Effect of dietary supplementation with Spirulina on the expressions of AANAT, ADRB3, BTG2 and FASN genes in the subcutaneous adipose and Longissimus dorsi muscle tissues of purebred and crossbred Australian sheep

  • Background: The demand for healthy, lean and consistent meat products containing low saturated fatty acid content and high quality polyunsaturated fatty acids (PUFA), especially long-chain (${\geq}C_{20}$) omega-3 PUFA, has increased in recent times. Fat deposition is altered by both the genetic background and dietary supplements, and this study aimed to assess the effect of dietary Spirulina supplementation levels on the mRNA expression patterns of genes controlling lipid metabolism in the subcutaneous adipose tissue (SAT) and Longissimus dorsi (ld) muscle of Australian crossbred sheep. Methods: Twenty-four weaned lambs belonging to four breeds under the same management conditions were maintained on ryegrass pasture and fed three levels of Spirulina supplement (control, low and high). In terms of nutrient composition, Spirulina is a nutrient-rich supplement that contains all essential amino acids, vitamins and minerals. It also is a rich source of carotenoids and fatty acids, especially gamma-linolenic acid (GLA) that infer health benefits. After slaughter, subcutaneous adipose tissue (SAT) and ld samples were subjected to mRNA extraction and reverse transcription using quantitative polymerase chain reaction (RT-qPCR) to assess the mRNA expression levels of the Aralkylamine N-acetyltransferase (AANAT), Adrenergic beta-3 receptor (ADRB3), B-cell translocation gene 2 (BTG2) and Fatty acid synthase (FASN) genes, which are associated with lipid metabolism. Results: Both low and high Spirulina supplementation levels strongly up-regulated the transcription of all the selected genes in both SAT and ld tissues (mostly in the subcutaneous adipose), but sheep breed and sex did not influence the gene expression patterns in these tissues. Conclusions: The evidence indicates that high Spirulina supplementation level resulted in a decrease in intramuscular fat content in Australian purebred and crossbred sheep due to the enhanced production of melatonin in sheep muscle tissues and strong up-regulation of mRNA expression of BTG2 in SAT which negatively affected fat deposition. In contrast, low Spirulina supplementation level strongly up-regulated the ADRB3 and FASN genes responsible for fat production. These findings are consistent with the observed phenotypic data suggesting that low Spirulina supplementation level can increase lamb production, with higher long-chain PUFA content.

Determination of Ametryn Residue in Agricultural Commodities Using HPLC-UVD/MS (HPLC-UVD/MS를 이용한 농산물 중 ametryn의 분석)

  • Ametryn is used in USA, China, and Japan, but not introduced in Korea yet. So, MRL (Maximum Residue Level), and analytical method of ametryn were not establishment in Korea. Therefore, this experiment was conducted to establish a determination method for ametryn residue in crops using HPLC-UVD/MS. Ametryn residue was extracted with acetone from representative samples of five raw products which comprised hulled rice, soybean, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and dichloromethane partition was followed to recover ametryn from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The ametryn was quantitated by HPLC with UVD, using a Tosoh ODS 120T ($4.6{\times}250$ mm) column. The crops were fortified with ametryn at 2 levels per crop. Mean recovery ratio were ranged from 83.7% for a 0.2 mg/kg in soybean to 91.1% for a 1.0 mg/kg in hulled rice. The coefficients of variation were ranged from 1.2% for a 1.0 mg/kg in hulled rice to 3.6% for a 1.0 mg/kg in soybean. Quantitative limit of amatryn was 0.02 mg/kg in representative 5 crop samples. A LC/MS with selected-ion monitoring was also provided to confirm the suspected residue. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of ametryne in agricultural commodities.

