A New Single Cross Maize Hybrid for Grain and Silage, 'Pyeongangok' (내도복 다수성 종실 및 사일리지 옥수수 신품종 '평강옥')

  • Pyeongangok, a new single cross variety, is an yellow dent maize hybrid (Zea mays L.) developed by the maize breeding team at the National Institute of Crop Science (NICS), RDA in 2011. This hybrid, which has a high yield of grain and dry matter, was produced by crossing two inbred lines, KS160 and KS155. KS160 is the seed parent and KS155 is the pollen parent of Pyeongangok. Silking date of Pyeongangok is 2 days earlier than that of check hybrid, Jangdaok, and equal to that of another check hybrid, Kwangpyeongok. Plant height of Pyeongangok is longer than that of Jangdaok and similar to that of Kwangpyeongok. Ear numbers per 100 plants of Pyeongangok is more than that of Jangdaok. Ear length of Pyeongangok is shorter than that of Jangdaok. 100 seeds weight of Pyeongangok is lighter than that of Jangdaok. Ear rate of Pyeongangok is lower than that of Kwangpyeongok. Stay-green of Pyeonganok is not greatly different with that of Kwangpyeongok. It has moderately resistance to southern corn leaf blight (Bipolaris maydis), black streaked dwarf virus (BSDV) and corn borer. It has strong resistance to northern corn leaf blight (Exserohilum turcicum). It has resistance to lodging. Pyeongangok was evaluated for the yields of grain and dry matter at four locations from 2009 to 2011. The yield of Pyeongangok in grain was 7.66 ton/ha. The yield of Pyeongangok in dry matter was 19.80 ton/ha. The yield of Pyeongangok in total digestible nutrient (TDN) was 13.32 ton/ha. Seed production of Pyeongangok has gone well due to a good match during crossing between the seed parent, KS160, and the pollen parent, KS155, in Yeongwol.

Antioxidant Activity and Grain Properties of Colored Rice Derived from Insertional Mutagenesis Progenies (벼 종피색 변이체에 대한 항산화 활성 분석과 미립특성)

  • This study examined the antioxidant activity of the dark purple rice seeds from the rice line, MGI079, derived from insertional mutagenesis. The contents of polyphenolic compounds were 1.3 and 1.9-fold higher in the MGI079-2-1 and MGI079-2-6 rice lines than in the donor cultivar MGI079. Flavonoid contents were 6.4-fold higher in the MGI079-2-1 line. The MGI079-2-1 line showed a 24.4-fold higher activity in DPPH free radical scavenging compared to the MGI079 line. The anthocyanin content of the MGI079-2-6 line was more than 106.4-fold higher than the MGI079 line and 1.4-fold higher than the Heugnam line. Anthocyanin content in colored rice grains was negatively correlated with Hunter's L, a, and b values, with the correlation coefficients of $-5.64^{**}$, $5.21^{**}$ and -1.15, respectively. The grain length/width of a mutant of MGI079 segregated to a medium and bold type compared to the medium type of MGI079. However, the 1,000 grain weight was decreased to 13.6~19.6 g compared to 19.8 g for MGI079. Amylose content of the endosperm was 5.6~23.8% higher than in the MGI079 line. The grain of mutants of MGI079 was distinguished by its starch characteristics. The higher antioxidant activity of the MGI079-2-1 and MGI079-2-6 lines indicated functional characteristics associated with high-value resources, so future breeding should focus on the development of pigments in colored rice in new varieties.

Agronomic Characteristics of Adzuki bean (Vigna angularis W.F. Wight) Germplasm in Korea (팥 유전자원의 작물학적 형질 특성)

