Fungi Associated with Soybean Seed, their Pathogenicity and Seed Treatment (콩 종자(種子)에서 분리(分離)한 사상균(絲狀菌), 그 병원성(病原性) 및 종자(種子) 소독(消毒)에 관한 연구(硏究))

  • Alternaria tenuis, Arthrobotrytis sp., Aspergillus spp., Cephalosporium sp., Cladosporium sp., Cylindrocarpon sp., Fusarium equiseti, F. moniliforme, F. semitectum, F. solani, Penicillium spp., Rhizopus sp. were saprophytic fungi and Cercospora kikuchii, Colletotrichum truncatum, Diaporthe phaseolorum var. sojae and Fusarium oxysporum were pathogenic fungi detected from 14 seed samples of soybean. Initial symptoms caused by C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum on seedlings from naturally infected seed by the test tube agar method have been described and discussed. Soybean seeds infected with C. truncatum, D. phaseolorum sojae and F. oxysporum reduced germination of seeds and have influenced on the growth of soybean seedling caused by C. kikuchii. Surface-sterilized soybean seedlings became diseased in the test tube agar artificially inoculated with C. kikuchii, C. truncatum and D. phaseolorum sojae isolated from naturally infected soybean seeds. F. oxysporum showed very weak pathogenicity. Seed disinfectants of Benlate-T, Homai, Tecto and Sisthane have effective to C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum. Arasan, Captan, Busan-30 and Mercron were inferior to C. kikuchii but effective against others. Seed disinfectants treated in this experiment have increased seed germination campared with non-treatment.

Identification and Etiological characteristics of Anthracnose Fungi Isolated from Soybean, Small Red Bean and Green Bean (콩, 팥 및 녹두에서 분리한 탄저병균류의 동정과 병원학적 특징)

  • Anthracnose fungi associated with soybean (Glycine max), small red bean (Vigna angularis), and green bean (Phaseolus radiatus var. aurea) were compared with respect to cultural characteristics, morphological aspects, and pathogenicity. From the legumes, four different species of Colletotrichum were identified based on their morphological characters : C. truncatum, C. destructivum, C. trifolii and C. gloeosporioides. Colonies of Colletotrichum spp.on culture media varied in colour and growth habits depending upon the media and isolates. In general, formation of conidia, setae, and perithecia on water agar leaf medium was better than that on potato dextrose agar medium under near ultraviolet light. In cross inoculation tests, isolates of C. truncatum, c. destructivum, C. trifolii and C. gloeosporioides could infect soybean, small red bean, and green bean at different growth stages. There were some differences in pathogenicity or susceptibility depending on crops tested. C. truncatum was the most virulent and major pathogenic species among the four species.

  • Pseudomonas sp. antagonistic to Colletotrichum truncatum and C. gloesporioies was selected as a biological control agent for soybean anthracnose. Pseudomonas sp. inhibited the mycelial growth of pathogens effectively as the funhicides such as benomyl and fluazinam in vitro tests without any adverse effects on soybean. Seed treatment with Pseudomonas sp. increased emergence rate of soybean seeds significantly after inoculaton with C. truncatum. When the suspension of Pseudomonas sp. was sprayed on soybean plants, the control efficacy was not different from that of fungicides, benomyl and fluazinam two weeks after treatment, however the efficacy did not last long enough after three weeks.

Influence of Temperature, Wetness Duration and Fungicides on Fungal Growth and Disease Progress of Soybean Anthracnose Caused by Colletotrichum spp. (콩 탄저병균의 생장 및 병 진전에 미치는 온도, 수분 존재시간, 살균제의 영향)

  • The effects of temperature, duration of wetness period, and fungicides on the spore germination, appressorium formation, acervulus formation and lesion development by Colletotrichum spp., cause of soybean anthracnose, and their pathogenicity were assessed in controlled environment. C. gloeosporioides was highly pathogenic on inoculated soybean seeds as high as C. truncatum, whereas remarkably low on the soybean leaves. Spore germination, appressorium formation and mycelial growth of C. gloeosporioides were best at $25^{\circ}C$, but C. truncatum was best at $30^{\circ}C$. It has also done at $15^{\circ}C$, even though it was much retarded. C. truncatum showed high sensitivity to the fungicides, fluazinam and benomyl, meanshile C. gloeosporioides showed to fluazinam and triflumizole. At least 8 hrs. of wetness period was requird for the pathogen to develop lesions at $30^{\circ}C$. When the wetness period was 32 hrs. lesion size of was larger at $25^{\circ}C$ than $30^{\circ}C$, however it was traceable at $20^{\circ}C$. Different sensitivity of Colletotrichum spp. to fungicides suggests that proper fungicide is required to effective control of soybean anthracnose ingected multiply with Colletotrichum spp.

