Cryptosporidium suis Infection in Post-Weaned and Adult Pigs in Shaanxi Province, Northwestern China

  • Cryptosporidium spp., ubiquitous enteric parasitic protozoa of vertebrates, recently emerged as an important cause of economic loss and zoonosis. The present study aimed to determine the distribution and species of Cryptosporidium in post-weaned and adult pigs in Shaanxi province, northwestern China. A total of 1,337 fresh fecal samples of post-weaned and adult pigs were collected by sterile disposable gloves from 8 areas of Shaanxi province. The samples were examined by Sheather's sugar flotation technique and microscopy at${\times}400$ magnification for Cryptosporidium infection, and the species in positive samples was further identified by PCR amplification of the small subunit (SSU) rRNA gene. A total of 44 fecal samples were successfully amplified by the nested PCR of the partial SSU rRNA, with overall prevalence of 3.3%. The average prevalence of Cryptosporidium infection in each pig farms ranged from 0 to 14.4%. Species identification by sequencing of SSU rRNA gene revealed that 42 (3.1%) samples were Cryptosporidium suis and 2 (0.15%) were Cryptosporidium scrofarum. C. suis had the highest prevalence (7.5%) in growers and the lowest in breeding pigs (0.97%). C. suis was the predominant species in pre-weaned and adult pigs, while C. scrofarum infected pigs older than 3 months only. A season-related difference of C. suis was observed in this study, with the highest prevalence in autumn (5.5%) and the lowest (1.7%) in winter. The present study provided basic information for control of Cryptosporidium infection in pigs and assessment of zoonotic transmission of pigs in Shaanxi province, China.

Saccharomyces yeast postbiotics supplemented in feeds for sows and growing pigs for its impact on growth performance of offspring and growing pigs in commercial farm environments

  • Objective: Three experiments were conducted to evaluate the effects of Saccharomyces yeast postbiotics (SYP) in feeds for sows on the growth of offspring (Exp. 1), for nursery pigs on their growth (Exp. 2), and for nursery and finishing pigs on their growth (Exp. 3). Methods: Exp. 1 had 80 sows at breeding assigned to 4 groups with SYP at 0, 0.050, 0.175, and 0.500 g/kg. Offspring were fed a common diet for 126 d. Exp. 2 had 144 barrows at 8 kg body weight (BW) allotted to CON (no SYP); YPC (SYP at 0.175 g/kg; d 0 to 42); and YPD (SYP at 1.25, 0.75, and 0 g/kg; d 0 to 7, d 8 to 21, and d 22 to 42, respectively) with 8 pens/treatment (6 pigs/pen). Exp. 3 had 96 barrows at 8 kg BW allotted to CON (no SYP); YPN (SYP at 0.175 g/kg; d 0 to 42); YPF (SYP at 0.100 g/kg; d 43 to 119); and YPA (SYP at 0.175 and 0.100 g/kg; d 0 to 42 and d 43 to 119, respectively) with 8 pens/treatment (3 pigs/pen). Results: In Exp. 1, increasing SYP increased (p<0.05, quadratic) the sow body score (maximum at 0.30% SYP), reduced (p<0.05, quadratic) the days-wean-to-estrus (minimum at 0.27% SYP), and increased (p<0.05) offspring BW at weaning and their average daily gain (ADG) and feed efficiency (G:F) at d 126. In Exp. 2, ADG, average daily feed intake (ADFI), and G:F of YPC were the greatest (p<0.05). The ADG and ADFI of YPD were greater (p<0.05) than CON. Fecal score of YPC and YPD was smaller (p<0.05) than CON. In Exp. 3, YPA had the greatest (p<0.05) ADG and YPN and YPF had greater (p<0.05) ADG than CON. Conclusion: SYP enhanced sow performance, offspring growth, growth of nursery and growing pigs with the greater efficacy at 0.27 to 0.32 g/kg feed.