키토산 처리 콩나물의 성장 중 당 및 유리아미노산의 함량변화

  • 키토산처리 콩나물에서 발아율이 높고 배축의 신장도가 높은 현상을 조사하기 위하여 전보에서와 동일한 방법으로 콩에 0.05% 키토산(493kDa, 금호화성, 한국)을 함유하는 0.05% 초산 용액 4배량을 가하여 8시간 침지한 후 2$0^{\circ}C$에서 2시간 간격으로 5일간 주수하면서 콩나물, 자엽, 배축으로 구분하여 전당, 환원당, 비환원당 및 유리아미노산의 함량변화를 조사하였다. 콩나물의 전당은 대조구, 키토산 처리구가 모두 성장에 따라 감소하였는데 대조구(1196$\longrightarrow$820 mg/100 g-f.w)보다 처리구(1241$\longrightarrow$670 mg/100 g-f.w)에서의 감소율이 높았다. 자엽의 전당함량은 성장에 따라 감소하였으며 키토산 처리구에서 현저하였고, 배축에서는 증가하는 경향을 나타내었다. 환원당 함량은 콩나물의 성장에 따라 감소하였으며 키토산 처리에 따른 뚜렷한 차이가 없었다. 그러나 자엽에서는 성장에 따라 감소하였으며 감소경향은 키토산처리구에서 높았다. 키토산 처리구에서 전당과 환원당의 감소율이 대조구보다 높은 현상은 키토산 처리에 의하여 생체 대사가 보다 활성화되기 때문이라 생각되며, 키토산처리에 의하여 발아율이 높고, 배축의 신장도가 높은 현상과 관련이 있는 것으로 판단된다. 유리아미노산은 재배 5일째 자엽과 배축의 유리아미노산 함량을 분석한 결과 aspartic acid의 함량이 가장 많았으며, 대조구의 경우 자엽에서는 25.4 mg/100 g-f.w, 배축에서는 88.2 mg/100 g-f.w이었다. 키토산처리구의 경우는 자엽에서는 23.12 mg/100 g-f.w, 배축에서는 120.18 mg/100 g-f.w이었다.높은 항 곰팡이 활성을 나타내었다. 이 외에도 황련의 물 추출물은 M. miehei 균주에서 우수한 항 곰팡이 활성을 나타내었다. 생약재의 70% Ethanol추출물을 농도별로 제조하여 항 곰팡이 활성을 살펴 본 결과, 70% ethanol추출물에서는 계피와 파고지, 초두구, 황련이 항 곰팡이 활성이 우수하였으며, 특히 계피의 70% ethanol 추출물에서는 5가지 균종에서 모두 우수한 항 곰팡이 활성이 나타났다. 균을 제외한 5가지 균종에서 높은 항균 활성을 보였으며, 이외에 생약재는 강황, 석곡, 황련, 호장근의 70% ethanol 추출물은 4가지의 균주에 대해서 항균 활성을 나타내었다. 또한 파고지, 오미자의 70% ethanol 추출물은 3가지 균주에 대해서 항균활성을 나타냈으며, 천궁, 육두구, 오수유, 갈근, 계피, 가시오가피의 70% ethanol 추출물은 2가지 균주에 대해 항균 활성을 나타내었다. 항균활성이 우수한 생약재를 농도별로 활성을 조사한 결과, 물 추출물과 10% Ethanol 추출물 모두 낮은 농도에서도 우수한 항균활성을 나타내었다.취와 함께 점질성 갈변물질이 생성되었다. 이와 같은 결과로 볼 때, BAAG의 처리는 BAAC의 경우보다 가격은 저렴하면서도 항균력은 우수한 천연 항균복합제재로써 농산물 식품원료에 적용하여 선도유지 기간을 연장할 수 있는 효과를 기대할 수 있었다. 과일 등의 포장제로서 이용할 가능성을 확인하였다.로 [-wh] 겹의문사는 복수 의미를 지닐 수 없 다. 그러면 단수 의미는 어떻게 생성되는가\ulcorner 본 논문에서는 표면적 형태에도 불구하고 [-wh]의미의 겹의문사는 병렬적 관계의 합성어가 아니라 내부구조를 지니지 않은 단순한 단어(minimal $X^{0}$ elemen

