Strategic Use of QTL Mapping to Improve the Palatability of Rice

  • The properties of starch play an important role in determining the palatability of rice. In addition, the gelatinization temperature (GT) of rice starch is an important factor in determining the quality of rice because it is related to the cooking time and texture of rice. For the development of high-quality rice, it is important to understand the genetic basis of palatability-related traits, and QTL analysis is an effective method to explain the genetic basis of variation in complex traits. QTL mapping related to alkali digestion value (ADV) of brown and milled rice was performed using the 120 Cheongcheong/Nagdong double haploid (CNDH) line. As a result, 12 QTLs related to ADV were detected, and 20 candidate genes were selected from the RM588-RM1163 region of chromosome 6 through screening by gene function analysis. The comparison of the relative expression level of candidate genes showed that OsSS1q6 is highly expressed in CNDH lines with high ADV in both brown rice and milled rice. In addition, OsSS1q6 has high homology with starch synthase 1 protein, and interact with various starch biosynthesis-related proteins, such as GBSSII, SBE, and APL. Therefore, we suggest that OsSS1q6 identified through QTL mapping could be one of the various genes involved in the GT of rice by regulating starch biosynthesis. This study can be used as basic data for breeding high-quality rice and provides a new genetic resource that can increase the palatability of rice.

Nutritional and Agronomic Characteristics of Super-Giant Embryo Mutant in Rice (벼 초거대배 돌연변이계통의 영양 및 작물학적 특성)

  • Super-giant-embryo mutant line (Hwacheong-ge$^s$) with high nutritional quality was developed from Hwacheongbyeo, a Korean Japonica cultivar. Grain weight of brown rice of Hwacheong-ge$^s$ line was lighter than of Hwacheongbyeo. However, embryo dry weight of the line was 3.22 times heavier than that of Hwacheongbyeo on the single grain base. Hwacheong-ge$^s$ line showed higher protein(8.99%) than the original variety(7.39%), without changes in storage protein. In amino acid composition, lysine content was greatly increased in the mutant, while the contents of methionine, serine and tyrosine were slightly decreased as compared with the original one. Lipid content of Hwacheong-ge$^s$ line was 1.5 times higher compared with that of Hwacheongbyeo. The contents of Vitamin Bl, B2, and E(${\alpha}$-tocopherol) were also greatly increased in Hwacheong-ge$^s$line. The grain yields of the mutant lines ranged from 69.2% to 78.8% compared with that of Hwacheongbyeo, 556kg /10a. Most of the mutant lines of M4 generation were homogeneous for growth characters, and some of them seemed to be readily applicable for farmer's field.

Comparison of Carcass Characteristics, Meat Quality, and Sensory Quality Characteristics of Male Laying Hens, Meat-Type Chickens under Identical Rearing Conditions (동일 사육 조건에서 산란계 수평아리 및 육용계의 도체 특성, 계육 품질 및 관능적 특성 비교)

  • This study was conducted to evaluate the potential of using laying hens as meat type chickens. Male broiler (Ross 308, R3), laying hens (Hy-Line Brown, HL), and Korean native chickens (Hanhyup-3, H3) were used, and 100 heads of each were prepared. Carcass characteristics, meat quality, and sensory quality characteristics were compared as analysis items. The rearing environment and feed for all treatments were identical to the broiler rearing manual, and the lighting system was maintained at 23L:1D. Feed and water were provided ad libitum. The test ended when the average weight of each treatment group reached 1.5 kg, and individuals of similar weight were randomly selected and compared. As a result of this study, the live weight of the selected individuals was approximately 1.5 kg, which was similar for all treatments (P>0.05). However, carcass weight and ratio and breast meat production were highest in R3, while HL had higher ratios of legs, wings, and neck (P<0.05). The H3 group showed high pH and WHC levels and low cooking loss, and R3 improved chicken meat color (P<0.05). In particular, the fat content in meat was lowest in HL (P<0.01). Nucleic acid substances ATP, Hx, ADP, AMP, and INO were abundant in R3, and IMP content was highest in HL (P<0.05). In sensory evaluation, all treatments showed similar characteristics and overall preferences (P>0.05). Based on the findings, it appears that HL, a male laying hen, produces meat with unique characteristics such as low fat content and high IMP content.

Comparison of Principle Components and Internal Quality of Eggs by Age of Laying Hens and Weight Standard (산란 주령 및 중량 규격에 따른 계란의 주요 성분 및 내부 품질 비교)

  • The objective of this study was to compare the principle components and internal quality of eggs by age of laying hens and weight standard. The eggs (2,140 in total) used were from Hy-Line Brown reared from 18 to 63 weeks of ages and all were higher than appearance grade B by animal grading standard of Korea. Eggs were investigated their principle components, internal quality index, and their relationship. The weights of whole egg, shell, yolk, and albumen were increased while albumen height and Haugh unit were decreased by age of laying hens. When the composition of egg was investigated after sorting by ages, the ratio of egg yolk increased while that of albumen decreased. Egg shell composition was not shown significant difference by age. Age of laying hens had positive correlation with the weights of whole egg, shell, yolk, and albumen while albumen height and Haugh unit showed negative correlation. Also, the albumen height and Haugh unit were decreased by increase of weight of whole egg or principle components of egg.

