Type VI secretion system (T6SS) has been discovered in a variety of gram-negative bacteria as a versatile weapon to stimulate the killing of eukaryotic cells or prokaryotic competitors. Type VI secretion effectors (T6SEs) are well known as key virulence factors for important pathogenic bacteria. In many Burkholderia species, T6SS has evolved as the most complicated secretion pathway with distinguished types to translocate diverse T6SEs, suggesting their essential roles in this genus. Here we attempted to detect and characterize T6SSs and potential T6SEs in target genomes of plant-associated and environmental Burkholderia species based on computational analyses. In total, 66 potential functional T6SS clusters were found in 30 target Burkholderia bacterial genomes, of which 33% possess three or four clusters. The core proteins in each cluster were specified and phylogenetic trees of three components (i.e., TssC, TssD, TssL) were constructed to elucidate the relationship among the identified T6SS clusters. Next, we identified 322 potential T6SEs in the target genomes based on homology searches and explored the important domains conserved in effector candidates. In addition, using the screening approach based on the profile hidden Markov model (pHMM) of T6SEs that possess markers for type VI effectors (MIX motif) (MIX T6SEs), 57 revealed proteins that were not included in training datasets were recognized as novel MIX T6SE candidates from the Burkholderia species. This approach could be useful to identify potential T6SEs from other bacterial genomes.
The use of recycled aggregate in concrete is gaining much attention due to the growing need for sustainability in construction. In the present study, Self Compacting Concrete (SCC) is made using both natural and recycled aggregate (crushed recycled concrete aggregate from building demolished waste) and performance of recycled aggregate based SCC for the bond behaviour of reinforcement is evaluated. The major factors that influence the bond like concrete compressive strength (Mix-A, B and C), diameter of bar ($D_b=10$, 12 and 16 mm) and embedment length of bar ($L_d=2.5Db$, $5D_b$ and full depth of specimen) are the parameters considered in the present study in addition to type of aggregates (natural and recycled aggregates). The mix proportions of Natural Aggregate SCC (NASCC) are arrived based on the specifications of IS 10262. The mix proportions also satisfy the guidelines of EFNARC. In case of Recycled Aggregate SCC (RASCC), both the natural coarse and fine aggregates are replaced 100% by volume with that of recycled aggregates. These mixes are also evaluated for fresh properties as per EFNARC. The hardened properties like compressive strength, split tensile strength and flexural strength are also determined. The pull-out test is conducted as per the specifications of IS 2770 (Part-1) for determining the bond strength of reinforcement. Bond stress versus slip curves were plotted and a typical comparison of RASCC is made with NASCC. The fracture energy i.e., area under the bond stress slip curve is determined. With the use of recycled aggregates, reduction in maximum bond stress is noticed whereas, the normalised maximum bond stress is higher in case of recycled aggregates. Based on the experimental results, regression analysis is conducted and an equation is proposed to predict the maximum bond stress of RASCC. The equation is in good agreement with the experimental results. The available models in the literature are made use to predict the maximum bond stress and compare the present results.
This study investigates experimentally into the design factors and quality variations having an effect on the properties of the combined high flowing concrete to be poured in the slurry wall of Inchon LNG in-ground receiving terminal. Especially, high belite cement and lime stone powder as cementitious materials and viscosity agent in order to improve self-compaction and hydration heat are used in this study. Water-cement ratio(W/C), fine aggregate volume ratio(Sr) and coarse aggregate volume ratio(Gv) as design factors of the combined high flowing concrete are applied to determine the optimum mix design proportion. Also quality variations for sensitivity test are selected items as followings. (1)Surface moisture(5cases) and (2)Fineness modulus of fine aggregate(5cases), (3)Concrete temperature(3cases), (4)Specific surface(3cases) and particle size of lime stone powder. As experimental results, water-cement ratio, fine and coarse aggregate volume ratio are shown as the optimum range 51%, 43% and 53% separately considering site condition of slurry wall. Also quality factors by sensitivity test should be controlled in the following ranges. (1) Surface moisture :to.67% and (2)Fineness modulus 2.6$\pm$0.2 of fine aggregate, (3)Concrete temperature l0-20t, (4) Specific surface 6,000$\textrm{cm}^2$/g and particle size 9.7$\pm$1.0${\mu}{\textrm}{m}$ of lime stone powder. Based on the results of this study, the optimum mix design proportion of the combined high flowing concrete are selected and poured successfully in the slurry wall of LNG in-ground tank.
PURPOSES : This study evaluated the field applicability and laboratory performance of warm-mix asphalt (WMA) as an alternative technology in asphalt pavement. METHODS : The pilot road using two different types of WMA mixture and one HMA mixture was constructed in Waegwan-Seokjeok road construction site and the mixtures were sampled at the asphalt plant for laboratory testings. The field applicability was assessed in environmental aspects, such as $CO_2$ emission, and in aspects of constructibility using the existing equipment and procedure, i.e., thickness and density measurement. The laboratory testings included the moisture susceptibility test by AASHTO T283, dynamic modulus test, triaxial repeated load permanent deformation test, and the fatigue test. RESULTS : The temperatures for production and compaction of WMA were $20{\sim}30^{\circ}C$ lower than those for HMA and therefore, the noxious gas emission were significantly reduced. The field density of WMA pavements was similar or better than that of HMA pavement. From the laboratory testings, it was found that WMA mixtures exhibit comparable performance to HMA mixture in moisture susceptibility, permanent deformation, and fatigue performance. CONCLUSIONS : With these results, it would be concluded that WMA could replace the existing HMA technology without any significant issue. To support this conclusion, it is necessary to track the long-term performance of WMA in pilot road.
Four Suffolk ewes were used in Latin Square switch over design to study the effects of varying levels and sources of protein on heat production and thermoregulatory responses at daytime high ($33^{\circ}C$ temperature. They were fed Italian ryegrass hay supplemented with fishmeal and/or urea, providing three different levels of crude protein (CP) (low/unsupplemented: 7.9, medium: 11.6, and high: 15.8%) at $1.5{\times}maintenance$. Feeds were distributed at 0900 (30%) and 1700 (70%). Urea diet caused higher heat production and increased vaginal temperature compared to fishmeal and fishmeal-urea mix diets. Time spent standing, skin temperature and respiration rate of sheep fed urea were similar with those of sheep fed fishmeal. Sheep fed diet with low CP level had higher heat production, increased vaginal and skin temperature than sheep fed diet with medium CP content. Sheep on high CP diet produced significantly more heat than sheep fed medium CP diets. Their vaginal temperatures were similar with those of sheep fed medium CP diet but lower than those of sheep fed low CP diet. Respiration rates of sheep and time spent by them for standing on all diets did not differ significantly. These results suggest that urea is inferior protein supplement for thermoregulation of animal at hot environment, as it induced higher heat production than fishmeal and fishmeal-urea mix. Thermoregulatory response on fishmeal-urea mix diet was similar to fishmeal diet. Increasing CP of the diet from low to medium gives advantage for thermoregulation of animal. Increasing CP further to high level was not beneficial as it resulted in the responses of sheep similar to those on low protein diet.