Screening of $\alpha$-amylase and $\alpha$-glucosidase Inhibitors from Brazilian Plant Extracts for Treatment of Rumen Acidosis (100종 브라질 식물 추출물로부터 반추동물 산독증 예방치료를 위한 $\alpha$-amylase 및 $\alpha$-glucosidase 저해제의 선별)

  • To develop anti-acidosis and anti-diabetes agentsfrom natural products, the inhibitory activities of Brazilian plant extracts against microbial $\alpha$-amylase and $\alpha$-glucosidase were evaluated. Among 100 different ethanol extracts tested, those of Acacia jurema Mart., Anacardium humile A. St.-Hil., Cedrela odorata L., and Guazuma ulmifolia Lam showed good inhibitoryactivities toward both enzymes. In addition, an extract of Plumeria drastica Mart. showed specific inhibition of $\alpha$-amylase, whereas that of Eugenia uniflora L. demonstrated strong inhibition of the enzyme. IC50 values of $\alpha$-amylase inhibition suggested that the extract of A. humile A. St.-Hil., which has been used as an anti-diabetes medicine in Brazil, had potent inhibitory activity. The IC50 for the A. humile A. St.-Hil. extract ($91.2{\mu}g/mL$) was similar to that of acarbose ($50.5{\mu}g/mL$). This activity of A. humile A. St.-Hil. was not reduced by heat or acid treatment. Moreover, treatment with HCl (0.01 M) for 1 h increased the inhibitory activity from 57.5% to 81.2%. Also, the extract did not cause hemolysis of human red blood cells at levels up to 1 mg/mL. The results indicate that the extract of A. humile A. St.-Hil. is potentially useful as an anti-acidosis and anti-diabetes agent.

Quality Characteristics of Tangor Jam including Fructo Oligosaccharide and Isomalto Oligosaccharide (프락토 올리고당과 이소말토 올리고당을 첨가한 국내산 한라봉잼의 품질특성 연구)

  • The present study relates to the development of a processed product for expansion of use of Tangor which is mostly eaten raw in winter. Furthermore, Tangor jam containing fructo oligosaccharide and isomalto oligosaccharide was prepared, and thus quality characteristics of the jam have been determined according to the sugar concentration by making a low-sugar jam by adding substitute sweeteners instead of sugar, because low-sugar products are recently preferred in the market. Furthermore, the general elements of flesh and rind of Tangor, DPPH free radical scavenging activity, chromaticity, sugar content, pH, acidity, and preference have been checked. As a result of the experiment, the chromaticity, luminosity (L), the sugar content, and pH were the lowest in the control group. In F1, F2, and F3, which gradually added fructo oligosaccharide, and I1, I2, and I3, which slowly added isomalto oligosaccharide, luminosity significantly increased with the addition of oligosaccharide. In contrast, as the amount of addition of oligosaccharide increases, red chromaticity (a), yellow chromaticity (b), and sugar content have significantly decreased. Lastly, in the preference test using a 9 point test scheme, F2 and I2 have been considered most appropriate when producing Tangor jam.

The Influences of Addition of Sugar with or without L. buchneri on Fermentation and Aerobic Stability of Whole Crop Maize Silage Ensiled under Anaerobic Silos

