Two Newly Recorded Estuarine Ciliates, Euplotes vannus and E. parawoodruffi (Ciliophora: Spirotrichea: Euplotida) from Korea

  • Two euplotid ciliates collected from the estuarine littorals in Korea were identified as Euplotes vannus ($M\ddot{u}ller$, 1786) and E. parawoodruffi Song and Bradbury, 1997. These species are reported taxonomically for the first time from Korea. These two species are redescribed with illustrations, photos and biometry based on live and silver impregnated specimens. Diagnostics of each species are as follows. E. vannus: size in vivo $94-111{\times}55-75{\mu}m$ (average $103{\times}60{\mu}m)$, adoral zone of membranelles (AZM) 70% of cell length with 57-74 adoral membranelles (AM) and terminating close to hook-shape, macronucleus (Ma) C-shaped with twisted foot-like, 10 frontoventral (FVC), 5 transverse (TC), 4-7 (average 5) caudal cirri (CC), 9-10 dorsal kineties (DK), mid dorsal kinety with 15-22 cilia; silver-line system single vannus type. E. parawoodruffi: size in vivo $125-163{\times}72-100{\mu}m$, (average $141{\times}87{\mu}m$), dorsally strongly arched, body shaped reserved triangular. AZM 67-83% of cell length with 60-85 AMs, 9 FVC, 5 TC, 4 CC, 9 DK; mid-dorsal kinety with 20-30 cilia, double-eurystomus type, T-shaped Ma with equal sized right and left arms or right arm shortened slightly.

Screening for Antitumor Efficacy from the Wild Plants in Korea (한국산 천연항종양성 자원의 Screening에 대하여)

  • This experiment was Conducted ta screening for the anti-cancer efficacy from the wild plants which are naturally growing in the Korea. The results are as follows. The results were shown thatZea may L. nad significantly effects on mediculal efficacy anganist anti-tumor by usulg the totalpacked cell volume methods and also, severals plants, such as Sofonum nigrum, Patrinia hispidoBunge, Eragrostis, ferrugenia Beauv, Salaginela pouzolgiana Spring, Platrycarya strobilacea Bunge,Codonopsis lanceolata Benth. et Hook fil. which are collected from Giri and Mooju mountain in Koreaand Nagano in Japan were showed effects on auti-tumor. But the pharmaceologial activities ofPharbitis nil Choisy was believed to strong effec on anti - cancer tumors, while toxicity of its wasshown high that induced te kill all used mice. Extraction of Patrinia hispida Bunge, Pharbitis nilChoisy, Toilis japonica DC, Eragrostis erruginea Beauv. and Forsythia koreana Nakai showed effec-tively supressed on growth rate of cancer tumor by the below 50 percent of T/C ratio at 30mg /mlof extraction from plant. That Is strong activity while Reynouxria japonica Houtt. was observed onlymild activities. The above results many possibly suggest that Patrinia hispido Bunge and Eragrostisferrugina Beauv. inhibited the growth of cancer tumor by the both total packed cell volume methodand cytotoxicity method. Although basic research is still going on, we will find out an accurate moth-od for developing useful medicinal plant to improve pharmacological activites against anti-cancertumor, especialy, in Eragrostis ferruginea Beauv.

Characterization of the Intestinal Corynosoma strumosum (Phylum: Acanthocephala) from the Harbor Seal, Phoca largha, from the East Sea, Korea

  • An acanthocephalan parasite, Corynosoma strumosum, was found in the intestine of harbor seal, Phoca largha, living in the East Sea, Korea. The probosci's hook and trunk spine patterns typical of the collected worms indicated it to be C. strumosum. The body lengths measured are 4.2-5.8 (4.9) mm in males and 5.3-6.8 (5.7) mm in females. The proboscis is bent ventrally, armed with 18 longitudinal rows of 9-13 hooks in males and 20 longitudinal rows of 11-13 hooks in females. The proboscis receptacle is double-walled with the robust trapezoidal neck being unarmed. The hind-trunk is pipe-shaped with posterior parallel sides. The characteristic hind trunk spines cover the anterior third of the ventral surface. The lemnisci are equal and slightly shorter than the double-walled proboscis receptacle, which is longer than the proboscis. This species is an acanthocephalan parasite reported for the first time in Korea.