  • Agronomic characteristics of 150 adzuki bean ($Vigna$ $angularis$ W.F. Wight) germplasm collected in Korea were investigated in order to establish basic data for the improvement of crop breeding. Growth types were classified as 3 types of indeterminate type, determinate type, and semideterminate type, of which indeterminate type was the highest percentage of 96.6%. Seed color were classified as 8 colors of which dark brown took 66.5% in 150 adzuki bean germplasm. Days from seeding to maturity showed the range from 89 to 142 days with 115 days in the average and the highest frequency proportion was 37.2% of the group from 131 to 140 days, and the next was 27.3% of the group from 121 to 130 days among 150 germplasm. Pod length showed the range from 5.9 to 10.1 cm with 7.8 cm in the average and the highest frequency distribution of it was the group from 7.1 to 8.0 cm with the proportion of 45.1% and the next was the group from 8.1 to 9.0 cm with the proportion of 32.2%. Number of pods per plant showed the range from 5.0 to 132.5 pods with 28.5 pods in the average and the highest frequency distribution of it was the group from 11.0 to 20.0 pods with the proportion of 30.8% and the next was the group from 21.0 to 30.0 pods with the proportion of 28.6%. Number of seeds per pod showed the range from 5.3 to 11.3 seeds with 8.2 seeds in the average and the highest frequency proportion of it was the group from 7.1 to 8.0 seeds, which occupied 32.2% among 150 germplasm. 89.3% of 150 germplasm showed the resistance to shattering of grains at harvest period. 100 grain weight showed the range from 5.7 to 23.0 g with 12.9 g in the average and the highest frequency proportion of it was the group from 9.01 to 13.0 g, which occupied 43.3% and the next was the group from 13.01 to 17.00 g with the proportion of 28.5% of 150 germplasm.

Effects of Growth Period and Cumulative Temperature on Flowering, Ripening and Yield of Soybean by Sowing Times (파종시기별 생육일수 및 적산온도 변화가 콩의 개화, 등숙 및 수량에 미치는 영향)

  • The purpose of this research was to analyze the effect of different sowing times on the flowering and maturing of major soybean cultivars by varying day length and temperature in the central plain region. The average of growth period and cumulative temperature in five test cultivars by sowing times were 121 days and 2,972℃ on June 1, respectively and gradually decreased to 85 days, 2,042℃, respectively on July 20. Analysis of the flowering response according to the sowing times showed that flowering was greatly influenced by the decrease of photoperiod until the sowing on July 10, and the minimum number of days for flowering were 27 days, 36 days, respectively in early and mid-rate maturing type in the central plain region. Daepung 2 is classified to the same ecotype with Daewonkong, the total number of growing days was not different between two cultivars, but ripening period (R2-R6) was longer by 5 days and yield was higher by 11% in Daepung 2. The maturity rate was also high and safe enough to maintain more than 90% through the entire sowing times. This ecological characteristic can be usefully applied as a section index for breeding environmental stress resistant and high yielding soybean varieties. The yield of 4 domestic cultivars (except TI196944) sowing on July 20 were 85~92% levels compared to sowing on June 20.

A New High-yielding Winter Oat Cultivar for Whole Crop Forage, 'Dahan' (내한 다수성 추파 조사료용 총체 귀리 신품종 '다한')

  • 'Dahan' (Avena sativa L.), a winter oat cultivar for forage use, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, National Institute Crop Science (NICS), Rural Development Administration (RDA) in 2011. It was derived from an original cross between $F_1$[Sprinter/73625] and 'Gwiri26'. Subsequent generations followed by the cross were handled in bulk and pedigree selection programs at Suwon. A line, 'SO99027-GB-B-113-4-4-3', was selected for cold tolerance and good agronomic characteristics and as a line name of 'Gwiri75'. The line 'Gwiri75' was subsequently evaluated for cold tolerance and forage yield during 3 years in four region such as Yesan, Iksan, Kimjae, and Jeju, from 2009 to 2011 and finally named as 'Dahan'. Though similar in heading date to the check cultivar Samhan, 'Dahan' had tall plant length and lodging resistance. It's average forage dry matter yield harvested at milk-ripe stage was 15.6 ton $ha^{-1}$, compared with 14.1 ton $ha^{-1}$ of check cultivar. Cultivar 'Dahan' was lower to the check cultivar 'Samhan' in protein content (8.4% and 9.9%, respectively), while it was superior to the check cultivar in total digestible nutrients (TDN) (60.8% and 59.3%, respectively), and in TDN yield $ha^{-1}$ (9.5 ton and 8.4 ton, respectively). Fall sowing of 'Dahan' is recommended only in the areas where daily minimum mean temperatures are averaged higher than $-7^{\circ}C$ in January, and excluded in mountain area where frost damage is presumable.

