Effects of Seaweeds on Matrix Metalloproteinases Derived from Normal Human Dermal Fibroblasts and Human Fibrosarcoma Cells (사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 대한 해조류의 효능)

  • In recent years novel potential pharmocological candidates have been looked for in animal, seaweed, sponge, fungi and marine bacteria resources. In this study, matrix metalloproteinases (MMPs) that play an important role in metastasis, arthritis, chronic inflammation and wrinkle formation were used as target enzymes to screen therapeutic agents. The inhibitory effects of several marine algae including green algae (5 species), red algae (18 species) and brown algae (4 species) methanolic extracts on MMPs were investigated in human dermal fibroblasts and human fibrosarcoma cell line (HT1080 cells) using gelatin zymography. In human dermal fibroblasts, the inhibition of MMP-2 was observed in Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate and Sinkoraena lancifolia of red algae. In contrast, MMP-2 activation was enhanced in Enteromorpha compressa and E. linza of green algae, and Peltaronia bighamiae and Sargassum thunbergii of brown algae. In human fibrosarcoma cells, MMP-9 activation was decreased in the presence of S. thunbergii of brown algae, Polysiphonia japonica in red algae and E. compressa and E. linza of green algae. The interesting finding is that E. compressa and E. linza of green algae, and S. thunbergii of brown algae exhibited a positive effect on MMP-2 in normal cells, but a negative effect on MMP-9 in cancer cell lines. These results suggest that E. compressa and E. linza of green algae, and S. thunbergii of brown algae contain potential therapeutic ingredients for cancer treatment.

Control of Rattus norvegicus on Uninhabitable Islands - Case of Sasudo Island - (무인도에서 집쥐 개체군의 포획과 제어 - 제주 사수도의 사례 -)

  • Brown rats (Rattus norvegicusBerkenhout, 1769) were eradicated from Sasudo Island (33°55'13.04" N, 126°38'19.98" E), an uninhabited island designated as Natural Monument No. 333 and the largest breeding site for the streaked shearwater (Calonectris leucomelasTemminck, 1835) in South Korea. Twelve eradication studies were conducted from November 2015 to February 2021. The survey was conducted using a line census method that entailed slow wandering throughout the island and identifying starting and returning points. Capture traps were installed around traces of Rattus norvegicus,such as excrement and identified holes. As a result, 2 to 6 individuals were captured at each instance, except for the last time, when no individuals were captured. It is hypothesized that brown rats arrived at Sasudo Island via vessels arriving at the island for leisure and fishing. After the damage to streaked shearwater caused by brown rats was confirmed in 2006, entry to Sasudo Island was strictly prohibited through security measures, and marine clean-up programs that began in 2013 and continuous capturing since 2015 have been successful in eradicating brown rats. To maintain and manage the condition in Sasudo Island, preservation and management measures, such as strict visitor control, are necessary to prevent the inflow of new brown rats in the future.

Breeding of Asiatic Hybrid Lily 'Redsky' with Orange Red Color Petals for Cut Flowers (절화용 주황색 아시아틱나리 '레드스카이' 품종 육성)

  • A new Asiatic hybrid lily 'Redsky' cultivar was selected from a cross between 'Vivaldi' cultivar with pale pink color flower as mother plant and 'Prato' with orange colored flower in 2000 year. The selected bulb line was propagated and cultivated for line nomenclature as 'Gonong-F-na-10'. The survey of the growth and flower characteristics of 'Gonong-F-na-10' were conducted from 2005 to 2007 on summer cropping system under structure in Highland Agriculture Research Center, NICS, RDA. After all 'Gonong-F-na-10' was named as 'Redsky'. The flowers of the 'Redsky' cultivar had not soft hair and fragrance, and had orange red (O-R N30D) colored petals with dark brown spots in center of flower. The stem color was dark pink and brown caused by anthocyanin pigment, the stigma and pollen colors were brown. Flowering direction was upward. The flowering date was July 7 so early flowering. The plant height was 97.2 cm so long, number of flowers was 4.9 ea, and length of flower bud was 9.3 cm. The bulb weight was about 34.8 g and its bulb size was about 14.8 cm in circumference. 'Redsky' cultivar was registered as a new variety with No. 3119 in Korea Seed and Variety Service on June of 2010.

