Sulforaphene Attenuates Cutibacterium acnes-Induced Inflammation

  • Acne is a chronic inflammatory disease of the sebaceous gland attached to the hair follicles. Cutibacterium acnes is a major cause of inflammation caused by acne. It is well known that C. acnes secretes a lipolytic enzyme to break down lipids in sebum, and free fatty acids produced at this time accelerate the inflammatory reaction. There are several drugs used to treat acne; however, each one has various side effects. According to previous studies, sulforaphene (SFEN) has several functions associated with lipid metabolism, brain function, and antibacterial and anti-inflammatory activities. In this study, we examined the effects of SFEN on bacterial growth and inflammatory cytokine production induced by C. acnes. The results revealed that SFEN reduced the growth of C. acnes and inhibited proinflammatory cytokines in C. acnes-treated HaCaT keratinocytes through inhibiting NF-κB-related pathways. In addition, SFEN regulated the expression level of IL-1α, a representative pro-inflammatory cytokine expressed in co-cultured HaCaT keratinocytes and THP-1 monocytes induced by C. acnes. In conclusion, SFEN showed antibacterial activity against C. acnes and controlled the inflammatory response on keratinocytes and monocytes. This finding means that SFEN has potential as both a cosmetic material for acne prevention and a pharmaceutical material for acne treatment.

개똥쑥 추출물의 항여드름 효능확인 (Anti-acne Properties of Artemisia annua Extract In Vitro)

  • 여드름은 털피지샘단위(pilosebaceous unit)와 관련된 만성 염증 피부 질환으로, 여드름 병변에서는 피지 과다분비(hyperseborrhea)나 이상분비(dysseborrhea), 염증반응, 그리고 다른 피부상재균들에 비해 증가된 Cutibacterium acnes (C. acnes)로 인한 피부 미생물 균총의 불균형이 관찰된다. 이 연구는 개똥쑥 추출물(Artemisia annua extract: AAE)의 항여드름 효과를 확인한 것으로, 피부 미생물 균총의 균형유지와 관련된 항균효과와 염증반응의 억제, 과도한 피지 분비의 감소 측면에서 실험을 진행하였다. C. acnes와 Staphylococcus epidermidis(S. epidermidis)를 AAE와 공동배양 하였을 때, S. epidermidis의 성장률은 저해되지 않았지만 C. acnes의 성장률은 저해된 것을 확인하였다. 또한 AAE를 처리하여 배양한 C. acnes 배양배지를 세포에 처리하였을 때, 인터루킨-1β(IL-1β), 종양괴사인자-α(TNF-α)와 인터루킨-6(IL-6) 같은 사이토카인 분비의 감소와 TLR2 활성 억제도 확인하였다. 마지막으로 피지세포에 AAE를 처리한 결과, 팔미트산에 의해 유도된 피지형성을 감소시키는 것을 확인하였다. 이 결과들은 AAE가 다양한 타깃을 지닌 천연추출물로써 여드름의 주요 원인들인 C. acnes의 선택적 성장저해와 C. acnes로부터 유도되는 염증반응을 억제할 수 있으며, 과도한 피지형성을 감소시켜 결과적으로 여드름을 완화시키는 물질로 사용될 수 있다는 것을 제시한다.

  • Cutibacterium acnes (이전 학명 = Propionibacterium acnes)은 혐기성, 그람 양성 간균으로 사람 피부 및 점막조직 표면의 정상 세균총에 해당할 뿐만 아니라 여드름, 유육종증, 뇌종양, 심내막염, 전립선암, 치주질환 및 치수감염질환과도 연관성이 있는 것으로 보고되고 있다. C. acnes KCOM 1861 (= ChDC B594) 균주가 사람 악골 골수염 병소에서 분리되었으며 그 유전체 염기서열을 해독하여 보고한다.

