  • Carbon-11 is one of the most sensitive and desirable positron emission tomography radio-isotope, which offers the capacity to be incorporated, through a covalent bond, into biologically active molecules without altering their biological properties. Carbon-11 can be obtained from the cyclotron with two different chemical forms: $[^{11}C]CO_2$ and $[^{11}C]CH_4$. [$^{11}C$]Methyl iodide has been widely used as a highly reactive labelling precursor that can be applied to label carbon-11 with biologically active molecules via alkylation of N-, O-, or S-nucleophiles. A more recent and still challenging labeling method is transition metal mediated $^{11}C$-carbonylation. Advances in organic chemistry, radiochemistry and improved automated techniques greatly encourage researchers to develop more carbon-11 labelled radiotracers for molecular imaging studies. This mini-review will introduce a historical track of carbon-11 chemistry combining with examples and its role in near future.

Beneficial Biological Activities of Conjugated Linoleic Acid (CLA의 생물학적 기능)

  • Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid with conjugated double bonds at C9,C11 and C10,C12 positions. Of possible CLA isomers, a naturally occurring CLA isomer is c9,t11-CLA which is produced from linoleic acid by linoleate isomerase from various rumen and lactic bacteria, and mushroom mycelia. Meanwhile, synthetically prepared CLA contained an equal amount of c9,t11-CLA and t10,c12-CLA isomers, and other isomers as minor constituents. CLA was firstly mentioned in 1939 during the elaidinization reaction of linoleic acid. Thereafter, CLA was not an attractant to scientists because it was not scientifically interested any more. However, since the anticarcinogenic action was driven from 7,12-dimethylbenz[a]anthracene (DMBA)-induced mouse skin carcinogenesis in 1987, CLA-related researches were drastically elevated, resulting in approximately 6,100 research papers in literature, so far. CLA exhibited the significant biological activities: anticarcinogenic, antidiabetic, antihypertensive, antiatherosclerotic, body-fat reducing, antioxidative, antiinflammatory, testosterone producing and other activities. Interestingly, two major CLA isomers, c9,t11-CLA and t10,c12-CLA, exhibited different biological activities. Meanwhile, t,t-CLA isomers which is minor constituent of chemically synthesized CLA from linoleic acid exhibited more potent anticarcinogenic activity in carcinogen-induced animal models and cancer cell lines than other CLA isomrs. In the present review, the significant biological activities of CLA were discussed along with historical studies of CLA since 1939.

Effect of Forage to Concentrate Ratio and Monensin Supplementation on cis-9, trans-11 Conjugated Linoleic Acid and trans-11 Octadecenoic Acid Concentrations of Ruminal Contents and Plasma in Sheep

  • Twenty-four cannulated Small-tailed Han${\times}$Poll Dorset wethers (BW $47.5{\pm}2.1kg$) were used to determine the effects of forage to concentrate ratio (40:60 vs. 70:30), monensin supplementation (0, 15 or 30 ppm, DM basis) and interactions of these two factors on cis-9, trans-11 conjugated linoleic acid (cis-9, trans-11 CLA) and trans-11 octadecenoic acid (trans11-$C_{18:1}$) concentrations in ruminal contents and plasma in sheep. The experiment was designed as a $2{\times}3$ factorial. The diet contained Chinese wild rye grass hay (Aneurolepidium Chinese), cracked corn, soybean meal, NaCl, limestone and trace mineral premix. Dietary crude fat and linoleic acid ($C_{18:2n-6}$) were adjusted with soybean oil to about 7.0% and 24.0 mg/g (DM basis), respectively. High forage diets increased (p<0.001) the concentrations of trans11-$C_{18:1}$ and cis-9, trans-11 CLA in ruminal contents and plasma. Monensin supplementation increased (p<0.001) the concentration of trans11-$C_{18:1}$ in ruminal contents, but had no effect on that of cis-9, trans-11 CLA. Concentrations of trans11-$C_{18:1}$ (p<0.019) and cis-9, trans-11 CLA (p<0.022) in plasma increased with dietary monensin levels. Interactions of forage: concentrate ratio and monensin level tended to affect the concentrations of trans11-$C_{18:1}$ (p<0.091) and $C_{18:2n-6}$ (p<0.083) in ruminal contents. Increasing forage levels increased the concentrations of trans11-$C_{18:1}$ and cis-9, trans-11 CLA in the rumen. Supplementing with monensin increased the ruminal production of trans11-$C_{18:1}$ and concentrations of trans11-$C_{18:1}$ and cis-9, trans-11 CLA in plasma.