Fishing investigation of vertical bottom longline fisheries in sea mount of central northern Pacific (북태평양 중부 해산어장에 있어서 저층 선주낙의 어획실태)

  • This study was conducted to survey the catches of vertical bottom longline fisheries in the sea mount of central northern Pacific($30^{\circ}-42^{\circ}N$, $170^{\circ}-175^{\circ}E$), during the period of July 1 to August 25,2004 by commercial fishing vessel. The number of 57 test fishing was carried out in the central northern Pacific during 43 days and the total catches were 21,092.4kg as 19 fish species, CPUE/day and catches/day were 185 baskets and 490.5kg, respectively. Main fish species caught from the experimental fishing were Squalus mitsukurii (66.3%), Coelorhyrchus asperocephaius (11.7%) and Helicolenus avius (9.8%) and, average inside diameter for fish mouth was 4.0cm over. Catch ratio according to each fishing ground was the order of F, D, J, B and C. Catch ratio fur water depth was the order of 450-500m, 350-400m, 300-350m, 400-450m, 1000-1100m and 500-550m and, main species by water depth was Squalus mitsukurii for 300-400m, Etmopterus lucifer for 300-550m, Coelorhyrchus asperocephaius far 1,000m over. Catch ratio according to the kind of hooks was higher at the hook no.6 for Squalus mitsukurii and no. 5 for Etmopterus lucifer and, catch ratio by baits was higher at squid for Squalus mitsukurii, saury and eel for Helicolemus avius and saury for Etmopterus lucifer. Accordingly, it is thought that the extension of fishing hours is needed with the reduction of damage and loss for fishing gears during fishing operation.

A taxonomic study on section Atratae of Carex L. in Korea (Cyperaceae) (한국산 사초속 감둥사초절의 분류학적 연구)

  • Morphological characters of section Atratae of the genus Carex (Cyperaceae) were reexamined. The epidermal patterns of perigynium, achene and leaf were investigated using by a scanning electron microscope (SEM) and a light microscope (LM). Morphological characters such as length and width of stem, leaf, bract, spike, scale, perigynium and achene, and shape of cross-sectioned stem, and leaf, spike, scale, apex of scale, perigynium, beak of perigynium and achene, and epidermal pattern of perigynium, achene and leaf (shape of fundamental epidermal cell and cell wall, type of silica body, shape of beak epidermal cell and cell wall in perigynium, subsidiary cell shape, size and frequency of stomatal complex of leaf) were useful for the identifications of the observed seven taxa. C. gmelinii, C. hancokiana and C. peiktusani have been confused each other due to similar morphological characters. Howerever these taxa were distinct with respect to shape of scale, apex of scale, perigynium, type of silica body, frequence of stomatal complex, subsidal cell shape, and epidermal patterns of achene and leaf.

Pleuronichthys sp. Fossils (Pleuronectidae) from the Duho Formation, Pohang Uhyeon-dong in Korea (포항시 우현동 두호층에서 산출된 Pleuronichthys sp. 화석)

  • Two specimens of the Cenozoic fish fossils were discovered from the Miocene Duho Formation of Uhyeon-dong, Pohang, Korea. These fossils are identified as Pleuronichthys sp. based on the following- firstly, front dorsal fin rays elongated to the upper part of neurocranium, Secondly, right sided orbit of neurocranium, Thirdly, presence of urohyal like fish-hook, Fourthly, curved sciatic part of the urohyal, Fifthly, presence of postcleithrum, Sixthly, over 27 centrum, Seventhly, elongated first pterygiophore of the anal fin rays, Eightly, c-shaped inner side of urohyal, Ninthly, small or few cardiac apophysis, and Tenthly, presence of many spots on body. These fossils of Pleuronichthys represent the first record in East-Asia. Two specimens are anatomically different in the extent of the asymmetry and the flatness of skull. This represents the unique ontogeny stage of the Pleuronectidae, because they accompany the above anatomical difference when they transform from pelagic lifestyle to benthic lifestyle.

  • This paper reports the improvements of the dialysis apparatus and operations since there are some problems to be improved in the dialysis method which is applied to determine the ionic and molecular weight of dissolved particles. The hook gauge was utilized in order to control the surface level of the dialysing solution and the solvent to be equal with the precision of 0.02 mm to minimize the osmotic and filtration effects. An accurate agitation velocity for both solutions was maintained and so was the same temperature for both solutions with ${\pm}0.01^{\circ}C$ precision. The dialysis membrane was fixed uniformly and flatly on one end of the dialysis cylinder by using a newly developed ring. The volume change of the dialysing solution during the dialysis was reduced to the range of -1.62 ~ +0.92%. Optimum duration of dialysis was searched from the relation between the osmotic and filtration effect and the pore size of the membrane.

