Assembly and Test of the In-cryostat Helium Line for KSTAR (KSTAR 저온용기 내부의 헬륨라인 설치 및 검사)

  • In-cryostat helium lines are under installation to transfer a cryogenic helium into cold components in KSTAR device. In KSTAR, three kinds of helium should be supplied into the cold components, which are supercritical helium Into superconduction(SC) magnet system, liquid helium into current lead system, and gas helium into thermal shields. Cryogenic helium lines consist of transfer lines outside the cryostat, in-cryostat helium lines, and electrical breaks. In-cryostat helium lines should be guaranteed of leak tightness for tong time operation at high internal helium pressure of 20 bar. We wrapped the helium line with multi-layer insulator(MLI) to reduce radiation heat and insulated the surface of the high potential part with prepreg tape. The electrical break was fabricated by brazing ceramic tube with stainless steel tube. To ensure the operation reliability at operation temperature, all the electrical break have been examined by the thermal cycle test at liquid nitrogen and by the hydraulic test at 30 bar. And additional surface insulation was prepared with prepreg tape to give structural safety. At present most of the in-cryostat helium lines have been installed and the final inspection test is progressing.

Anti-metastatic Activity of Water Extract of Samguikoeuitang via Enhancement of Natural Killer Cells (삼귀고의탕 물추출물의 자연살해세포 활성을 통한 항전이효과)

  • The anti-metastatic activity of water extract of Samguikoeuitang (WSGKE) consisting of Ginseng Radix, Angelicae Radix, Sophora flavescens and Coicis Semenwas examined. Ethanol extract of Samguikoeuitang (ESGKE) showed significant cytotoxicity against colon 26-M3.1 carcinoma cells, while WSGEK did not. However, WSGKE significantly increased the production of IL-6 and IL-12 in thioglycollate-induced macrophages from Balb/c mice, whereas ESGKE did not. WSGKE significantly increased natural killer (NK) cell cytotoxicity against effecter YAC-1 cells in an Effecter cells/Target ratio dependent manner. Also, WSGKE significantly suppressed lung metastasis after i.v. injection of colon26-M3.1 carcinoma cells. Inhibitory effect of WSGKE on lung metastasis totally abolished in NK cells-deficient mice by treatment with anti-asialo GM1 serum. In addition, the combination treatment of cisplatin and WSGKE (100 ${\mu}g$/mouse) prolonged the lifespan of mice inoculated by colon26-M3.1 cell. These findings suggest that WSGKE can inhibit lung metastasis and prolong life span via immunological enhancement as a Biological Response Modifier.

Transfer of Porcine Embryos Injected with Sperm Carrying with Exogenous DNA

  • The main goal of this study was to produce transgenic piglets by the method of injection of sperm-mediated exogenous DNA. Spermatozoa (1$\times$106 sperm of final concentration) obtained from caudal epididymis were mixed with pBC1-hEPO (20 ng/${mu}ell$) or pcDNA3 LAC Z (20 ng/${mu}ell$), and followed by electroporation (500 V, 25 ㎌). Matured oocytes having the first polar body and dense cytoplasm were selected and centrifuged at 12,000g for 6 min. After sperm injection, the oocytes were activated electrically (1.7 ㎸/cm, 30 $\mu$ sec, single pulse) in 0.3 M mannitol solution. Eggs injected sperm were cultured in NCSU 23 medium (0.4% BSA) at 39$^{\circ}C$, 5% $CO_2$ in air for 192 h. This study were comprised 3 experiments. Experiment 1 compared the developmental efficiencies between the sperm-injected oocytes (Group 1) and further activated electrically (Group 2). Experiment 2 compared the expression of pcDNA3 LAC Z in the embryos produced by Group 1 and Group 2. Finally, experiment 3 carried out transfer of embryos (1-8 cell stage) transfected with pBC1 -hEPO into surrogate recipients synchronized by injection of combination of PG600 with hCG. The rates of cleavage and development into blastocyst stage in Group 2 were significantly higher than those of Group 1 (71.3% and 28.1% vs. 43.3% and 10.3%, respectively, p<0.05). Thirty (24.2%) out of 124 embryos analyzed in Group 2 were positive by X-gal. Similarly, in Group 1, 16.3% (8/49) were positive. After transfer of 789 embryos to 7 recipient gilts, three out of them examined by ultrasound became pregnant. One recipient is in day 50 pregnancy. On day 54 of gestation, two were carried out uterotomy in order to confirm the pregnancy One had 7 and another had 2 fetuses. We conclude that injection of sperm-mediated gene transfer will be used as a valuable tool for the production of transgenic piglets.