Resolution of the Triacylglycerols Containing Conjugate Trienoic Acids into Their Molecular Species by HPLC in the Reversed-phase and Silver Ion Mode (Reversed-phase 및 $Ag^{+}$-HPLC에 의한 Conjugate Trienoic Acid 함유(含有) Triacylglycerol 분자종(分子種)의 상호분리(相互分離))

  • Conjugate trienoic acids (CTA) occurred in triacylglycerols (TGs) of the seed oils of Trichosanthes kirilowii, Momordica charantia and Aleurites fordii, and they were easily converted to their methyl esters in a mixture of sodium methoxide-methanol without any structural destruction. The main fatty acids in triacylglycerol (TG) fraction of the seed oils of Trichosanthes kirilowii are $C_{18:2{\omega}6}$ (32.2 mol %), $C_{18:3{\;}9c.11t,13c}$ (38.0 mol %) and $C_{18:1{\omega}9}$ (11.8 mol %), followed with $C_{16:0}$ (4.8 mol %) and $C_{18:0}$ (3.1 mol %). The TG fraction was resolved into 20 TG molecular species according to the partition number (PN) by reversed-phase (RP)-HPLC. The main TG species were $DT_{c2}$, $MDT_{c}$ and $D_{2}T_{c}$, of which amounts reached 63 mol % of total TG molecular species. The TG sample was fractionated into 11 fractions according to the number of double bond in the molecule by $Ag^{+}-HPLC$ and the species of $DT_{c2}$, $MDT_{c}$ and $D_{2}T_{c}$ were also eluted as main components. The TG species containing CTA showed unusual behaviours in the order of elution by HPLC ; first, TG moleular species of $DT_{c2}$ (D; dienoic acid, $T_{c}$; punicic acid, $T_{ci}$; ${\alpha}-eleostearic$ acid, M ; monoenoic acid, $S_{t}$; stearic acid) was eluted earlier than $Mt_{c2}$, although they have the same PN number of 40, and, secondly, the species of $DT_{ci2}$ with eight double bonds was eluted earlier than that of $D_2T_{ci}$ with seven double bonds. Intact TG of the seed oils of Momordica charantia contained mainly fatty acids such as $C_{18:3{\omega}9c,11t,13t}$ (57.7 mol %), $C_{18:1{\omega}9}$ (17.4 mol %), $C_{18:0}$ (12.3 mol %) and $C_{18:2{\omega}6}$ (10.6 mol %), and was classified into 13 fractions by RP-HPLC. The main TG species were as follows ; $MT_{ci2}$ [$(C_{18:1{\omega}9})(C_{18:3\;9c,11t,13t})_{2}$, 39.1 mol %] and $S_{t}T_{ci2}$ [$(C_{18:0})(C_{18:3\;9c,11t,13t})_2$, 33.9 mol %] comprising about 73 mol % of total TG species, accompanied by $DT_{ci2}$ [$(C_{18:2{\omega}6})(C_{18:3\;9c,11t,13t})_{2}$, 7.3 mol %], $D_{2}T_{ci}$ [$ (C_{18:2{\omega}6})_{2}(C_{18:3\;9c,11t,13t})$, 3.6 mol %] and $MDT_{ci}$ [$(C_{18:1{\omega}9})(C_{18:2{\omega}6})(C_{18:3\;9c,11t,13t})$, 3.5 mol %]. Simple TG species of $T_{ci3}$ [$(C_{18:3\;9c,11t,13t})_3]$ was present in a small amount of 1.4 mol %, but other simple TG species were not detected. The TG was also resolved into 11 fractions according to the number of double bond by $Ag^{+}-HPLC$, and the species were mainly occupied by $MT_{ci2}$ [$(C_{18:1{\omega}9})(C_{18:3\;9c,11t,13t})_{2}$, 39.4 mol %] and $S_tT-{ci2}$ [$(C_{18:0})(C_{18:3\;9c,11t,13t})_{2}$, 35.4 mol %] $DT_{ci2}$ species with eight double bonds was also developed faster than $D_2T_{ci}$ one with seven double bonds as indicated in the analysis of TG of the seed oils of T. kirilowii, and $MT_{ci2}$ species with cis, trans, trans-configurated double bond was eluted earlier than $MT_{c2}$ species with cis, trans, cis-configurated double bond. The main components of fatty acid in total TG fraction isolated from the seed oils of of Aleurites fordii were in the following order ; $C_{18:3\;9c,11t,13t}$ (81.2 mol %)> $C_{18:2{\omega}6}$ (8.5 mol %)> $C_{18:1{\omega}9}$ (5.4 mol %)$. With resolution of the TG by RP-HPLC, eight fractions such as $T_{ci3}$, $Dt_{ci2}$, $D_{2}T_{ci}$, $MT_{ci2}$, $PT_{ci2}$ (P; palmitic acid), $PMT_{ci}$, $PDT_{ci}$ and $S_{t}T_{ci2}$ ($S_{t}$; stearic acid) were isolated, respectively. TG species of $T_{ci3}$ [$(C_{18:3\;9c,11t,13t})_{3}$, 54.2 mol %], $DT_{ci2}$ [$(C_{18:2{\omega}6})(C_{18:3\;9c,11t,13t})_{2}$, 15.0 mol %] and $MT_{ci2}$ [$(C_{18:1{\omega}9})(C_{18:3 9c,11t,13t})_{2}$, 14.8 mol %] were present as main species.