부산지방에서 사육중인 애완견으로부터 분리한 피부사상균에 관한 연구 (Studies on Distribution and Pathogenicity of Dermatophytes Isolated from Pet Dogs in Pusan Area)

  • This study were performed to distribution of dermatophytes isolated from pet dogs in Pusan area and pathogenicity of the isolates. During the period from March 1993. to April 1994, 201 pet dogs of house breeding were examined. The result of obtained were summarized as followes; 1. Isolation rate of dermatophytes were 4.7%(7/149) in no lesion and 23.1%(12/52) in lesion of dog's skin. 2. According to breeding conditions, isolation rate of dermatophytes were 1) 3.8% in no lesion and indoor breeding, 2) 16.7% in lesion and indoor breeding, 3) 13.5% no lesion and outdoor breeding, and 4) 37.5% in lesion and outdoor bleeding. 3. Among the 201 Pet dogs, 16 strains of M. cams(84.2%), 1 of M. gypseum(5.3%), and 2 of T. mentagrophytes (10.5%) were isolated. 4. For Pathogenicity test, one strain of each M. canis, M, gypseum, T. mentagrophytes isolated from skin lesions were inoculated to the skin of the posterior back of guinea pigs. All of dermatophytes tested were induced skin lesions. Among them, M. cams and T. mentagrophytes were more virulent than M. gypseum.

Mapping Quantitative Trait Loci for Meat Quality on Pig Chromosome 3, 4 and 7

  • The objective of this study was to localize QTL affecting meat quality in a pig family of three generations. All animals were genotyped for twenty-four microsatellites on SSC3 (Sus scrofa chromosome 3), SSC4 and SSC7. One hundred and forty $F_2$ offsprings were scored for eleven meat quality traits. Least square regression interval mapping revealed quantitative trait loci (QTL) effect for meat pH (m. Semipinalis Capitis, SC) on SSC4 and SSC7; for moisture (m. Longissimus Dorsi, LD) on SSC3. Furthermore, there was suggestive evidence for a QTL on SSC4 affecting intramuscular fat (IMF) content that nearly approached the chromosomewise (p=0.05) significance threshold.

A genome-wide association study on growth traits of Korean commercial pig breeds using Bayesian methods

  • Objective: This study aims to identify the significant regions and candidate genes of growth-related traits (adjusted backfat thickness [ABF], average daily gain [ADG], and days to 90 kg [DAYS90]) in Korean commercial GGP pig (Duroc, Landrace, and Yorkshire) populations. Methods: A genome-wide association study (GWAS) was performed using single-nucleotide polymorphism (SNP) markers for imputation to Illumina PorcineSNP60. The BayesB method was applied to calculate thresholds for the significance of SNP markers. The identified windows were considered significant if they explained ≥1% genetic variance. Results: A total of 28 window regions were related to genetic growth effects. Bayesian GWAS revealed 28 significant genetic regions including 52 informative SNPs associated with growth traits (ABF, ADG, DAYS90) in Duroc, Landrace, and Yorkshire pigs, with genetic variance ranging from 1.00% to 5.46%. Additionally, 14 candidate genes with previous functional validation were identified for these traits. Conclusion: The identified SNPs within these regions hold potential value for future marker-assisted or genomic selection in pig breeding programs. Consequently, they contribute to an improved understanding of genetic architecture and our ability to genetically enhance pigs. SNPs within the identified regions could prove valuable for future marker-assisted or genomic selection in pig breeding programs.

Enzyme-linked immunosorbent assay for detection of Trichinella spiralis antibodies and the surveillance of selected pig breeding farms in the Republic of Korea

  • Trichinellosis is a parasitic zoonosis of public health importance. It is caused by Trichinella spiralis which has a wide host range including humans. In the present communication, the ELISA technique was employed on a total of 803 blood samples from 7 selected pig breeding farms in 1996 for diagnosis and surveillance of trichinellosis. Out of the entire 803 samples, nine were found to be suspected while one was positive by ELISA. But western blot analyses employed for further confirmation have shown that all of 10 samples did not react to larval excretory-secretory product antigens. These results indicate that pig breeding farms included in the present study are free from trichinellosis . However, it does not mean Korea is free from trichinellosis since human trichinellosis has recently been reported. The necessity of continued surveillance for trichinellosis in both pigs and wild animals was discussed.

