Determination of Dietary Fiber Contents in Dodok ( Codonopsis lanceolata traut (Beneth et Gook)) and Ginseng (Panax ginseng C. A. Meyer) (더덕과 인삼의 식이섬유소 함량의 측정)

  • The total dietary fiber (TDF) contents in cultivated dodok, wild dodok and ginseng were determined by AOAC method and Mongeau et al's method. Also, the nonstarch polysaccharide (NSP) contents of them were analyzed by Englyst et al's gas chromatographic (GC) method. The TDF values by AOAC method and Mongeau et al's method were 34.50% and 35.92% for wild dodok, 46.40% and 47.55% for cultivated dodok, and 14.93% and 14.03% for ginseng, respectively. The ratios of soluble dietary fiber (SDF) contents to TDF contents by Mongesu et al's method were 64.17% for wild dodok, 75.77% for cultivated dodok, and 53.74% for ginseng, respectively. The major sugar constituents of NSF in wild dodok, cultivated dodok and ginseng by GC were glucose, galactose and uronic acid. The Enalyst's TDF contents, i.e., NSP plus lignin contents in wild dodok, cultivated dodok, and ginseng were 20.65%, 20.03% , and 9.72%, respectively.

  • This experiment was Conducted ta screening for the anti-cancer efficacy from the wild plants which are naturally growing in the Korea. The results are as follows. The results were shown thatZea may L. nad significantly effects on mediculal efficacy anganist anti-tumor by usulg the totalpacked cell volume methods and also, severals plants, such as Sofonum nigrum, Patrinia hispidoBunge, Eragrostis, ferrugenia Beauv, Salaginela pouzolgiana Spring, Platrycarya strobilacea Bunge,Codonopsis lanceolata Benth. et Hook fil. which are collected from Giri and Mooju mountain in Koreaand Nagano in Japan were showed effects on auti-tumor. But the pharmaceologial activities ofPharbitis nil Choisy was believed to strong effec on anti - cancer tumors, while toxicity of its wasshown high that induced te kill all used mice. Extraction of Patrinia hispida Bunge, Pharbitis nilChoisy, Toilis japonica DC, Eragrostis erruginea Beauv. and Forsythia koreana Nakai showed effec-tively supressed on growth rate of cancer tumor by the below 50 percent of T/C ratio at 30mg /mlof extraction from plant. That Is strong activity while Reynouxria japonica Houtt. was observed onlymild activities. The above results many possibly suggest that Patrinia hispido Bunge and Eragrostisferrugina Beauv. inhibited the growth of cancer tumor by the both total packed cell volume methodand cytotoxicity method. Although basic research is still going on, we will find out an accurate moth-od for developing useful medicinal plant to improve pharmacological activites against anti-cancertumor, especialy, in Eragrostis ferruginea Beauv.

Development of Transgenic Plant (Codonopsis lanceolata Trautv.) Harboring a Bialaphos Resistance Gene, bar (Bialaphos 저항성 유전자 bar를 이용한 형질전환 더덕개발)

  • Codonopsis lanceolata ("Deoduck" in Korea) is a perennial herb, and belongs to family, Campanulaceae. Its taproot is used a good source of a wild vegetable as well as an herbaceous medicine. In this study, to develop a bialaphos-resistant transgenic Codonopsis, seed germination mechanism and somatic embryogenesis of the plant were investigated, and Agrobacterium-mediated transformation with bar gene encoding phosphinothricin acetyltransferase (PAT) was performed. Attempt were made to regenerate plant from cells via somatic embryogenesis. When the cotyledons, nodes and leaf disks were cultured on MS medium containing 2,4-D and zeatin, embryogenic calli were induced. Upon transferring the somatic embryos to N6 solid medium without plant growth regulators, they developed into plantlets under continuous illumination. All plants were dead on MS basal medium containing 10 mg/L phosphinothricin (PPT) and Basta, respectively. The explants did not produce calli in the medium containing 200 mg/L kanamycin. The explants were cocultured with Agrobacterium tumefaciens for 2 days, and transformants were selected in MS basal medium containing 1.0 mg/L 2,4-D, 100 mg/L kanamycin and 500 mg/L carbenicillin. After the selection, embryogenic calli were induced and then somatic embryos were produced by subsequent subculturing. The somatic embryos were germiated on N6 basal medium containing 200 mg/L kanamycin and 500 mg/L carbenicillin. PCR analysis showed that nptII and bar genes were introduced in the Deoduck transformants. After the confirmation of bar gene expression in RNA and protein level, the transgenic Deoduck will be used to study the genetics of filial generation with the herbicide control gene, bar.gene, bar.

