Quantitative Analysis of Transient Expression in Tah Tasai Chinese Cabbage (Brassica campestris var. narinosa) Seedlings Following Agrobacterium-Mediated Transformation (다채 (Brassica campestris var. narinosa) 유묘의 형질전환 및 일시발현의 정량적 분석)

  • Tah tasai chinese cabbage (Brassica campestris var. narinosa), a vegetable plant popularly consumed as several-days-old seedlings in oriental countries, can be easily cultivated using a simple appliance. We demonstrated that Agrobacterium-mediated transformation via vacuum infiltration (agroinfiltration) resulted in a successful transient GUS gene expression in tah tasai chinese cabbage seedlings. Pre-germinated seeds were found to be more susceptible to Agrobacterium infection than one-day-old or two-days-old seedlings. We also demonstrated that hydrogen peroxide (HPO) treatment increased GUS expression especially for two-days-old seedlings. In ELISA using seedlings transformed with hepatitis B surface antigen (HBsAg) DNA by agroinfiltration, HBsAg protein synthesis increased more than two folds by HPO treatment to two-days-old seedlings in comparison to the mock-treated pre-germinated seeds.

Production and Characterizations of Somatic Hybrids between Brassica campestris L. ssp pekinensis and Brassica of oleracea L. var capitata

  • Protoplasts isolated from inbred lines of Brassica oleracea L. var capitata (cabbage) and Brassica campestris L. ssp. pekinensis (Chinese cabbage) were fused by PEG-mediated method, and somatic hybrid cells were differentiated into plants. for the identification of somatic hybrid plants, ploidy level, plant morphology, and cytological analysis were performed. All of the regenerated plants derived from fused protoplasts were shown to be 2X-4X, or higher ploidy level, presumably due to somatic hybridization or chromosome doubling. The morphology of leaves, petioles, and flowers showed an intermediate phenotype between Chinese cabbage and cabbage. Chromosome numbers in these somatic hybrids ranged mostly from 33 to 38. According to Genomic in situ hybridization (GISH) pattern, signals from both fusion parents of B.campestris or B.oleracea were detected in different colors when chromosomes of putative somatic hybrids were observed.

  • Objectives : Brassica campestris var narinosa (BCN), Canavalia gladiata DC semen (CGD) and their combinational prescription (BCN+CGD) have been use to demonstrate to regulate hematopoiesis. In the current study, we investigated whether Brassica campestris var narinosa, Canavalia gladiata DC semen and their combinational prescription is related to hemato-potentiating function using Sca-$1^+$ hematopoietic stem cells (Sca-$1^+HSCs$) as a testing system. Methods : Sca-$1^+HSCs$ isolated from femur in C57bl/6 mice with leukopenia and thrombocytopenia induced by cyclophosphamide (CTX). Then, Real-time PCR was performed to measure the mRNA expression, ELISA and haematopoiesis-related gene (EPO, TPO, IL-3, SCF, c-kit, GM-CSF), the phosphorylation of JAK2, GATA-1 and STAT-5a/b were observed by western blot, and the numbers of $CD117^+/Sca-1^+$ cell and the number of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E), semisolid clonogenic assay was performed. Result : When Sca-$1^+HSCs$ were treated with Brassica campestris var narinosa, Canavalia gladiata DC semen and their combinational prescription with rIL-3/rSCF, the expression of haematopoiesis-related (EPO, TPO, IL-3, SCF, c-kit, and GM-CSF) were significantly increased at the levels of mRNA as well as production in Sca-$1^+HSCs$. Additionally, CGS enhanced phosphorylation of JAK2, GATA-1, and signal transducer and activator of transcription-5a/b (STAT-5a/b) in Sca-$1^+HSCs$. Furthermore, their combinational prescription (BCN+CGD) significantly enhanced the growth rate of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E) in vitro. Conclusion : These result suggest that Brassica campestris var narinosa (BCN) and Canavalia gladiata DC have hematopoietic enhancement via hematopoietic cytokine-mediated JAK2/GATA-1/STAT-5a/b pathway, and their combinational prescription (BCN+CGD) has superior hematopoietic enhancement to those of individual extracts.

