Objectives : We performed an in vivo experiment to investigate the effect of $^{166}Holmium$ and $^{166}Holmium$-chitosan complex($^{166}Ho$-CHICO) on the normal brain of rats and to determine the sublethal dose of $^{166}Ho$-CHICO. Materials and Methods : $^{166}Ho$ is a beta and gamma ray emitter. $^{166}Ho$-CHICO is a novel radio-pharmaceutical complex with chitosan to facilitate the transport of $^{166}Ho$ obtained from Korea Atomic Energy Research Center(Taejon, Korea). It is in acidic form and becomes gel state at alkaline pH. One hundred and seventy consecutive rats were divided into four groups : $^{166}Ho$ treated(n=50), $^{166}Ho$-CHICO treated(n=57), saline treated(n=5) and chitosan treated(n=5) groups. $^{166}Ho$ and $^{166}Ho$-CHICO were injected into the rat brain stereotactically with various doses of 0.1mCi/$20{\mu}l$, 0.2mCi/$20{\mu}l$, 0.3mCi/$20{\mu}l$, and 0.4mCi/$20{\mu}l$ using an automated microinjector. Nuclear imaging, histopathological and hematological studies were performed in 10 rats in each group at 1 day, 3days, 7 days, 1 month and 3 months after the injections. Results : An infiltration of inflammatory cells and necrotic changes were noted in $^{166}Ho$ treated group at 1 week after the injection. A wedge-shaped tissue defect due to necrosis, lined with infiltrated glial cells in $^{166}Ho$ treated group and a cystic defect lined with reactive astroglial cells in $^{166}Holmium$-CHICO treated group at 3 months after the injection were observed. $^{166}Ho$ alone without chitosan leaked out and caused necrotic lesion on the cerebral surface but $^{166}Holmium$-CHICO treated group did not show this feature. As the dose of $^{166}Ho$ increased, the mortality rates were also increased. The mortality rate of the $^{166}Holmium$-CHICO group was higher than the $^{166}Ho$ treated group at a dose of 0.4mCi/$20{\mu}l$/300g. There was no detectable radioactivity due to the leakage or extravasation from the injected site of the brain on the scintigraphy performed at 1 hour, 24 hours and 48 hours after the injection. There was also no detectable activity of $^{166}Holmium$-CHICO in other organs including spleen, liver and kidney. Conclusions : $^{166}Ho$-CHICO did not leak out to the critical cortical surface of the brain from the injection site and induced radiation changes of the parenchyma around the injection site without cortical damage. The sublethal dose of $^{166}Ho$-CHICO for the normal brain in rats was determined to be 0.2mCi/$20{\mu}l$/300g.
The kinesin proteins (KIFs) make up a large superfamily of molecular motors that transport cargo such as vesicles, protein complexes, and organelles. KIF5 is a heterotetrameric motor that conveys vesicles and plays an important role in neuronal function. Here, we used the yeast two-hybrid system to identify the neuronal protein(s) that interacts with the tail region of KIF5 and found a specific interaction with ${\beta}III$ spectrin. The amino acid residues between 1394 and 1774 of ${\beta}III$ spectrin were required for the interaction with KIF5C. ${\beta}III$ spectrin also bound to the tail region of neuronal KIF5A and ubiquitous KIF5B but not to other kinesin family members in the yeast two-hybrid assay. In addition, these proteins showed specific interactions, confirmed by GST pull-down assay and co-immunoprecipitation. ${\beta}III$ spectrin interacted with GST-KIF5 fusion proteins, but not with GST alone. An antibody to ${\beta}III$ spectrin specifically co-immunoprecipitated KIF5s associated with ${\beta}III$ spectrin from mouse brain extracts. These results suggest that KTF5 motor proteins transport vesicles or organelles that are coated with ${\beta}III$ spectrin.