Dietary Effects of Herbal Mixture on the Laying Performance, Blood Parameters and Immune Response in Laying Hens (사료 내 생약물질의 첨가 급여가 산란계의 생산성, 혈액성상 및 면역반응에 미치는 영향)

  • The objective of this experiment was to investigate the effect of dietary supplementation of herbal medication (HM) on laying performance, blood parameter, and immune response in laying hens. A total of 800 Hy-Line Brown laying hens of 60 weeks of age were randomly allotted to one of four dietary treatments with four replicates per treatment. Three additional diets were prepared by adding 0.5, 1.0 or 1.5 % of HM to the basal diet. Feeding trial lasted 7 weeks under 16L:8D lighting regimen. The diet and water were available ad libitum. Hen-day egg production in groups fed diets with 0.5% and 1.5% HM was significantly (P<0.05) higher than that of control. Feed conversion ratio in group fed diet with 0.5% HM showed the lowest value (P<0.05). Egg weight, feed intake, broken and shell-less egg production were not affected HM supplementation levels. Egg yolk color was significantly greater in all HM supplemented groups than basal treatment. However, eggshell strength, eggshell thickness, eggshell color, and Haugh units were not affected by HM supplementation level. There were no differences in leukocyte counts among the treatments. Plamsa IgM concentration was (P<0.05) higher in all HM treated groups. In conclusion, dietary HM improved egg production, and IgM of laying hens.

  • This study was designed to investigate the effects of dietary isoflavones(IF) on bone ash content in young chicks. Daidzein(DE) and genistein(GE), aglycone forms of IF, were added to purified-type basal diet containing calcium at 50% of NRC requirement. The two IF at two different levels(1.0 and 2.0 mM) were added to the diets prepared to be very low in IF using soy protein concentrate(SPC) as the only source of protein. The diets Including control with no IF-added were fed for three weeks. One hundred, Hy-Line Brown, day-old male chicks were divided into five dietary groups with four replicates and five chicks per replication. General performances of the chicks were not affected by the dietary treatments. Serum alkaline phosphatase(ALP) activities, an indicator for bone formation, of the birds fed DE diet were lowered compared to those fed GE and control diets. However, the levels of IF did not show any differences in the responses. Bone ash o/o of birds fed the IF diets were significantly increased compared to that of control group (p〈0.05), and, however, not affected by levels of IF in diets. Bone breaking strength measured with an Instron appeared to be equivalent among the birds of all treatments. The observations that equivalences in bone ash o/o and bone breaking strength even with lowered activity of ALP by DE ingestion compared to GE suggest a necessity of further investigation.

계란의 저장기간과 저장온도 및 암탉의 수정이 계란의 품질에 미치는 영향

  • A total of 1.200 eggs obtained from 312-day-old Hy-line Brown layer breeder hens and 319-day-old Hy-line Brown commercial layer hens (600 eggs obtained from each ones) were used to investigate the effects of storage period, storage temperature, and insemination of hens on the change of albumen height, Haugh unit (HU), albumen pH, shell strength, and yolk color. Eggs were stored up to 14 days after lay at $3\;^{\circ}C\;or\;10\;^{\circ}C$ and sampled one day after stored and then 24 hours interval. Longer periods of storage resulted in lower albumen height and HU at both storage temperatures, but in higher albumen pH. The eggs stored at $3\;^{\circ}C$ were generally higher in HU and lower in albumen pH than the ones stored at $10\;^{\circ}C$. There was no statistically difference although the eggs obtained from the non-inseminated-hens were slightly higher in albumen height and HU than the eggs obtained from the inseminated-hens. Whereas, the eggs obtained from the non-inseminated-hens in the albumen pH of eggs stored at $3\;^{\circ}C$ was significantly (P<0.05) higher than the ones obtained from the inseminated-hens, but the albumen pH of eggs stored at $10\;^{\circ}C$ did not differ each other. The mean shell strength of the eggs obtained from the inseminated-hens was significantly (P<0.05) stronger than that of the eggs obtained from the non-inseminated-hens at both storage temperatures. Albumen height and albumen pH were negatively correlated(P<0.01~0.001) in both inseminated and non-inseminated-hen's egg groups. The degree of yolk colors were not significantly changed overall of the experimental periods in both storage temperatures. The study suggests that the change of egg freshness such as albumen height and HU are relatively more associated with storage period and storage temperature than insemination or non-insemination of hens.

