Fatty Acid Profiles of Supraspinatus, Longissimus lumborum and Semitendinosus Muscles and Serum in Kacang Goats Supplemented with Inorganic Selenium and Iodine

  • Fat and fatty acids in muscle and adipose tissues are among the major factors influencing meat quality particularly nutritional value and palatability. The present study was carried out to examine the effects of supplementing inorganic selenium (Se), iodine (I) and a combination of both on fatty acid compositions in serum, and supraspinatus (SS), longissimus lumborum (LL), and semitendinosus (ST) muscles in goats. Twenty-four, 7 to 8 months old, Kacang male goats with a mean live weight of $22.00{\pm}1.17kg$ were individually and randomly assigned into four groups of six animals each for 100 d of feeding prior to slaughter. The animals were offered the same concentrate (basal) diet as 1% of body weight with ad libitum amount of fresh guinea grass. The four groups were as follows: T1 (control) - basal diet without supplementation; T2 - basal diet with 0.6 mg Se/kg DM; T3 - basal diet with 0.6 mg I/kg DM; T4 - basal diet with combination of 0.6 mg Se/kg DM and 0.6 mg I/kg DM. The major fatty acids (FAs) detected in the serum were palmitic (C16:0), stearic (C18:0), oleic (C18:1n9) and linoleic (C18:2n-6), while the major FAs in the selected muscles were C16:0, C18:0 and C18:1n9 acids. The main polyunsaturated fatty acids (PUFA) detected in muscles and serum were (CI8:2n-6), linolenic acid (C18:3n-3), and arachidonic acid (C20:4n-6). No significant differences (p>0.05) were observed in the concentration of total saturated fatty acids (SFA) among the four groups. PUFA concentrations in the goats supplemented with Se (T2) were significantly higher (p<0.05) than the goats of the control group (T1). The PUFA: SFA ratio was significantly higher in the animals supplemented with dietary Se (T2) than those of control ones (T1). It is concluded that dietary supplementation of inorganic Se increased the unsaturated fatty acids in muscle. The supplementation of iodine with or without Se had negligible effects on muscle fatty acid content of Kacang crossbred male goats.

한국 주요 어종의 지방산 조성 및 ${\omega}-3$ 고도 불포화 지방산의 함량 (Fatty Acid Composition and Content of Omega-3 Polyunsaturated Fatty Acids of Major Fishes Caught in Korean Seas)

  • 우리나라에서 어획되는 주요 어류 10종에 대해 지방질함량과 총지방질, 중성 및 극성지방질의 지방산 조성을 분석 하였다. 시료어종의 지방질을 구성하는 주된 지방산은 $C_{16:0}(14{\sim}24%)$, $C_{16:1}(4{\sim}13%)$, $C_{18:1}(8{\sim}29%)$, $C_{20:5}(4{\sim}18%)$, $C_{22:6}(6{\sim}33%)$ 등이었다. ${\omega}-3$계 고도불포화지방산은 어종에 따라 총지방산중 $18{\sim}48%$로 큰 차이를 나타냈다. 한편 어체를 기준으로 EPA를 다량 함유하고 있는 어종은 정어리 (1.55%), 전어 (1.11%),임연수어 (0.93%)등이고 DHA의 함유량이 높은 어종은갈치 (1.68%), 임연수어 (1.11%). 정어리 (0.95%)등으로 나타났다. 어유 기준으로는 정어리유(16.0%), 멸치유(15.6%), 명태유, 임연수어유, 전어유 등에서EPA의 함량이 10.0%이상이었으며 DHA의 함량은 꽁치유가 29.2%로 가장 많고 명태유, 멸치유, 고등어유, 가자미유도 l6.0%이상의 높은 함유량을 나타냈다.

Chemometric Approach to Fatty Acid Profiles in Soybean Cultivars by Principal Component Analysis (PCA)