Genetic Diversity and Differentiation of Colletotrichum spp. Isolates Associated with Leguminosae Using Multigene Loci, RAPD and ISSR

  • Genetic diversity and differentiation of 50 Colletotrichum spp. isolates from legume crops studied through multigene loci, RAPD and ISSR analysis. DNA sequence comparisons by six genes (ITS, ACT, Tub2, CHS-1, GAPDH, and HIS3) verified species identity of C. truncatum, C. dematium and C. gloeosporiodes and identity C. capsici as a synonym of C. truncatum. Based on the matrix distance analysis of multigene sequences, the Colletotrichum species showed diverse degrees of intera and interspecific divergence (0.0 to 1.4%) and (15.5-19.9), respectively. A multilocus molecular phylogenetic analysis clustered Colletotrichum spp. isolates into 3 well-defined clades, representing three distinct species; C. truncatum, C. dematium and C. gloeosporioides. The ISSR and RAPD and cluster analysis exhibited a high degree of variability among different isolates and permitted the grouping of isolates of Colletotrichum spp. into three distinct clusters. Distinct populations of Colletotrichum spp. isolates were genetically in accordance with host specificity and inconsistent with geographical origins. The large population of C. truncatum showed greater amounts of genetic diversity than smaller populations of C. dematium and C. gloeosporioides species. Results of ISSR and RAPD markers were congruent, but the effective maker ratio and the number of private alleles were greater in ISSR markers.

Identification and Pathogenicity of Colletotrichum Species Causing Anthracnose of Korean Radish (Rhaphanus sativus) in Korea (무 탄저병을 일으키는 Colletotrichum속의 종 동정 및 병원성)

  • In August 2017, anthracnose symptoms were observed on the petioles and leaf veins of Korean radish (Rhaphanus sativus) in Hongcheon, Jeongseon, and Pyeongchang of the Gangwon province, Korea. Many grayish to dark-brown spots of 1-2 mm in diameter, appeared on the lower surface and leaf veins of the radish leaves. The spots gradually enlarged and coalesced to form dark-brown irregular lesions. Ten Colletotrichum isolates were obtained from the affected tissues of the Korean radish. Out of them, eight isolates were identified as C. higginsianum and two isolates were identified as C. truncatum based on morphological characteristics and multi-locus molecular phylogenetic analysis using the internal transcribed spacers and intervening 5.8S rDNA (ITS), partial beta-tubulin gene (TUB2), partial actin gene (ACT), and partial chitin synthase-1 gene (CHS1). The pathogenicity test was carried out on wounded and unwounded Korean radish (cv. Siraegimu and Osarimu), and Chinese cabbage (cv. Chuno and Smart) by inoculating with a spore suspension. All isolates except one C. truncatum isolate developed typical symptoms on both wounded and unwounded Korean radish. In Chinese cabbage, only the plants inoculated with C. higginsianum isolates developed symptoms regardless of the wound. This is the first report of anthracnose caused by C. truncatum on Korean radish in Korea.

Antifungal Properties of Streptomyces bacillaris S8 for Biological Control Applications

  • Soybean (Glycine max), a crucial global crop, experiences yearly yield reduction due to diseases such as anthracnose (Colletotrichum truncatum) and root rot (Fusarium spp.). The use of fungicides, which have traditionally been employed to control these phytopathogens, is now facing challenges due to the emergence of fungicide-resistant strains. Streptomyces bacillaris S8 strain S8 is previously known to produce valinomycin t through a nonribosomal peptide synthetase (NRPS) pathway. The objective of this study was to evaluate the antifungal activity of S. bacillaris S8 against C. truncatum and Fusarium sp., assessing its efficacy against soybean pathogens. The results indicate that strain S8 effectively controlled both above-ground and underground soybean diseases, using the NRPS and NRPS-related compound, suggesting its potential as a biological control in plant-microbe interactions. These findings underscore the pivotal role of the stain S8 in fostering healthy soybean microbial communities and emphasize the significance of microbiota structure studies in unveiling potent biocontrol agents.

