The Effects of the Breeding site of the Egrets and Herons by Road Construction, Palhyun-ri, Daegu, Korea (대구광역시 팔현리 지역 도로공사가 백로류 번식지에 미치는 영향)

  • This survey was conducted to analyze the effect of road construction near the group breeding places of the egrets and herons located at Palhyun-ri, Daegu, on their breeding during four years from 2003 to 2006. With the number of the individuals of the whole egrets and herons have been decreased due to the three-year road construction work(2003-2005). When the road construction was temporally stopped in 2006, the number of individuals of egrets and herons except Cattle egret Bubulcus ibis increased and partocilarly, the Gray heron Ardea cinerea was found to have been influenced the most by this construction in comparison with other egrets and herons. The number od individuals of large egrets, little egrets and herons was found to decrease during the construction period. After cutting off the nest trees and removing some parts of the breeding site, the herons and egrets tended to move their nesting place from roadsides to the inside of the breeding area. Trees for the nests decreased due to the increase in fellen trees caused by bird nests per tree increased gradually. As for the formation ratio of trees for the nests, the ratio of pine trees lowered yearly while that of acacia and oak trees was increasing during the survey.

Genetic Analysis on the Bacterial Blight Resistance of Suweon497, a Rice Breeding Line Developed through Wide Hybridization (벼 종간교잡 후대계통 '수원497호'의 흰잎마름병 저항성에 대한 유전분석)

  • Rice wild relatives have been recognized as reservoirs of genetic reinforcements to improve cultivating rice against biotic and abiotic stresses. A wild relative, Oryza. minuta(BBCC; Acc. 101141), was hybridized with a Korean Japonica cultivar, 'Hwaseong'(AA), followed by ovule culture and several times of back crossings to overcome high level of sterility. During evaluation of the introgression lines, breeding line exhibited resistance to bacterial blight with reasonable agronomic performances, and nominated as an elite breeding line, the 'Suweon497'. A mapping population, to dissect genetic basis of the resistance, was constructed by using $F_2$ progenies of the 'Suweon497' ${\times}$ 'Milyang23'. Association analysis between SSR marker genotypes and pathogenisity levels of each $F_2$ progeny revealed the end terminal region of rice chromosome 11 as the nesting place for the wild rice derived bacterial blight resistance gene, where at least four other genes, Xa3, Xa4, Xa26 and Xa31, have been reported.

Agronomic and Genetic Evaluation on a Dull Mutant Line Derived from the Sodium Azide Treated 'Namil', a Non-Glutinous Japonica Rice (남일벼 돌연변이 유래 중간찰 계통의 작물학적 특성 및 배유특성 지배유전자위 표지)

  • Developing rice lines with various amylose contents is necessary to diverse usages of rice in terms of raw materials for processed food production, and thereby to promote rice consumption in Korea. A rice mutant line, 'Namil(SA)-dull1' was established through sodium azide mutagenesis on 'Namil', a non-glutinous Korean Japonica rice cultivar. Namil(SA)-dull1' had dull endosperm characteristics and the evaluated amylose content was 12.2%. A total of 94 F2 progenies from a cross between 'Namil(SA)-dull1' and 'Milyang23', a non-glutinous Tongil-type rice cultivar, was used for genetic studies on the endosperm amylose content. Association analyses, between marker genotypes of 53 SSR anchor markers and evaluated amylose contents of each 94 F2:3 seeds, initially localized rice chromosome 6 as the harboring place for the modified allele(s) directing low amylose content of 'Namil(SA)-dull1'. By increasing SSR marker density on the putative chromosomal region followed by association analyses, the target region was narrowed down 0.94 Mbp segment, expanding from 28.95 Mbp to 29.89 Mbp, on rice chromosome 6 pseudomolecule. Among the SSR loci, RM7555 explained 84.2% of total variation of amylose contents in the $F_2$ population. Further physical mapping on the target region directing low amylose content of 'Namil(SA)-dull1' would increase the breeding efficiency in developing promising rice cultivars with various endosperm characteristics.