Tau proteins, which stabilize the structure and regulate the dynamics of microtubules, also play important roles in axonal transport and signal transduction. Tau proteins are missorted, aggregated, and found as tau inclusions under many pathological conditions associated with neurodegenerative disorders, which are collectively known as tauopathies. In the adult human brain, tau protein can be expressed in six isoforms due to alternative splicing. The aberrant splicing of tau pre-mRNA has been consistently identified in a variety of tauopathies but is not restricted to these types of disorders as it is also present in patients with non-tau proteinopathies and RNAopathies. Tau mis-splicing results in isoform-specific impairments in normal physiological function and enhanced recruitment of excessive tau isoforms into the pathological process. A variety of factors are involved in the complex set of mechanisms underlying tau mis-splicing, but variation in the cis-element, methylation of the MAPT gene, genetic polymorphisms, the quantity and activity of spliceosomal proteins, and the patency of other RNA-binding proteins, are related to aberrant splicing. Currently, there is a lack of appropriate therapeutic strategies aimed at correcting the tau mis-splicing process in patients with neurodegenerative disorders. Thus, a more comprehensive understanding of the relationship between tau mis-splicing and neurodegenerative disorders will aid in the development of efficient therapeutic strategies for patients with a tauopathy or other, related neurodegenerative disorders.
Background: Wilson disease (WD) is an autosomal recessive disorder of copper transport and characterized by degenerative changes in the brain, liver dysfunction, and Kayser-Fleischer rings due to toxic accumulation of copper. Since the identification of Wilson disease gene (ATP7B), more than 80 mutations have been detected among the different ethnic groups. Methods: Twenty three children with Wilson disease were included in this study. They were all diagnosed by low serum ceruloplasmin and increased 24 hour urinary copper excretion with characteristic clinical findings. We analysed WD gene mutation by assessing the nucleotide sequence of exon 7, 8, 9 and 10 including intron-exon boundaries of ATP7B gene from genomic DNA. Results: Arg778Leu mutation was identified in 16 WD patients; three were homozygous and 13 were heterozygous for this mutation. Of the 46 alleles, 19 alleles had a Arg778Leu mutation (19/46=41%). Homozygote patients had neurologic forms of WD. Arg778Leu mutation was not found among 50 normal healthy persons. Conclusion: Arg778Leu mutation is a common mutation in Korean WD gene. Arg778Leu mutation screening might be used as a useful supplementary diagnostic test in some patients to confirm Wilson disease in Korea.
Dose monitoring in CT patients requires accurate dose estimation but most of the CT dose calculation tools are based on Caucasian computational phantoms. We established a library of organ dose conversion coefficients for Korean adults by using four Korean adult male and two female voxel phantoms combined with Monte Carlo simulation techniques. We calculated organ dose conversion coefficients for head, chest, abdomen and pelvis, and chest-abdomen-pelvis scans, and compared the results with the existing data calculated from Caucasian phantoms. We derived representative organ doses for Korean adults using Korean CT dose surveys combined with the dose conversion coefficients. The organ dose conversion coefficients from the Korean adult phantoms were slightly greater than those of the ICRP reference phantoms: up to 13% for the brain doses in head scans and up to 10% for the dose to the small intestine wall in abdominal scans. We derived Korean representative doses to major organs in head, chest, and AP scans using mean CTDIvol values extracted from the Korean nationwide surveys conducted in 2008 and 2017. The Korean-specific organ dose conversion coefficients should be useful to readily estimate organ absorbed doses for Korean adult male and female patients undergoing CT scans.
Kinesin-2는 heterotrimeric kinesin-2와 homodimeric kinesin-2의 두종류가 존재한다. 세포내 미세소관을 따라 이동하는 분자 모터 단백질인 heterotrimeric kinesin-2는 motor 활성을 가진 kinesin superfamily protein (KIF) 3A와 3B (KIF3A와 KIF3B) 그리고 kinesin associated protein 3 (KAP3)로 구성되어져 있다. 특히 섬모내에서 heterotrimeric kinesin-2는 미세소관의 plus방향으로 운반체를 운반하는 역할을 수행한다. Heterotrimeric kinesin-2는 다양한 운반체 그리고 단백질들과의 결합이 알려져 있지만 heterotrimeric kinesin-2와 결합하는 결합단백질에 대하여서는 아직 충분히 밝혀지지 않았다. 본 연구에서는 KIF3A의 C-말단 영역과 결합하는 단백질을 효모 two-hybrid system을 사용하여 탐색한 결과, brain expressed X-linked 2 (Bex2)를 분리하였다. Bex2는 KIF3A의 cargo-binding domain (CBD)을 포함하는 C-말단 영역과 결합하지만, KIF3B와 kinesin-1의 motor 단백질인 KIF5A와는 결합하지 않았다. 그리고 KIF3A는 Bex2의 다른 isoform인 Bex1과는 결합하지만, Bex3와는 결합하지 않았다. KIF3A은 GST-Bex1, GST-Bex2와는 결합하지만 GST와는 결합하지 않았다. HEK-293T세포에 Bex2을 발현시켰을 때 Bex2와 KIF3A는 세포 내의 같은 부위에 발현하며, Bex2 그리고 KIF3A을 면역침강한 결과 KIF3A와 KIF3B도 같이 침강함을 확인하였다. 이러한 결과들은 Bex2는 heterotrimeric kinesin-2와 운반체를 매개하는 단백질로의 가능성을 시사한다.