  • The purpose of this study was to investigate the fatty acid profiles in 18 soybean cultivars grown in Korea. A total of eleven fatty acids were identified in the sample set, which was comprised of myristic (C14:0), palmitic (C16:0), palmitoleic (C16:1, ${\omega}7$), stearic (C18:0), oleic (C18:1, ${\omega}9$), linoleic (C18:2, ${\omega}6$), linolenic (C18:3, ${\omega}3$), arachidic (C20:0), gondoic (C20:1, ${\omega}9$), behenic (C22:0), and lignoceric (C24:0) acids by gas-liquid chromatography with flame ionization detector (GC-FID). Based on their color, yellow-, black-, brown-, and green-colored cultivars were denoted. Correlation coefficients (r) between the nine major fatty acids identified (two trace fatty acids, myristic and palmitoleic, were not included in the study) were generated and revealed an inverse association between oleic and linoleic acids (r=-0.94, p<0.05), while stearic acid was positively correlated to arachidic acid (r=0.72, p<0.05). Principal component analysis (PCA) of the fatty acid data yielded four significant principal components (PCs; i.e., eigenvalues>1), which together account for 81.49% of the total variance in the data set; with PC1 contributing 28.16% of the total. Eigen analysis of the correlation matrix loadings of the four significant PCs revealed that PC1 was mainly contributed to by oleic, linoleic, and gondoic acids, PC2 by stearic, linolenic and arachidic acids, PC3 by behenic and lignoceric acids, and PC4 by palmitic acid. The score plots generated between PC1-PC2 and PC3-PC4 segregated soybean cultivars based on fatty acid composition.

통조림에 주입된 유지중 Trans 지방산 함량에 관한 연구 (A Study on Trans Fatty Acids Contents of Vegetable Oils Added to Can)

  • 참치기름담금통조림과 굴, 홍합훈제기름담금통조림을 구입하여 Glass Capillary Gas Chromatography (Hewlett Packard, 5880 A)로 분석한 trans 지방산 함량과 산가, 과산화물가, 요오드가, 검화가 측정으로 관찰된 이화학적 성질의 변화는 다음과 같다. 1. 면실유가 주입된 참치, 굴, 홍합통조림에서 검출된 trans지방산의 대부분은 t,c-18:2이며 대두유가주입된 참치통조림에서 검출된 trans 지방산은 t,c-18:2 뿐만 아니라 c,c,t-18:3과 t,t,c-18:3도 소량 존재하였다. 2. Trans 지방산의 총평균 함량은 대두유가 주입된 참치통조림에서 2.26%로 가장 높았다. 또한 참치통조림의 경우 주입된 면실유나 대두유 모두 유통기간 긴 것에서 총 trans지방산 함량이 다소 더 높은 값을 보인 반면, 굴, 홍합통조림에서는 총 trans지방산 함량도 낮으면서 유통기간 중 변화도 거의 없는 것으로 나타났다. 3. 통조림에 주입된 유지의 유통기 간 중 산패도는 1년 내외의 기간에서는 산가가 0.5∼2.0 정도, 과산화물가는 0.6∼3.5 meq/kg oil 정도로 나타나 안정된 상태를 유지하고 있는 것으로 나타났다.

수산물의 지질에 관한 연구(제5보) -쏘가리, 꺽지, 누치 및 메기의 근육지질 조성의 비교- (Studies on the Lipid of Aquatic Products(Part 5) -Comparison of flesh Lipid Composition of Some Fresh Water Fishes Mandarin Fish Korean Perch Cornet Fist and Cat Fish-)

  • The lipid compositions including fatty acid sterol lipid class and the lipid component of the total lipid extracted from the flesh of some fresh water fishes I. e. mandarin cornet cat fish and Korean perch were compared. The levels of total lipid of flesh tissues from the fresh water fishes were high(7.4%) in cat fish but low(1.4-2.2%) in mandarin fish Korean perch and cornet fish. On the contrary the content of unsaponifiable matters found in total lipid was low(2.6%) in cat fish but high(6.0%-6.5%) in mandarin fish Korean perch and cornet fish. Total lipids were mainly composed of triglyceride(74.6-86.5%) as major component in each sample and the other lipid components of total lipid e. g. polar lipid free fatty acids and free sterol were the minor components, The major fatty acids in total lipid of each sample were{{{{ {C }_{16 { }:_{ }0 } }}}}(19.6-29.2%) {{{{ {C }_{16 { }:_{ }1 } }}}}(17.3-30.7%) and {{{{ {C }_{18 { }:_{ }1 } }}}}(16.8-29.2%) and additionally it chiefly consisted of {{{{ {C }_{14 { }:_{ }0 } }}}} and {{{{ {C }_{18 { }:_{ }2 } }}}} Particularly the contents of polyenoic acids in total lipid of cat fish were higher than those of the other fish samples. The level of cholesterol in total lipid was low (8.3mg/g) in cat fish but were high(36.9-59.9mg/g) in mandarin fish Korean perch and cornet fish. The contents of fractionated neutral lipid(NL) were higher than those of polar lipid(PL) in each sample. Particularly phospholipid content in PL was low(6.0%) in cat fish but were high(23.1-36.3%) in mandarin fish Korean perch and cornet fish. Neutral lipids were mainly consisted of triglyceride(84.5-93.4%) as amjor component in each sample and the other lipid components of neutral lipid e, g, free fatty acids and free wterol were exhibited as a minor components, The fatty acid compositon of neutral lipids was very resembled to total lipids. The phospholipid in mandarin cornet and cat-fish were mainly composed of phosphatidyl serine(23.1-49.8%) and phosphatidyl choline(20.8-45.3%) The relatively higher amounts of phosphatidyl serine were observed in mandarine cornet and cat-fist than in Korean perch, But phosphatidyl ethanolamine(42.3%) and phosphatidyl choline (49.9%) were the main phospholipid in Korean perch. The extraordinary high content of phos-phatidyl ethanolamine compared to other fishes was characteristics in phospholipid composition of Korean perch. The major fatty acids in phospholipid of each sample were {{{{ {C }_{16 { }:_{ }0 } }}}} (38.3-46.5%) {{{{ {C}_{18 { }:_{ }0 } }}}}(14.2-21.7) and C16:1(11.6-13.8%) and additionally it chiefly consisted of C18:2, C18:0 and C17:0 The major fatty acids in glycolipid of each sample were C16:0(28.8-40.1%) C18:1(5.4-29.9%) C18:0(5.1-28.9%) and C16:1(8.2-20.1%) and additio-nally it chiefly consisted of C14:0 and C20:1.