Changes in pathogenic characters of Pseudomonas tolaasii 6264 strain by storage period (저장 기간에 따른 Pseudomonas tolaasii 6264 균주의 병원 특성 변화)

  • Pseudomonas tolaasii strain No. 6264 has been isolated from mushroom tissue and identified as one of the major pathogen causing brown blotch disease. It secretes peptide toxins, known as tolaasin and its analogue peptides. P. tolaasii 6264 has been used as a typical pathogenic strain to study the brown blotch disease for last 20 years after confirming its blotch-forming ability, hemolytic activity, and white line formation. In this study, the characteristics of P. tolaasii 6264 strain were analyzed and compared according to storage period. Strains of P. tolaasii 6264 stored annually since 2012 were cultured and their pathogenic characters were analyzed. When the 16S rRNA sequences were compared, all strains were divided into two groups. Pathogenic characters including hemolytic activity, blotch-forming ability, and white line test were also investigated. The strains, P. tolaasii 6264-15-2 and P. tolaasii 6264-17, had all three activities; however, the rest of stored strains showed only blotch-forming ability losing other pathogenic characters. Tolaasin peptides were purified from the bacterial cultures and analyzed by mass spectrometry. The strains, P. tolaasii 6264-15-2 and P. tolaasii 6264-17, secreted Tol I (1987 Da), Tol II (1943 Da), and its analogues (1973 Da, 2005 Da) while some of these peptides were not found in the media cultured other strains. These results indicate that the pathogenicity of P. tolaasii could be varied during the storage period.

Time Trends in Estimates of Genetic Parameters in a Population of Layer Breeders (난용종계 집단에서의 선발에 의한 유전모수 변화 양상)

  • This study was carried out to investigate the time-trends of genetic parameters of the dosed flock population which selected for improving egg production. Data for two layer pure lines, Line-W (Single Comb White Leghorn) and Line-B (brown layer) which have been maintained at the Mani Breeding Farm were collected from 1980 to 1985 during 5 generations. The effective number of parents per generation ranged from 148 to 366 in Line-W and 85 to 355 in Line-B, and the cumulative expected inbreeding coefficients during 5 generations of selection were 15% and 1.6%. So inbreeding could not be considered a critical factor on estimating the genetic parameters, heritabilities and genetic correlations Heritabilities of EN 300 and EN 400, primary two selected traits were significantly decreased during 5 generations but the estimates of the other 03its not showed the consistent decreasing pattern significantly. No time trends of probable consequence were evident in the genetic correlation coefficients of the traits studied. The reason for that situation was attributed to the fact that selection was conducted for multiple objectives and the relative importance of selection for the studied traits were not consistent by generations.

The change of amino acid content in fruit-body of winter mushroom according to the storage period and strains (팽이버섯의 저장기간 및 품종별 자실체내의 아미노산 함량 변화)

  • Quality change of winter mushroom were investigated during storage and distribution phase as influenced by storage temperature. According to storage period and temperature, amino acids, were analysed and quantified with the mushroom fruiting bodies using HPLC and morphological characteristics were investigated. Characteristic changes of winter mushroom fruiting bodies were described as follows during storage according to the storage temperature. Looking at the results of the analyzed amino acid contents, temperature between $-1^{\circ}C$ and $4^{\circ}C$ was optimal condition for the storage. At $4^{\circ}C$, the chemical composition tended to be maintained. On the other hand, the results indicate the rapid loss of nutrition at $-1^{\circ}C$ within 7 days of storage. Exceptionally, proline was shown to be increased. Brown line mushroom had a larger loss than white line mushroom. Based on this result, brown line mushroom have shown significant differences among varieties. Therefore, winter mushroom should be stored at $4^{\circ}C$ to minimize nutrient loss and to maintain freshness and mushrooms should be consumed within 14 days after harvest.