Changes in Enzyme Activity and Sensory Characteristics of Kochujang with Different Ratios of Added Deoduk (Codonopsis lanceolata) Root Powder (더덕 분말 첨가량을 달리한 고추장의 효소력 변화 및 관능적 특성)

  • Deoduk (Codonopsis lanceolata) root powder was added to traditional kochujang to improve the quality of traditional kochujang. The microbial characteristics, enzyme activities, and sensory characteristics were investigated during fermentation. The aerobic bacterial count in kochujang was not remarkabley different, and yeast and mold decreased during fermentation. Yeast and mold in the kochujang with 3 and 5% added deoduk root powder were 5 log scale at 8 weeks, whereas those in the control reached the same level at 6 weeks. ${\alpha}$, ${\beta}$-Amylase activity was at its highest level at 4 weeks during fermentation, and glucose and fructose contents showed the same results. The major free sugars in kochujang were glucose and fructose, and their contents increased rapidly at 2 weeks. Free sugar contents of kochujang with added deoduk was higher than that in the control. The sensory evaluation results showed that 1 and 3% deoduk kochujang had higher scores for taste and overall acceptance than those in the control. In particular, 1% deoduk kochujang had the highest scores.

Epilithic Diatom Communities in Streams of Jeju Island (제주도내 하천의 부착규조 군집)

  • To identify distribution of epilithic diatom assemblages and the relationship between diatoms and water chemistry, samples were collected twice a year from 2008 to 2012, from at 6 streams in Jeju Island, South Korea. A total of 171 diatom taxa were identified, and Achnanthes lanceolata, Gomphonema pumilum, Melosira varians, Navicula minima and Nitzschia palea were dominantly observed. BOD ranged 0.0~1.6 mg $L^{-1}$, TN 0.230~13.240 mg $L^{-1}$ (average 4.432 mg $L^{-1}$), TP 0.020~0.316 mg $L^{-1}$ (average 0.092 mg $L^{-1}$), and N/P rate ranged 2.5~178.5. Average of epilithic algal chlorophyll-a and AFDM were 1.1 ${\mu}gcm^{-2}$ and 0.6 mg $cm^{-2}$, respectively. According to BOD standard, the water qualities of study area sites were at 'Good' level. However, the healthiness levels assessed from DAIpo and TDI were 'Fair' to 'Poor'.

Vegetation Structure of Urban Forests on Mt. Goehwa, Sejong-Si (세종시 괴화산 도시숲의 식생구조)

  • Phyto-sociological methods were used in this study to assess the vegetation structure of a forest stand at Mt. Goehwa in Sejong-Si with the aim of providing vegetation information for urban forest utilization and management plans. The actual forest vegetation was classified into two types of community groups (Quercus serrata-Lindera obtusiloba and Coreopsis lanceolata community groups) at the highest hierarchical level. The Q. serrata-L. obtusiloba community group was classified into six units, which included artificial forest and natural forest vegetation. Artificial forests were classified into three communities (Pinus rigida, Castanea crenata, and Robinia pseudoacacia), whereas natural forests were classified into three communities (Quercus variabilis, Quercus acutissimaa, and Pinus densiflora). The Coreopsis lanceolata community group, which exhibited vegetative characteristics of urban forest edge areas, was categorized into four units. The urban forest edges were classified into four communities (Indigofera bungeana, Lespedeza bicolor, Amorpha fruticosa, and Lespedeza cuneata). Accordingly, the vegetation structure of Mt. Goehwa was categorized into 10 vegetation unit systems. An importance value analysis showed the highest importance value for C. crenata at 6.7%, followed by P. rigida at 6.4%, and R. pseudoacacia at 6.3%, indicating that the ecological impact of plantation species can be significant on Mt. Goehwa. A community coefficient of similarity analysis revealed that the artificial and natural forests had similar species compositions; however, both forests differed from the urban forest edge. This variation was further confirmed by Detrended correspondence analysis(DCA), with similar results. Canonical correspondence analysis(CCA) showed that the artificial forest and natural forest community types were positively correlated with altitude, bare rock, and the present species. By contrast, the urban forest edge community types were negatively correlated with these factors.

Effect of $GA_3$ and Light Quality on Seed Germination in Three Campanulan Plants ($GA_3$ 및 파종(播種) 후(後) 광질(光質) 처리(處理)에 따른 도라지, 더덕, 만삼의 발아율(發芽率))

  • Gibberellin (GA) has been widely used to break seed dormancy for better stand establishment. The experiment was done to clarify the effect of $GA_3$ (concentration; period) and light quality (red; white; dark) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum; Codonopsis lanceolata; C. pilosula) to get an information on their field emergence. The germination test was carried out with 12 hours irradiation for 9 days after priming treatment. In the darkness, the mean germination rate of all the species was decreased in the order to P. grandiflorum, C. lanceolata, C. pilosula. Their mean germination rates and radicle lengths were increased with increased concentration to 0.1mM of $GA_3$. Earlier germination rate was higher but later one was less in 4-day $GA_3$ treatment than in 1- or 2-day $GA_3$ treatment. Light treatment. especially red light given after $GA_3$ treatment. eliminated the $GA_3$ treatment effect. Red light done after $GA_3$ treatment nearly blocked the germination of P. grandiflorum and C. pilosula but delayed that of C. pilosula compared to the other light quality treatments having the similar rate. In addition. the radicle elongation of all three species affected by light quality treatment showed the same result as the germination rate.