Effect of Individual Fatty Acids on Synthesis and Secretion of Apolipoprotein and Lipoprotein in hep-G2 Cells

  • The effects of individual fatty acids, differing in their degree of unsaturation(18:0, 18:1, 18:2 and 18:3) on the biosynthesis and secretion and lipids were investigated in Hep-G2 cells. Synthesis of apolipoprotein was measured by the incorporation of 3H-leucine into apolipoprotein(d<1.21g/ml) and synthesis of lipids was measured by the incorporation of 3H-glycerol and 14C-acetate into various lipid classes. Inclusion of 1.0mM of each fatty acids into the culture medium significantly increased the synthesis of total apolipoprotein and Apo B(p<0.05). However, addition of fatty acid did not affect the synthesis of cellular and medium protein. Among different fatty acids tested, oleic acid had the greatest effect on Apo B synthesis. While stearic, linoleic and linolenic acid, all had similar effects. The secretion of triglyceride into the medium markedly increased in all fatty acid groups being 5-6 times over the albumin control. The triglyceride secretion was the highest int he oleic acid group. The secretion of phospholipid and cholesterol also increased with triglyceride output. A positive relationship existed between the output of lipoprotein-triglyceride and Apo B. Since the synthesis of Apo B was significantly increased when various fatty acids were included into the culture medium, part of the apparently stimulated synthesis of the apolipoprotein may be in response to the increased formation and secretion of lipoprotein lipids.

  • 뱀장어 근육유의 지방질 및 sterol 조성을 GLC로 분석하였고 특히 sterols조성을 IR, NMR, GLC-MS로 동정하여 다음과 같은 결과를 얻었다. 1) 뱀장어 근육유의 지방산 조성은 $C_{18:1}(40.98\%),\;C_{16:0}(23.71\%),\;C_{16:1}(13.37\%)$산이 가장 높은 비율로 함유하며, 다음은 $C_{14:0}(7.19\%),\; C_{18:0}(5.55\%),\;C_{20:1}(1.83\%),\;C_{18:2}(1.42\%)$산 이였고, $C_{18:3}(0.92\%),\;C_{15:0}(0.73\%),\;C_{14:1}(0.50\%)$산은 그 함량이 가장 적었다. 2) sterols 조성은 cholesterol$(96.58\%)$이 대부분이 였고, 24-methylene cholesterol$(3.43\%)$이 있음을 알았다.

대두유의 탈취과정에서 생성되는 trans 지방산의 정량 (Effect of Deodorizing Conditions on Formation of trans-Fatty Acids of Soybean Oil)