Radiolabeling of 11C-sertraline by fast and easy loop method with [11C]CH3OTf

  • Cis-(1S,4S)-4-(3,4-dichlorophenyl)-1,2,3,4-tertrahydro-N-methyl-1-naphthalenamine (sertraline) hydrochloride from among selective serotonin reuptake inhibitors (SSRIs) is a treatment of major depression. For the differential diagnosis by metabolizing serotonin in a patient with neurological disorders, the radiolabeled $^{11}C$-sertraline was developed for non-invasive positron emission tomography in living brain and use the evaluation of new drug for SSRIs. We release the results of a fast and easy radiolabeling method applied a one-step loop method with $[^{11}C]CH_3OTf$ for routine clinical applications of $^{11}C$-sertraline. 1 mg of a precursor for $^{11}C$-sertraline in 0.1 mL DMF and $5{\mu}L$ of 1N NaOH, were injected into the loop of semi-prep high-performance liquid chromatography (HPLC). $[^{11}C]CH_3OTf$ was passed through the loop at room temperature (RT). The $^{11}C$-sertraline was separated by the semi-preparative HPLC. $^{11}C$-sertraline was eluted at 28.0 min was collected and evaluated by analytical HPLC and mass spectrometer. The total radiolabeling efficiency of $^{11}C$-sertraline was $30.7{\pm}8.7%$. The specific activity was $64.8{\pm}51.4GBq/{\mu}mol$. The radiochemical and chemical purities were higher than 99%. The mass spectrum of the product showed m/z peaks at 307.1 (M+1), indicating the mass of sertraline. By the one-step loop method with $[^{11}C]CH_3OTf$, $^{11}C$-sertraline could be quickly and easily prepared for clinical application.

A new purification method for the Fab and F(ab)2 fragment of 145-2C11, hamster anti-mouse CD3ε antibody

  • Recombinant protein G has been utilized in the purification of antibodies from various mammalian species based on the interaction of antibodies with protein G. The interaction between immunoglobulin and protein G may not be restricted to the Fc protion of antibodies, as many different $F(ab)_2$ or Fab fragments can also bind to protein G. I found both FAb $F(ab)_2$ of 145-2C11, a hamster anti-mouse $CD3{\varepsilon}$ antibody, bound to the protein G-sepharose. Interestingly, Fab and $F(ab)_2$ of 145-2C11 did not bind to the protein A-sepharose. The binding of Fab and $F(ab)_2$ of 145-2C11 to protein G provided a useful method to remove proteases, chopped fragments of the Fc region, and other contaminating proteins. The remaining intact antibody in the protease reaction mixture can be removed by using a protein A-sepharose, because the Fab and $F(ab)_2$ portions of 145-2C11 did not bind to protein A-sepharose. The specific binding of Fab and $F(ab)_2$ portions of 145-sC11 to a protein G-sepharose (though not to a protein A-sepharose) and binding of intact 145-2C11 to both protein A- and G-sepharose will be useful in developing an effective purification protocol for Fab and $F(ab)_2$ portions of 145-2C11.

