Prediction of Fatigue life of Composite Laminates using Micromechanics of Failure (미시역학적 파손이론을 이용한 복합재 적층판의 피로수명 예측)

  • Many tests are required to predict the fatigue life of composite laminates made of various materials and having different layup sequences. Aiming at reducing the number of tests, a methodology was presented in this paper to predict fatigue life of composite laminates based on fatigue life prediction of constituents, i.e. the fiber, matrix and interface, using micromechanics of failure. For matrix, the equivalent stress model which is generally used for isotropic materials was employed to take care of multi-axial fatigue loading. For fiber, a maximum stress model considering only stress along fiber direction was used. The critical plane model was introduced for the interface of the fiber and matrix, but fatigue life prediction was ignored for the interface since the interface fatigue strength was presumed high enough. The modified Goodman equation was utilized to take into account the mean stress effect. To check the validity of the theory, the fatigue life of three different GFRP laminates, UDT[$90^{\circ}2$], BX[${\pm}45^{\circ}$]S and TX[$0^{\circ}/{\pm}45^{\circ}$]S was examined experimentally. The comparison between predictions and test measurements showed good agreement.

Gossypol Induces Apoptosis of Human Pancreatic Cancer Cells via CHOP/Endoplasmic Reticulum Stress Signaling Pathway

  • Gossypol, a natural phenolic aldehyde present in cotton plants, was originally used as a means of contraception, but is currently being studied for its anti-proliferative and anti-metastatic effects on various cancers. However, the intracellular mechanism of action regarding the effects of gossypol on pancreatic cancer cells remains unclear. Here, we investigated the anti-cancer effects of gossypol on human pancreatic cancer cells (BxPC-3 and MIA PaCa-2). Cell counting kit-8 assays, annexin V/propidium iodide staining assays, and transmission electron microscopy showed that gossypol induced apoptotic cell death and apoptotic body formation in both cell lines. RNA sequencing analysis also showed that gossypol increased the mRNA levels of CCAAT/enhancer-binding protein homologous protein (CHOP) and activating transcription factor 3 (ATF3) in pancreatic cancer cell lines. In addition, gossypol facilitated the cleavage of caspase-3 via protein kinase RNA-like ER kinase (PERK), CHOP, and Bax/Bcl-2 upregulation in both cells, whereas the upregulation of ATF was limited to BxPC-3 cells. Finally, a three-dimensional culture experiment confirmed the successful suppression of cancer cell spheroids via gossypol treatment. Taken together, our data suggest that gossypol may trigger apoptosis in pancreatic cancer cells via the PERK-CHOP signaling pathway. These findings propose a promising therapeutic approach to pancreatic cancer treatment using gossypol.

Application of Decision Trees for Prediction of Sugar Content and Productivity using Soil Properties for Actinidia arguta 'Autumn Sense'

  • Environmental conditions are important in increasing the fruit sugar content and productivity of the new cultivar Autumn Sense of Actinidia arguta. We analyzed various soil properties at experimental sites in South Korea. A Pearson's correlation analysis was performed between the soil properties and sugar content or productivity of Autumn Sense. Further, a decision tree was used to determine the optimal soil conditions. The difference in the fruit size, sugar content, and productivity of Autumn Sense across sites was significant, confirming the effects of soil properties. The decision tree analysis showed that a soil C/N ratio of over 11.49 predicted a sugar content of more than 7°Bx at harvest time, and soil electrical capacity below 131.83 µS/cm predicted productivity more than 50 kg/vine at harvest time. Our results present the soil conditions required to increase the sugar content or productivity of Autumn Sense, a new A. arguta cultivar in South Korea.

Breeding of 'Seeberry' F1 Hybrid Strawberry (F1 종자 딸기 '씨베리' 육성)

  • 'Seeberry' is a short-day $F_1$ hybrid strawberry for seed propagation, that was released by the Protected Horticulture Experiment Station (RDA, Republic of Korea) in 2013. Inbred lines derived from domestic germplasms and selected varieties were used to develop this cultivar. 'Seeberry' originated from a cross of two inbred lines: a male parent 'Wongyo 3115' with high firmness and a female parent 'Wongyo 3116' with excellent fruit shape and high yield. 'Seeberry' cultivar should be grown with a crown diameter of over 10 mm to reveal its unique characteristics. 'Seeberry' has an upright plant shape with elliptic leaves, 12-15 flowers per cluster, vigorless growth habit, and average flower bud differentiation compared to vegetatively propagated cultivars. Fruits of 'Seeberry' are conical, having light skin color and approximately 15-16 g in average weight. Yield of this cultivar is about 92% lower than that of 'Akihime', but 'Seeberry' has excellent taste, sugar content of $9.7^{\circ}Bx$, acidity of 0.6%, and good texture. With regard to disease and pest resistance, 'Seeberry' is sensitive to powdery mildew, aphids, and the two-spotted spider mite, and is resistant to anthracnose.