  • Trans 지방산의 생성은 탈취온도 $240^{\circ}C$에서 2시간 탈취시부터, $250^{\circ}C$에서 1시간 탈취시부터 생성되었으며, 검출된 함량은 각각 0.70%, 2.15%이었다 한편 이들 trans 지방산을 GC-MS 의하여 동진한 결과 $C_{18:2}\;{\Delta}9-cis,\;{\Delta}12-trans,\;C_{18:2}\;{\Delta}9-trans,\;{\Delta}12-cis,\;C_{18:2}\;{\Delta}9-cis,\;{\Delta}12-cis,\;{\Delta}15-trans,{\;},C_{18:3}\;{\Delta}9-trans,\;{\Delta}12-cis,\;{\Delta}15-cis,\;C_{18:3}\;{\Delta}9-cis,\;{\Delta}12-trans,\;{\Delta}15-cis$로 추정하였다. Trans 지방산의 생성 억제 여부를 조사하기 위하여 첨가된 citric acid및 catechin의 효과를 관찰한 결과 trans 지방산의 생성 억제에는 효과가 없었다. 탈취된 대두유의 품질을 평가하기 위하여 산값, 과산화물값 등을 측정한 결과 $240^{\circ}C$에서 2시간 이하 또는 $250^{\circ}C$에서 1시간 이하의 탈취조건에서 가장 좋은 대두유의 성상을 나타냈다. 이상의 모든 결과를 종합하면 $240^{\circ}C$에서 2시간 또는 $250^{\circ}C$에서 1시간의 탈취조건에서 trans 지방산의 생성을 억제할 수 있으며 이 조건에서 탈취된 대두유의 품질은 양호한 것으로 판단되었다.

Effects of C18 Fatty Acids on Intracellular $Ca^{2+}$ Mobilization and Histamine Release in RBL-2H3 Cells

  • To investigate the underlying mechanisms of C18 fatty acids (stearic acid, oleic acid, linoleic acid and ${\alpha}$-linolenic acid) on mast cells, we measured the effect of C18 fatty acids on intracellular $Ca^{2+}$ mobilization and histamine release in RBL-2H3 mast cells. Stearic acid rapidly increased initial peak of intracellular $Ca^{2+}$ mobilization, whereas linoleic acid and ${\alpha}$-linolenic acid gradually increased this mobilization. In the absence of extracellular $Ca^{2+}$, stearic acid ($100{\mu}M$) did not cause any increase of intracellular $Ca^{2+}$ mobilization. Both linoleic acid and ${\alpha}$-linolenic acid increased intracellular $Ca^{2+}$ mobilization, but the increase was smaller than that in the presence of extracellular $Ca^{2+}$. These results suggest that C18 fatty acid-induced intracellular $Ca^{2+}$ mobilization is mainly dependent on extracellular $Ca^{2+}$ influx. Verapamil dose-dependently inhibited stearic acid-induced intracellular $Ca^{2+}$ mobilization, but did not affect both linoleic acid- and ${\alpha}$-linolenic acid-induced intracellular $Ca^{2+}$ mobilization. These data suggest that the underlying mechanism of stearic acid, linoleic acid and ${\alpha}$-linolenic acid on intracellular $Ca^{2+}$ mobilization may differ. Linoleic acid and ${\alpha}$-linolenic acid significantly increased histamine release. Linoleic acid (C18:2: ${\omega}$-6)-induced intracellular $Ca^{2+}$ mobilization and histamine release were more prominent than ${\alpha}$-linolenic acid (C18:3: ${\omega}$-3). These data support the view that the intake of more ${\alpha}$-linolenic acid than linoleic acid is useful in preventing inflammation.

수산물의 지질에 관한 연구 (제2보) -해조류 지질의 지방산조성에 대하여- (STUDIES ON THE LIPID OF AQUATIC PRODUCTS (PART 2))

  • 8종의 식용해조류 지질의 지방산을 GLC로 분석하고 해조류 종속간에 어떤 특징이 있나, 검토하여 다음과 같은 결과를 얻었다. 1) 해조류의 지질함량은 녹조류 $1.51\%$, 갈변류 $2.81\%$, 강조류 $1.02\%$로서 갈조류의 지질함량이 가장 높았다. 2) 지방산조성의 해조류 종속간의 함량 차이을 보면, 녹조류는 $C_{16:0}$산의 함량이 가장 높은 치를 나타내며, 동시에 $C_{18:1},\;C_{18:2},\;C_{18:3}$산의 함량도 높았다. 갈조류는 $C_{16:0}$$C_{18:1},\;C_{18:2},\;C_{18:3}$산의 함량이 비교적 높은 반면, $C_{18:0}$산은 다른 해조류에 비해 아주 낮은 함량치를 나타내고 있다. 홍조류는 $C_{16:0}$산의 함량이 높은 반면, $C_{14:0},\;C_{18:1},\;C_{18:2}$산의 함량이 매우 낮았다. 3) 조성지방산의 탄소수 $C_{14},\;C_{15},\;C_{16:},\;C_{18},\;C_{22}$의 분포를 보면, 녹조류는 $C_{16},\;C_{18}$산을, 녹조류는 $C_{16},\;C_{18},\;C_{22}$산을 각각 주요성분으로 하고 있다.
