Separation and Purification of Lipase Inhibitory Peptide from Fermented Milk by Lactobacillus plantarum Q180

  • In this study, we separated and purified lipase inhibitory peptide from fermented milk by Lactobacillus plantarum Q180 with the aim of developing a new functional anti-lipase activity yogurt product. L. plantarum 180 was inoculated into 10% reconstituted skimmed milk and incubated at 37℃ until the pH of the culture reached pH 4.4. The lipase activity was measured using porcine pancreatic lipase. The lipase inhibitory peptides were gradually isolated by ultrafiltration, reversed phase column chromatography (RPC), reversed phase high-performance liquid chromatography (RP-HPLC), and gel permeation high-performance liquid chromatography (GP-HPLC) from the fermented milk by L. plantarum Q180. An ODS-AQ column was used for the RPC, a Vydac C18 column for the RP-HPLC, and a Superdex Peptide HR column for the GP-HPLC. The peptide was composed of Asp, Thr, Ile, Ser, Ala, and Gln, and the anti-lipase activity (IC50) was 2,817 ㎍/mL.

진핵생물 개시인자 유래 펩타이드의 세포 성장 억제 효능 (Effect of cell growth inhibition by eukaryotic initiation factor 2 derived peptides)

  • In the process of protein transcription and translation, various protein complexes bind to DNA, and all processes are precisely controlled. Among the proteins constituting this complex, a peptide derived from eukaryotic initiation factor (eIF) 2 was synthesized. In addition, in order to increase the efficiency of transduction of this peptide into cells, peptides with polyarginine, one of the protein transduction domains (PTD), were synthesized. Cell growth inhibition was confirmed in HER2 positive breast cancer (SK-Br-3) and HER2 negative breast cancer (MDA-MB-231), and cardiomyocytes (H9c2). The peptide with polyarginine had high transduction efficiency in all cells, and had excellent cancer cell growth inhibitory effects. The peptide used in this study might be useful peptide therapeutics for the treatment of cancer through future research.

DNA와 상호작용에서 T4 endonuclease V의 C-말단 부위의 역할에 관한 분광학적 연구: 핵자기공명과 형광 실험 (The Spectroscopic Study on the Role of C-terminal Region of T4 endonuclease V in the Interaction with DNA: NMR and Fluorescence Experiment)

  • In order to study the role of C-terminal aromatic region of T4 endonuclease V in the interaction with substrate DNA, NMR and Fluorescence spectrum were recorded. Analysis of flu orescence emission spectra showed that C-terminal region of T4 endonuclease V is in or very near the binding site. In the HSQC spectrum of $^{15}N$-Tyr-labeled T4 endonuclease V*DNA complex, the broadening of a peak was observed. It is presumed that this peak corresponds to one among three tyrosine residues which belong to the WYKYY segment of C-terminal region of T4 endonuclease V. Interactions of peptide fragments consisting of C-terminal residues of T4 endonuclease V with DNAs(TT-, T^T-DNA) were investigated by NMR and Fluorescence experiment. The results suggest that two peptide fragments themselves bind to DNAs and their binding pattern is not an intercalation mode.

식단의 당부하량에 따른 20대 성인의 체중 감량 효과 연구 (Effects of a low glycemic load diet on body weight loss in overweight or obese young adults)

  • 본 연구에서는 한국인이 평소 섭취하는 일반식사와 유사한 수준의 당부하량을 가진 high GL 식사와 당부하량이 낮게 설계된 low GL 식사를 섭취한 후 체중, 체지방, 혈압 및 혈액 내 생활습관병 위험 지표 등의 변화를 분석 및 비교하였다. 20대 성인남녀를 대상으로 체중 감량을 위해 wash-out 기간인 중간 2주를 제외하고 개인 필요 열량의 30%를 줄인 low GL 식사와 high GL 식사를 각 2주간 제공하였다. Low GL 식사와 high GL 식사의 효과를 비교한 결과 low GL 식사가 high GL 식사에 비해 체중 (p < 0.001), 체지방량 (p = 0.024), 체질량지수 (p < 0.001)에서 유의적으로 감소량이 더 컸다. 하지만 혈압, 혈액 내 비만 질환 위험 지표에서는 유의한 차이가 없었다. Low GL 식사의 전·후를 비교한 결과 체중, 체지방량, 체질량지수, 체지방률, 이완기 혈압, 중성지방, 총콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤, 공복혈당, C-peptide HOMA 지수가, high GL 식사에서는 체중, 체지방량, 체질량지수, 총콜레스테롤, HDL-콜레스테롤, 공복혈당, C-peptide, C-peptide HOMA 지수에서 각 식사 전보다 후에 유의적으로 감소되었다. 또한 실험 식사 섭취 전과 섭취 후를 비교한 결과 체중, 체지방량, 체질량지수, 체지방률, 혈압 (수축기, 이완기), 총콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤, 공복혈당, 당화혈색소, C-peptide, C-peptide HOMA 지수가 모두 유의적으로 감소되었다. 본 연구 결과 체중감량 및 생활습관병 위험을 줄이기 위해서는 단순히 섭취 열량을 낮추기보다는 열량과 함께 당부하량을 고려하여 섭취하는 것이 체중을 감소시키는 데에 더 효과적일 것으로 사료된다.

