2-Methylbutanoic, 3-methylbutanoic, 4-methylpentanoic, 2-ethylhexanoic, 4-methylhexanoic, 4-methyloctanoic, 6-methyloctanoic, 4-ethylhexanoic. 4-methylhexanoic 및 2-ethylhexanoic acids를 포함하는 다수의 volatile branched-chain fatty acids(VBCFAs)가 쇠고기, 돼지고기, 염소고기(미국산 흰염소고기, 한국산 흑염소고기), 양고기로부터 분리, 동정되었다. 쇠고기 향은 amino acid, carbohydrate, 그리고 lipid의 반응에 의해 생성된 향기성분에 의해 이루어지는 기본 meaty flavor로 결정되어 지는 반면, 염소고기가 양고기의 경우는 쇠고기의 기본향에 4-ethyloctanoic acid와 4-methyloctanoic acid의 독특한 노린내가 더해지므로 염소고기나 양고기 특유의 향을 내었다. 돼지고기는 3-methylbutanoic acid 함량이 다른 세종류에 비해 높고 3-methylbutanoic acid 특유의 꼬린냄새는 돼지고기의 기름층으로부터 나는 unclean flavor 에 중요한 영향을 미치는 것으로 생각된다. 염소고기는 C5-C7 chain length의 volatile branched 및 n-chain fatty acids를 적게 함유하고 있는 반면 4-ethyloctanoc acid는 염소고기의 노린내를 내는 결정적인 화합물이었다. 4-Methyloctanoic acid는 양고기의 sweaty-muttony flavor를 내는 중요한 화합물이었으며 염소고기와는 달리 C5, C6, 그리고 C7 branched-chain fatty acids 도 상당량 함유되어 있었다. 한국산 흑염소고기와 미국산 흰염소고기 중에 들어있는 VBCFAs의 종류는 같으나, 한국산 흑염소고기가 미국산 흰염소고기 보다 노린내가 강하며 4-ethyloctanoic 의 함량도 높았다.
The lubricating performance of 23 kinds of polyol ester base oils 〔POEs〕 having different branch shapes was investigated by using a four ball tribometer under boundary lubrication condition. All the polyol ester base oils used in this study were made up of polyhydric alcohols of two-four valence and normal or branched fatty acids of different carbon number. The wear characteristics of polyol ester base oils are different from those of mineral oil, strongly affected by the branch shapes of fatty acids in their molecles. In particular, the polyol ester base oils having normal fatty acids such as n-octanoic acid, n-nonanoic acid etc. show much better wear performance than POEs having branched fatty acids such as 2-ethylhexanoic acid, 3,5,5-trimethyl hexanoic acid, etc. As the carbon chain length of normal fatty acids, in case of POEs of normal fatty acids, is increased, their wear rate is decreased and, in case of POEs of branched fatty acids, as the degree of branch of branched fatty acids is decreased, their wear rate is decreased. All the wear results of polyol ester base oils could be reasonably explained by comparing cohesive ability among fatty acid molecules in adsorption film by fatty acids obtained as POEs were decomposed.

Objective: This study aims to identify the relationship between odd- and branched-chain fatty acids (OBCFAs) and microbial nucleic acid bases in the rumen, and to establish a model to accurately predict microbial protein flow by using OBCFA. Methods: To develop the regression equations, data on the rumen contents of individual cows were obtained from 2 feeding experiments. In the first experiment, 3 rumen-fistulated dry dairy cows arranged in a $3{\times}3$ Latin square were fed diets of differing forage to concentration ratios (F:C). The second experiment consisted of 9 lactating Holstein dairy cows of similar body weights at the same stage of pregnancy. For each lactation stage, 3 cows with similar milk production were selected. The rumen contents were sampled at 4 time points of every two hours after morning feeding 6 h, and then to analyse the concentrations of OBCFA and microbial nucleic acid bases in the rumen samples. Results: The ruminal bacteria nucleic acid bases were significantly influenced by feeding diets of differing forge to concentration ratios and lactation stages of dairy cows (p<0.05). The concentrations of OBCFAs, especially odd-chain fatty acids and C15:0 isomers, strongly correlated with the microbial nucleic acid bases in the rumen (p<0.05). The equations of ruminal microbial nucleic acid bases established by ruminal OBCFAs contents showed a good predictive capacity, as indicated by reasonably low standard errors and high R-squared values. Conclusion: This finding suggests that the rumen OBCFA composition could be used as an internal marker of rumen microbial matter.