Effect of light on conidial formation in several anthracnose fungi (몇가지 탄저병균의 분생포자형성에 미치는 광선의 영향)

  • Effect of light on conidial formation of Colletotrichum panacicola, C. gloeosporioides, C. runcatum and C. nicotianae was examined on potato dextrose agar on which each conidial suspension was seeded. The light source was Mitsubishi day light fluorescent lamps wi th an intensity of 100-120 ft-c at the colony surface. The treatments were (i)constant light, (ii) constant darkness, and (iii) alternating light (11hr.) and darkness {13hr.) at $26\{\circ}C\pm2$. With the exception of C. nicotianae, constant light gave the greatest sporulation over that obtained in the other two treatments; there were on significant differences in number of sporulation between under constant darkness and alternating light and darkness. C. nicotianae resulted in maximum. inter mediate and minimum sporulation under alternating light and darkness. constant darkness and constant light, respectively. These differences were statistically significant at $5\%$ level. C. truncatum and C. nicotianae produced abundant conidia in the entire test; C. gloeosporioides was intermediate, and C. panacicola was the least. Differences in number of sporulation due to light. organisms. and t he interaction of these two were all statistically significant.

Efficacy of Fluazinam and Iprodione+Propineb in the Suppression of Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii, the Causal Agents of Seed Decay in Soybean

  • Seed decay of soybean caused by Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii is a serious disease when soybean is harvested under warm and wet weather conditions. Benomyl has been used for controlling the disease, however, benomyl application may be limited due to common occurrence of resistance. The efficacy of 21 fungicides against the pathogens was evaluated in vitro. Among the fungicides tested, benomyl, carbendazim, fluazinam, iprodione+propineb, thiophanate-methyl, and triflumizole were found effective and were evaluated for their ability to control the seed pathogens. Fluazinam completely inhibited mycelial growth at a concentration of 100 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for D. phaseolorum; and at a concentration of 500 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for C. truncatum and C. kikuchii. $EC_90$ values of fluazinam were similar to that of benomyl. Because fluazinam, iprodione+propineb, and triflumizole were found effective against the seed pathogens, these were subjected for field-testing. Suppression of pod and seed infection by fluazinam and iprodione+propineb was as high as that of benomyl without any reduction in agronomic characters of soybean. This study shows that fluazinam and iprodione+propineb may be used in combination with benomyl to control seed pathogens, manage resistance, and ensure production of high quality soybean seeds.

Effect of Environmental Factors on Appressorium Formation of Colletotrichum truncatum (환경조건(環境條件)이 대두(大豆) 탄저병균(炭疽病菌)(Colletotrichum truncatum)의 부착기형성(附着器形成)에 미치는 영향(影響))

  • The results investigated of the effect of light, pH, and nutrients in vitro and the levels of fertilizer(P,K and Ca) in vivo in order to find out the effect of environmental factors in forming an appressorium of Colletotrichum truncatum were as follows; The ratio of appressorium formation promoted more in a light treatment than in a dark treatment according to investigation after three and six hours passed away. But after the period, the ratio of appressorium formation promoted highly in both treatments. Reasonable pH of appressorium formation was pH 6-pH 8. The ratio of appressorium formation was the most favorable at none nutrient level in every treatment of C-source and N-source, and suppressed in a high concentration of C-source and N-source. But according as the time passed away, the ratio of appressorium formation promoted in C-source and N-source. When their quantities were increased in P and K treatments, the ratio of appressorium formation was somewhat suppressed but it was highly suppressed in the gradual order of none fertilizer level < standard fertilizer level < doubled fertilizer level in Ca treatment according to investigation after the first three hours passed away. But after the period, the ratio of appressorium formation promoted highly regardless of the level of fertilizer.
