Prevalence of Centrocestus formosanus Metacercariae in Ornamental Fish from Chiang Mai, Thailand, with Molecular Approach Using ITS2

  • The prevalence of Centrocestus formosanus metacercariae was investigated in ornamental fish purchased from a pet shop in Chiang Mai, Thailand, including Carassius auratus (goldfish), Cyprinus carpio (Koi), Poecilia latipinna (Sailfin Molly), Danio rerio (Zebrafish), and Puntigrus tetrazona (Tiger barb). The parasite species was identified by the morphology of worms as well as by a molecular approach using ITS2. The results showed that 50 (33.3%) of 150 fish examined were infected with the metacercariae. The highest prevalence was found in C. auratus (83.3%), and the highest intensity was noted in C. carpio (70.8 metacercariae/fish). The most important morphological character was the presence of 32-34 circumoral spines on the oral sucker. The phylogenetic studies using the rRNA ITS2 region revealed that all the specimens of C. formosanus in this study were grouped together with C. formosanus in GenBank database. This is the first report on ornamental fish, C. carpio, P. latipinna, D. rerio, and P. tetrazona, taking the role of second intermediate hosts of C. formosanus in Thailand. Prevention and control of metacercarial infection in ornamental fish is urgently needed.

Ichthyofauna and Structure of the Fish Community in Hapcheon Lake on the Hwang River (합천호의 어류상과 어류 군집구조)

  • The ichthyofauna and structure of the fish community of Hapcheon Lake on the Hwang River were surveyed at six localities from May 2004 to Feb. 2005. There were 36 species belonging to 31 genera and 12 families in the Hapcheon Lake system. Of these, 24 species (66.7%) were cyprinids; cobitids and centropomids were next with 2 species (5.6%) each. The dominant species in the Hapcheon Lake system was Hemiculter eigenmanni (relative abundance 28.4%), the subdominant species Hemibarbus labeo (24.1%). Ten species (28.3%) of the 36 were Korean endemic species. Among the endemic species, H. eigenmanni and Squalidus chankaensis tsuchigae were abundant at each locality except site 6, but the eight other species were rare. In biomass, H. labeo comprised about 36.5% of the total weight, H. eigenmanni 22.3%, Cyprinus carpio 7.5%, Hypomesus nipponensis 6.9%, Culter brevicauda 3.7% and Lepomis macrochirus 3.6%. Average dominance, diversity, and evenness index of the fish community collected from six localities in the Hapcheon Lake system were 0.21, 0.85, and 0.67, respectively.

Process Optimization for Cookies with Low-marketable Israeli Carp Cyprinus carpio Using Response Surface Methodology (RSM) (반응표면분석법을 활용한 저상품성 향어(Cyprinus carpio) 쿠키 제조 공정 최적화)

  • This study was conducted to optimize the cookie preparation using Israeli crap Cyprinus carpio paste (C-ICP). The results of response surface methodology suggested the selection of other supplementary materials/[soft flour (SF) + ICP] (X1) and SF/ICP (X2) as independent variables, and hardness (Y1), yellowness (Y2), amino acid nitrogen (Y3), and overall sensory acceptance (Y4) as dependent variables. The optimal conditions of OS, SF, and ICP were 35.0%, 40.3% and 24.7%, respectively, and the predicted values of the multiple response optimal conditions Y1, Y2, Y3, and Y4 were 2,006.2 N/m2, 21.1, 30.0 mg/100 g, and a score of 6.6, respectively. Under the optimum conditions, experimental values of Y1, Y2, Y3, and Y4 were 2,010.5 ± 22.3 N/m2, 21.6 ± 0.5, 29.6 ± 0.7 mg/100 g, and a score of 6.9 ± 0.3, respectively, which were not significantly different from the predicted values (P < 0.05). The results on hardness, moisture, VCI yellowness, and lightness suggested that the optimum heating period was 20 min. C-ICP prepared under the optimum conditions was superior in sensory evaluation to cookies without the Israeli carp paste.