Physicochemical Properties of Dried Burdock (Arctium lappa L.) Powder in the Peeling Process (박피 유무에 따른 우엉 분말의 이화학적 특성)

  • This study evaluated the qualitative properties of burdock (Arctium lappa L) peels as food material. Proximate composition, sweetness, pH, total acidity, mineral, fatty acid, free sugar, and organic acid contents of preprocessed burdock were measured. Crude ash, protein, fat, and total dietary fiber contents of non-peeled treated lotus roots were higher than those of peeled treated lotus roots (p<0.05). $^{\circ}Bx$, pH, and total acidity were not different in the peeling process. Redness and yellowness of lotus roots were reduced by the peeling process, whereas lightness increased (p<0.05). The mineral contents of Ca, K, Fe, and P were higher in non-peeled lotus roots, whereas non-peeled treated lotus root showed higher K contents (p<0.05). Maltose content was not significantly different, whereas fructose, sucrose and maltose contents were significantly different. Total organic acid contents were higher in non-peeled lotus roots (p<0.01). However total free amino acid contents were higher in peeled lotus roots (p<0.01).

Expression and Production of Human Granulocyte Colony Stimulating Factor (G-CSF) in Silkworm Cell Line (누에세포를 이용한 인간 G-CSF의 발현 및 생산)

  • Granulocyte colony stimulating factor (G-CSF) is a hematopoietic cytokine that stimulates bone marrow cells to proliferate and differentiate into granulocytes. G-CSF is approved and used for therapeutic purposes. The endoplasmic reticulum (ER) signal peptide of hG-CSF was replaced with silkworm-specific signal peptides to express and efficiently secrete recombinant hG-CSF by silkworm cells. Plasmids that contain cDNAs for hG-CSF and hG-CSF fused with silkworm- specific signal peptides of prophenoloxidase activating enzyme (PPAE), protein disulfide isomerase (PDI), and bombyxin (BX) were constructed. The G-CSF protein was expressed in insect cell line BM5 and was detected by western blot analysis. The cells transfected with plasmids containing rhG-CSF genes with silkworm-specific signal sequences released mature rhG-CSF protein more efficiently than the cells transfected with pG-CSF, the plasmid containing human G-CSF gene, including its own signal sequence. The production of hG-CSF reached maximal level at four days post-transfection and remained at a high level until 7 days post-transfection. These data demonstrate that the modification of the human G-CSF mimic to insect proteins synthesized in ER greatly improves the production of the protein.

Development of Bulgogi Sauce added with the Concentrate of Jujube Flesh Left around Seed (대추씨 주변 과육 농축물을 활용한 불고기 소스 개발)

  • This study was carried out to develop concentrate of residual jujube flesh around the seed as a thickening agent of Bulgogi sauce. Quality characteristics and sensory test of Bulgogi sauces added with jujube concentrate (30%, 35%, 40%, 45%, 50%) were evaluated in comparison with control sauce containing xanthan gum as a thickening agent. Moisture content of Bulgogi sauce added with 30% jujube concentrate was the highest (74.59%). As content of jujube concentrate increased, moisture contents in Bulgogi sauces significantly decreased. Sugar content of control sauce ($30.82^{\circ}Bx$), was lowest among the sauces. As content of jujube concentrate increased, Sugar content of sauces significantly increased ($33.00{\sim}34.38^{\circ}Bx$). Salinity (%) was highest in control sauce (1.38%), and there was no difference in salinity among the sauces depending on the amount of jujube concentrate. Viscosity was highest in sauce added with 50% jujube concentrate (43.33 cP) and lowest in sauce with 30% jujube concentrate (13.20 cP). As content of jujube concentrate increased, viscosity tended to increase significantly. Spreadability was highest in sauce added with 30% jujube concentrate (8.88 cm). As the amount of jujube concentrate increased, spreadability of sauce tended to decrease significantly. Color value was different according to the contents of jujube concentrate in sauces. L-, a-, and b-values of control sauce were highest at 40.19, 1.5, and 24.57, respectively. The results of the sensory test for Bulgogi sauce showed that overall preference was significantly highest in sauce added with 35% jujube concentrate. Moreover, sauce added with 35% jujube concentrate was preferred the most in terms of appearance, taste, and fluidity. Meanwhile, the results of the differentiation test showed a significant difference in color intensity, jujube flavor and taste, sweet taste, and salty taste. However, no significant difference was observed in fluidity. The results of the preference test for Bulgogi seasoned with sauces added with jujube concentrate showed that overall preference and flavor, taste, and texture preference were highest in Bulgogi seasoned with sauce added with 35% jujube concentrate. However, no significant difference was observed in preference towards appearance among the samples.