Effect of Dietary Acid Hydrolysates of Cocoon on Performance, Egg Quality, Blood Immune Substance and Serum Cholesterol in Laying Hens (산란계 사료 내 누에고치 산가수분해물 첨가 시 산란율, 계란 품질, 혈중 면역 물질 및 혈청 콜레스테롤 함량에 미치는 영향)

  • The study was conducted to evaluate the effects of dietary acid hydrolysates of cocoon on laying performance, egg quality, blood level of immune substance and cholesterol in laying hens. The animals used in the experiment were a total of 240 Hy-Line Brown laying hens (41 weeks old). Dietary treatments included 1) CON (basal diet), 2) SP1 (basal diet + acid hydrolysates of cocoon 0.05%), 3) SP2 (basal diet + acid hydrolysates of cocoon 0.1%) and 4) SP3(basal diet + acid hydrolysates of cocoon 0.2%) twelve laying hens were allotted to a block (pen) with five replicated. Through the whole period of experiment, egg production, egg weight, egg shell breaking strength and egg shell thickness were no significant difference. However, acid hydrolysates of cocoon treatments (SP1, SP2, SP3) significantly increase their egg yolk height and Haugh unit compared to control (P<0.05). IgG and WBC (white blood cell) contents in blood were higher in SP1, SP2, SP3 treatment compared to control (P<0.05). However, albumin and total protein content in blood were no significant difference. Phospholipid, HDL-cholesterol, LDL-cholesterol and total cholesterol contents in serum were higher in SP1, SP2, SP3 treatment compared to control (P<0.05). In conclusion, supplement dietary acid hydrolysates of cocoon improved egg yolk height, Haugh unit, and blood level of phospholipid, HDL-cholesterol, LDL-cholesterol, total cholesterol in laying hen.

Effect of Stocking Density on the Feather Condition, Egg Quality, Blood Parameters and Corticosterone Concentration of Laying Hens in Conventional Cage (케이지 사육시 사육밀도가 산란계의 깃털손상도, 계란품질, 혈액성상 및 스트레스 호르몬 농도에 미치는 영향)

  • The purpose of this study was to evaluate the effects of stocking density on feather condition, egg quality, blood parameters, and corticosterone levels of laying hens reared in conventional cages. A total of 500, 28 weeks-old Hy-Line Brown laying hens were evenly assigned to two stocking densities (750 and 500 ㎠/bird). There were 50 replicates of each treatment, each consisting of 750 ㎠/birds (n=6) or 500 ㎠/bird (n=4) birds in a cage, respectively. We observed an increase in the laying period of, the group that was assigned 500 ㎠/bird, however, this group experienced significant feather damage when compared with the group allotted 750 ㎠ bird. There was no effect of stocking densities on egg quality. These results suggest that the 500 ㎠/bird stocking density in the conventional cage can negatively affect feather condition and cause the birds to experience more stress than the group allotted 750 ㎠/bird.

Inhibitory Activity of Pigmented Rice Bran Extract to the Allergic Inflammation in Basophilic Cell Line and Peritoneal Mast Cells (호염구세포주와 복강 비만세포에서 유색미 겨 추출물의 알레르기 염증 억제활성)

  • The effects of the extracts from the bran part of pigmented rices on inflammation was evaluated by determining their inhibitory action on the histamine and ${\beta}-hexosaminidase$ release, together with inflammatory cytokine productions ($IL-1{\beta},\;TNF-{\alpha}$ and IL-6). Examination of the inhibitory effects on the histamine and ${\beta}-hexosaminidase$ release from a basophilic cell line RBL-2H3 cells and rat peritoneal mast cells (RPMC) showed that the pigmented rice extract inhibited these inflammation-mediating substances (10.19% and 110.03% inhibition in histamine and ${\beta}-hexosaminidase$ release, respectively), while normal brown rice extract rather increased their release. For RPMC, the pigmented rice extract was found to have 8 or 3-fold stronger inhibitory activity than normal brown rice toward histamine or ${\beta}-hexosaminidase$ release, respectively. Expression of $IL-1{\beta},\;TNF-{\alpha}$ and IL-6 was measured as the representative inflammatory cytokine species showed that the pigmented rice extract had a higher inhibitory activity than the normal rice counterpart. ELISA analysis for determining cytokine release demonstrated a more effective blockading ability of the pigmented rice to the release of $IL-{\beta},\;TNF-{\alpha}$ and IL-6 compared to normal rice. These results showed us the superiority of the pigmented rice bran extract not only in suppressing the release of inflammation-mediating substances such as histamine and ${\beta}-hexosaminidase$, but also in repression of the inflammatory cytokine expression.