Antibacterial potential of the extracts derived from leaves and in vitro raised calli of medicinal plants Pterocarpus marsupium Roxb., Clitoria ternatea L., and Sanseveiria cylindrica Bojer ex Hook

  • Pterocarpus marsupium, Clitoria ternatea, and Sanseveiria cylindrica are some of the important and endangered medicinal plant species of India. Despite of medicinal properties, antibacterial potential of the plants have not yet been explored. The present study was designed to optimize the in vitro technique for micropropagation and to screen the extracts from leaves and in vitro raised calli for antibacterial properties. Excised leaf-explants from the parent plants were surface sterilized and cultivated on Murashige & Skoog's (MS) medium containing $N^6$-benzyladenine (BA) in concentrations of 1, 2, 5, and $10{\mu}M$. Optimal growth of calli was noticed at a concentration of $5{\mu}M$, therefore the extracts from calli grown at this concentration were further studied for antibacterial activity. Both alcoholic and aqueous extracts from leaves of respective plants, and their in vitro raised calli were tested for antibacterial activity by agar well diffusion method against a range of Gram-positive and Gram-negative bacteria. Aqueous extracts showed antibacterial activity against limited number of bacterial species; notably the extracts of C. ternatea which showed antibacterial activity against Streptococcus pyogenes, Bacillus subtilis and Bacillus cereus. Alcoholic extracts of all three plants showed antibacterial activity against a wider range of bacteria. Among the Gram-positive bacteria, extracts from C. ternatea showed strong antibacterial activity against Bacillus spp., whereas the extracts of S. cylindrica showed good antibacterial potential for Staphylococcus aureus, S. epidermidis and S. pyogenes. The extracts from all three plants showed antibacterial activity against Gram-negative bacteria, including, Salmonella spp. and Shigella dysenteriae; organisms causing enteric fever and dysentery. In most of the cases, the extracts from respective calli showed comparable, and in some cases better, result in comparison to the extracts from parent leaves. To the best of our knowledge this is the first preliminary report on antibacterial potential, especially through calli extracts, of these plants; and in vitro cultivation of the explants may be used to obtain phytotherapeutic compounds.

Anti-Oxidant and Anti-Inflammatory Complex Health Functional Analysis of Chinese Resource Plant Boehmeria, Carlemannia and Clausena (중국 자원식물 보메리아, 칼라마니아, 클로세나의 항노화 항염 복합 건강관리 기능성 연구)

  • Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.

A Study on the Fishing Efficiency of the Jigging Gear Neon Flying Squid , Ommastrephes Bartrami in the North Pacific (북태평양 빨강오징어 채낚기의 조획성능에 관한 연구)

  • Drift gillnet fishery for neon flying squid in the North pacific was one of the major pelagic fisheries of Korea until 1992, its annual catch was 79, 000M/T as average during 1988-1992, but moratoriumed since 1993 according to the decision of UN. Therefore, for the developing of the new fishing gear for the squid, the seven types of rip hook by automatic squid jigging machine were experimented by the korean research vessel Pusan 851 (G/T 1.126, 2.600 PS) in the North Pacific (38 $^{\circ}$30'-43 $^{\circ}$N, 152 $^{\circ}$E-178 $^{\circ}$W) from July 6. 1993 to August 31. 1993. The investigation on catch rate, dropout rate, and catch condition of the rip hooks related to the fishing lamp power for aggregating the squid were carried out during the period. The results obtained are as follows: The composition of catch by automatic squid jigging machine was 83.9% for neon flying squid. 15.5% for boreopacific gonate squid. 0.6% for boreal clubhook squid, and 0.01% for luminous flying squid. The catch rate of neon flying squid was 94.6% in 13.6-18.3$^{\circ}C$ of surface water temperature and 5.4% in others. The higher catch rate of neon flying squid was made in the range 13.6-18.3$^{\circ}C$ of temperature at the surface and about 1$0^{\circ}C$ of temperature at the 100m layer. The CPUE of neon flying squid in the 13.6-18.3$^{\circ}C$ of surface water temperature was ranged 0.8-11.8kg (8.7kg as average). The mantle length and body weight of neon flying squid caught in the experiment were ranged 18.3-51.3 cm, 140-3, 980g and mean mantle length and mean body weight were 29.4cm, 972g respectively. The catch rate of neon flying squid was the highest at dawn with a value of 25.0% of the total catch. The body weight of neon flying squid caught by the D type hooks was 1.7 times more than that of the A type hooks. The dropout rate of neon flying squid caught by the seven types hooks was 7.9-57.5% (19.0% as average), and dropout rate of the D type hooks was 7.9% with 2.7 times decrease than that of the A type hooks. The catch efficiency of small sized neon flying squid in case of using on-off switch method on fishing lamp in 15 minutes intervals was 2.6 times higher than that of the on-switch method with same fishing lamp power.