Ecotoxicities of emulsifiable concentrate and granules of cinnamon (Cinnamomum zeylanicum) essential oil against Cyprinus carpio and Danio rerio (계피(Cinnamomum zeylanicum) 정유의 유제 및 입제의 잉어(Cyprinus carpio) 및 제브라피쉬에 대한 생태독성)

  • In this study, cinnamon essential oil (CEO) was formulated as emulsifiable concentrate (EC) and a granule. For the evaluation of their negative effects on the ecosystem, acute toxicities against Cyprinus carpio was determined in a static condition. The formulations were made using CEOs extracted by 3 different methods (steam distillation (SD), solvent extraction and supercritical fluid extraction (SFE)) and were tested to obtain $LC_{50}$ values. Among the ECs, EC including CEO extracted by SFE showed highest acute toxicity against C. carpio. Among the granules, a granule including CEO extracted by SD showed highest acute toxicity against C. carpio. Nevertheless, $LC_{50}$ of EC and a granule formulation with CEOs was higher than toxicity level III of pesticide standardized by Korea rural development administration. These results were similar to those using zebrafishes. Chronic toxicities were not found for 45 days in zebrafishes until $500{\mu}gL^{-1}$ level of EC formulation including CEO obtained by the SD. Based on these results, EC formulation of CEOs may be considered to be used as environmental-friendly natural insecticides in accordance with the standards.

Changes of Gill Structure and Identification of Genes by Muddy Water Exposure in Cyprinus carpio (잉어(Cyprinus carpio)에서 탁수 노출에 의한 아가미 미세구조 변화와 유전자 확인)

  • Present study aimed to investigate morphological change and gene expression in the gill of Cyprinus carpio after exposure against muddy water caused by riverbed disturbance with various rearing condition. The gill of C. carpio showed abnormal shapes in its secondary lamellae and a rough surface with impure debris after exposed to muddy water for 80 days. In addition, the gills showed the edema, the exfoliation of epithelial cell, and the fusion of the secondary lamellae. Using 20 ACPs, 24 differentially expressed genes (DEGs) exposed to muddy water for 80 days were identified. 17 genes among them were up-regulated, while 7 genes were down-regulated in preterm deliveries. A BLAST searches revealed that 3 genes were revealed known genes as calcium transporter 1 (TRPV6) mRNA, macha mRNA for putative puroindoline b protein, and Efnb3 protein-like. Therefore, it is considered that Efnb3 gene from gill would be a useful indicator for neurobehavioral changes in fish influenced by muddy water.

Fish Community and Upstream of Glass Eels (Anguilla japonica) in the Imjin River Estuary, Korea (임진강 하구역의 어류 군집과 실뱀장어 소상)

  • The fish community and upstream of glass eels (Anguilla japonica) in the Imjin River estuary Gyeonggi-do, Korea was investigated from 2008 to 2012. During the surveyed period 43 species belonging to 18 families were collected. Korean endemic species was Microphysogobio jeoni which showed a ration of 0.04% in collected species. Exotic fishes were Carassius cuvieri and Ctenophayngodon idellus, and comprised 0.06% of the total individual number. Among the fish species observed, 17 species of the freshwater fish (40.5%), 6 species of migration fish (14.2%), 8 species of the brackish water fish (19.1%) and 11 species of sea water fish (26.1%). In terms of composition ration, Anguilla japonica (glass eels, 49.8%), Coilia ectens (23.3%), Saurogobio dabryi (8.1%), Repomucenus olidus (3.3%), Lateolabrax maculata (2.9%) and Chelon haematocheilus (2.8%) were found to display high individual number. On the other hand, C. ectens (33.6%), C. haematocheilus (14.2%), Cyprinus carpio (10.5%), Mugil cephalus (9.1%) and S. dabryi (7.6%) were dominated of biomass. The dominant species of individual was A. japonica (glass eels), and subdomint species included C. ectens. The fish biomass catched of fisherman in the Imjin River (Paju-si) were 83.6~240.3 t by each year. The year 2011 and 2012 were decreased rapidly. Among these, M. cephalus (48.0~80.0%), C. carpio (6.7%), Carassius auratus (4.9%), Silurus asotus (3.9%), Takifugu obscurus (2.5%), Anguilla japonica (adult, 2.2%) and Pseudobagrus fulvidraco (2.1%) dominated of biomass. Body weight of glass eels at each year were 0.03~ 1.13 t, average 212.0 kg and 1,325,000 individual by 0.16 g each one individual.

Nutritional Properties of Cookies Made with Israeli Carp Cyprinus carpio (향어(Cyprinus carpio) 함유 쿠키의 영양특성)

  • This study was conducted to compare the nutritional properties of cookies made with Israeli carp Cyprinus carpio (C-IC) to those made without Israeli carp (control). The proximate composition of C-IC per 100 g was 4.1 g moisture, 9.7 g protein, 29.2 g lipid, 1.4 g ash, and 55.6 g carbohydrates. Moisture, protein and ash contents were significantly higher and the carbohydrate content was significantly lower (P<0.05) in C-IC than control, but there was no difference in lipid content (P>0.05). The total amino acid content of C-IC per 100 g was 9.46 g and the major amino acid was glutamic acid (2.49 g). The mineral contents of C-IC per 100 g were 216.6 mg calcium, 193.2 mg phosphorus, 170.9 mg potassium, and 18.2 mg magnesium, which were all significantly higher than the contents of the control (P<0.05). The major fatty acids of C-IC were 16:0, 18:1n-9, and 18:2n-6. The digestibility of C-IC in the small intestine was 51.3%, which was higher than the digestibility of the control. These results suggest that C-IC have better nutritional properties than the control.