저온 환경에서의 생장에 영향을 주는 지방산 합성 관련 유전자 bkdR, sigL, yplP, des들의 역할을 알아보기 위하여 각각 유전자들이 상실된 Bacillus subtilis CU1065와 JH642 돌연변이들을 제조하였다. 이들 유전자들의 저온 민감성을 확인하기 위해 $37^{\circ}C$와 $15^{\circ}C$에서 세포들의 생장을 측정하였다. $37^{\circ}C$에서 야생형과 결실 돌연변이 균주는 거의 유사한 정도의 생장을 보였으나, $15^{\circ}C$에서 오직bkdR 결실 돌연변이만이 야생형에 비해 매우 느린 생장이 관찰되었으며 sigL, yplP 결실의 경우 야생형에 비해 다소 느리거나 유사한 생장을 보였다. bkdR, sigL, yplP 결실에 대한 이중, 삼중 돌연변이를 만들어 LB agar에서 $20^{\circ}C$로 키워 저온생장을 조사한 결과, bkdR 결실이 포함되지 않은 어떤 이중, 삼중 결실들에서는 저온에 민감한 생장을 보이지 않았다. 온도 민감성 특성을 보다 잘 알아보기 위하여 $37^{\circ}C$에서 $OD_{600}=0.4$까지 키워 $15^{\circ}C$로 온도를 내리는 저온충격 조건에서 생장하는 실험을 진행하였다. 이 실험에서 오직bkdR 결실 돌연변이만이 현저히 낮은 생장을 보였으며 추가적인 des 결실은 저온 민감성을 증가시킨다. bkdR은 branched-chain fatty acid을 합성하는 전구물질인 isoleucine, valine, leucine 아미노산을 생산하는 bkd operon을 활성화한다. bkdR 결실 돌연변이의 저온생장에서 이들 아미노산의 저온생장에 미치는 영향을 조사한 결과 isoleucine은 bkdR 결실에 대한 저온 민감성을 회복시켜주나 valine은 저온 민감성을 회복시켜 주지 못하는 결과를 보였다. isoleucine은 분해되어 anteiso-branched 지방산 합성의 전구물질로 만들어지는 반면에, valine은 iso-branched 지방산 합성의 전구물질로 만들어진다. 따라서 저온생장에서 branched-chain fatty acid 중 anteiso-branched 지방산이 중요한 역할을 하고 있음을 알 수 있었다.
Objective: This study aimed to investigate the effect of dietary supplementation with Allium mongolicum Regel extracts on the growth performance, carcass characteristics, fat color, and concentrations of three branched-chain fatty acids related to flavor in ram lambs. Methods: Sixty 3-month-old, male, small-tailed Han sheep were selected and randomly allocated into four groups in a randomized block design. Four feeding treatments were used: i) a basal diet without supplementation as the control group (CK); ii) the basal diet supplemented with 10 g/lamb/d Allium mongolicum Regel powder as the AMR group; iii) the basal diet supplemented with 3.4 g/lamb/d Allium mongolicum Regel water extract as the AWE group; and iv) the basal diet supplemented with 2.8 g/lamb/d Allium mongolicum Regel ethanol extract as the AFE group. Results: The results demonstrated that the dry matter intake was lower for the AFE group than that in other groups (p = 0.001). The feed conversion ratio was greater for the AFE than that in other groups (p = 0.039). Dietary supplementation with Allium mongolicum Regel powder and its extracts decreased the concentrations of 4-methyloctanoic acid (MOA) (p<0.001), 4-ethyloctanoic acid (EOA) (p<0.001), and 4-methylnonanoic acid (MNA) (p = 0.044) in perirenal adipose tissue compared to those observed in the CK lambs. Dietary supplementation with Allium mongolicum Regel powder and its extracts decreased the concentrations of MOA (p<0.001) and EOA (p<0.001) in dorsal subcutaneous adipose tissue compared to those in the CK lambs. The concentrations of MOA (p<0.001) and EOA (p = 0.002) in omental adipose tissue were significantly affected by treatment, although there was a tendency for lower MNA (p = 0.062) in AMR, AWE, and AFE lambs than that in CK lambs. Conclusion: This study demonstrated that Allium mongolicum Regel and its extracts could significantly promote feed efficiency, although dry matter intake decreased and could decrease the MOA and EOA concentrations related to characteristic flavor and odor of body fat in lambs, except for tail adipose tissue.

NdgR, a global regulator in soil-dwelling and antibiotic-producing Streptomyces, is known to regulate branched-chain amino acid metabolism by binding to the upstream region of synthetic genes. However, its numerous and complex roles are not yet fully understood. To more fully reveal the function of NdgR, phospholipid fatty acid (PLFA) analysis with gas chromatography-mass spectrometry (GC-MS) was used to assess the effects of an ndgR deletion mutant of Streptomyces coelicolor. The deletion of ndgR was found to decrease the levels of isoleucine- and leucine-related fatty acids but increase those of valine-related fatty acids. Furthermore, the defects in leucine and isoleucine metabolism caused by the deletion impaired the growth of Streptomyces at low temperatures. Supplementation of leucine and isoleucine, however, could complement this defect under cold shock condition. NdgR was thus shown to be involved in the control of branched-chain amino acids and consequently affected the membrane fatty acid composition in Streptomyces. While isoleucine and valine could be synthesized by the same enzymes (IlvB/N, IlvC, IlvD, and IlvE), ndgR deletion did not affect them in the same way. This suggests that NdgR is involved in the upper isoleucine and valine pathways, or that its control over them differs in some respect.