A Research for Shorebirds on the Southernmost of Nakdong Estuary (낙동강 하구 최남단 사주의 도요.물떼새류에 관한 연구)

  • Estuary islets should be managed systematically because they are important places for birds to rest and breed. This paper investigates the environmental properties of islets where the sand banks are located from east to west on the southernmost of Nakdong estuary during a year (September 2003 $\sim$ August 2004). The research showed that 59 species and 19,148 individuals were found in the area A (Jangja Shinja-Do) and 61 species and 28,394 individuals in the area B (Saja-Do; Beakhapdeung Doyodeung). Totally, 74 species and 47,539 individuals were observed in both of this estuary. Shorebirds are the most observed species in both areas. In area, most of individuals were especially observed in the spring when Shorebirds migrate northward for breeding. In B area, various species were also observed in the fall when they migrate southward for wintering. Therefore, many groups of birds are observed around Shinja-Do in the spring. They passed the winter in Doyodueng, associated with feeding in the main stream of Nakdong river mainly. Although the southernmost sand bar is the breeding place to access to outside without interception, invasion by human beings and predators (mice, weasels and etc.) gives a fatal blow for birds to breed or rest. The reed which is a tall herbaceous plant is flourished according to the process of ecological succession. Therefore, the sand bar becomes a land. It results in reduction of habitats and breeding grounds for birds. In conclusion, these areas where birds can use the islets as habits and breeding places must be preserved by restraining luxuriance of reeds and systematic management of human beings and predators is necessary.

  • As Korean ginseng is hybrid, an individual variation is very severe, and it takes long times in new breeding because it is required 4 years to pick the seed. But, transformation technique makes the high-functional breeding in short time. The focus of these ginseng studies is to find and secure the useful gene. And it is urgent to accumulate the fundamental data for the molecular breeding and secure the useful genes. Therefore, transformation and soil acclimatization technique are necessary to molecular breeding in use of the introduction of functional genes. In this study, it add to secure of new regulation gene and useful gene as to accumulate the fundamental data for the place where it will contribute to raise the national competitive power. To analyze the useful genes in large scale, we constructed CDNA libraries with various tissues, species, and treated tissue. EST analysis of ginseng perform in large scale and build the EST database of ginseng. We perform the full length sequencing about the selected lots of clones that include the entire open reading frame of the amino acid residues and construct cDNA chip with the parental EST clones. Establishment of the transformation and a soil acclimatization system throuth the re-introduction of the selected ginseng gene that related with the secondary metabolism and anti-stress into the ginseng.

Biotechnology of Reproductive Processes in Cereals

  • Sexual reproduction is an essential process in the propagation of flowering plants. Recent advances in plant cell biology and biotechnology have brought new and powerful methodologies to investigate and manipulate the reproductive processes of angiosperms including agronomically important crop plants. Successful cryopreservation of maize, rye and triticale pollen and young embryos of microspore-and zygote-origine contributes to long term preservation of important plant germ-lines in gene banks. Discovering morphogenetic characteristics of the different developmental pathways taking place in wheat and maize androgenesis in vitro helps to influence the procedure to produce genetically and phenotipically stable homozygous doubled haploid plants for breeding purposes. Detailed ultrastructural and cell-biological studies on the developmental sequences of male and female gametophyte development in wheat, experimental protocols developed to isolate and micromanipulate egg cell protoplasts, make it possible to use plant gametes and the sexual route itself to produce genetically improved organisms. Plant gametes can become useful tools for crop improvement in the near future. Recent achievements by our laboratory in this field are reviewed in the present paper

Studies on the Pure-bred Korean Native Cattle of Chonnam Area - Hematology and Biochemistry Values - (전남지방 순수번식단지의 한우에 관한 연구 - 혈액학식 및 혈액화학식 -)

  • Samples of blood were taken from 293 cows and heifers in Koheung, Chonnam, which was dsignated as a place for breeding pure-bred Korean native cattle. Each animal was examined clinically and parasitologically and with the history available considered to be normal at the time of blood collection. The hematology and biochemistry values were determinded, and comparisons were made between three age groups, using the results obtained. All the values obtained were within physiological range : except the alanine aminotransferase values which were higher(P<0.05) in young than in mature or old group of cattle, no significant differences were notred between age groups.