  • 여드름은 사춘기와 젊은 연령층에서 나타나는 염증성 피부질환이다. Propionibacterium acnes (P. acnes)는 여드름에서 염증을 유발하는 주요한 균으로 알려져 있다. P. acnes 는 피부에서 과잉생산된 피지에 의해 막힌 모낭 내에서 증식하며, 피부 내의 단핵구를 활성화시키고 염증성 사이토카인(pro-inflammatory cytokine)의 분비를 촉진하여 염증반응을 증가시키는 것으로 알려져 있다. 본 논문에서는 Cnestis palala (Lour.) Merr. 추출물이 P. acnes 에 의한 염증반응을 감소시킬 수 있는지 확인하고자 하였다. 마우스 대식세포주인 RAW264.7 에서 C. palala 추출물은 P. acnes 에 의해 증가하는 염증성 사이토카인인 $IL-1{\beta}$의 단백질 발현량과 IL-6, $TNF-{\alpha}$, COX-2 의 mRNA 발현량을 감소시켰다. 또한 염증성 사이토카인 발현의 주요 전사인자(transcription factor)인 $NF-{\kappa}B$ 의 전사 활성 역시 C. palala 추출물에 의해 감소하는 것을 확인하였다. 따라서 C. palala 추출물이 P. acnes 에 의해 발생하는 여드름의 치료제나 그 보조제로 사용될 가능성이 있음을 제안 할 수 있다.

항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과 (Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

  • Cutibacterium acnes는 사람의 피부, 결막, 장관, 외이도 및 구강의 정상 세균 총의 하나이다. 이 세균 종은 여드름, 심내막염 감염, 유육종증, 뇌 농양, 치주염 및 골수염과 관련된 기회감 염성병원균으로 확인되었다. C. acnes KCOM 1315 (= ChDC KB81)는 사람 악골골수염 병소로부터 분리되었다. 여기에서 C. acnes KCOM 1315 균주 완전 유전체 염기서열을 해독하여 보고한다.

  • According to the traditional chinese herbal Cuscuta japonica Choisy and C. australis R.Be (original plant of Semen Cuscutae in Korea) have been used to cure pimple. In order to prove their bacteriostatic effects against Propionibacterium acnes, we performed the micro broth dilution method. The minimal inhibitory concentration (MIC) of C. japonica juice was 27.3 mg/ml, but C. australis juice showed no effect. The MIC of water extracted C. japonica seed and Wontosa and Bupjetosa were $960\;{\mu}g/ml$. The MIC of both water and ethanol extracts of C. australis seed was $9240\;{\mu}g/ml$. But ethanol extracts of C. japonicaseeds, Wontosa and Bupjetosa had no bacteriostatic activity. We also performed the disk diffusion test, C. japonica juice had prominent effect. Water extracts of C. japonica seed, Wontosa and Bupjetosa showed bacterio-static effect in 10 mg/ml, and C. australis seed in 10 mg/ml. The ethanol extract of C. japonica seed, Wontosa and Bupjetosa, and C. australis seed showed bacteriostatic effect in 20 mg/ml.

여드름균(Cutibacterium acnes)에 대한 해조류 추출물의 항균효과 (In vitro Antimicrobial Activities of Edible Seaweeds Extracts Against Cutibacterium acnes)

  • The study was aimed at developing an alternative therapeutic agent against acne vulgaris, one of the most common skin diseases, to meet the continuing demand for new therapies. Acne vulgaris is often associated with the acne-causing bacteria such as Cutibacterium acnes. To investigate the safety of agents against acne vulgaris, we evaluated the potential antibacterial activities of edible seaweeds against C. acnes in Korea. Forty-one edible seaweeds, including the brown, green, and red varieties, were selected for the antibacterial test. In comparison with other seaweeds, 70% ethanolic extracts of brown seaweeds, such as Cladophora wrightiana var. minor, Eisenia bicyclis, Ecklonia cava, Ishige foliacea, Ishige okamurae, Sargassum filicinum, and Sargassum miyabei Yendo, exhibited potential antibacterial activity against C. acnes with minimum inhibitory concentrations ranging between 64 and 128 ㎍/mL. To investigate the active anti-acne agents and to enhance our understanding of the antibacterial activities against C. acnes, further solvent-fractionation experiments are warranted. The findings imply that brown seaweeds can be a potential source of natural agents against acne vulgaris.

STUDIES OF ACNE TREATMENT USING ORIENTAL HERBS(New Approach to select anti-acne agents)

  • Acne vulgaris, the most common skin disease. can be formed as only a few comedons or severe inflammatory lesions. The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.