The purpose of this study was to develop a new way of delivery system of chlorhexidine using self-curing acrylic resin. Different preparations of chlorhexidine, such as chlorhexidine varnish($Chlorzoin^{(R)}$) and chlorhexidine diacetate crystalline, were mixed into self-curing acrylic resin with different methods. Every resin plate was made and was immersed in 100ml of distilled water individually, and kept in an incubator at $37^{\circ}C$. Solution(0.8ml) was collected from the each container at every 24 hours, and the amount of released chlorhexidine in the solution was measured in an ultraviolet spectrophotometer at 255nm. Flexural strength of all of the resin plates in the Experiment 2-A and 2-B were measured using Instron at the end of the experimental periods. The results were as follows: 1. It was found that chlorhexidine was released from the experimental groups in the Experiment 1, 2-A, and 2-B. And the release of chlorhexidine from all of the experimental groups showed a pattern of sustained-release preparation. 2. It seemed likely that a condition of "dryness" reduced a release of chlorhexidine from the chlorhexidine varnish. 3. It may be stated that a method of "chlorhexidine diacetate mix" with the polymer be more efficient than a method of "Chlorzoin mix" with the monomer. 4. Although it was evident that a flexural strength of the acrylic resin plates be reduced by a mix of either Chlorzoin or chlorhexidine diacetate crystalline, it seemed likely that the resin plates except Group 4 and 5 in the Experiment 2-B may be usable in the clinical situation.

This experiment was carried out to select suitable organic matter in To-jik nursery (self soil nursery) for complement To-jik nursery's defects that are deterioration of raw material by poor quality of seed ginseng and reduction of the quantity in seed ginseng production. Organic matter used were Yacto, rice bean, defatted rice bran, soybean cake and their mixture. As follows, bulk density in soil physical property by treating organic composts was the greatest in soybean cake and the next was followed by mix, Yacto, defatted rice bran, and rice bran treatment in order. Soil pore space ratio was totally the opposite; that was rice bran the first and followed by defatted rice bran, Yacto, mix and soybean cake treatment. The incidence rate of damping off by treating organic composts was 1.5% in both soybean cake and mix while the others was 1.0%. Emergence time was the same among treatment on April 16 and Emergence rate was the highest at 73% in Yacto. There was no significant differences among treatment in the growth of aboveground part but it was a little better in defatted rice bran treatment. In Yacto treatment, the growth of underground part, total root number per kan, rate of first grade ginseng seedling, and rate of usable ginseng seedling etc. were entirely higher but there was little differences. Using defatted rice bran was slightly lower in productivity compared to Yacto, but the possibility was high as a alternative for Yacto in a view of managing cost down.

In order to evaluate service life of RC (Reinforced Concrete) structures exposed to chloride attack, chloride penetration analysis is required referred to the chloride diffusion coefficient from the actual mix proportions. In this work, accelerated diffusion coefficients are obtained from NT BUILD 492 and ASTM C 1202 and the related apparent diffusion coefficients are derived via the previously proposed relationship for RC structures near to sea shore. Considering the properties of the mix proportions and the most conservative analysis conditions like critical and surface chloride contents, service lifes in column and exterior wall member are evaluated through conventional program LIFE 365 ver.2. The different built-up period of 10 and 15 years has no significant effect on service life. The results from mix proportions with slag show longer than 75 years of service life with the help of higher time dependent parameter and lower initial diffusion coefficient.

In this study, the application results of high volume fly ash concrete(HVFAC) on the construction site were reported. The structures were the mat foundations of 3m and 2m thickness with design strength of 40MPa. The replacement ratio of fly ash was 50%, and the pre-mix type binder was used. As a result, it appeared that the temperature increases of concrete foundations were about $39^{\circ}$C for 3m thickness and $36^{\circ}$C for 2m thickness.