Quality Characteristics of Wheat Nuruks by Storage Conditions of Liquid Starters Using Rhizopus oryzae N174 (Rhizopus oryzae N174를 이용한 액체종국 저장조건에 따른 밀누룩의 품질특성)

  • Production of liquid starters using wheat bran as a medium for Rhizopus oryzae N174 and the changes in their characteristics noted during storage were investigated in this study. The optimal culture conditions of the liquid starters were determined to be 5~15% (w/v) wheat bran and 48~72 hrs of incubation. The effects of liquid starters with different storage periods and temperatures (-18, 4, 10 and $25^{\circ}C$) on the quality of wheat nuruk were evaluated. According to the results of the pH, acidity, reducing sugar and enzyme activities, it was found that liquid starter using wheat bran preserved for one day, at any temperature, is the best method of storage for seed cultures for R. oryzae N174.

Utilization of Dietary Nutrients, Retention and Plasma Level of Certain Minerals in Crossbred Dairy Cows as Influenced by Source of Mineral Supplementation

  • Nutrient utilization and plasma level of minerals were studied in lactating crossbred cows fed diet supplemented from different source of minerals. Twelve crossbred cows of first to third lactation during their mid stage of lactation were distributed equally under two groups and were fed concentrate mixture, green fodder (para grass) and finger millet straw as per requirement. Cows in group I were fed concentrate mixture supplemented with 1% mineral mixture whereas cows in group II were not supplemented with mineral mixture in the concentrate mixture but were offered additional quantity of green fodder (1 kg DM/cow/day) to compensate for the requirement of minerals. Balance study conducted towards the end of 120 days of feeding experiment indicated that the total dry matter intake in both the groups did not differ significantly. The cows in group II offered additional quantity of green fodder consumed significantly (p<0.05) more of green fodder (5.11 vs. 4.51 kg /animal/day) and the cows in group I consumed significantly (p<0.01) more of finger millet straw (1.71 vs. 0.92 kg/animal/day). The digestibility of major nutrients did not differ between the groups except for ether extract which was significantly (p<0.05) lower in cows fed additional green fodder. The total daily intake of P, Cu, Fe and Co did not differ significantly in both the groups whereas significantly higher intakes of Ca, Mg, Zn and Mn were observed in cows supplemented with inorganic source of minerals. However, supplementation through both the sources could meet the mineral requirement in group I and group II. The gut absorption (%) of all the minerals was comparable between the groups except for Mg which was significantly (p<0.05) higher in cows supplemented mineral mixture. The net retention of all the minerals was significantly more in the group supplemented with inorganic source of minerals. Except for P, Mg and Co the retention as percentage of total intake was comparable for all minerals in both the groups. Irrespective of source of mineral supplementation the average monthly blood plasma mineral levels (Ca, P, Mg, Cu, Zn, Fe) were within the normal range and comparable between the groups. Lower level of Cu observed at the start of the experimental feeding in both the groups increased with the advancement of supplemental feeding. The plasma Zn and Fe content in cows of both the groups did not vary. The blood plasma level of some minerals (Ca, P, Mg and Cu) was significantly higher towards the end of experimental feeding as compared to the initial values due to the reduction in milk yield with advancement of lactation or due to supplemental effect of minerals. It could be concluded that supplementation of minerals through inorganic source is better utilized in terms of retention as compared to green fodder (para grass), which is a moderate source of most nutrients.

Using Implementation Science to Advance Cancer Prevention in India

  • Oral, cervical and breast cancers, which are either preventable and/or amenable to early detection and treatment, are the leading causes of cancer-related morbidity and mortality in India. In this paper, we describe implementation science research priorities to catalyze the prevention and control of these cancers in India. Research priorities were organized using a framework based on the implementation science literature and the World Health Organization's definition of health systems. They addressed both community-level as well as health systems-level issues. Community-level or "pull" priorities included the need to identify effective strategies to raise public awareness and understanding of cancer prevention, monitor knowledge levels, and address fear and stigma. Health systems-level or "push" and "infrastructure" priorities included dissemination of evidence-based practices, testing of point-of-care technologies for screening and diagnosis, identification of appropriate service delivery and financing models, and assessment of strategies to enhance the health workforce. Given the extent of available evidence, it is critical that cancer prevention and treatment efforts in India are accelerated. Implementation science research can generate critical insights and evidence to inform this acceleration.

Types of Weed Community in Transplanted Lowland Rice and Relationship between Yield and Weed Weight in Weed Communities (논 잡초의 군락형 종류와 군락형별 잡초발생과 수도수량과의 관계)

  • Eight different weed community types growing in association with transplanted rice were identified in the experimental fields used in these studies. The Importance Value (LV.) of weed species growing in association with rice changed with time. The I. V. of Echinochloa glabrescens Munro ex Hook f. increased from 40 days after transplanting (DAT) until heading and then, decreased from heading to maturity of rice. However, the LV. of Monochoria vaginalis (Burm.) Pres!. and Echinochloa crus-galli ssp. hispidula (Retz.) Honda increased from 40 DAT to maturity while Scirpus maritimus L. decreased from 40 DAT. There was a high negative correlation between grain yield and weed weight at rice heading in all weed communities. The competitive ability of rice against weeds varied depending upon weed species. The highest yield decrease due to weed competition was obtained from the E. crus-galli ssp. hispidula-S. Maritimus community. Forty-two percent yield reduction was caused by competition of 100g dry weight of weeds per square meter of this community type at rice heading. The yield decrease for the same amount of weeds was 10% for the E. glabrescens community, 15% for the M vaginalis community and the M vaginalis-Scirpus supinus L. community and 21% for the M vaginalis-E. glabrescens-Fimbristylis littoralis Gaud.-S. maritimus community type.