Correlation and Sensitivity of Acute Toxicity of Pesticides on the Common Carp(C. carpio) and Killifish(O. latipes) (농약에 대한 잉어(C. carpio)와 송사리(O. latipes)의 급성독성 상관성 및 감수성)

  • The recommended species in fish acute toxicity test for pesticide registration in korea are common carp (Cyprinus carpio), killifish(Oryzias latipes) and loach(Misgurnus anguillicaudatus). But most of fish toxicity was evaluated using common carp. The common carp has several problems such as difficulty of room breeding, difference of sensitivity to breeding condition, limited experimental period, etc. For that reason, it is necessary to reexamine of test species. The present study investigated the sensitivity and correlation of $LC_{50}$ values that determined using the acute toxicity test with common carp and killifish. And this study is a realistic approach to evaluated appropriate species for fish acute toxicity test of pesticide. As a result, correlation coefficient(R) of the commom carp and killifish was high correlation of 0.8480 and the $LC_{50}$ values was not significant in the interspecies sensitivity. When the $LC_{50}$ values of common carp and killifish was compared with fish toxicity guideline in Korea, fish toxicity class has fluctuated in 18 pesticides with not very special tendency. Therefore, it is necessary to review of killifish has more advantage to standardization of fish culture and in the side of correlation and sensitivity of acute toxicity.

Fish and Efficiency on Attached Fish Eggs of Artificial Floating Island in Lake Soyang, Korea (소양호 인공식물섬의 어류 서식 및 어란 부착 효과)

  • The fish and attached fish eggs by artificial floating island in lake Soyang was investigation from May to June 2013. The observed fish at the floating island during the survey period were Zacco platypus (5.2 %), Siniperca scherzeri (1.0 %), Lepomis macrochirus (90.8 %) and Micropterus salmoides (3.0 %) at the Cheongpyeong-ri, Cyprinus carpio (0.4 %), Z. platypus (54.6 %), L. macrochirus (20.3 %) and M. salmoides (24.7 %) at the Ungjin-ri. The collected species at the neighboring facility were 22 species belonging the seven families. Among them, exotic species such as Carassius cuvieri, L. macrochirus and M. salmoides. Also, Z. platypus (30.4 %), L. macrochirus (18.9 %), Opsariichthys uncirostris amurensis (15.4 %) and Hemibarbus labeo (9.3 %) were numberous. Attached fish eggs number were 708,800 (800 per $m^2$) at the Cheongpyeong-ri and 974,500 (1,100 per $m^2$) at the Ungjin-ri. The fish eggs number were 504,920 of the Carassius auratus (212,640 at the Cheongpyeong-ri, 292,280 at the Ungjin-ri) and 1,178,380 of the C. carpio (496,160 at the Cheongpyeong-ri, 682,220 at the Ungjin-ri). The hatched eggs number were 350,505 of the C. auratus(102,067 at the Cheongpyeong-ri, 248,438 at the Ungjin-ri) and 811,222 of the C. carpio (238,157 at the Cheongpyeong-ri, 573,065 at the Ungjin-ri).

In Vivo Metabolic studies on Carbofuran Degradation in carp(Cyprinus carpio L.) (Carbofuran 의 잉어(Cyprinus carpio)체내 대사)

  • Absorption, distribution, metabolism and excretion of $^{14}C-carbofuran$(2,3-dihydro-2,2-dimethyl-7-benzofuranyl methylcarbamte) were studied in carp(Cyprinus carpio L.) after the treatment of carbofuran at the dose level of 43 parts per billion. Maximum radioactivities in tissues(liver, kidney, gut, gall bladder) and blood of carp were shown 12hrs after the treatment of $^{14}C-carbofuran$. Carbofuran was metabolized to 3-hydroxycarbofuran and 3-ketocarbofuran in liver and kindney of carp, and the major metabolite was 3-hydroxycarbofuran. Most radioactivity absorbed into the carp tissues was eliminated 3hrs after transfer of the carp to fresh water. The excretory metabolites were 3-ketocarbofuran(32.3%), 3-hydroxycarbofuran(52.8%) and an unknown metabolite(2.6%) during the period of 3hrs of the excretory experiment.
