Anti-inflammatory and Cytotoxic Activities of Phenolic Compounds from Broussonetia kazinoki

  • The phytochemical investigation of Broussonetia kazinoki roots led to the isolation of ten compounds, including six flavonoids (1-6), two lignans (7 and 8), and two coumarins (9 and 10) by comparing their 1H and 13C NMR spectra with reference values. To the best of our knowledge, compounds 9 and 10 were isolated from this plant for the first time. Among the ten isolates, compounds 2, 4, and 6 exhibited inhibitory effects against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in macrophage RAW264.7 cells with IC50 values of 11.98, 10.16, and 24.06 μM, respectively. Furthermore, compounds 2, 4, and 6 reduced LPS-induced inducible nitric oxide synthase (iNOS) expression in a dose-dependent manner. Pre-incubation of cells with these compounds also significantly suppressed LPS-induced COX-2 protein expression. Compounds 2, 4, and 6 also showed cytotoxic activity against HL-60 cells with IC50 values ranging between 46.43 and 94.06 μM.

The Leaves of Broussonetia kazinoki Siebold Inhibit Atopic Dermatitis-Like Response on Mite Allergen-Treated Nc/Nga Mice

  • Broussonetia kazinoki Siebold. (B. kazinoki) has long been used in the manufacture of paper in Asian countries. Although B. kazinoki leaves (BK) have been employed in dermatological therapy, use of BK has not been tested in patients with atopic dermatitis (AD). Using Nc/Nga mice, which are genetically predisposed to develop AD-like skin lesions, we confirmed the efficacy of BK in AD treatment. BK extract was applied topically to Dermatophagoides farinae-induced AD-like lesions in Nc/Nga mice, and the effects were assessed both clinically and by measuring skin thickness on the back and ears. We measured the effects of BK extract on plasma levels of IgE and IL-4. We also measured the ability of BK extract to inhibit the secretion of hTARC in HaCaT cells after stimulation by TNF-${\alpha}$ and IFN-${\gamma}$. We found that BK extract significantly reduced ear and dorsal skin thickness and the clinical signs of AD, as well as significantly down-regulating the plasma levels of IgE and IL-4 (p<0.01 for each comparison). Moreover, $500{\mu}g/mL$ of BK extract inhibited hTARC secretion in HaCaT cells by activated TNF-${\alpha}$/IFN-${\gamma}$ by about 87%. These findings suggest that topical application of BK extract has excellent potential in the treatment of AD.

Nondestructive Bending Strength Evaluation of Woodceramics Made from Woody Part of Broussonetia kazinoki Sieb. -Effect of Carbonization Temperature- (닥나무의 목질부로 만든 우드세라믹의 비파괴휨강도평가 -소성온도의 영향-)

  • Nondestructive evaluation (NDE) technique method using a resonance frequency mode was carried out for woodceramics made by different carbonizing temperature (600, 800, 1000, $1200^{\circ}C$) for Broussonetia kazinoki Sieb. Dynamic modulus of elasticity increased with increasing carbonizing temperature. There was a close relationship of dynamic modulus of elasticity and static bending modulus of elasticity to MOR. Therefore, the dynamic modulus of elasticity using resonance frequency mode is useful as a nondestructive evaluation method for predicting the MOR of woodceramics made by different carbonizing temperature for B. kazinoki Sieb.

Effect of the powder from fruit of paper mulberry (Broussonetia Kazinoki Siebold) on tenderness and palatability of cooked ground beef (닥나무 열매(楮實子)분말이 마쇄육의 연화와 맛에 미치는 효과)

  • This investigation was undertaken for the purpose of studying the effect of the addition of the powder(0, 1, 2, 3%)from fruit of paper mulberry(Broussonetia Kazinoki SieboId) on the hardness, cohesiveness, elasticity, gumminess, chewiness, cooking loss and sensory characteristics of cooked ground beef with or without soysauce. The results obtained were summerized as follows; Hardness, gumminess, chewiness, and cooking loss were decreased by increasing the percent of adding the powder from the fruit of paper mulberry. As a result of sensory evaluation for cooked ground beef, sample with powder from the fruit of paper mulberry showed the higher scores in most of the characteristics. And cooked ground beef added the powder from the fruit of paper mulberry with soysauce showed the higher decreasing rate of hardness than those of without soysauce.