Association of single-nucleotide polymorphisms in dual specificity phosphatase 8 and insulin-like growth factor 2 genes with inosine-5'-monophosphate, inosine, and hypoxanthine contents in chickens

  • Objective: This study aimed to identify the single-nucleotide polymorphisms (SNPs) in the dual-specificity phosphatase 8 (DUSP8) and insulin-like growth factor 2 (IGF2) genes and to explore their effects on inosine-5'-monophosphate (IMP), inosine, and hypoxanthine contents in Korean native chicken -red-brown line (KNC-R Line). Methods: A total sample of 284 (males, n = 127; females n = 157) and 230 (males, n = 106; females, n = 124) aged of 10 weeks old KNC-R line was used for genotyping of DUSP8 and IGF2 genes, respectively. One SNP (rs313443014 C>T) in DUSP8 gene and two SNPs (rs315806609A/G and rs313810945T/C) in IGF2 gene were used for genotyping by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and KASP methods, respectively. The Two-way analysis of variance of the R program was used to associate DUSP8 and IGF2 genotypes with nucleotide contents in KNC-R chickens. Results: The DUSP8 (rs313443014 C>T) was polymorphic in KNC-R line and showed three genotypes: CC, CT, and TT. The IGF2 gene (rs315806609A/G and rs313810945T/C) was also polymorphic and had three genotypes per SNP, including GG, AG, and AA for the SNP rs315806609A/G and genotypes: CC, CT, and TT for the SNP rs313810945T/C. Association resulted into a strong significant association (p<0.01) with IMP, inosine, and hypoxanthine. Moreover, the significant effect of sex (p<0.05) on nucleotide content was also observed. Conclusion: The SNPs in the DUSP8 and IGF2 genes might be used as genetic markers in the selection and production of chickens with highly flavored meat.

Taxonomic Review of the Genus Platycephalus (Platycephalidae) from Korea (한국산 양태속 어류의 분류학적 검토)

  • Two sibling species of the genus Platycephalus collected from the South and West Sea of Korea were reviewed based on the several meristic and morphological characters, electrophoresis of muscle protein, and LDH and MDH isozyme. Counts and morphometric characters of Platycephalus indicus were different from those of Platycephalus sp. by having 18~20 pectoral fin rays (16~19 in Platycephalus sp.), 70~80 scales of lateral line (83~100), 10.2~13.1% eye diameter (10.9~15.5%), 14.5~18.1% interorbital width (11.8~17.8%), dark brown of body color (light brown) and dark brown of pectoral and ventral fin outside (light brown and pale). The number of soluble protein bands in muscle separated by polyacrylamidegel electrophoresis was 16 and 18 from P. indicus and Platycephalus sp, respectively, and each bands showed their characteristics in activated degree. The number of bands separated from LDH isozyme were 2 and 1 in P. indicus and Platycephalus sp. and in the case of MDH isozyme, 5 and 4 bands, respectively. So we were thought that P. indicus and Platycephalus sp. from Korea were different species each other.

Whitening Effects of Angelica dahurica Radix Ethanol Extract (백지 에탄올추출물의 미백효능 연구)

  • To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ${\mu}g/m{\ell}$ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/$cm^2$ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450~500g) and then the test agent of $30{\mu}{\ell}$ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ${\mu}g/m{\ell}$, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.001) than the vehicle control group and this resultsagreed with the results of observation under microscope. From these results, weconcluded that ADEE had positive whitening effect.

Improvement of Memory by Dieckol and Phlorofucofuroeckol in Ethanol-Treated Mice: Possible Involvement of the Inhibition of Acetylcholinesterase

  • Phlorotannins, the polyphonic compounds found in brown Eisenia and Ecklonia algae, have several pharmacologically beneficial effects such as anti-inflammation. In addition, our recent data show that these compounds may improve the cognitive functions of aged humans suggesting the potential ability to enhance memory in several neurodegenerative disorders. To examine the experimental hypothesis that two effective components of Ecklonia cava, dieckol and phlorofucofuroeckol (PFF), have memory-enhancing abilities, both were administered orally to mice before a passive avoidance test. The repeated administration of either dieckol or PFF dose-dependently reduced the inhibition of latency by the administration of ethanol. To investigate the mode of memory-enhancing actions, the levels of major central neurotransmitters in three different regions (striatum, hippocampus, and frontal cortex) of the mouse brain were measured. The levels of some of the neurotransmitters were significantly changed by ethanol. Both dieckol and PFF altered the levels of some neurotransmitters modified by the ethanol treatment. It is noteworthy that both dieckol and PFF increased the level of acetylcho-line, and they exerted anticholinesterase activities. Overall, the memory-enhancing abilities of dieckol and PFF may result from, at least in part, the increment of the brain level of acetylcho-line by inhibiting acetylcholinesterase.
