Multiple Shoot Induction and Bulb Mass Proliferation System by in Vitro Immature Spathe Culture of Elephant Garlic (Allium ampeloprasum L.) (코끼리마늘(Allium ampeloprasum L.)의 기내 미숙총포 배양을 통한 다신초유도와 종구대량증식 시스템)

  • This study was performed to develop the mass propagation system using tissue culture technique to supply the seeds of Elephant garlic (Allium ampeloprasum L.) which has difficulty in propagation. Immature spathe of Elephant garlic was cultured on Murashige & Skoog (MS) medium supplemented with two plant growth regulators, naphthaleneacetic acid (NAA) and kinetin. After 6 weeks of culture, the highest number of shoot (14.9/explant) was obtained when the immature spathe with 10 cm length was cultured right after harvesting. In MS medium supplemented with 2 mg/L kinetin and 0.5 mg/L NAA, the most vigorous growth characteristics was observed, the shoot number was 14.9/explant, its length was 11.3 cm, and its fresh weight was 2.5 g. When the bulblets were cultured in MS medium with 2 mg/L kinetin and 0.5 mg/L NAA, the addition of 30 mg/L adenine improved their proliferation and growth significantly, the highest bulblet formation rate (48%) was obtained. The addition of 7% sucrose also increased the bulblet formation rate at the highest frequency of 98.2%. The shoots were shown be more vigorously proliferated at the secondary subculture stage rather than primary culture stage, their propagation rate was 80% after subculture.

Large-scale Culture of Plant Cell and Tissue by Bioreactor System

  • Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

  • Regulation of organ differentiation by growth regulators was investigated through the shoot-tip and bulb-scale cultures of Lilium longiflorum (cv Georgia). When shoot tips were placed on MS medium supplemented with 0.1 mg/L NAA alone or 0.1 mg/L NAA and 0.1 mg/L BA, axillary shoots were proliferated. Root diffentiation and growth were stimulated on the basal medium. Although growth regulation did not seem to be necessary when bulb scales were used as explants for shoot differentiation, its differentiation was promoted vigorously by 0.2 mg/L NAA, but suppressed by BA. Bulblets were formed from bulb-scale-derived plantlets cultured on MS medium supplemented with 0.1 mg/L IBA. And more bulblets were formed from the plantlet in MS medium supplying 0.2 mg/L NAA with 6% than3% sucrose

  • The present study was carried out to determine the effects of lighting directions and light intensities on the growth of in vitro plantlets of strawberry, potato and lily. The growth of plantlets was affected by two different lighting systems, downward and sideward lighting system. There were no difference for the plantlets of strawberry regardless of lighting directions. However, leaf area, fresh weight and dry weight have increased as the light intensity increased. Also, plant height and root length on node culture of potato decreased at high intensity of the sideward lighting, while thickness of stem and root was thicker, fresh weight and dry weight were heavier, and leaf area was increased. Also, bulblet formation on scale culture of lily has been abundant, and fresh weight and dry weight were increased. Thus, the sideward lighting system which loaded three stories for culture vessels with raising light intensity into culture vessels instead of conventional downward lighting system promoted growth and raised the efficiency of production of high quality microplant.

Plant Regeneration from Leaf and Root Cultures of Lycoris chejuensis via Bulblet Formation (제주상사화 (Lycoris chejuensis K. Tae et S. Ko) 잎 및 뿌리 절편으로부터 소자구 형성을 통한 식물체 재생안)

  • Plant regeneration system from leaf and root segments of Lycoris chejuensis via bulblet formation was established. Surface-sterilized leaf and root segments were cultured on the B5 medium containing 2,4-D. After 12 weeks of culture onto B5 medium containing 2,4-D, white globular structures and white calluses were formed on the cut surface of the explants. The highest frequency of globular structures and calluses formation from leaf explants was 32.1% when leaf explants were cultured onto B5 medium supplemented with 1 mg/L of 2,4-D. However, the higher concentration of 2,4-D (over than 3 mg/L) resulted in decrease of the frequency. In comparison to leaf explants, root segments showed the highest frequency at a rate of 36.1% when root explants were cultured onto B5 medium supplemented with 3 mg/L of 2,4-D. These structures and calluses were sub-cultured and proliferated onto the same culture medium. Upon transfer to B5 basal medium, white globular structures were developed into bulblets and normal plantlets. After 4 weeks of incubation in the light, plantlets were successfully rooted over the frequency of approximately 90%. Rooted plantlets were successfully transferred to potting soil and acclimatized in the growth chamber. The plant regeneration system of Lycoris chejuensis established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.

Bulblet Regeneration through the Callus Culture induced from Bulb Scales of Lillium longiflorum‘Gelria’. (나리‘Gelria’의 기내인편에서 유도된 callus 배양을 통한 자구의 재분화)

  • This study was conducted to establish a regeneration system of plantlets through callus culture induced from bulb scales of Lillium‘Gelria’. Friable callus was induced very easily from bulb scales, and grew vigorously on medium lacking growth regulators. In media with 0.5∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA, 100% of explants produced callus. Proliferation of callus was actively occurred on media containing 0.1 ∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA. Callus proliferation and regeneration of bulblets from callus were occurred simultaneously. Light condition was more effective for the callus proliferation and solid medium was better than liquid medium. Althrough callus was proliferated vigorously on media containing 0.1 ∼ 1.0 mg/L BA and NAA, the frequncy of plantlet regeneration was better on medium without growth regulators, then on medium with 0.1 mg/L BA and NAA.

