A Study with $P^{32}$ on Availability of phosphorus in Pasture Soils of Jeju Island ($P^{32}$에 의(依)한 제주목야토양(濟州牧野土壤)의 유효인산(有效燐酸)에 관(關)한 연구(硏究) -목초별(牧草別) 토양(土壤) 인산(燐酸)의 이용력(利用力)에 관(關)하여-)

  • A pot experiment with $P^{32}$ was carried out to investigate the soil phosphorus availability to four leguminous forage crops and three graminaceous, of black volcanic ash soil and red one. Soil phosphorus was extracted with 6 different extractants and also fractionated in Fe, Al and Ca phosphorus. The results were: 1) Soil phosphorus availability was in decreasing order of Italian rye grass${\gg}$ soybean> cassia> corn> weeping love grass${\gg}$ Korean lespedeza> Red clover and they might be grouped into three levels by A-value, over 1000, 200-500 and below 40 $p_{2}O_{5}\;kg/ha$. 2) The amount of various available phosphorus and phosoborus fraction in the black soil was higher than that in the red soil. No difference in phosphorus availabiliy to forage crops was shown between two soils. Therefore an extractant able to draw out similar amount of phosphorus from two soil will be suitable for determining the phosphorus availability index. 3) Two extractants, one extracting 20 ppm as maximum and the other extracting 100 ppm as minimum will be recommendable for determining the availability of phosphorus; the former for red clover and Lespedeza and the latter for others. Truog method may be good for the former but no appropriate method for the latter was found in the methods used. 4) T/R ratios of legumes were negatively correlated at 5% level with % phosphorus from fertilizer (% pdF). Legumes showed below 50 of % pdF over 5 of T/R ratio and over 80 of % pdF below 5 of T/R.

Au-Ag Minerals and Genetic Environments from the Yeongdeog Gold-Silver Deposits, Korea (영덕(盈德) 금(金)-은광상(銀鑛床)에서 산출(産出)되는 금(金)-은광물(銀鑛物)과 광상(鑛床)의 생성환경(生成環境))

  • The Yeongdeog gold-silver deposits at Jipum, Gyeongsangbugdo, is of a middle Paleogene $(45.52{\pm}1.02Ma)$ vein type, and is hosted in shale and sandstone of Cretaceous age. Based on mineral paragenesis, vein structure and mineral assemblages, the ore mineralization can be divided into two distinct depositional stages. The early stage is associated with base-metals such as pyrite, arsenopyrite (27.99~30.99 at%), hematite, rutile, pyrrhotite, sphalerite (10.53~18.42 FeS mole%), chalcopyrite and galena with wallrock alteration such as chlorite, sericite and pyrite. The late stage is characterized by the Au-Ag mineralization such as electrum, Ag-bearing tetrahedrite, freibergite, pyrargyrite, unidentified mineral, pyrite, sphalerite (1.08~5.57 FeS mole%), chalcopyrite and galena. Fluid inclusion data indicate that fluid temperatures and salinities range from 343 to $227^{\circ}C$ and from 8.3 to 5.7 wt% eq. NaCl in early stage, respectively. Temperatures and salinities of NaCl eq. wt% range from 299 to $225^{\circ}C$ and from 12.9 to 4.3 in late stage, respectively. They suggest that complex cooling histories were occured by the mixing of the fluids. Sulfur fugacity $(-logfs_2)$ deduced by mineral assemblages and composition ranges from 8.3 to 14.7 atm. in early stage, and from 8.8 to 14.5 atm. in late stage. It suggests that the mineralization was related to decrease of temperature in early stage and fluctuations of $fS_2$ with decrease of temperature in late stage. Sulfur and oxygen isotope compositions are 4.48~5.60‰ and 9.25~10.8% in early stage, and late stage is 4.84~7.00‰ and 5.7‰, respectively. It indicated that hydrothermal fluids may be magmatic origin with some degree of mixing of another water during paragenetic time.