Effect of Water Temperature on Ammonia Excretion of Juvenile Pacific Cod Gadus macrocephalus (대구 Gadus macrocephalus 치어의 암모니아 배설에 미치는 수온의 영향)

  • A study was carried out to examine the effect of water temperature on daily pattern and rate of total ammonia nitrogen (TAN) excretion in juvenile Pacific cod Gadus macrocephalus (mean body weight: $36.5{\pm}0.8\;g$) under fasting and feeding conditions. Fish were acclimated over 10 days under three different water temperatures (9, 11 and $13^{\circ}C$), and transferred to TAN measuring system under each water-temperature condition. After 72 hours of starving, fasting TAN excretion was measured at each temperature. To investigate post-prandial TAN excretion, fish were hand-fed with a commercial diet containing 40.6% crude protein for 7 days, two times daily at 08:00 and 16:00 h. Water was sampled from both the inlet and outlet of the fish chamber every 2 h over a 24-h period. Both fasting and post-prandial TAN excretion increased with increased water temperatures (p<0.05). Mean fasting TAN excretion rates at 9, 11 and $13^{\circ}C$ were 9.3, 11.0 and $11.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. The value of $9^{\circ}C$ was lower than those of 11 and $13^{\circ}C$ (p<0.05), but there was no significant difference between $11^{\circ}C$ and $13^{\circ}C$. Mean post-prandial TAN excretion rates at 9, 11 and $13^{\circ}C$ were 23.0, 31.6 and $45.4\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. A peak value of post-prandial TAN excretion rate occurred after 2 h from each feeding, and the second value is always higher than the first value. Maximum post-prandial TAN excretion rate occurred after 10 h from the first feeding at $9^{\circ}C$ (mean $38.0\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), $11^{\circ}C$ ($52.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$) and $13^{\circ}C$ ($77.5\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), respectively. The TAN loss for ingested nitrogen at $9^{\circ}C$ (43.9%) was lower than those of $11^{\circ}C$ (46.4%) and $13^{\circ}C$ (48.4%). The overall results indicate that water temperature exhibits a significant effect on the nitrogen excretion of juvenile Pacific cod.

The Phase Transition of Pb5Ge3O11 Single Crystal by the Thermal Conduction Measurement (열전도 측정에 의한 Pb5Ge3O11 단결정의 상전이)

  • The thermal diffusivity of lead germanate single crystal was measured from room temperture up to the $200^{\circ}C$ by heating method. The thermal diffusivities were measured as k=0.0117, 0.0105, and $0.0112cm^2/^{\circ}C$ at $30^{\circ}C$, $177^{\circ}C$, and $180^{\circ}C$, respectively. It is identified that the heat capacity of the lead germanate single crystal is maximum at $177^{\circ}C$, and this point is phase transition temperature of $Pb_5Ge_3O_{11}$ single crystal.

Effect of Temperature on the Quality and Storability of Cherry Tomato during Commercial Handling Condition (유통중 온도관리가 방울토마토의 품질과 저장성에 미치는 영향)

  • This study was carried out in order to investigate the effect of temperature of treatment and storage on the longevity of 'Unicorn' tomatoes of light red maturity stage during commercial handling conditions encountered while exporting over long distances. Tomato stored at $5^{\circ}C$ and $11^{\circ}C$ temperature with 85% relative humidity by pre-treating handling temperature that was using from field to before shipment as a winter temperature $5^{\circ}C$, spring temperature $11^{\circ}C$ and summer temperature $25^{\circ}C$ for 3 days. On the final storage day, $25^{\circ}C/11^{\circ}C$ (treated/stored) tomatoes showed the highest respiration and ethylene production rate; whereas the lowest respiration and ethylene production rate was found for $5^{\circ}C/5^{\circ}C$ treated and stored tomatoes. Tomatoes treated and stored at $5^{\circ}C/5^{\circ}C$ showed higher marketability, without evidence of fungal rot, decay or spots for 23 days. The fresh weight loss under all treatment conditions increased gradually during $5^{\circ}C$ and $11^{\circ}C$ storage temperatures. The higher firmness and soluble solids were determined from $5^{\circ}C/5^{\circ}C$ and $5^{\circ}C/11^{\circ}C$ treated and stored tomatoes repectively, than from others tomatoes on the final day of storage. In addition, $5^{\circ}C/5^{\circ}C$ tomatoes showed higher vitamin C contents than tomatoes stored at other temperatures, on the final day of storage. As the ripening and storage period progressed, the titratable acidity increased, but declined (P < 0.05) thereafter, due to over ripe tomatoes under all treatment conditions. These results show that $5^{\circ}C/5^{\circ}C$ treated and stored light red maturity stages of 'Unicorn' tomatoes are optimum to export because they show the highest storability and marketability. Moreover, the marketability of light red maturity stage of 'Unicorn' tomato maintained for 2 weeks in $25^{\circ}C/11^{\circ}C$ treated and stored temperature that might be the export temperature from Korea to Japan in summer season. This research result could be useful in helping tomato growers and exporters to get optimum market value by satisfying the buyer and consumer with a fresher product.
