Solid Phase Extraction(SPE) of Curcuminoids from Turmeric by Optimization Analytical Condition (최적 분석조건에 의한 강황으로부터 Curcuminoids의 고체상추출(SPE))

  • Turmeric is a medicinal herb with various pharmacological activities. Curcumin, demethoxycurcumin(DMC) and bisdemethoxycurcumin(BDMC) were extracted from turmeric by dipping and ultrasonic wave method. And extraction efficiency was identified using solid phase extraction(SPE). The extracted sample were simultaneously separated and analyzed from three kinds of commercial $C_{18}$ reversed-phase high performance liquid chromatography using water and acetonitrile as mobile phase with isocratic elution mode. Flow rate 1.0mL/min, injection volume $10{\mu}L$ and column oven temperature $40^{\circ}C$ at 425nm wavelength has been conducted. From the experimental results, the optimum mobile phase composition of water/acetonitrile 50/50vol% using RS tech column. It is evident that the amount of curcuminoids extracted(extraction time 4h) by 100% MeOH was higher than any aqueous MeOH composition. Finally, in 100% water extraction, the amount(mAU${\times}$mim) of curcuminoids extracted by SPE was 14.3 and 24.5 times respectively higher than ultrasonic wave and dipping method. The shown results can be applied as sources for pharmaceuticals and functional material.

Effect of Cosurfactant on Microemulsion Formation and Cleaning Efficiency in Systems Containing Alkyl Ethoxylates Nonionic Surfactant, D-Limonene and Water (보조계면활성제 첨가가 Alkyl Ethoxylates계 비이온 계면활성제, D-limonene, 물로 이루어진 시스템에서의 마이크로에멀젼 형성 및 세정력에 미치는 효과)

  • In this study, the effect of sosurfactant on microemulsion phase behavior was investigated in ternary systems containing alkyl ethoxylates nonionic surfactant, water and d-limonene. The addition of a cosurfactant produced a microemulsion phase over a wide range of temperature and promoted formation of a microemulsion phase at lower temperatures. In particular, small amounts of n-propanol, as a cosurfactant, were found to be the most effective in extending a microemulsion phase region over a wide range of temperature. Temperature sensitivity of a nonionic surfactant system was effectively relieved by addition of the anionic surfactant sodium dodecyl sulfate. And the formation of one phase microemulsion was not affected by pH, hardness concentration and addition of an antioxidation agent. The cleaner candidates were determined from microemulsion phase behavior study, and their cleaning efficiency was tested using a dipping method. All the cleaner candidates selected during this study showed excellent removal efficiency for abietic acid over a temperature range from 30 to $40^{\circ}C$ presumably due to a decrease in interfacial tension.

  • Objectives: In homeopathy, it is claimed that more homeopathically-diluted potencies render more protective/curative effects against any disease condition. Potentized forms of Condurango are used successfully to treat digestive problems, as well as esophageal and stomach cancers. However, the comparative efficacies of Condurango 6C and 30C, one diluted below and one above Avogadro's limit (lacking original drug molecule), respectively, have not been critically analyzed for their cell-killing (apoptosis) efficacy against lung cancer cells in vitro, and signalling cascades have not been studied. Hence, the present study was undertaken. Methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays were conducted on H460-non-small-cell lung cancer (NSCLC) cells by using a succussed ethyl alcohol vehicle (placebo) as a control. Studies on cellular morphology, cell cycle regulation, generation of reactive oxygen species (ROS), changes in mitochondrial membrane potential (MMP), and DNA-damage were made, and expressions of related signaling markers were studied. The observations were done in a "blinded" manner. Results: Both Condurango 6C and 30C induced apoptosis via cell cycle arrest at subG0/G1 and altered expressions of certain apoptotic markers significantly in H460 cells. The drugs induced oxidative stress through ROS elevation and MMP depolarization at 18-24 hours. These events presumably activated a caspase-3-mediated signalling cascade, as evidenced by reverse transcriptase-polymerase chain reaction (RT-PCR), western blot and immunofluorescence studies at a late phase (48 hours) in which cells were pushed towards apoptosis. Conclusion: Condurango 30C had greater apoptotic effect than Condurango 6C as claimed in the homeopathic doctrine.