메기의 껍질로부터 항균성 펩타이드의 정제 (Purification of Antibacterial Peptide from the Skin of the Catfish Silurus asotus)

  • 메기(Silurus asotus)의 껍질로부터 항균성 펩타이드를 정제하였다. 메기 껍질의 산 추출물은 단계적 농도구배조건으로 Sep-Pak C18에 의해 부분적으로 정제되었으며, 그 중에서 특히 60% 메탄올 분획(RM60)이 Escherichia coli D31에 대해 가장 좋은 항균활성을 나타내었다. 이 RM60을 사용하여 이온교환 및 5단계의 연속적인 역상 HPLC로 정제하였다. 정제된 펩타이드의 분자량과 아미노산 서열분석은 MALDI-TOF MS와 에드만 분해법으로 분석하였다. 이 펩타이드의 분자량은 약 4182.1 [M+H]+이었으며, 분석된 이 물질의 부분적인 일차구조서열은 다음과 같다; PALXXKARREAKVKF. 이러한 결과는 메기의 껍질에서 존재하는 이 펩타이드가 메기의 껍질에서 반응하는 선천성 방어 시스템에서 중요한 역할을 하고 있다고 여겨진다.

Changes of Atrial Natriuretic Peptide System in Rats with Puromycin Aminonucleoside-Induced Nephrotic Syndrome

  • Sodium retention is a hallmark of nephrotic syndrome. We investigated whether sodium retention is associated with changes of natriuretic peptide system at different stages (i.e., a sodium retaining stage and a compensatory stage) of nephrotic syndrome. At day 7 after PAN(puromycin aminonucleoside) injection, the urinary excretion of sodium was decreased, along with the development of ascites and positive sodium balance. The plasma and urinary ANP(atrial natriuretic peptide) immunoreactivities were increased. ANP mRNA expression was increased in the heart and kidney, whereas that of NPR(natriuretic peptide receptor)-A and NPR-C mRNA was decreased in the kidney. The expression of NEP was decreased in the kidney. At day 14, urinary excretion of sodium did not differ from the control. The plasma ANP level and heart ANP mRNA expression returned to their control values. The expression of ANP mRNA in the kidney was increased in association with increased urinary ANP immunoreactivities. The expression of NPR-A in the kidney became normal, whereas that of NPR-C kept decreased. The expression of NEP(neutral endopeptidase) remained decreased. These findings suggest that the increased renal ANP synthesis in association with decreased metabolism via NEP and NPR-C may play a compensatory role against the development of sodium retention in nephrotic syndrome. The decreased of NPR-A expression in the kidney may contribute to the ANP resistance at day 7. The subsequent recovery of NPR-A expression may play a role in promoting sodium excretion in later stage(at day 14).

항-펩타이드 항체를 이용한 암유전자 N-myc 산물의 면역조직화학적 검출 (Immunohistochemical Detection of N-myc Gene Product by Using Antiserum Against Synthetic Peptide)

  • N-myc, a DNA sequence related to the oncogene c-myc, was found to be amplified in untreated primary neuroblastomas and the amplification appeared to be associated with advanced disease at diagnosis and rapid tumor progression. Synthetic peptides have been useful immunogens for generating antisera and monoclonal antibodies to a number of native proteins. In order to identify myc-related protein in the tumor cells, an antiserum against a synthetic hexapeptide (-Glu-Asp-Ile-Trp-Lys-Lys-), whose sequence corresponds to a part of the exon 2 of oncogene N-myc, was prepared by immunizing a rabbit with BSA-conjugated peptide. After ammonium sulfate precipitation and affinity column chromatography, it appeared to be specific to the peptide. Strong nuclear staining in immunoperoxidase method using this serum was observed in both human promyeloid leukemic cell line, HL-60(containing high c-myc copy number), and human neuroblastoma cell line, LA-N-5 (containing high N-myc copy number), whereas LA351 (human lymphoid cell line) cells did not react with the serum. This reaction was completely abrogated by incubating the antiserum with soluble excess peptide. These data suggest that the protein encoded by N-myc could be localized in the nucleus as c-myc protein and this antiserum can be used to detect myc-related tumor cells in clinical samples and to determine if the N-myc expression correlates with genomic amplification in cell lines, untreated primary tumors, and untreated metastases.

Molecular Characterization of a Defensin-like Peptide from Larvae of a Beetle, Protaetia brevitarsis

  • A cDNA encoding a defensin-like peptide (Protaetiamycine) from the larvae of a beetle, Protaetia brevitarsis was cloned. The DNAs encoded the deduced propeptide of 79 amino acid residues with the predicted molecular weight of 8.4 kDa and PI of 8.24. Overall amino acid sequence of this protein has 39% similarity to that of Rhodnius prolixus defensin, 43% similarity to that of Acalolepta luxuriosa defensin, and 72% similarity to that of Oryctes rhinoceros defensin, suggesting that this gene is an insect defensin. In an attempt to apply the anti-bacterial peptide to the development of therapeutic agents, a 12-mer peptide amidated at its C-terminus, ACAAHCLAIGRG-$NH_2$ (Ala55-Lys66-$NH_2$, 12Pbn) was synthesized. This peptide showed some antifungal activity against Candida albicans. To increase antifungal activity, six 9-mer peptides were synthesized by modifying amino acid sequences of 12Pbn fragment. Among these peptides, 9Pbm3-9Pbm6 exhibited strong activity compared with Cecropin B and mellitin.