  • The purpose of this study was carried out to investigate the effects of plant growth regulators on in vitro propagation of Liliem cernum Komarov. Small bulblets were poliferated from callus explants after 2 weeks and leaf, root and bulb were formed after 4 weeks culture. Leaf differentiation was promoted vigorously by the combination of TDZ 0.1 mg/L and NAA 0.01 mg/L(87.5%). The rate of root differentiation was the greatest at BA 0.2 mg/L alone(81.8%). The rate of callus formation was the high in medium containing TDZ. The number of bulblets and leaves formed in bulb scales was the greatest at TDZ 0.1 mg/L(5.7). Also, the longest length of total length, leaf and root length were in Zeatin 1.0 mg/L + NAA 0.1 mg/L(10.5 cm). However the longest bulblet was in TDZ 0.1 mg/L(1.4 cm).

Effect of Bulblets Size Oriented from Tissue Culture on Growth and Bulb Enlargement of Lilium Oriental Hybrids Grown in Highlands

  • This study was conducted to investigate the effect of bulblet size on growth and bulb enlargement of Lilium Oriental hybrids, such as 'Sorbonne', 'Siberia', and 'Casa Blanca', grown in areas of high altitude (780 m), in 2009. The bulblets were oriented from tissue culture and planted to produce virus-free bulblets. All the three cultivars showed high vitality, with more than 93% producing shoots, even from small bulblets with a bulb circumference (BC) of less than 6 cm. Bulblets with BC 9-10 cm showed an increased height and number of leaves rapidly to induce the phase conversation from vegetative growth to reproductive growth. The bulblets of BC 9-10 cm in 'Sorbonne', 'Siberia', and 'Casa Blanca' had 2.0, 2.9, and 2.8 flower bud sets per plant, respectively. However, the flowers from those bulblets were not adequate as standard bulbs to produce cut-flowers for the domestic market. The larger bulblets showed more prosperous characteristics in growth and development of Lilium Oriental hybrids. Small bulblets had a high occurrence of viruses and leaf blight symptoms during cultivation, indicating the aggravated disease symptoms in the previously infected bulbs. 'Sorbonne' cultivars showed a high rate of enlargement of bulbs, and small bulblets under BC 6 cm produced more than 23% of the standard bulbs of BC 14-16 cm, which could produce cut-flowers for export. However, 'Casa Blanca' bulblets with BC 6-9 cm produced low bulbs of BC 14-16 cm at 21% level. 'Casa Blanca' cultivars had more bulb roots, longer root lengths, and higher weights than 'Sorbonne' and 'Siberia' cultivars. 'Casa Blanca' cultivars with BC over 9-10 cm grew rapidly and produced a high quantity of bulbs. In these results, 'Sorbonne' bulblets of BC 9-10, 'Siberia' bulblets of BC 10-12 cm, and 'Casa Blanca' bulblets of BC 10-12 cm produced 89%, 86%, and 93% of the standard bulbs of BC 14-16 cm, respectively. It is recommended that bulblets larger than the above sizes be used to produce cut-flowers for the export market. Experiment results suggest that production of bulbs larger than BC 18 cm requires bulblets that are larger than BC 12-14 cm of the three cultivars for the highest quality cut-flowers.

  • Since garlics (Allium sativum L.) are propagated through cloves, infection by virus or other pathogens may become severe problem if not using high quality seed bulbs every year resulting in the reduction of yield and bulb quality, In order to solve this problem, the establishment of virus-free bulb production and its supply system have been required because no chemicals were found to eliminate viruses from seed bulbs. This experiment was conducted to develop an effective production technique of high quality seed bulbs using shoot-tip culture. Over 90% of shoot-tips explanted on January L 1990 were survived at constant temperature of either 20, 24 or 28$^{\circ}C$, wheres 88% at alternate temperature (28/20$^{\circ}C$). The growth of shoot and root was most vigorous at constant 24$^{\circ}C$, and least at alternate temperature (28/20$^{\circ}C$) condition. When shoot-tips were explanted June 21 to August 1,1991, survival and growth of shoot-tips was most vigorous on MS medium supplymented with 0.1 mg/L NAA and 2 mg/L kinetin and least 1 mg/L Gh$_3$. The shoot-tips taken from the seed bulbs stored at 4$^{\circ}C$ for 15 to 60 days were placed on MS medium, shoot growth and in vitro bulblet formation increased slightly as affected by the increase of told treatment period at 4$^{\circ}C$.

  • This study was conducted to select an excellent goods and to estabilish the plant density of Amorphohpallus konjac. K about 3 local varieties and 2 introduced varieties through each fertilizer level. 1. Plant height and leaf width were increased according to increasing fertilizer level, and tuber yield was the highest at N-P$_2$ $O_{5}$ -K$_2$ O (14-10-14kg) per 10a as 390kg in Geumsan. 2. The more tuber size was large, the more yields per plant due to increasing the plant height and leaf width at low densities, while yields per 10a was decreased. 3. Among 5 varieties used, Jechon cultivar was good as a propagating variety because of increasing number of tuber and bulblet and Japan cultivar was high tuber yield per 10a.