  • Bast fibers of paper mulberry ( Broussonetia Kazinoki Sieb) were cooked with a weak alkaline salt, potassium carbonate which has been known as a major inorganic component of the traditional lye, and its cooking characteristics were investigated . The bast fiber was easily cooked by potassium carbonate. The pulp yield was rapidly decreased up to 20-30 mol of potassium carbonate, but the Kappa number was slowly decreased with increasing of potassium carbonate. The potassium carbonate pulps were easily defibered at low cooking chemical charge of 25mm on and high pulp yield of about 80%. These results were confirmed that pectin was easily removed during the potassium carbonate cooking. In contrast, when sodium carbonate was used as a cooling agent, the bast fiber was only partially defibered. Thus, sodium carbonate was a less effective cooling chemical of the bast fiber. The results of this experiment indicated that potassium carbonate could not only be used as a good cooling agent of bast fiber, but also as an alternative agent of sodium hydroxide.

Antioxidative Activity and Component Analysis of Broussonetia kazinoki SIEB Extracts (닥나무 추출물의 항산화 활성 및 성분 분석)

  • In this study, the antioxidative activities and component analysis of Broussonetia kazinoki SIEB (B. kazinoki). extracts were investigated. B. kazinoki extract showed the effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=8.53{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the ethyl acetate fraction of B. kazinoki. extracts in the luminol-dependent $Fe^{3+}-EDTA/H_2O_2$ system was $1.69{\mu}g/mL$. The ethyl acetate fraction of B. kazinoki. extracts also exhibited more prominent cellular protective effects (${\tau}_{50}$, 183.3 min at $10{\mu}g/mL$) than that of typical antioxidant $\alpha$-tocopherol (${\tau}_{50}$ = 38.00 min) in the $^1O_2$-induced photohemolysis of human erythrocytes. Components of the ethyl acetate fraction obtained from B. kazinoki extracts were analyzed by TLC, HPLC chromatogram, LC/ESI-MS/MS and $^1H$-NMR. Consequently, Components, components were identified as the kazinol J of kazinol series and luteolin (2-(3,4-dihydroxyphenyl)-5,7-dihydroxy-4-chromenone) of flavonoid series having antioxidant activities. These results indicate that extract/ fraction of B. kazinoki can be used as antioxidants in biological systems, particularly skins exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS. Thus, the extract/fraction of B. kazinoki could be applicable to new cosmeceuticals.

Broussonetia kazinoki Siebold stimulates immune response in ovalbumin-immunized mice

  • Objective: To evaluate the immune-stimulatory potential of extracts of Broussonetia kazinoki Siebold (BK) on specific cellular and humoral immune responses in ovalbumin (OVA)-immunized mice. Material and Methods: C57BL/6 mice were immunized intraperitoneally with OVA/alum ($100{\mu}g/200{\mu}g$) on days 1, 8, and 15. BK (100, 300 or 1000 mg/kg) was given to mice orally for 21 days (from day 1 to day 21). At day 22, OVA-, lipopolysaccharide (LPS)- and concanavalin A (Con A)-stimulated splenocyte proliferation and OVA-specific and total antibodies were measured in plasma. Further, the effects of BK on expression of cytokine mRNA in OVA-immunized mice splenocytes were evaluated by RT-PCR analysis. Results: BK significantly enhanced OVA-, LPS-, and Con A-induced splenocyte proliferation in OVA-immunized mice (p<0.01). BK also significantly enhanced total IgM and OVA-specific IgG1 levels in plasma compared with the OVA control group. Moreover, BK up-regulated significantly the expression of mRNA level of IL-2 and IFN-${\gamma}$ in splenocytes. Conclusions: BK has immune-stimulating activity in an OVA-immunized mouse model system, enhancing the Th1 immune response. BK showed no cytotoxicity in this system, suggesting that BK may be a safe and effective adjuvant in humans.