Growth Characteristics and Productivity of New Single Cross Maize Hybrid for Grain, 'Singwangok' (종실용 옥수수 신품종 '신광옥'의 생육특성과 수량성)

  • Singwangok, a new single cross variety, was developed by the maize breeding team at the National Institute of Crop Science (NICS), RDA in 2012. This hybrid, consisting of a high yield of grain, was produced by crossing two inbred lines, KS172 and KS173. KS172 is the seed parent and KS173 is the pollen parent of Singwangok. It is a yellow-orange intermediate maize hybrid (Zea mays L.). After the preliminary yield trial and advanced yield trial of Singwangok (Suwon185) in Suwon for 2 years, the regional yield trial of that was subsequently carried out for its growth characteristics and yield at 3 different locations from 2010 to 2012. It was named as Singwangok. The silking date of Singwangok is similar to the check hybrid, Jangdaok. The plant height of Singwangok is similar to Jangdaok, but its ear height ratio is lower than that of Jangdaok. Moreover, it has a resistance to lodging. The ear number per 100 plants of Singwangok is more 17 than that of Jangdaok, whereas the ear length of Singwangok is similar to Jangdaok. Further, the weight of 100 seeds of Singwangok is similar to Jangdaok. It has a moderate resistance to southern leaf blight (Bipolaris maydis) and a strong resistance to northern leaf blight (Exserohilum turcicum). Furthermore, it has a moderate resistance to the black streaked dwarf virus (BSDV), ear lot and corn borer. The grain yield of Singwangok, 7.81 ton/ha, was similar to that of Jangdaok. The seed production of Singwangok was well processed due to the good match during crossing between the seed parent, KS172 and the pollen parent, KS173, in Yeongwol; the F1 seed yield was 3.84ton/ha. Singwangok would be a suitable cultivar to all plain areas in Korea.

Effect of the Sowing and Harvesting Dates on the Agronomic Characteristics and Feed Value of Corn and Sorghum×Sorghum Hybrid in Youngnam Mountain Area (영남산간지역에서 옥수수와 수수×수수 교잡종의 파종시기와 수확시기가 조사료 생육 특성과 사료가치에 미치는 영향)

  • This study was carried out from 2015 to 2016 to identify the suitable sowing and harvesting dates of summer crops in the mountain of Yeongnam, South Korea. The experimental design consisted of the different sowing and harvesting dates as follows: corn hybrid (Z. mays, Kwangpyeongok) of sowing (May 8, 19, and 27) and harvesting (August 10, 20, and 30); sorghum${\times}$sorghum hybrid (Sorghum bicolor ${\times}$ Sorghum bicolor, SS405) of sowing (May 27, June 19 and June 27) and harvesting (August 10, 20, and 30). In corn hybrid, ear rate and dry matter (DM) yield decreased (p<0.05) with the postponement of sowing date. Otherwise, ear rate and DM yield increased (p<0.05) with the postponement of harvesting date. Crude protein content decreased (p<0.05) with the postponement of sowing date, but neutral detergent fiber content increased (p<0.05). In sorghum${\times}$sorghum hybrid, plant length and DM yield with the postponement of harvesting date increased (p<0.05), while crude protein content with the postponement of harvesting date decreased (p<0.05). This study concluded that sowing corn hybrid in early May and sorghum${\times}$sorghum hybrid in early June then harvest in the middle of August was recommend to increase dry matter yield and feed value.

A Comparative Analysis of the Illumina Truseq Synthetic Long-read Haplotyping Sequencing Platform versus the 10X Genomics Chromium Genome Sequencing Platform for Haplotype Phasing and the Identification of Single-nucleotide variants (SNVs) in Hanwoo (Korean Native Cattle) (일루미나에서 제작된 TSLRH (Truseq Synthetic Long-Read Haplotyping)와 10X Genomics에서 제작된 The Chromium Genome 시퀀싱 플랫폼을 이용하여 생산된 한우(한국 재래 소)의 반수체형 페이징 및 단일염기서열변이 비교 분석)