수영만에서 분리된 Bacillus cereus group LS-1 의 형태학적, 생화학적 성상 및 지방산 조성 분석을 통하여 세균학적인 특성을 밝혔다. B. cereus group LS-1 은 면양혈구배지에서 비용혈성의 점조성 집략과 convex 하고 가장자리가 규칙적인 둥근형태로 운동성이 없고 glucose, maltose, sucrose 와 gelatin 을 이용하고 trehalose 와 salicin 을 분해하지 않으며 6.5% NaCl 에서 자라지 않는 Gram 양성의 중심성 아포형성간균으로 표준균주 B. cereus group 과 다소 차이를 나타내었다. 지방산 조성 분석에서 chain 의 길이가 $C_{12}$ 에서 $C_{17}$로 iso $C_{15}$와 iso $C_{13}$의 branched chain 이 우점하는 B. cereus group 의 전형적인 특징을 나타내었으며 $nC^{15}$가 검출되지 않는 B.mycoides GC subgroup B 로 0.312 의 similarity index(SI) 를 지칭하여 다른 연구 결과와도 일치하였다. 한편 API system (API 50 CHB & API 20E) 의 ATB computer profile 에서 "Doubful Profile" 99.7% 의 B. firmus 로 나타내어 큰차이를 나타내었다. 67 개의 biochemical character 로 B. mycoids S-12 는 각각 42%, 42% 59% 와 52% 의 similarity matrix 를 나타내었다. B. cereus group 간의 아주 낮은 similarity 를 나타내어 상당한 차이가 있음을 인식하였다. 따라서 key test 와 지방산 조성등을 종합하여 볼때 B. ceresu group 중의 B. mycoides 로 새로운 biotype 인 것으로 사료되며 지방산 조성 분석으로 동정함이 훨씬 용이하였다.
Severe protein-calorie malnutrition, common in patients with advanced liver disease, can seriously undermine the capacity for regeneration and functional restoration of liver. Nutritional supplementation for these patients can improve biochemical and hormonal abnormalities. However, these effects were not identified in patient with nonalcoholic liver cirrhosis. To determine effects of nutritional supplementation in patients with nonalcoholic liver cirrhosis, 77 subjects aged 29 to 69 years participated in this study for 12 weeks and were subdivided into three groups; normal diet group (Control group, n = 16), branched-chain amino acid supplementation group (BCAA group, n = 31), nutritional supplementation group (NS group, n = 30). Anthropometric parameters, hemoglobin, hematocrit, blood cell counts, serum levels of lipids, vitamins, minerals and fatty acid composition, and plasma amino acids were examined. The mean values of age and height, and the initial values of weight and body mass index (BMI) were not different among all groups. After 12 weeks, there were no significant changes in these values in Control group. Only NS group showed significant increases in weight, lean body mass, midarm circumference, triceps skinfold thickness. Serum transferrins were increased both in BCAA and NS groups. Plasma levels of branched-chain amino acids, urea amino acids and glutamic acid were also significantly increased in these groups, but plasma levels of ammonia, serum LDL cholesterol and atherogenic index were decreased. However, there were no significant changes in serum levels of vitamin and mineral and composition of fatty acids in phospholipids in these groups. These results showed that the nutritional supplementation for patients with nonalcoholic liver cirrhosis can more improve nutritional status in these people together with increases of weight, body fat and lean body mass, compared to only BCAA supplementation. To ascertain and investigate the appropriate nutritional supplementation for patients with nonalcoholic liver cirrhosis, further studies are necessary.
This study was performed to investigate the effect of different levels of dietary crude protein (CP) on composition of odorous compounds and bacterial communities in pig manure. A total of 48 male pigs (average initial body weight 45 kg) fed diets containing three levels of dietary CP (20%, 17.5%, and 15%) and their slurry samples were collected from the pits under the floor every week for one month. Changes in composition of odorous compounds and bacterial communities were analyzed by gas chromatography and 454 FLX titanium pyrosequencing systems, respectively. Levels of phenols, indoles, short chain fatty acid and branched chain fatty acid were lowest (p<0.05) in CP 15% group among three CP levels. Relative abundance of Bacteroidetes phylum and bacterial genera including Leuconostoc, Bacillus, Atopostipes, Peptonphilus, Ruminococcaceae_uc, Bacteroides, and Pseudomonas was lower (p<0.05) in CP 15% than in CP 20% group. There was a positive correlation (p<0.05) between odorous compounds and bacterial genera: phenol, indole, iso-butyric acid, and iso-valeric acid with Atopostipes, p-cresol and skatole with Bacteroides, acetic acid and butyric acid with AM982595_g of Porphyromonadaceae family, and propionic acid with Tissierella. Taken together, administration of 15% CP showed less production of odorous compounds than 20% CP group and this result might be associated with the changes in bacterial communities especially whose roles in protein metabolism.