Gamma-Radiation Sensitivity of Pathogenic Bacteria in Beef (우육에 오염시킨 병원세균의 방사선 감수성)

  • The gamma-radiation sensitivity of eight kinds of pathogenic bacteria related to beef was investigated in frozen cells $(-18^{\circ}C)$ with 0.1 M phosphate buffer and inoculated cells in beef. In frozen cells, D10 values of pathogenic bacteria related to beef were $0.07{\sim}0.69$ kGy, and inactivation factors were $2.90{\sim}42.86$ at the radiation doses of $2{\sim}3$ kGy. Beef was inoculated with Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimurium, Enterobacter aerogenes, Vibrio parahaemolyticus, Staphylococcus aureus, Listeria monocytogenes, and Aeromonas hydrophila. Inoculated beef samples were packaged in air and irradiated at 0.005 to 3.0 kGy. Ninety percent of the viable pathogenic bacteria in beef was eliminated by doses of $0.1{\sim}0.61$ kGy at room temperature, and the inactivation factors were $3.28{\sim}30.0$ kGy at the radiation doses of $2{\sim}3$ kGy. Therefore, irradiation is considered to be an effective method to control pathogenic bacteria in beef.

Effect of Lysine to Digestible Energy Ratio on Growth Performance and Carcass Characteristics in Finishing Pigs

  • This experiment was performed to investigate the effects of lysine (Lys) to DE ratio on growth performance, and carcass characterics in finishing barrows. Ninety six cross-bred finishing barrows ((Landrace${\times}$Yorkshire)${\times}$Duroc, average BW $58.25{\pm}0.48$ kg) were assigned as a randomized complete block design by 2 energy levels and 4 Lys:DE ratios on the basis of BW to one of 8 treatments with 3 replications with 4 animals per pen. The levels of DE and Lys:DE ratio for each treatment were i) DE 3.35 Mcal/kg, 1.5 g Lys/Mcal DE, ii) DE 3.35 Mcal/kg, 1.8 g Lys/Mcal DE, iii) DE 3.35 Mcal/kg, 2.1 g Lys/Mcal DE, iv) DE 3.35 Mcal/kg, 2.4 g Lys/Mcal DE, v) DE 3.60 Mcal/kg, 1.5 g Lys/Mcal DE, vi) DE 3.60 Mcal/kg, 1.8 g Lys/Mcal DE, vii) DE 3.60 Mcal/kg, 2.1 g Lys/Mcal DE, viii) DE 3.60 Mcal/kg, 2.4 g Lys/Mcal DE. During finishing period from 58 kg to 103 kg of BW, increased energy density in the diet increased (p<0.05) ADG and gain:feed ratio, but did not influence ADFI. As Lys:DE ratio was increased, ADG, ADFI and gain:feed ratio were improved in finishing barrows (p<0.05). There were positive interactions (p<0.05) between carcass weight, grade, and backfat thickness and energy density and Lys level (p<0.05). In conclusion, data from our current study suggest that maximum yields including ADG, gain:feed ratio, carcass weight and grade can be achieved by administrating finishing pigs with an ideal Lys:DE ratio, Lys 2.1 g/DE Mcal.

Fabrication and Dosimetry Characteristics of Intracavitary Cones for Radiotherapy (방사선 강내치료를 위한 소조사면 전자선cone의 선량분포 특성)

  • The intracavitary cones were designed which were made of stainless steel and have scratched inside cone to be generated electron scatter and designed to be attached easily to the LINAC collimator and controlled cones length to be contacted smoothly between the patient and the cone tip. Two types of intracavitary cones were designed. One is the straight end cones with circular opening on the distal end and the other is 30 degree beveled end cones with elliptical opening on the distal end. Each type of intracavitary cone ranged in daimeter from 2.5 cm to 3.5 cm and required a separate set of lower trimmer annulias cone diameter. The film phantom was designed with an internal cassette that accurately aligned the film edge with the film phantom surface. Film optical density data were measured by photodensitometer(Wellhofer 700i) Dosimetry measurements were made to commission the LINAC for 6 - 20 MeV electron using the intracavitary cones. Isodose curves were measured for all energy and cones combinations. Output is defined as the maximum dose per MU along the clinical central axis in water at 113 cm SSD. Calibration output, defined to be the output for the 15cm$\times$15cm diameter straight cone, was adjusted to 1.00 cGy/MU at each energy according to the TG-21 protocol.