Depletion of Cytoplasmic Tail of UL18 Enhances and Stabilizes the Surface Expression of UL18

  • Background: Human cytomegalovirus UL18, a MHC class I homologue, has been considered a natural killer (NK) cell decoy. It ligates LIR-1/ILT2 (CD85j), an NK inhibitory receptor, to prevent lysis of infected target cells. However, precise role of UL18 to NK cell cytotoxicity is yet elusive. Difficulty in clarifying the function of UL18 lies in complication in detecting UL18 mainly due to low level expression of UL18 on the surface and gradual loss of its expression. Methods: To overcome this hurdle, cDNA of cytoplasmic tail-less UL18 was constructed and expressed in swine endothelial cell (SEC). The expression level and its stability in the cell surface were monitored with FACS analysis. Results: Surface expression of UL18 is up-regulated by removing cytoplasmic tail portion from UL18F (a full sequence of UL18). SECs transfected with a cDNA of UL18CY (a cytoplasmic tail-less UL18) stably expressed UL18 molecule on the surface without gradual loss of its expression during 6 week continuous cultures. In the NK cytotoxicity assay, UL18 functions either inhibiting or activating NK cell cytotoxicity according to the source of NK cells. We found that there is individual susceptibility in determining whether the engagement of NK cell and UL18 results in overall inhibiting or activating NK cell cytotoxicity. Conclusion: In this study, we found that cytoplasmic tail is closely related to the regulatory function for controlling surface expression of UL18. Furthermore, by constructing stable cell line in which UL18 expression is up-regulated and stable, we provided a useful tool to clarify exact functions of UL18 on various immune cells having ILT2 receptor.

Molecular Cloning and Enzymatic Characterization of Cyclomaltodextrinase from Hyperthermophilic Archaeon Thermococcus sp. CL1

  • Genome organization near cyclomaltodextrinases (CDases) was analyzed and compared for four different hyperthermophilic archaea: Thermococcus, Pyrococcus, Staphylothermus, and Thermofilum. A gene (CL1_0884) encoding a putative CDase from Thermococcus sp. CL1 (tccd) was cloned and expressed in Escherichia coli. TcCD was confirmed to be highly thermostable, with optimal activity at $85^{\circ}C$. The melting temperature of TcCD was determined to be $93^{\circ}C$ by both differential scanning calorimetry and differential scanning fluorimetry. A size-exclusion chromatography experiment showed that TcCD exists as a monomer. TcCD preferentially hydrolyzed ${\alpha}$-cyclodextrin (${\alpha}$-CD), and at the initial stage catalyzed a ring-opening reaction by cleaving one ${\alpha}$-1,4-glycosidic linkage of the CD ring to produce the corresponding single maltooligosaccharide. Furthermore, TcCD could hydrolyze branched CDs (G1-${\alpha}$-CD, G1-${\beta}$-CD, and G2-${\beta}$-CD) to yield significant amounts (45%, 40%, and 46%) of isomaltooligosaccharides (panose and $6^2$-${\alpha}$-maltosylmaltose) in addition to glucose and maltose. This enzyme is one of the most thermostable maltogenic amylases reported, and might be of potential value in the production of isomaltooligosaccharides in the food industry.

Slat Noise Source Modeling of Multi-element Airfoil in High-lift Configuration

  • We investigate the slat noise generation mechanism by using large-eddy simulation (LES) and simple source modeling based on linearized Euler equations. An incompressible LES of an MD 30P30N three-element airfoil in the high-lift configuration is conducted at $Re_c=1.7{\times}10^6$. Using the total derivative of the hydrodynamic pressure (DP/Dt) acquired from the incompressible LES, representative noise sources in the slat cove region are characterized in terms of simple sources such as frequency-specific monopoles and dipoles. Acoustic radiation around the 30P30N multi-element airfoil is effectively computed using the Brinkman penalization method incorporated with the linearized Euler equation. The directivity pattern of $p^{\prime}_{rms}$ at $r=20c_{slat}$ in the multiple sources is closely compared to that obtained by the application of the LES/Ffowcs-Williams and Hawking's methods to the entire flow field. The power spectrum of p' at ${\theta}=290^{\circ}$ is in good agreement with the data reported in BANC-III, especially the broadband part of the spectrum with a decaying slope ${\propto}f^{-3}$.