Properties of Woodceramics Made from Broussonetia kazinoki Sieb. -Effect of Carbonization Temperature- (닥나무로 제조된 우드세라믹의 성질 -탄화온도의 영향-)

  • This study was carried out to investigate the properties of woodceramics made from woody part of Broussonetia Kazinoki at different carbonizing temperatures of $600^{\circ}C$, $800^{\circ}C$, $1000^{\circ}C$and $1200^{\circ}C$. The physical and mechanical properties increased with increasing carbonizing temperature. The highest mean values of density, bending strength, Brinell hardness and compressive strength were $0.62g/cm^3$, $79kgf/cm^2$, $203kgf/cm^2$ and $129kgf/cm^2$, respectively. There were close correlations between density and bending strength, Brinell hardness and compressive strength and MOE and MOR.

Germination Characteristics in Seeds of Broussonetia kazinoki Siebold ex Siebold & Zucc (Moraceae) Native to East Asia (동아시아 특산식물 애기닥나무 Broussonetia kazinoki Siebold ex Siebold & Zucc (Moreaceae) 종자의 발아특성)

  • This research was conducted to develop a seed propagation protocol for the use of Broussonetia kazinoki Siebold ex Siebold & Zucc, which native to East Asia. Light and temperature conditions, GA3 treatment, and low-temperature stratification were performed to evaluate the germination characteristics. The germination percentage at 4, 15/6, 20/10 and 25/15℃ was 0, 42. 5, 44. 4 and 91.7%, respectively. Thus, the optimal germination temperature of the B. kazinoki seeds was 25/15℃. Although germination was delayed by relatively low temperature conditions, GA3 treatment allowed for increased germination rates even under the low temperature conditions. Cold stratification reduced time to germination. Relatively low germination percentages at 20/10℃ and 15/6℃ indicated that the seeds had been under the state of conditional dormancy. Through this study, B. kazinoki seeds have conditional dormancy because they can be germinated through GA3 treatment even under relatively low temperature conditions and it is judged that it will support the commercial propagation practice in the future.

Inhibitory effects of Broussonetia kazinoki twig extract on allergic inflammatory reactions in TNF-𝛼/IFN-𝛾-stimulated HaCaT and IgE-sensitized RBL-2H3 cells (TNF-𝛼/IFN-𝛾로 자극된 HaCaT 및 IgE로 감작된 RBL-2H3 세포에서 닥나무 가지 추출물의 알러지 염증반응 억제 효과)

  • Broussonetia kazinoki twig extract (BKT) is recognized for its antioxidant and anti-cancer effects and natural whitening properties. So, it is used as a raw material for cosmetics. B. kazinoki twig is also an edible raw material. B. kazinoki has been used in Asia for paper production and oriental medicine, has anti-diabetic effects, and contains various flavonoids and alkaloids. In this study, to evaluate the efficacy of BKT on allergic skin inflammatory responses, we investigated its effects on factors related to skin inflammation in HaCaT keratinocytes and allergic responses in RBL-2H3 cells. There was no cytotoxicity of the 70% ethanol extract against HaCaT and RBL-2H3 cells. In HaCaT cells, stimulation with tumor necrosis factor-alpha (TNF-𝛼) and interferon-gamma (IFN-𝛾) increased the production of several chemokines, including thymus and activation-regulated chemokine (TARC), macrophage-derived chemokine (MDC), and regulated on activation, normal T cell expressed and secreted (RANTES). However, it was observed that this elevation was notably mitigated in a concentration-dependent manner upon treatment with BKT. Furthermore, BKT treatment demonstrated a significant reduction of 𝛽-hexosaminidase and inflammatory cytokines TNF-𝛼 and IL-4 in IgE-sensitized RBL-2H3 cells. Thus, it is expected that BKT can be used as a natural cosmetic and food ingredient that effectively suppresses allergic inflammatory reactions.