모체효과 모형을 이용한 돼지 품종 간의 성장형질 및 등지방두께에 대한 유전모수 추정 (Estimation of genetic parameters for growth traits and backfat thickness using Maternal animal model in pigs)

  • 국내에서는 우리 고유의 재래돼지가 보존되고 있으나 성장 및 번식능력이 낮고 등지방두께가 두꺼워 경제성이 떨어진다는 이유로 농가에서 사육을 기피하고 있다. 하지만, 나고야의정서 발효 등에 따라 고유 유전자원의 중요성은 커지고 있으며 따라서, 고유 유전자원인 재래돼지를 활용한 신 계통 개발의 중요성은 점차 커지고 있다. 본 연구는 개체모형을 이용하여 돼지의 경제적 형질에 대한 유전모수 및 유전적 변화의 추정을 통해 모체유전효과가 돼지의 경제 형질에 미치는 영향을 알아보기 위해 수행되었다. 자료는 국립축산과학원으로부터 2000년부터 2015년까지 수집한 두록종, 재래돼지 그리고 합성종(재래돼지${\times}$두록종)의 자료를 사용하였다. 본 연구에 사용된 경제형질은 일당증체량과 등지방두께를 이용하였으며 분석에 사용된 Model식은 개체효과만을 고려한 Model 1과 모체효과를 추가한 Model 2 그리고 모체 영구환경효과가 포함된 Model 3을 사용하여 분석에 이용하였다. 분석 결과 상가적 유전력이 모체효과 유전력보다 높은 유전력을 나타냈으며 상가적 유전분산과 모체 유전분산 간의 유전상관은 모든 품종에서 강한 부(-)의 상관관계를 가지는 것을 확인할 수 있었다. 따라서, 좀 더 효율적인 개량 효과를 거두기 위해서는 품종 및 형질 또는 개량 방향에 따라 개체 효과와 모체유전효과 그리고 유전상관을 적절히 고려해야 할 것으로 사료된다.

Assessment of the Contribution of Poultry and Pig Production to Greenhouse Gas Emissions in South Korea Over the Last 10 Years (2005 through 2014)

  • The goal of this study was to estimate the emissions of greenhouse gases (GHG), namely methane ($CH_4$), nitrous oxide ($N_2O$), and carbon dioxide ($CO_2$) from poultry and pig production in South Korea over the last 10 years (2005 through 2014). The calculations of GHG emissions were based on Intergovernmental Panel on Climate Change (IPCC) guidelines. Over the study period, the $CH_4$ emission from manure management decreased in layer chickens, nursery to finishing pigs and gestating to lactating sows, but there was a gradual increase in $CH_4$ emission from broiler chickens and male breeding pigs. Both sows and nursery to finishing pigs were associated with greater emissions from enteric fermentation than the boars, especially in 2009. Layer chickens produced lower direct and indirect $N_2O$ emissions from 2009 to 2014, whereas the average direct and indirect $N_2O$ emissions from manure management for broiler chickens were 12.48 and $4.93Gg\;CO_2-eq/yr$, respectively. Annual direct and indirect $N_2O$ emissions for broiler chickens tended to decrease in 2014. Average $CO_2$ emission from direct on-farm energy uses for broiler and layer chickens were 46.62 and $136.56Gg\;CO_2-eq/yr$, respectively. For pig sectors, the $N_2O$ emission from direct and indirect sources gradually increased, but they decreased for breeding pigs. Carbon dioxide emission from direct on-farm energy uses reached a maximum of $53.93Gg\;CO_2-eq/yr$ in 2009, but this total gradually declined in 2010 and 2011. For boars, the greatest $CO_2$ emission occurred in 2012 and was $9.44Gg\;CO_2-eq/yr$. Indirect $N_2O$ emission was the largest component of GHG emissions in broilers. In layer chickens, the largest contributing factor to GHG emissions was $CO_2$ from direct on-farm energy uses. For pig production, the largest component of GHG emissions was $CH_4$ from manure management, followed by $CO_2$ emission from direct on-farm energy use and $CH_4$ enteric fermentation emission, which accounted for 8.47, 2.85, and $2.82Gg-CO_2/yr$, respectively. The greatest GHG emission intensity occurred in female breeding sows relative to boars. Overall, it is an important issue for the poultry and pig industry of South Korea to reduce GHG emissions with the effective approaches for the sustainability of agricultural practices.