Effect of Low Salt Concentrations on Microbial Changes During Kimchi Fermentation Monitored by PCR-DGGE and Their Sensory Acceptance

  • Various salt concentrations (1.0%, 1.3%, 1.6%, 1.9%, and 2.1% labeled as sample A, B, C, D, and E, respectively) were investigated for microbial diversity, identification of Lactic Acid Bacteria (LAB) in salted kimchi cabbage, prepared under laboratory conditions. These samples were stored at 4°C for 5 weeks in proper aluminum-metalized pouch packaging with calcium hydroxide gas absorber. A culture-independent method known as polymerase chain reaction - denaturing gradient gel electrophoresis was carried out to identify LAB distributions among various salt concentration samples that had identified 2 Weissella (W. confusa and W. soli), 1 Lactobacillus (Lb. sakei), and 3 Leuconostoc (Lc. mesenteroides, Lc. lactis, and Lc. gelidum) in the overall kimchi samples. The pH, titratable acidity, viable cell counts, and coliform counts were not affected by salt variations. In order to assess sensory acceptance, the conducted sensory evaluation using a 9-point hedonic scale had revealed that samples with 1.3% salt concentration (lower than the manufacturer's regular salt concentration) was more preferred, indicating that the use of 1.3% salt concentration was acceptable in normal kimchi fermentation for its quality and safety. Despite similarities in pH, titratable acidity, viable cell counts, coliform counts, and LAB distributions among the various salt concentrations of kimchi samples, the sample with 1.3% salt concentration was shown to be the most preferred, indicating that this salt concentration was suitable in kimchi production in order to reduce salt intake through kimchi consumptions.

Simultaneous Biocatalytic Synthesis of Panose During Lactate Fermentation in Kimchi

  • As a functional additive for intestinal microflora, panose ($6^2-{\alpha}$-D-glucopyranosylmaltose) was synthesized during kimchi fermentation using the glucose transferring reaction of glucansucrase from Leuconostoc mesenteroides. For the glucose transferring reaction, sucrose and maltose were added ($2\%$ each, w/v) to dongchimi-kimchi as the glucosyl donor and acceptor molecule, respectively. After five days of incubation at $10^{\circ}C$, referring to the initial phase for the production of lactic acid in kimchi, over $60\%$ (w/v) of the total sugars were converted into panose and other branched oligosaccharides. Thereafter, the kimchi was stored at $4^{\circ}C$ and the amount of panose remained at a constant level for three weeks, thereby indicating the stability of panose to microbial degradation during the period of kimchi consumption. The use of maltose as the acceptor molecule in the kimchi also facilitated a lower viscosity in the kimchi-juice by preventing the synthesis of a dextran-like polymer which caused an unfavorable taste. Accordingly, the application of this new method of simultaneous biocatalytic synthesis of oligosaccharides during lactate fermentation should facilitate the extensive development of new function-added lactate foods.

The Effects of Drying Method and Spice Extracts Added to Beef Jerky on the Quality Characteristics of Beef Jerky (건조방법과 향신료 추출물 첨가가 육포의 품질특성에 미치는 영향)

  • To develope Hanbang beef jerky as health food, six kinds of beef jerky were prepared by adding sugar (A), licorice (B) and three kinds of spice extracts (clove: C, fennel fruit: D and Chungyang green pepper extract: E) and mixture of all extracts (F). The effects of the drying method and added spice extracts on the quality characteristics of the beef jerky were evaluated. In general, the air-blast dried beef jerky contained $31{\sim}33%$ moisture, $50.0{\sim}51.2%$ crude protein, $7.2{\sim}7.8%$ crude lipid and $3.0{\sim}3.3%$ crude ash. For the mineral content of the air-blast dried jerky, the most prevalent mineral was Na $(1540.08{\sim}1838.17$ mg%), followed by K, P, Mg, and Ca. The Ca content of the beef jerky was highest in the mixed extract group (88.53 mg%), and the lowest content was in sugar-added group (53.12 mg%). For the color properties, the L-value (lightness), a-value (redness) and b-value (yellowness) were higher in the air-blast dried beef jerky than in the hot air dried samples. The drying methods showed their greatest affect on the redness (a) for all six jerkies (p<0.001). For the preference by sensory evaluation, the beef jerky samples with added sugar (A) and licorice extract (B) had significantly higher scores than the beef jerky samples with the added spice extracts, for both of air-blast drying and hot air drying (p<0.05). Preference for the air-blast dried beef jerkies with added clove (C) and fennel fruits (D) were significantly higher, in terms of taste, color, softness and aftertaste as compared to the respective hot air dried jerkies (p<0.01). Considering all the obtained results, we concluded that licorice and spice extracts can be used as natural preservatives in the development of health foods and the air-blast drying method is recommended to improve the quality characteristics of beef jerky.