미생물이 이용할 수 있는 nitrogen source는 di-, tri-, oli- go-peptide 또는 amino acid의 형태로 세포내로 uptake되어 대사과정에 사용되고 있다. 이와같은 peptide는 특이한 transport system에 의해서 이동되고 있는데 oligo peptide(Opp) transport system에는 binding protein, permease protein, energy 생성을 위한 ATP 분해에 관여하는 protein 이 관여하고 있으며 염색체 상에서 이들 단백질들은 operon 형태의 유전자로부터 발현되고 있다. 본 연구는 gram 음성 세균이며 수해양 서식 세균인 V, fluvialis로부터 얻어진 Opp operon 유전자 가운데 oligopeptide binding protein을 coding하고 있는 oppA 유전자가 deletion된 mutant를 사용하여 여러 환경변화에 따른 생육을 wild type과 비교한 연구 결과 이다. 생육을 위한 완전배지인 brain heart infusion (BHI) 배지와 최소배지인 M9 minimal 배지를 사용한 결과 OppA protein의 생성 결핍에 따라 초기 및 대수증식기 과정 중에는 mutant의 생육이 늦어지고 있으나 Opp system이 아닌 다른 peptide전달 경로로 추정되는 system을 이용하여 대수 증식기 후반에서는 wild type과 거의 같은 생육 형태를 보여 주고 있었다. pH의 변화에 따른 생육은 pH 7에서는 생육정도가 비슷하였으나 약알칼리 부근에서는 oppA mutant의 생육이 wild type에 비하여 낮아지고 있었다. 또한 5 mM $H_2O_2$를 사용하여 $OD_{600}=1.2$농도의 세포들에 대한 영향을 검토한 결과 두 균 모두 높은 생존율을 보여 주었으며 이는 대수증식기 세포들을 사용한 결과와는 매우 다른 형태를 보여 주고 있었다. 항생제 내성에 대한 연구에서는 mutant가 streptomycin과 tetracycline 에 대해서는 wild type과는 다르게 매우 낮은 농도에서도 생육이 되고 있지 않으나 polymyxin B에 대해서는 wild type과 같이 $10{\mu}g/ml$의 농도에서도 잘 자라고 있었다.
캡사이신 채널로 알려진 바닐로이드 수용체 TRPV1 (캡사이신채널, Transient Receptor Potential Vanilloid 1)은 통증발현에서 중요한 역할을 하는 것으로 알려져 있다. 하지만 TRPV1의 활성조절에 관여하는 단백질에 대하여는 알려진 바가 많지 않다. 최근 rat TRPV1과 직접적으로 결합하는 단백질을 탐색하여 mouse Rab11-FIP3 (rab11-family interaction protein 3)가 rat TRPV1과 직접적으로 결합한다는 것이 보고되었다. Rab11은 여러 가지의 세포내 이동에 관여하는 것으로 보고되었다. 그러므로 Rab11-FIP3과의 결합을 통해 TRPV1의 세포막으로의 이동에 관여할 것으로 추측할 수 있다. 본 연구에서는 전에 보고된 연구가 mouse와 rat 이라는 다른 종의 단백질끼리의 결합이기 때문에 같은 종에서의 상호작용을 확인하고 Rab11-FIP3의 TRPV1의 세포막으로의 이동에서의 역할을 알아보고자 현재까지 동정되지 않은 rat의 Rab11-FIP3의 유전자를 GenBank 서열을 바탕으로 rat 뇌의 RNA 로부터 cDNA 를 클로닝하여 유전자를 분리하고 TRPV1 과의 관계를 세포생물학적으로 알아보았다. 연구결과 rat의 Rab11-FIP3는 489개의 아미노산 서열을 가지고 있으며 human과는 80%, mouse와는 90% 이상 아미노산 서열의 상동성을 보였다. 조직별 분포는 심장, 뇌, 간, 콩팥, 정소에서 발현되고 있는 것을 northern blot assay와 western blot assay 로 확인하였다. rat 의 뇌조직에서 TRPV1 과 Rab11-FIP3 단백질이 결합하여 colocalize 하는 것을 면역화학방법으로 확인하였다. 이 결합은 같은 family 의 TRPV2 와는 결합하지 않는 특이적 결합이므로 Rab11-FIP3 가 TRPV1 과 상호작용하여 세포막으로의 이동에 관여할 것이라는 것을 시사한다.