Resistance of Newly Introduced Vegetables to Meloidogyne arenaria and M. incognita in Korea (새로운 채소류의 고구마뿌리혹선충과 땅콩뿌리혹선충에 대한 저항성)

  • To select resistant vegetables against two species of root-knot nematodes, M. incognita and M. arenaria, 39 vegetables belongs to 7 families, 13 genera, 25 species were screened in greenhouse pot test. Susceptible vegetables to both nematodes were amarath and leaf beet in Amaranthaceae, Malabar spinach in Basellaceae, Moroheiya in Tiliaceae, and Water-convolvulus in Convolvulaceae, Pak-choi in Brassica campestris var. chinensis, Tah tasai in B. campestris var. narinosa, B. campestris var. chinensis x narinosa, Leaf mustard, Mustard green in B. juncea, Kyona in B. juncea var. laciniate, Choy sum in B. rapa subsp. arachinenesis, Kairan in B. oleracea var. alboglabra, Arugula in Eruca sativa, Garland chrysanthemum in Chrysanthemum coronarium, Endive in Cichorium endivia, Artichoke in Cynara cardunculus var. scolymus, Lettuce in Lactuca sativa. Resistant to M. arenaria but susceptible to M. incognita were B. oleracea cv. Matjjang kale, B. oleracea var. gongyloides cv. Jeok kohlrabi, and C. intybus cv. Radicchio. Resistant vegetables to both nematodes were C. intybus cv. Sugar loaf, Grumoro, Radichio treviso, B. oleracea cv. Manchu collard, Super matjjang, B. oleracea italica, B. oleracea var. botrytis italiana, and Perilla in Lamiaceae. Vegetables resistant to both species of root-knot nematodes could be used as high-valued rotation crops in greenhouses where root-knot nematodes are problem.

Selection of Promising 'Ssam' Vegetable for Summer Production in Highland (고랭지 여름출하용 유망 쌈채소 선발)

  • Field investigations were initiated to determine possible new 'ssam' vegetable that could be grown during the summer season in highland. Korean 'ssam' means cooked rice in vegetables. The 'ssam' vegetable is called that those leaves has been used for wrapping at meal. Seventeen leafy vegetables were studied, including chard (Beta vulgaris L. var. flavescens DC.), leaf broccoli (Brassica oleracea L. var. italica), Chinese leek (Allium tuberosum Rottl.), Japanese hornwort (Cryptotaenia japonica Hassk), red leaf mustard (Brassica juncea L.), green leaf mustard (Brassica juncea L.), leaf parsley (Petroselinum crispum Nijim), pakchoi (Brassica campestris L. ssp. chinensis Jusl.) tatsoi (Brassica campestris var. narisoma), kale (Brassica oleracea var. acephala), collard (Brassica oleracea var. acephala), Korean mint (Agastache rugosa O. Kuntze), romaine lettuce (Lactuca sativa L. var. longifolia Lam.), red leaf chicory (Cichorium intybus L. var. folisum), red chicory (Cichorium intybus L. var. folisum), green leaf chicory (Cichorium intybus L. var. folisum), and sprouting broccoli (Brassica oleracea L. var. italica). Date of the first harvest in 1999 was July 10th in chard, leaf broccoli, red mustard, mustard, leaf parsley, pakchoi, tatsoi, kale, collard, Korean mint, red leaf chicory, red chicory, green leaf chicory, and sprouting broccoli. Date of the first harvest for Chinese leek, Japanese hornwort, and romaine lettuce was from the middle of June to the late of August. Based on ratings on marketable yield, we found good leaf shape, taste and yield in chard, red leaf mustard, green leaf mustard, pakchoi, tatsoi, romaine lettuce, red leaf chicory, red chicory, and green leaf chicory. Even though the most of all these vegetables were obtained high scores by test panels in shape, taste and quality, but some of them revealed inadequate leaf size as a 'ssam' vegetable. That problem will be solved by control of harvesting time. On the basis of this observation, chard, red leaf mustard, green leaf mustard, pakchoi, tatsoi, romaine lettuce, red leaf chicory, red chicory, and green leaf chicory were rated as promising 'ssam' vegetable by analysis of profits, adaptation, and quality.