Physicochemical Properties and Biological Activities of Angelica gigas Fermented by Saccharomyces cerevisiae (유용 효모균주를 이용한 발효참당귀분말 추출물의 이화학적 특성 및 생리활성 효과)

  • The purpose of this study was to investigate the biological activities of an aqueous extract of Angelica gigas (Ag) fermented by Saccharomyces cerevisiae (Sc). First, the soluble solids of the F/3 group, in which the Ag was fermented by Sc for 3 days, decreased from $1^{\circ}Bx$ to $0.9^{\circ}Bx$. On the other hand, the pH increased with the number of days of fermentation. The result of a TLC experiment confirmed that it gradually decomposed into a low-molecular weight sugar form upon fermentation. The total phenolic compounds and flavonoid contents were higher in the fermented group than in the non-fermented group. K and Ca contents were increased by fermentation in the following order: F/3, NF, and F/0 groups. Decursin and decursinol angelate contents were highest in the F/3 group. The DPPH (${\alpha}$, ${\alpha}{\prime}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging activity of the NF, F/0, and F/3 groups were 41.89%, 39.51%, and 60.26%, respectively. The inhibition activities of tyrosinase and lipoxygenase were stronger in the F/3 group than in the NF group. This experiment showed that the fermentation of Ag Nakai can lead to an increase in its antioxidant ability, physiological activity, whitening and anti-inflammatory effects. Thus, this oriental herbal medicine can be developed into a functional material that can be utilized in the development of cosmetic products in future.

The Effect of Vigor of a Bearing Shoot in ‘Bluecrop’ Highbush Blueberry (Vaccinium corymbosum L.) on Growth Characteristics of Shoots and Fruits (하이부시 블루베리 ‘블루크롭’의 결과지 세력이 신초와 과실 생장에 미치는 영향)

  • The aim of this study was to elucidate the influence of vigor of a bearing shoot in ‘Bluecrop’ Highbush Blueberry (Vaccinium corymbosum L.) on growth characteristics of shoots and fruits. Bearing shoots were classified with BS (bearing shoot) and BMB (bearing mother branch). The vigor of bearing shoots were divided into four arbitrary categories; A was thin (< 6.0 ㎜) BMB and short (< 10 ㎝) BS, B was thin BMB and long (≥ 10 ㎝) BS, C was thick (≥ 6 ㎜) BMB and short BS and D was thick BMB and long BS. Shoots from D were longer (6.5 ㎝) and thicker (1.70 ㎜) than those from the others. Shoots of D had more leaves (5.8 ea) than those of the others. Leaf area of D was larger (13.5 ㎠) than those of the others. The first harvest of D was one week faster than the others. Ratio of big berry (>14 ㎜) from the long BSs was higher (B : 41.7, D : 46.8%) than that from the short BSs. Soluble solid content of small berrys did not show any different according to vigor of bearing shoots, but soluble solid content of big berrys of the long BSs was higher (B : 16.2, D : 15.6°Bx) than those of the short BSs. The thickness of BMB did not affect ratio of fruit size and soluble soild content. The long BSs would be proper than the short BSs for bearing bigger fruits.

Development of a Rapid Detection Method for Pectobacterium carotovorum subsp. carotovorum Using the Loop-Mediated Isothermal Amplification (LAMP) (Loop-Mediated Isothermal Amplification (LAMP)법을 이용한 Pectobacterium carotovorum subsp. carotovorum의 신속 진단법 개발)

  • Pectobacterium carotovorum subsp. carotovorum is the causative agent of soft rot in crops such as potato and cabbages. Loop-mediated isothermal amplification (LAMP) is a simple DNA amplification method, as well as isothermal PCR technique. In this study, a new method for the rapid detection of Pectobacterium carotovorum subsp. carotovorum was developed using LAMP that named PCC-LAMP. Based on lytic murein transglycolase gene of Pectobacterium carotovorum subsp. carotovorum, a set of four primers for LAMP was designed. The optimal PCC-LAMP reaction temperature was established at $61^{\circ}C$. Under standard conditions, PCC-LAMP amplified $1{\times}10^3$ copies of clone PCC-pBX437 per reaction. Further, this method can also assay directly by SYBR Green I without electrophoresis. Amplification was not detected for five other bacterial species. In conclusion, PCC-LAMP may be a useful method for the detection Pectobacterium carotovorum subsp. carotovorum in the field.