비육돈의 도체형질과 A-FABP Promoter 유전자형의 연관 분석 (The Association between A-FABP Promoter Genotype and Carcass Traits in Pigs)

  • 본 연구는 비육돈에서 지방세포지방산결합단백질(A-FABP) 유전자의 유전자형과 도체형질의 상관관계를 조사하였다. 유전자 A-FABP의 프로모터 영역에 대해 HinfI-RFLP를 이용하여 유전자형을 조사한 결과 두 가지 유전자형(-406T/T T/C)들이 발견되었고 C/C는 발견되지 않았다. 유전자형 중 -406C/C 동형접합자가 발견되지 않는 이유는 비육돈 생산에 관여하는 돼지 품종들 중에서 Duroc 품종만이 이 유전자에 대한 고도의 다형성을 나타내기 때문으로 추정되었다. 유전자형 -406C/-를 나타내는 돼지들은 근내지방과 도체중이 통계적으로 유의적인 증가함을 나타내었으나 (p<0.05), 그 외의 도체형질들(조지방, 육색, 조직감, 수분 삼출도, 근육분리도)은 유의한 차이를 나타내지 않았다 (p>0.05). 본 연구를 통해 돼지 A-FABP 유전자의 -406C/- 유전자형이 근내지방에 대한 유용한 분자 마커가 될 수 있을 뿐만 아니라 다형성이 비육돈의 근내지방에 대한 유용한 분자 마커가 될 수 있으며, 특히 Duroc 품종을 최종 종모돈으로 이용할 경우 이 유전자형을 보유한 돼지를 이용하는 분자육종체계에서 marbling이 잘 된 돼지의 생산을 통한 육질 개선에 기여할 것으로 사료된다.

Efficacy of Cr (III) Supplementation on Growth, Carcass Composition, Blood Metabolites, and Endocrine Parameters in Finishing Pigs

  • The study was conducted to evaluate the effects of trivalent chromium from different sources on growth, carcass composition, and serum parameters in finishing pigs. Ninety-six crossbred pigs with an initial average body weight of 65.57${\pm}$1.05 kg were blocked by body weight and randomly assigned to four treatments with three replicates. Pigs were offered one of four diets including a control diet or the control diet supplemented with 200 ${\mu}g/kg$ chromium from either chromium chloride ($CrCl_{3}$), chromium picolinate (CrPic) or chromium nanocomposite (CrNano) for 40 days. After completion of the feeding trial, eight pigs from each treatment were selected to collect blood samples, and slaughtered to measure carcass composition. The results showed that supplemental chromium had no significant effect on growth performance, while CrNano increased carcass lean proportion and loin Longissimus muscle area (p<0.05), and decreased carcass fat proportion and 10th rib backfat depth (p<0.05). CrPic supplementation also resulted in lower fat proportion and larger Longissimus muscle area (p<0.05). The addition of Cr from CrNano or CrPic decreased serum glucose (p<0.05) and increased concentrations of total protein and free fat acid in serum (p<0.05). Serum urea nitrogen, triglyceride and cholesterol were decreased (p<0.05), and serum high density lipoprotein and lipase activity were increased (p<0.05) with the supplementation of CrNano. Serum insulin was decreased (p<0.05) by supplemental Cr from CrNano or CrPic, and serum insulin-like growth factor I was increased significantly in the CrNano treated group. These results suggest that chromium nanocomposite has higher efficacy on carcass composition in pigs compared to the traditional chromium sources.
