Expression of Cytochrome P450 1A1, 1A2, 2C8, 2E1 and 3A4 in Human Brain

  • We have carried out PCR reactions to investigate if cytochrome P450 (P450) enzymes (1A1, 1A2, 2C8, 2E1 and 3A4), which are well hewn to be the key enzymes in detoxification process and/or synthesis of steroids in the liver, are expressed in the human brain, too. P450 1A1, 2C8 and 2E1 were expressed clearly. However, P450 1A2 and 3A4 were not detectable. Their expression levels in the human brain could be extremely low or they were not expressed at all. One base substitution at nucleotide 290 (A->G) was identified in P450 1A1. It is suspected to be an individual polymorphism. Our results should contribute to the better understanding of the role of cytochrome P450 enzymes in the human brain.

Isolation and Characterization of Catalase-producing Bacteria from Soil (토양으로부터 카탈라제 생산균의 분리 및 특성)

  • This study investigated the production of catalase from Bul-kyo soil bacteria through fermentation process. Isolation and selection of bacteria was performed through chemical and physiological analysis. Catalases were produced from bacteria which belong to 3 different species (Bacillaceae bacterium BKBChE-1, Bacillus sp. BKBChE-2, Bacillus flexus BKBChE-3) confirmed by using 16S rDNA sequence method. The catalases were found to be stable in the temperature range of $30^{\circ}C-60^{\circ}C$ for BKBChE-1, BKBChE-2 and BKBChE-3 and also in the pH range of 9.0-12.0 for BKBChE-1 and BKBChE-3. Long-term stability of the catalases was about 20 days at $4^{\circ}C$. However, BKBChE-2 has kept its activity over 30 days at $4^{\circ}C$.

The Effects of Temperature, pH, and Bactericides on the Growth of Erwinia pyrifoliae and Erwinia amylovora

  • In this comparative study, the effects of temperature, pH, and bactericides on the growth of Erwinia pyrifoliae and Erwinia amylovora were investigated. The maximum temperature for the growth of both Erwinia species was estimated to be $36{\circ}C$. The maximum specific growth rates of E. pyrifoliae and E. amylovora were observed at $27{\circ}C$ and $28{\circ}C$, respectively, and no significant growth differences were shown at their optimum temperatures. However, at lower temperatures ranging from 12-$21{\circ}C$, E. pyrifoliae showed higher growth rates with doubling times shorter than those of E. amylovora. Distinct growth rates at these temperatures revealed that E. pyrifoliae is more cold-tolerant than E. amylovora. The optimum pH for the growth of both pathogens was 7.5 and growth was not seen at pH ${\le}$ 5.0 and ${\ge}$ 10.0. These results showed that the effect of pH on the growth of E. pyrifoliae and E. amylovora was similar. Minimum inhibitory concentrations (MICs) of copper sulfate, oxolinic acid, streptomycin, and tetracycline, which inhibited growth of E. pyrifoliae and E. amylovora, were determined. The strains of both pathogens were able to grow at 0.08-0.32 mM copper sulfate, but not at higher concentrations. However, none of the tested strains grew in the presence of oxolinic acid (0.001 mM), streptomycin (0.1 mM), and tetracycline (0.01 mM) concentrations. These results suggested that all strains of both Erwinia species were sensitive to tested bactericides and indicated no occurrence of resistant strains of E. pyrifoliae in Korea.

Effects of a New-Nurse Education Program Utilizing E-learning and Instructor Demonstration on Insulin Injection Practices (이러닝 교육(인슐린 주사방법)을 통한 신규 간호사 교육 프로그램의 효과)

  • Purpose: The purpose of this study was to develop and evaluate a new-nurse education program utilizing both e-learning and instructor demonstration. Methods: From August to December in 2009, the e-learning education program about insulin injection was developed. The control (C) group was educated via instructor demonstration from April 15 to October 6 in 2009, and the experimental (E) group was educated via both e-learning and instructor demonstration from January 5 to October 13 in 2010. After each education, knowledge and educational effectiveness were checked. Results: Satisfaction with the education contents in the E group was significantly higher than those of the C group (Z=-3.72, p<.001), and satisfaction with the education method in the E group was higher than those of the C group (Z=-2.98, p=.003). Usefulness (Z=-3.33, p=.001), application (Z=-2.62, p=.009), and confidence (Z=-2.61, p=.009) in the E group were all higher than those of the C group. 78.9% in the E group reused the e-learning program after the experimental education. Conclusion: Combined educational program with e-learning and instructor demonstration had both merits of online efficiency and face-to-face education. It would be useful especially for new-nurses to improve their nursing skills in accomplishing their roles.