소 뇌 미세혈관에서 분리한 내피세포(BBMECs)를 직경 $3.0{\mu}m$및 $0.4{\mu}m$인 구멍을 가지는 다공성막(Transwell)에서 일차배양하였을 때의 특징을 전자현미경을 사용하여 살펴보았다. 분리된 모세혈관의 작은 조각과 분리된 내피세포들은 콜라겐으로 도포한 배양기구의 표면에 고착되어 성장하였다. BBMECs들은 직경 $0.4{\mu}m$인 다공성막의 위쪽 구획에서만 성장하였으나 직경 $3.0{\mu}m$인 구멍을 가진 막에서는 세포들이 구멍을 통해 막의 반대쪽으로 이주하여 다공성막의 아래쪽 구획에서도 성장하여 세포단층을 형성하였다. 이상의 결과로 효소 처리에 의해 분리한 BBMECs는 직경 $0.3{\mu}m$의 다공성막을 통과하나 직경 $0.4{\mu}m$의 다공성막을 통과할 수 없음을 알 수 있었으며, 약물이동도를 관찰하는 실험이나 전기저항을 측정을 목적으로 하는 실험에서는 $3.0{\mu}m$의 다공성막을 사용하는 대신 $0.4{\mu}m$ 크기의 다공성막을 사용해야 한다는 것을 알 수 있었다.
페닐케톤뇨증(PKU)은 전세계적으로 가장 잘 알려지고 중요한 유전성 대사질환이다. 1950년대 이후 단백제한을 이용한 식사치료를 처음으로 시도하여 성과가 있었던 질환이며 1960년대 이후 신생아선별검사를 통해 조기진단과 조기치료가 가능하게 된 최초의 유전성대사질환이기 때문이다. 단백제한 식사치료의 효과가 좋지만 학동기, 사춘기 이후 성인시기까지 유지하는 것의 어려움이 있고 이시기에 조절이 잘 되지 않았을 경우 경련, 여러가지 정신과적인 문제들, 삶의 질의 감소 등이 문제가 되어서 오랜 기간 치료를 위한 여러 방법들이 제시되었다. 더해서 2014년 미국의학유전학회(American Medical College of Medical Genetics and Genomics, ACMG)에서 전 연령에서 혈중 페닐알라닌 수치를 120-360 umol/L로 제시를 한 이후 더욱 치료의 중요성이 올라갔다. 2000년대 페닐알라닌수산화 효소(phenylalanine hydroxylase, PAH)의 조효소인 tetrahydrobiopterin (BH4)가 치료 승인되어서 약물반응을 보이는 환자에서 치료가 시작되었으며 4세미만에서도 허가가 되어서 이른 시기부터 약물치료를 병행하여 효과를 보게 되었다. 높은 혈중 페닐알라닌수치가 혈액-뇌 장벽(Blood-brain barrier, BBB)을 통하여 뇌로 넘어가서 회백질의 변성을 나타내게 되는 문제를 막기 위해 거대중성아미노산(LNAA)를 이용한 치료가 시도되고 있다. 오랫동안 연구되었던 페닐알라닌을 trans-cinnamic acid와 암모니아로 변화를 시키는 phenylalanine ammonialyase (PAL)을 이용한 효소치료는 최근 약제로 개발되어서 2018년 이후 성인환자를 대상으로 치료가 시작되었고 잘 조절되지 않는 환자들에게 효과를 보이고 있다. PAL을 경구용으로 개발하는 것이 빠르게 진행 중이며 유전자치료에 대한 연구들도 활발하게 진행이 되고 있어 다양한 치료들이 앞으로 기대된다.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