  • The whole plant of crop maize was chopped and ensiled in airtight 1-L capacity glass jars to determine the influence of residual sugar on anaerobic yeast growth and on the fermentation of lactic acid by L. buchneri in whole crop maize silage. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g L. buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1+L.b.$ and $g_2+L.b.$ All silos were stored in the dark at $20^{\circ}C$ until end of experiment. Jars were opened on duplicates at day 2, 7, 14, 28, 56 or triplicates at day 91 after ensiling for measuring the pH, microbiological enumeration and fermentative products. Results indicated that acidification rates for all silages were very fast, no difference occurred among treatments before day 28. After day 28 the pH values for silages inoculated by L. buchneri. with or without sugar tended to increase especially for treated only with L. buchneri, resulting in higher (p<0.01) finial pH than uninoculated silages. Compared with control silage, the added sugar significantly (p<0.01) increased dry matter (DM) loss, L. buchneri enhanced (p<0.01) DM loss further at different sugar existence. Silages inoculated by L. buchneri only or in combination with sugar addition contained less (p<0.01) lactic acid than the correspondent silages without inoculation with L. buchneri. In comparison with control, ethanol production is about 3 or 6 fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The added sugar resulted in less acetic acid concentration (p<0.01) than control, but inoculation with L. buchneri increased (p<0.01) acetic acid than correspondent uninoculated silages at different sugar levels. No butyric acid and propionic acid were found in uninoculted silages, silages inoculated with L. buchneri. produced more propionic acid, 1-propanol and butyric acid. Lactobacilli counts were not influenced by added sugar, but increased (p<0.01) with inoculation of L. buchneri. The added sugar increased significantly (p<0.01) the yeast count, whereas L. buchneri showed the contrary effect. No differences were found in the aerobic stability among all treatments. In conclusions, 1) the added sugars encourage the growth of yeast and yeasts convert extra sugar into ethanol in maize silages. 2) The added sugars and L. buchneri do not influence the aerobic stability of silages stored in anaerobic silos.

Farm Study of Direct Seeding Cultivation of the Korean Ginseng(Panax ginseng C. A. Meyer) (고려인삼의 직파재배에 관한 조사 연구)

  • Direct seeding is a cultivation method of Korean ginseng which can cut down production cost and increase productivity. This study was conducted to obtain detailed information about this method and to aid the development of it in Korea. Major pre-crops before ginseng cultivation were rice, corn and rye, and organic matters, such as rice straw, rye straw and poultry feces were used for basal fertilization. In direct seeding of the ginseng field, seeding density was 196 to 210 seeds per $3.3m^2$ and germination ratio was 67%. Survival ratio of 4-year-old ginsengs which were products of direct seeding was 51%, and more than 100 plants per $3.3m^2$ survived. The most critical diseases in the directly seeded ginseng field were grey mold, damping off, and stem diseases including stem spot disease. Plant growth of the ginseng cultivated by the direct seeding method was not different from the ginseng cultivated by transplanting method. But the root of the ginseng cultivated by the direct seeding method took the shape of a shorter main root compared to the ginseng cultivated by the transplanting method. Root yield per $3.3m^2$ of the direct seeding ginseng field was larger compared to the field of the transplanting cultivation.

Anti-proliferative Effects of the Isothiocyanate Sulforaphane on the Growth of Human Cervical Carcinoma HeLa Cells (Sulforaphane에 의한 HeLa 인체자궁경부함세포의 증식 억제 기전 연구)

  • Sulforaphane, an isothiocyanate derived from hydrolysis of glucoraphanin in broccoli and other cruciferous vegetables, was shown to induce phase II detoxification enzymes and inhibit chemically induced mammary tumors in rodents. Recently, sulforaphane is known to induce cell cycle arrest and apoptosis in human cancer cells, however its molecular mechanisms are poorly understood. In the present study, we demonstrated that sulforaphane acted to inhibit proliferation and induce morphological changes of human cervical carcinoma HeLa cells. Treatment of HeLa cells with $10{\mu}M\;or\;15{\mu}M$ sulforaphane resulted in significant G2/M cell cycle arrest as determined by flow cytometry. Moreover, $20{\mu}M$ sulforaphane significantly induced the population of sub-G1 cells (9.83 fold of control). This anti-proliferative effect of sulforaphane was accompanied by a marked inhibition of cyclin A and cyclin-dependent kinase (Cdk)4 protein and concomitant induction of Cdc2, Cdk inhibitor p16 and p21. However, sulforaphane did not affect the levels of cyelooxygenases and telomere-regulatory gene products. Although further studies are needed, the present work suggests that sulforaphane may be a potential chemoprevetive/ chemotherapeutic agent for the treatment of human cancer cells.

