Cell Cycle Regulation in the Budding Yeast

  • Cell cycle is regulated cooperatively by several genes. The dynamic regulatory mechanism of protein interaction network of cell cycle will be presented taking the budding yeast as a sample system. Based on the mathematical model developed by Chen et at. (MBC, 11,369), at first, the dynamic role of the feedback loops is investigated. Secondly, using a bifurcation diagram, dynamic analysis of the cell cycle regulation is illustrated. The bifurcation diagram is a kind of ‘dynamic road map’ with stable and unstable solutions. On the map, a stable solution denotes a ‘road’ attracting the state and an unstable solution ‘a repelling road’ The ‘START’ transition, the initiation of the cell cycle, occurs at the point where the dynamic road changes from a fixed point to an oscillatory solution. The 'FINISH' transition, the completion of a cell cycle, is returning back to the initial state. The bifurcation analysis for the mutants could be used uncovering the role of proteins in the cell cycle regulation network.

Effects of the Myosin ATPase Inhibitor, 2,3-Butanedione-2-Monoxime, on Growth and Dimorphic Switches of Candida albicans

  • Dimorphic yeast Candida albicans reversibly switches between the form of yeast and hyphae depending on external conditions. We investigated possible roles of the myosin family in the growth and dimorphic switches of C. albicans with a general myosin ATPase inhibitor, 2,3-butanedione-2-monoxime (BDM). Transition to hyphae as well as proliferation by budding was completely inhibited by BDM at 16 mM. Presence of 16 mM BDM did not affect hyphae-to-bud transition but it blocked budding. The effects of BDM on yeast growth and dimorphic switches were reversible. More than 70% of the BDM-treated cells demonstrated defects in the amount and the polarized localization of F-actin as well as in the shape and migration of the nucleus, suggesting that myosin activities are needed in these cellular processes of C. albicans.

Roles of Budding Yeast Hrr25 in Recombination and Sporulation

  • Hrr25, a casein kinase $1{\delta}/{\varepsilon}$ homolog in budding yeast, is essential to set up mono-orientation of sister kinetochores during meiosis. Hrr25 kinase activity coordinates sister chromatid cohesion via cohesin phosphorylation. Here, we investigated the prophase role of Hrr25 using the auxin-inducible degron system and by ectopic expression of Hrr25 during yeast meiosis. Hrr25 mediates nuclear division in meiosis I but does not affect DNA replication. We also found that initiation of meiotic double-strand breaks as well as joint molecule formation were normal in HRR25-deficient cells. Thus, Hrr25 is essential for termination of meiotic division but not homologous recombination.

Bifurcation analysis of budding yeast cell cycle

  • Bifurcation analysis of cell cycle regulation in the budding yeast is performed basedon the mathematical model by Chen et al [Molecular biology of cell, 11:369-391, 2000]. On the bifurcation diagram, locations of both stable and unstable solutions of the nonlinear differential equations are presented by taking the mass of cell as a controlparameter. Based on the bifurcation diagram, dynamic mechanism underlying the 'start' transition, initiation of a new round of cell cycle, and the 'finish' transition, completion of cell cycle and returning back to the initial state, is discussed: the 'start' transition is a transition from a stable fixed solution for a small mass and to an oscillatory state for a large mass, and the 'finish' transition is a switching back to the stable fixed solution from the oscillatory state. To understand the role of the genes during the cell cycle regulation, bifurcation diagrams for the mutants are compared with that of the wild type.

  • 뽕잎 두께가 비교적 두껍고 수량성이 높은 후엽녹의 생장점에 Colchicine용액 적하처리를 하여 창성한 4배성인 4원뽕 28호의 주요 특성은 다음과 같다. 1. 체세포의 염색체수는 2n=56인 4배체이다. 2. 봄철 발아개엽기는 중생뽕이고 잎모양은 삼각형에 가까운 타원형으로 폭광형이고 잎두께는 매우 두께우며 잎면적당 염록소 함량이 많다. 3.가지길이는 비교적 짧고 가지수도 다소 적으며 가지굵기는 굵은 편이고 기부불발아장비율이 높고 내동성은 약하다.

  • Abnormal phenotypes resulting from haploinsufficiency (HI) are due to the loss of one allele. Recent studies in budding yeast have shown that HI originates from insufficient protein levels or from a stoichiometric imbalance between subunits of protein complexes. In humans, however, HI often involves transcription factors. Therefore, the species differences in HI and the molecular mechanisms of species-specific HI remain under investigation. In this study, HI in fission yeast was systematically surveyed. HI in fission yeast affected genes related to signaling and to basic cellular processes, as observed in budding yeast. These results suggest that there are species differences in HI and that the HI that occurs in fission yeast is intermediate to HI in budding yeast and humans.

  • 선체구조 부재에는 이중저의 거더 및 늑판등에서 유공을 가진 판이 많이 사용되고 있고, 이는 중량 경감, 사람 및 화물의 이동, 배관 등의 목적으로, 보통은 강도상 큰 문제가 없는 부위에 위치하지만, 때로는 불가피하게 높은 응력이 작용하는 부위에 설치해야 한 경우도 있다. 이러한 판에 유공의 존재는 면내 하중에 의한 탄성좌굴강도 및 최종강도에 큰 영향을 주게 된다. 따라서, 유공판의 탄성좌굴강도 및 최종강도 평가는 선박의 초기 구조설계단계에서 구조부재 치수를 결정할 때 검토해야 할 중요한 설계기준 중의 한가지 이다. 그러므로, 유공판에 대한 합리적인 신뢰적인 탄성좌굴강도 및 최종강도 설계식이 필요시 되고 있다. 본 연구에서는 다양한 종횡비와 유공의 치수비 그리고 세장비의 영향을 고려하여 탄소성대변형 유한요소법을 근간으로 한 해석코드인 ANSYS를 사용하여 수치계산을 수행하였다.