Phenotypic Stability of a Temperature-Controllable Expression Vector on Phenylalanine Production by Escherichia coli (대장균을 이용한 Phenylalanine 생산에 있어서 온도조절형 발현 Vector의 안정성)

  • The plasmid pSY130-14 for the high production of phenylalanine is a temperaturecontrollable expression vector composed of the $P_R$ and the $P_L$ promoter and a temperature sensitive repressor, $cI_{857}$ of bacteriophage lambda. Strain AT2471 harbouring plasmid pSY13O- 14 is induced the phenylalanine production by shifting up the incubation temperaure to $38.5^{\circ}C$. Plasmid stability of E. coli AT2471 harbouring pSY130-14 was very low, it was about 30% after 48 h cultivation at $38.5^{\circ}C$ without kanamycin. The plasmid disappeared immediately at $40^{\circ}C$ without kanamycin, and at $40^{\circ}C$ adding kanamycin, the plasmid stability decreased at the beginning, but rose with the extension of the culture time. For the improvement of plasmid stability, the plasmid obtaind was designated as pSY15O-1 by changing origin region (ori) pACYC 177 of pSY130-14 for ori pSC101. E. coli AT2471 harbouring pSY150-1 was stable at $38.5^{\circ}C$ without tetracycline, and the plasrnid stability was about 40% after 48 h cultivation at $40^{\circ}C$.

A Study for Regulation of Ethanol-inducible $P_{450}$(CYP2E1) on $CCI_4$-induced Hepatic Damage

  • Previous study showed that $CCl_4$ administration evoked a rapid decrease in cytochrome $P_{450}$ 2E1 protein soon after the exposure due to posttranslational inhibition(Biochem. Biophys. Res. Commun. 179:449-454, 1991). In this report, aniline hydroxylase and the amounts of immunoreactive $P_{450}$ 2E1 were rapidly decreased during day 1 to 2 and recovered during day 3 to 4 after a single dose of $CCl_4$. The activity of pentoxyresorufin-O-dealkylase was also suppressed at day 1 and began to repair from day 2. However, the decrease in immunoreactive $P_{450}$ 2C content was not observed. The decreases in $P_{450}$ 2E1 enzyme activity and immunoreactive protein by acute $CCl_4$ treatment were accompanied by a decline in $P_{450}$ 2E1 mRNA level. The data thus suggested a pretranslational reduction of $P_{450}$ 2E1 during day 1 to 2 after acute $CCl_4$ treatment.

Construction of a Corynebacteriurn glutarnicum-Escherichicr coli Shuttle Vector and Cloning the Homoserine ehydrogenase Gene from C. glutamicum (Corynebacterium glutamicum-Escherichia coli Shuttle Vector 개발과 C.glutamicum 의 Homoserine Dehydrogenase Gene Cloning)

  • A 7.5 kilobases hybrid plasmid, designated as pCE1301, was constructed by combining Eschurichia cwli plasmid pBELl which carries the kanamycin resistance gene of Tn5 with a cryptic plasmid, pSRl of Corynebacterium glutamicum. pCE1301 was transformed C. glutaicum by PEG-mediated protoplast method and its transformation efficiency was about $3.0\times 10^3$ transformants per $\mu g$ of the hybrid plasmid DNA. The physical map reveals that pCE1301 has single restriction sites for SalI and EcoRl, respectively. 'The kanamycin resistance of pCE1301 was stably maintained in C. glutamicum up to 25 generations and any segregation was not detected. pCI31301 was also introduced into Brevibacterium flavum and E coil, and replicated in those strains. pCE1301 was proved to be useiul in cloning the homoscrine dehydrogenase gene from C. glutamicum.