Comparison of Radish Cultivars for Physicochemical Properties and Kakdugi Preparation (무 품종별 이화학적 특성 및 깍두기 가공적성)

  • For scientification of commercial fermented radish products the study on physicochemical and processing properties of various radish cultivars should be proceeded and needed. Moisture contents of 3 parts of root ranged from 91.3 to 94.0%. Although, the upper part showed less content of moisture and ash than other parts, the upper part contained higher amount of crude protein and soluble solids. The sugar contents of Baekkwang grown on spring was the lowest$(5.0^{\circ}Brix)$ and that of Taebaek grown on autumn was the highest $(6.6^{\circ}Brix)$ among 6 cultivars. Significant difference of acidity was shown in various cultivars but not in each parts. The most abundant soluble sugars in radish root was glucose ranging from 15.8 to $27.3\;{\mu}mole/g\;f.w.$ fresh weight, f.w. and followed by fructose ranging from 16.4 to $23.1\;{\mu}mole/g\;f.w.$ However, the content of sucrose ranging from 0.7 to $2.7\;{\mu}mole/g\;f.w.$ was the lowest compared to others. Hardness of fresh root was the highest in Taebaek $[93.4{\sim}156.9N/m^2(\times10^3)]$, followed by Dongja and Chudong, and the lowest in Baekkwang. Changes in rigidity of roots during brining were determined. Although the rigidity was drastically reduced during initial 60 min., it was reversely increased during further storage. Sensory acceptabilities of Taebaek and Dongja were the best among 7 cultivars. Optimal time of radish fermentation at pH of 4.2 and acidity of 0.6% was between 24 and 28 days of storage.

Dietary Risk Factors Associated with Hypertension in Patients (고혈압 환자의 식이관련 위험요인 분석에 관한 연구)

  • This study was performed to determine the dietary risk factors associated with hypertension. The hypertensive group were composed of 112 hypertensive patients (male 53, female 59) who first visited the hypertension clinic and had been diagnosed as having primary hypertension ($SBP{\ge}140mmHg\;or\;DBP{\ge}90mmHg$). The regular visitors or the subjects on special diets or medical therapies were excluded. The normal group consisted of as subjects (male 41, female 54) matched with age and socioeconomic levels. The subjects having higher intakes (above the 75 percentile) in energy, protein, iron, vitamin A or C showed significantly higher hypertension risk estimated with odds ratio after the covariance factors (age, sex and BMI) were adjusted. More than 2400 mg of sodium (6 g of salt) intake was associated with significantly higher risk of hypertension (odds ratio: 1.773, CI: 1.014-3.014 for $SBP{\ge}140mmHg$; odds ratio: 2.373, CI: 1.359-4.215 for $DBP{\ge}90mmHg$). Hypertensive group showed significantly increased intakes of vegetables and fish and shell fish compared to the normal group. When the vegetable intakes were classified into Kimchi, fresh vegetables and cooked vegetables with seasoning, the hypertensive group was observed as having higher intakes of Kimchi and cooked vegetables with seasoning. The intakes of highest quartile for vegetables (${\ge}327g/day$) (odds ratio: 3.164, CI: 1.740-5.752), fish and their products (${\ge}102g/day$) (odds ratio: 2.756, CI: 1.486-5.109), grains(${\ge}311g/day$) (odds ratio 2.393, CI: 1.186-4.832), meats and their product (${\ge}106g$) (odds ratio: 2.210, CI: 1.225 - 3.987) compared to the lower were significantly associated with the higher risk of hypertension estimated with DBP (${\ge}90mmHg$) after covariance factors were adjusted. In conclusion, our findings confirm that higher intake of energy or sodium are associated with the increased risk of hypertension. Because increased intake of vegetable or fish was associated with the higher risk of hypertension, in contrast with the finding of western countries, choosing or preparation of vegetables or fish with reduced salt is recommended.