Evaluation of Chinese Brown Rice as an Alternative Energy Source in Pig Diets

  • A total of six crossbred barrows ($Duroc{\times}Landrace{\times}Large$ White, $44.17{\pm}1.94kg$ BW) were housed conducted to evaluate apparent fecal digestibilities of Brown Rice (BR) as an alternative energy source in growing pigs. Pigs were housed individually on metabolism crate on the basis of body weight. Four treatments contained: 1) 100% of corn-soybean meal (C100; Control diet), 2) 75% of corn-soybean meal diet plus 25% of corn meal (C25), 3) 100% of brown rice-soybean meal diet (BR100), 4) 75% of brown rice-soybean meal diet plus 25% of brown rice meal (BR25). Brown rice has an excellent gross energy and crude protein composition compared to corn. The BR used had 3,801 kcal of gross energy/kg, 8.0% crude protein, 2.6% of ether extract, 0.035% calcium and 0.35% total phosphorus. The best digestibilities of energy (87.75%), DM (81.71%) and CP (78.57%) were observed in BR 100 group and the worst were found in Corn 25 group. The nutrient digestibility was not significantly different in most nutrients. Through this experiment, BR appeared a good alternative energy source that can replace corn yellow to 100% in growing pigs. Therefore, the price relationship between corn and BR may provide an excellent opportunity for pork producers to use BR in order to reduce feed costs provided that diet has been balanced for digestible amino acids.

Seasonal Prevalence and Altitudinal Distribution of the Flies in Mt. Hallasan, Jejudo Island, Korea (제주도 한라산 일대 파리류의 계절적인 발생소장 및 수직분포)

  • This study focuses on the fauna, seasonal prevalence and altitudinal distribution of the flies from March to November, 2008 in Mt. Hallasan, Jejudo Island, Korea. The fauna of flies was 39 species belonging to 5 Families which consist of 12 Calliphoridae, 19 Muscidae, 5 Sarcophagidae, 1 Dryomyzidae 1, Scathophagidae and 1 Anthomyiidae. The total number of flies collected was 21,605 with sex ratio of 57.98% in Mt. Halla. The ratio is higher than that of the residential area and other mountain survey sites. The average number of per trap individuals collected on the trapping day is 267, which is similar with the average of Mts. Jiryong, Juwang, Palgong and Gumo. The five dominant species showed a distinctive seasonal prevalence of 92.6% of the total flies: Chrysomyia pinguis (34.2%), Calliphora lata (25.3%), Hydrotaea dentipes(23.1%), Fannia scalaris (5.1%), and Lucilia caesar (4.9%). The seasonal prevalence of the flies was from the beginning of March to the end of November, and the highest peak time of the whole flies, males and females were different respectively. In the similarity index of the flies collected in different altitude, that of between 1,000 meters and 1,500 meters was the highest among the indices of 500 meters and above 1,000 meters. The index at each altitude was above 80% because of the same area. The diversity index of fly species was between 0.00 and 0.88 through altitudinal traps monthly. The highest index in Trap 1 was 0.88 in October, in Trap 2, 0.82 in July and in Trap 3, 0.63 in July. The diversity above 1,000 meters was most various in July. The five dominant species of C. pinguis, C. lata, H dentipes, F. scalaris, and L. caesar showed distinctive seasonal prevalence, relative abundance and altitudinal distribution respectively.