Effects of Heat Treatment History of Skim Milk Powder on the Quality Characteristics of RTD Coffee with High Protein (탈지분유의 열처리 공정이 고단백질 RTD 커피의 품질 특성에 미치는 영향)

  • This study investigated the quality of characteristics of high-protein RTD coffee using domestic and imported skim milk powder with different heat treatment. Skim milk powders (A, B) had high-heat treatment, C had medium-heat, and D and E had low-heat treatment. The transmittance of A and B were higher than that of C, and that of C were higher than that of D and E (p<0.05). The precipitate attached on bottom of container of RTD coffee using A and B were 2.993~3.053% and higher than those (0.753~0.803%) of RTD coffee using C, D and E (p<0.05), but there was no difference between those of RTD coffee using C, D and E (p<0.05). The centrifuged precipitate of RTD coffee using A and B were 1.987~2.040% and higher than those (0.820~0.830%) of RTD coffee using C, D and E (p<0.05), but there was no difference between those of RTD coffee using C, D and E (p<0.05). The proximate composition of precipitate attached on bottom of container of RTD coffee using A, which showed the highest amount of precipitate, showed 65.7% of carbohydrate, 19.0% of protein, 4.8% of fat and 4.8% of ash in dry basis, that of RTD coffee being 72.7%, 15.1%, 7.9% and 4.3% in dry basis respectively. Protein and fat content of precipitate were lower and protein and ash content were higher than those of RTD coffee. But seeing that the most increased portion was protein, precipitation of RTD coffee appears to be attributed to heat-denatured proteins.

Synergistic Effects of Dietary Vitamins C and E on Methylmercury-Induced Toxicity in Juvenile Olive Flounder Paralichthys olivaceus

  • This experiment was conducted to evaluate the synergistic effects of vitamin C and E on methylmercury (MeHg) toxicity in juvenile olive flounder Paralichthys olivaceus. In a $3{\times}3$ factorial design, 9 experimental diets containing three different vitamin C (0, 200 or 400 mg/kg diet in the form of l-ascorbyl-2-monophosphate) and vitamin E (0, 100 or 200 mg/kg diet in the form of dl-${\alpha}$-tocopheryl acetate) levels with the Hg toxicity level (20 mg/kg diet in the form of MeHg) were formulated. Triplicate groups of fish averaging $2.3{\pm}0.05g(mean{\pm}SD)$ were fed one of the 9 diets in a flow through system for 8 weeks. Fish fed 400 mg vitamin C/kg diet with 100 or 200 mg vitamin E/kg diet showed significantly (P < 0.05) higher weight gain (WG) than did fish fed the other diets. Fish fed 400 mg vitamin C/kg diet at all vitamin E levels and those which fed vitamin C and E equally at a rate of 200 mg/kg diet showed significantly (P < 0.05) higher feed efficiency (FE), specific growth rate (SGR) and protein efficiency ratio (PER) than did fish fed the other diets. Fish fed 200 and 400 mg vitamin C/kg diet exhibited significantly (P < 0.05) lower Hg concentration in their muscle as well as kidney than did fish fed the other diets. Therefore, these results clearly indicated that the synergistic effects of these two vitamins on MeHg toxicity by supplementing dietary vitamin C (200 and 400 mg/kg diet) with vitamin E (100 and 200 mg/kg diet) in juvenile olive flounder.

  • Let E be a uniformly convex Banach space with a uniformly $G{\hat{a}}teaux$ differentiable norm, C a nonempty closed convex subset of E, and $T:C{\rightarrow}{\mathcal{K}}(E)$ a multivalued nonself-mapping such that $P_T$ is nonexpansive, where $P_T(x)=\{u_x{\in}Tx:{\parallel}x-u_x{\parallel}=d(x,Tx)\}$. For $f:C{\rightarrow}C$ a contraction and $t{\in}(0,1)$, let $x_t$ be a fixed point of a contraction $S_t:C{\rightarrow}{\mathcal{K}}(E)$, defined by $S_tx:=tP_T(x)+(1-t)f(x)$, $x{\in}C$. It is proved that if C is a nonexpansive retract of E and $\{x_t\}$ is bounded, then the strong ${\lim}_{t{\rightarrow}1}x_t$ exists and belongs to the fixed point set of T. Moreover, we study the strong convergence of $\{x_t\}$ with the weak inwardness condition on T in a reflexive Banach space with a uniformly $G{\hat{a}}teaux$ differentiable norm. Our results provide a partial answer to Jung's question.
