 Single Nucleotide Polymorphism with Spontaneous Resolution of Hepatitis C Virus - Genotype 4a Infection in Egyptian Patients

  • Background: HCV is a major global health problem. IL-27 is a member of the IL-6/IL-12 cytokine family with a broad range of anti-inflammatory properties. Recent studies highlighted the effect of a SNP in the IL-27 promoter region on modulating the progression of infectious diseases and individual responses to therapy. Aim of the work: The present study investigated the potential role of (-964 A/G) SNP in the promoter region of IL-27p28 gene (alleles rs153109) on the outcome of HCV infection among genotype 4a infected patients. Materials and Methods: HCV genotyping confirmed that all of the HCV-infected patients had genotype 4a infection. Genomic DNA was extracted from 111 patients with chronic HCV infection, 42 spontaneous resolvers (SR) and 16 healthy controls. IL- 27p28.rs153109 genotyping was assessed using PCR-RFLP then confirmed by DNA sequencing. Results: The frequency of IL-27-p28.rs153109AA, AG, and GG genotypes among chronically infected subjects were 74.8 %, 25.2%, and 0% while among the SR, they were 57.1%, 35.7%, and 7.14%, respectively. Our data show the unique presence of G/G genotype in the SR group (3 patients; 7.14%). Moreover, the "G" allele frequencies among chronic and resolved subjects were 12.6% and 25.0%, respectively (p=0.0136). Importantly, subjects with the GG genotype were more likely to clear their HCV infection than those with the AA genotype (p=0.0118). Conclusions: HCV genotype 4a subjects with the IL-27-p28.rs153109 A/G and G/G genotype were more likely to clear their HCV infection. Therefore, we propose IL- 27p28.rs153109SNPas a genetic biomarker for predicting HCV infection outcome.

A Study on Estimation of Individual Growth Curve Parameters and their Relationships with Meat Quality Traits of Crossbred between Korean Native Boars and Landrace Sows (재래돼지와 랜드레이스 교잡종의 개체별 성장곡선 추정 및 육질형질과의 상관관계 추정에 관한 연구)

  • This study was conducted to estimate the growth curve parameters of crossbreds between Korean native boars and Landrace sows and their relationships with meat qualities. The data used were weight-age data and carcass data from 131 males and 122 females raised at N.L.R.I in Korea. Growth curve parameters were estimated from nonlinear regression using Gompertz model individually. Average mature weight(A), average maturing rate(k), and average inflection point(u) showing maximum growth rate estimated were 179.54${\pm}$6.06kg, 0.3154${\pm}$0.0059, and 5.50${\pm}$0.11 months in females, and 179.84${\pm}$6.33kg, 0.3049${\pm}$0.0061, and 5.24${\pm}$0.13 months in males, respectively. For the growth curve parameters and derived statistics, the phenotypic correlations of maturing rate with gain rate at inflection, mature weight, and inflection point were - .30, - .77, and - .93 in male, and - .31, - .78 and - .94 in female, respectively. Matrure weight was positively correlated to the inflection point as + .89 in both male and female, indicating that late maturing pigs with lower k had longer maturing period with increasing gain rate and reached point of inflection later than early maturing pigs with higher k, and grew to larger mature weight. Backfat thickness and erode fat contents were correlated with mature weight positively in male and negatively in female, and correlated with gain rate at inflection point positively in both male and female, of which coefficients were as high as .42 and .50 in male, respectively.

Comparison of the accuracy of digitally fabricated polyurethane model and conventional gypsum model

  • PURPOSE. The accuracy of a gypsum model (GM), which was taken using a conventional silicone impression technique, was compared with that of a polyurethane model (PM), which was taken using an iTero$^{TM}$ digital impression system. MATERIALS AND METHODS. The maxillary first molar artificial tooth was selected as the reference tooth. The GMs were fabricated through a silicone impression of a reference tooth, and PMs were fabricated by a digital impression (n=9, in each group). The reference tooth and experimental models were scanned using a 3 shape convince$^{TM}$ scan system. Each GM and PM image was superimposed on the registered reference model (RM) and 2D images were obtained. The discrepancies of the points registered on the superimposed images were measured and defined as GM-RM group and PM-RM group. Statistical analysis was performed using a Student's T-test (${\alpha}=0.05$). RESULTS. A comparison of the absolute value of the discrepancy revealed a significant difference between the two groups only at the occlusal surface. The GM group showed a smaller mean discrepancy than the PM group. Significant differences in the GM-RM group and PM-RM group were observed in the margins (point a and f), mesial mid-axial wall (point b) and occlusal surfaces (point c and d). CONCLUSION. Under the conditions examined, the digitally fabricated polyurethane model showed a tendency for a reduced size in the margin than the reference tooth. The conventional gypsum model showed a smaller discrepancy on the occlusal surface than the polyurethane model.