  • Factors Affecting the Survival of Frozen Thawed Bovine In Vitro Produced Blastocysts. The effect of some factors on the post-thaw survival of a total of 240 in vitro produced bovine blastocysts was investigated using logistic regression analysis. The explanatory variables tested were: type of culture medium before freezing (TCM 199 supplemented with BSA, BSAITS (BSA+insulin+transferrin+selenium), ECS (estrous cow serum) with or without BOEC (bovine oviductal epithelial cells), age of the blastocyst (Day 7, Day 8+9), morphological appearance before freezing (distinct=Q1 or indistinct=Q2 inner cell mass) and type of cryoprotectant (glycerol, 1.0 M or ethylene glycol, 1.6 M). The survival after thawing based on the post-thaw quality and the development after co-culture with BOEC for 24 and 48 hours. Day 7 blastocysts had an almost three times better chance of survival than Day 8+9 blastocysts. Q1, Day 8+9 blastocysts had higher odds to survive after 48 hours in culture than Q2 blastocysts (p<0.05). Blastocysts produced in BSAITS medium had the best chances of survival; however, the odds were not always significant. Blastocysts frozen in glycerol had a better post-thaw quality rating than those frozen in ethylene glycol; however, the difference in post-thaw development at culture was not significant. The relationship between post-thaw quality and post-thaw development at culture was significant (p<0.05). The developmental stage and/or age of the embryo and culture medium where development up to blastocyst takes place affect the post-thaw survival of the bovine embryos.

  • Embryo transfer (ET) technology is of high importance in modern cattle breeding programs. ET is one step in the process of removing one or more embryos from the reproductive tract of an outstanding donor female and transferring them to one or more recipient females. Embryos also can be produced in the laboratory via techniques such as in vitro fertilization (IVF). But the actual transfer of an embryo is only one step in a series of processes that may include some or all of the following: superovulation and insemination of donors, collection of embryos, isolation, evaluation and short-term storage of embryos, micromanipulation and genetic testing of embryos, freezing of embryos and embryo transfer. Cryopreservation and direct transfer of frozen-thawed embryos is common-place with pregnancy rates near that of fresh embryos. Polymerase chain reaction (PCR) technology is currently being used for sexing embryos, and this technology will be used for "embryo diagnostics" and "embryo genomics" in the future. Although, many limitations and problems remain to overcome, these and other new technologies promise to change livestock breeding drastically in the next decade.

Integrated Network System of Milk Cow Stock-Farming Facilities for Stockbreeding Management (사양관리를 위한 젖소 목장 시설 통합 네트웍 시스템)

  • This paper introduces the method to make management network about milking cow farm tasks. The object of this research was to design of biological measuring system and managing network system in a livestock farm. This auto-management system provides informations about individual cows' temperature, conductivity of milk and weight for efficient management of feeding, and milking works by a micro-processor and RS -485 type serial COM. ports. And measured bio-data which are basic informations for remote raising management are saved to user PC by serial communication between the PLC and user PC. Milking cow farm is divided into three working place to each measurement work and feed. The first working place is milking station which has two thermometers, a conduct meter and a scale set. The second working place is feeding station, and the third place is cattle cage. These are combined by network system and the PLC which is used to drive network and sub-modules. Sub-modules have a micro-process to control the sensor and to interface with network. The PLC which drive network and control sequence has two serial communication port to be linked with user PC for sending the measured data and for receiving data. Above all, in this study tells the sequence operating method by the driving scenario of breeding milk cow for livestock auto-management using the PLC and network system.