  • In Hanwoo cattle (Korean native cattle), there is a scarcity of comparative analysis papers using highdepth sequencing and haplotype phasing, particularly a comparative analysis of the Truseq Synthetic Long-Read Haplotyping sequencing platform serviced by Illumina (TSLRH) versus the Chromium Genome Sequencing platform serviced by 10X Genomics (10XG). DNA was extracted from the sperm of a Hanwoo breeding bull (ID: TN1505D2184/27214) provided by Hanwoo research canter and used for the generation of sequence data from both the sequencing platforms. We then identified SNVs using an appropriate analysis pipeline tailored for each platform. The TSLRH and 10XG platforms generated a total of 355,208,304 and 1,632,772,004 reads, respectively, corresponding to a Q30 (%) of 89.04% and 88.60%, respectively, of which 351,992,768(99.09%) and 1,526,641,824(93.50%) were successfully mapped. For the TSLRH and 10XG platforms, the mean depth of the sequencing was 13.04X and 74.3X, the longest phase block was 1,982,706 bp and 1,480,081 bp, the N50 phase block was 57,637 bp and 114,394 bp, the total number of SNVs identified was 4,534,989 and 8,496,813, and the total phased rate was 72.29% and 87.67%, respectively. Moreover, for each chromosome, we identified unique and common SNVs using both sequencing platforms. The number of SNVs was directly proportional to the length of the chromosome. Based on our results, we recommend the use of the 10XG platform for haplotype phasing and SNV identification, as it generated a longer N50 phase block, in addition to a higher mean depth, total number of reads, total number of SNVs, and phase rate, than the TSLRH platform.

Breeding and characteristics of Uram, a New Variety of Pleurotus nebrodensis (백령느타리 신품종 '우람'의 육성 및 특성)

  • This study was carried out to breed new variety of Pleurotus nebrodensis. We have collected and tested characteristics of genetic resources from domestic and abroad since 2015. The varieties of P. nebrodensis from China are grown by farmers, but those have been unstable fruiting and are weak against bacterial diseases. To solve this problem, we bred the unique domestic variety 'Uram' of P. nebrodensis and the results of the characteristic test for the new 'Uram' are as follows. The proper temperature for the mycelial growth was 26~29℃ and fruit body growth temperature was 15~18℃. It was similar to the control variety KME65035 of P. nebrodensis in the pileus form of a flat and white color. The number of days required for initial fruting was 5 days for bottle cultivation and 6 days for bag cultivation which was 2-4 days shorter than that of the control variety. The pileus diameter was 32.6-37.0 mm which was smaller but the fruit body length was 130.4 mm, which was longer than those of the control variety. The effective number of fruit bodies was 1.8 in bottle cultivation and 2.9 in bag cultivation, which was more than those of the control variety. The yield rate was 93.3-100%, which was more stable than those of the control variety. In bottle cultivation and bag cultivation, the yield was 173.1 g/bottle (1100 cc) and 283.4 g/bag (1.2 kg), respectively, which was 25-44% higher than those of the control variety 138.0 g/bottle (1100 cc) and 197.4 g bag (1.2 kg). When incubating the parent and control varieties of 'Uram', the replacement line was clear and as a result of mycelial DNA RAPD-PCR reaction, the band pattern was different from that of the parent and control varieties, confirming the hybrid species.

Characteristics of a new Hypsizigus marmoreus variety 'Yunseul' with little bitterness and high yield (쓴 맛이 적은 다수성 느티만가닥버섯 신품종 '윤슬')

  • This study was conducted to diversify the cultivation of mushroom items and develop a competitive variety of Hypsizigus marmoreus. We focused on developing Hypsizigus marmoreus with lower bitterness, likable shape, and high yield. We have collected and tested characteristics of genetic resources from domestic and abroad since 2019. Breeding (2019), characterization (2020), productivity test (2021), and farm demonstration test (2021) have been sequentially conducted. We bred the new variety 'Yunseul' with unique traits. The optimal temperature for mycelial and fruit body growth were 22-25℃ and 15-18℃, respectively. It was similar to the control variety (Mangadak-2Ho) in the pileus form (hemispherical shape) and the cultivation period. However, it was thinner and longer than the control variety with the pileus diameter, thickness, stipe diameter, and length being 19.9 mm, 7.0 mm, 9.4 mm, and 86.3 mm, respectively. The effective number of fruit bodies was 47.8 in bottle cultivation, which was more than that of the control variety. The yield was 197.4 g/bottle (1,100 cc), which was 30% higher than the yield of the control variety, 151.9 g/bottle (1,100 cc). The parent and control varieties were also incubated alongside the new variety. The somatic incompatibility line was distinct. The band pattern in the mycelial DNA PCR reaction was different from that of the parent and control varieties, confirming the hybrid species.