  • In this paper, an experimental technique using a reference accelerometer has been developed to measure the in-plane vibration intensity of a beam. It has the advantages of shortening measurement time and reducing accelerometer phase error comparing with the cross spectral Intensity measurement technique using an accelerometer array. Solubility and Physicochemical Stability of Caroverine Hydrochloride in Aqueous Solution (수용액중 염산카로베린의 용해성 및 안정성)

  • The solubility and physicochemical stability of caroverine hydrochloride (CRV), an antispasmodic, in buffered aqueous solutions were studied using a reverse phase high performance liquid chromatography. The solubilty of the drug at pH 2.76-5.40 was similar at the range 31.9-36.2 mg/ml $(34^{circ}C)$, but, at the pH higher than 6.0, markedly decreased. The use of polyethylene glycol 400 as a cosolvent did not increase the solubility at any compositions examined. Moreover. increasing molar concentration of aqueous phosphate buffer from 0 to 0.5 M remarkably decreased the solubility. The degradation of CRY followed the apparent first-order kinetics. The degradation was accelerated with decreasing pH and increasing storage temperature. The half-lives for the degradation of CRY (1.0 mg/ml) at pH 1.28. 4.01 and 5.93 $(45^{\circ}C)$ were 2.8, 31.4 and 124 hr. respectively. The pHs of incubated solutions were to some extent lowered perhaps due to the formation of acidic degradation products. The addition of disodium edetate (0.01%) to the CRY solution (pH 4.95) retarded 2.5 times the degradation rate at $45^{\circ}C$, but the use of sodium bisulfite (0.1%) accelerated 2.9 times the rate. The activation energy for the CRY solution (20 mg/ml. pH 5.4) containing 0.01% EDTA was calculated to be 5.98 kcal/mole. When the solution was stored under nitrogen displacement in ampoule, there was no significant degradation even after 3 months at $40^{\circ}C$, indicating that protection from oxidation by air (oxygen) is essential for the complete stabilization of CRY solution.

  • The magnetoresistance (MR) of $La_{1-\chi}S_{\chi}CoO_{3-\delta}$ films prepared by pulsed-laser deposition were investigated in order to clarify the magnetotransport properties around the metal-insulator transition. For the films in the metallic state ($\chi$ > 0.25), the MR(T) manifests a small peak at the Curie temperature due to the spin-disorder scattering. The transition of the film into the insulating state ($\chi\;\leq$ 0.25) is accompanied by an essential growth of the MR and results in a significant increase in the MR(T) with decreasing temperature, due to a phase separation into the ferromagnetic-metal clusters and the insulating matrix.

  • A neutral section is installed around feeding substation(SS) and sectioning post(SP) that M phase and T phase are isolated in AC feeding system. Electric Train under "Notch-OFF" is operated by inertia within the neutral section. It causes disturbing the operation of electric trains for speed drop and driver's mistakes.   • In this paper, we describe a UHF-band CMOS fractional-N frequency synthesizer using a ring - type VCO. It has been designed using $0.18{\m}m$ CMOS technology. First, The newly designed charge-pump circuit includes an OTA for matching between the upper current and the lower current In addition, a ring - type VCO is also used for small chip sire. The simulation results show that the designed circuit has a phase noise of -109.53dBc/Hz at 1MHz offset and consumes 19.4mA from a 1.8V supply. The lock time is less than 30usec and the chip size is $0.45mm{\times}0.5mm$.

  • Fluid cooling is one of the manufacturing processes used to control mechanical properties, and is recently used for hot stamping of automobile parts. The formed part at room temperature is heated and then cooled rapidly using various fluids in order to obtain better mechanical properties. The formed part may undergo excessive thermal deformation during rapid cooling. In order to predict the thermal deformation during fluid cooling, a coupled simulation of different fields is needed. In this study, cooling simulation of boron steel square sheet was performed. Material properties for the simulation were calculated from JMatPro, and three convection heat transfer coefficients such as water, oil and air were obtained from the experiments. It was found that the thermal deformation increased when the difference of cooling rate of sheet face increased, and the thermal deformation increased when the thickness of sheet decreased.