Flora and Community Structure of Benthic Marine Algae in Ilkwang Bay, Korea (한국 일광만 저서 해조류의 해조상과 군집구조)

  • Marine algal flora and community structure were investigated seasonally at three sites in Ilkwang Bay on the southern east coast of Korea from May 2007 to February 2008. Total 103 species including 10 of green algae, 17 of brown algae, 76 of red algae were collected and identified. Among these species, 21 species were found throughout the year. Ulva pertusa, Enteromorpha linza, Grateloupia lanceolata, Chondracanthus intermedia and Caulacanthus ustulatus were distributed dominantly in upper intertidal zone. By contrast, crustose coralline algae, Grateloupia spp., Chondracanthus tenellus, Prionitis cornea and Sargassum spp. occurred predominantly in middle intertidal zone. Grateloupia spp., Sargassum spp., Ecklonia cava and Ulva pertusa were dominant in lower intertidal zone. Annual mean biomass in wet weight was 478.3 g m$^{-2}$. Maximum biomass was recorded in site 1 (731.8 g m$^{-2}$), and minimum was recorded in site 3 (78.5 g m$^{-2}$). The R/P, C/P and (R + C)/P value reflecting flora characteristics were 4.47, 0.59 and 5.06, respectively. Two groups produced by cluster analysis, one including sites 1, 2 and the other including site 3, showed meaningful difference in similarity, each other. Site 3 showed the limited species composition due to inflow of fresh water and absence of solid substratum. However, there was no significant difference between site 1 and site 2. In conclusion, the number of marine algae species and biomass in Ilkwang Bay were markedly reduced comparing with the previous studies. These suggest that a solution for reconstruction of the poor marine algal vegetation is considerably demanded.

Effect of washing methods on the quality of freshly cut sliced Deodeok (Codonopsis lanceolata) during storage (세척방법에 따른 신선편이 슬라이스 더덕의 저장 중 품질 특성 변화)

  • There is increasing interest in freshly cut products, that is, foods produced without washing and cutting. In this study, the quality of freshly cut sliced Deodeok was compared with that of what based on its washing methods. In bubble washing, the Deodeok rises to the water surface apace and is broken into centimeter sizes. Microbubble washing calls for the production of a great number of 0.1 mm-sized bubbles in anions-bearing water and their passing through a trumpet-shaped hole at a high pressure. To compare the product deterioration rates of the specimens, they were stored at $10^{\circ}C$ for 10 days. In the specimens washed with the control method and with hand washing, the deterioration rate was 80%; and in the specimens washed with bubble and microbubble washing, 20~30%. The L-value (an index of browning) was higher in the bubble and microbubble washing than in the control and the hand washing, which implies that browning was minimized during the storage. As for the viable cell and coliform group counts that were measured during the storage, the specimens washed with the control method showed the highest values. In contrast, the specimens washed with microbubble washing showed the lowest values. In the sensory test, the specimens washed with microbubble were highest in storage preference. In conclusion, the Deodeok that was stored after it was washed with microbubble washing was found to have had the best quality.

Studies on the Environmental Condition, the Search and the Response to Temperature and Photoperiods of the Plants for Urban Forest Aromatic Bath (도시형(都市型) 삼림욕장(森林浴場)의 환경(環境), 욕장식물(浴場植物)의 탐색(探索)과 온도(溫度) 및 광주기(光週期) 반응(反應))

  • The studies were carried out to develop an urban forest aromatic bath in the idle space within city area for handicapped persons, patients and city workers, who having less chance to visit the natural forest aromatic bath. The environmental conditions were checked in the experimental double vinyl house where native and exotic aromatic woody and herbaceous plants were growing with minimum energy input in the periods of mid-winter and mid-summer. The growth and photoperiodic responses of the plants to the low and high temperatures were observed during winter and summer respectively. The most of herbaceous plants in the double vinyl house with the controlled minimum night temperature of $-2^{\circ}C$ had no winter injury but did not grow and started the growth in April. The exotic herbs such as Catnip faassen's(Nepeta x faassenia) and Lemon bergamot(Monarda citriodora) grew better in the longer photoperiodic conditions. The growth rates of Abies koreana, Thuja koraiensis and Pinus sylvestriformis in the early spring were affected mainly by the night minimum temperature and less affected by the photoperiodic conditions during winter. The native aromatic herbs such as Robdosia japonica, Molsa punctulata, Agastache rugosa and Codonopsis lanceolata showed stimulatory growth at the 16 hours day-length in case of the high temperature vinyl hose. The height and root collar diameter growth of pine seedlings were inhibited in maxim temperatures over $40^{\circ}C$ during summer.