Host Preference of Gray Field Slug, Deroceras reticulatum Müller, and Its Development and Longevity on Host Plants (작은뾰족민달팽이의 기주 선호성 및 기주에 따른 발육과 수명)

  • This study was conducted to investigate food preference and developmental characteristics of the gray field slug, $Deroceras$ $reticulatum$ M$\ddot{u}$ller (Stylommatophora: Limacidae) using various foods. The food preference of $D.$ $reticulatum$ was most highest on Chinese cabbage ($Brassica$ $campestris$ L., followed on cucumber ($Cucumis$ $sativus$ L.), and significantly low on beet ($Beta$ $vulgaris$ var. Ruba), kale($Brassica$ $oleracea$ L. var. $acephala$ D.C), carrot (Daucus carota L.) and Chinese cabbage ($Brassica$ $campestris$ L.) which were not different significantly. The total number of eggs laid by $D.$ $reticulatum$ was the highest (n = 109.6) on cucumber and the lowest (n = 10.1) on leaf perilla ($Perilla$ $frutescens$ var. $japonica$ Hara). Hatchability was the highest (92.4%) on cucumber and the lowest (62.5%) on leaf perilla. The egg developmental period was not significantly different from 12 days to 13 days among host plants. Juvenile period and adult longevity ranged from 75 days to 111 days and 66 days to 187 days, respectively. Thereby, life span from egg to adult longevity of $D.$ $reticulatum$ was the longest on cucumber with 273 days and the shortest on 190 days on leaf perilla.

Morphological Characteristics and Composition of Cell Wall Polysaccharides of Brassica campestris var. pekinensis (Baechu) (배추조직의 형태학적 특성과 세포벽 다당류의 조성)

  • This study was conducted to examine morphological characteristics and the content of cell wall polysaccharides of Brassica campestris var. pekinensis(baechu). First of all, the variety of scientific name and naming of parts of baechu in the literatures of kimchi showed, which will unify marks. So, we propose not so much mid-rib and leaf blade of baechu leaf as white part and green part, respectively. On the other hand, the forms of vessel elements of white part in baechu consist in ring, sclariform and reticulate thickening. The proximate compositions and contents of cell wall polysaccharides of baechu has significant differences between its cultivars. The cell wall pectin from baechu exhibited four peals with molecular weights of 2,000,000, about 100,000 and less than 10,000 by gel filteration chromatography and hemicellulose did two peaks with molecular weights of 2,000,000 and 10,000.

Thioltransferase (Glutaredoxin) from Chinese Cabbage: Purification and Properties

  • Thioltransferase, also known as glutaredoxin, was purified from Chinese cabbage (Brassica campestris ssp. napus var. pekinensis) by a combination of ion-exchange chromatography and gel filtration. Its purity was confirmed by SDS-polyacrylamide gel electrophoresis and its molecular weight was estimated to be about 12,000 which is comparable with those of most known thioltransferases. The enzyme utilizes 2-hydroxyethyl disulfide, S-sulfocysteine, ${\alpha}-chymotrypsin$, insulin, and trypsin as substrates in the presence of reduced glutathione. The enzyme has Km values of 0.03-0.97 mM for these substrates. It appeared to contain dehydroascorbate reductase activity. The pH optimum of the enzyme was 8.5, when 2-hydroxyethyl disulfide was used as a substrate. It was greatly activated by reduced glutathione. Its activity was not significantly lost when stored at high temperature, indicating its thermostable character. It may play an important role in thiol-disulfide exchange in plant cells.
