PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 지실의 세포보호효과 연구 (Protective Effect of Aurantii Immaturus Fructus on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil)

  • Object : This research was performed to investigate the protective effect of Aurantii Immaturus Fructus against ischemic damage using PC12 cells and global ischemia in gerbils. Methods : To observe the protective effect of Aurantii Immaturus Fructus on ischemia damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Aurantii Immaturus Fructus during ischemic insult. Gerbils were divided into three groups : a normal group, a 5-min two-vessel occlusion (2VO) group, and an Aurantii Immaturus Fructus administered after 2VO group. The CCAs were occluded by microclip for 5 minutes. Aurantii Immaturus Fructus was administered orally for 7 days after 2VO. The histological analysis was performed at 7 days after the surgery. For histological analysis, the brain tissue was stained with 1% cresyl violet solution. Results : The results showed that 1. Aurantii Immaturus Fructus had a protective effect against ischemia in the CAI area of the gerbil hippocampus 7 days after 5-minute occlusion, 2. In the hypoxia/reperfusion model using PC12 cells, the Aurantii Immaturus Fructus had a protective effect against ischemia in the dose of $0.2{\;}\mu\textrm{g}/ml,{\;}2{\;}\mu\textrm{g}/ml{\;}and{\;}20{\;}\mu\textrm{g}/ml$ 3. Aurantii Immaturus Fructus increased the activities of glutathione peroxidase and catalase, 4. The activity of superoxide dismutase (SOD) was increased by ischemic damage, which might represent self protection. This study suggests that Aurantii Immaturus Fructus has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils, and it also has protective effects on a hypoxia/reperfusion cell culture model using PCq2 cells. Conclusions : Aurantii Immaturus Fructus has protective effects against ischemic brain damage at the early stage of ischemia.

Doxorubicin에 의해 활성화된 미세 아교세포의 면역반응으로 인한 신경손상에 Noni가 미치는 영향 (Noni Inhibits Neuronal Damage Caused by the Immune Reaction of Microglial Cells Activated by Doxorubicin)

  • Microglial cells function as major immune cells in the brain, playing an important role in the protection and damage of neurons. BV2 microglia, activated by drug stimulation, secrete inflammatory cytokines by activating the nuclear factor kappa-light-chain-enhancer of the activated B cells pathway and are involved in neuroinflammatory and immune responses. The overactivation of microglia by stimuli can cause neuronal damage, leading to brain disease. Noni, a natural product, reduces the activity of microglia to prevent neuronal damage and is a potential natural medicine because it exerts excellent regeneration and anti-inflammatory effects on damaged cells. In this study, when noni was used to treat BV2 cells stimulated by the anti-cancer drug doxorubicin, it reduced the release of pro-inflammatory cytokines from BV2. On the other hand, neuronal damage is a side effect of doxorubicin. Therefore, the cytokines released from doxorubicin-stimulated BV2 cells treated with noni had a positive effect on the neuronal viability compared to those released from doxorubicin-stimulated BV2 cells not treated with Noni. Thus, Noni increases neuronal viability. These results suggest that noni inhibits the release of cytokines by regulating the nuclear factor kappa-light-chain-enhancer of the activated B cells pathway of BV2, thereby inhibiting neuronal damage.

사지 실행증의 평가 및 신경생리학적 고찰 (Assessment and Neurophysiology of the Limb Apraxia: Review Article)

  • The purpose of this study was to review the limb apraxia. It includes the evaluation and neurophysiological aspects for limb apraxia. Limb apraxia comprised a wide spectrum of higher-order motor disorders that results from acquired brain disease affecting the performance of skilled and/or learned movements with the forelimbs and is a common sequela of left brain damage that consists of a deficit in performing gestures to verbal command or to imitation. There are two forms in limb apraxia; ideational apraxia and ideomotor apraxia. A assessment of limb apraxia typically includes pantomiming and imitation of transitive, intransitive, and meaningless gestures. Limb apraxia has been attributed to damage confined to the cerebral cortex, cortico-cortical connecting pathways, and basal ganglia.

Post-Traumatic Cerebral Infarction Following Low-Energy Penetrating Craniocerebral Injury Caused by a Nail

  • Post-traumatic cerebral infarction (PTCI) is a secondary insult which causes global cerebral hypoxia or hypoperfusion after traumatic brain injury, and carries a remarkable high mortality rate. PTCI is usually caused by blunt brain injury with gross hematoma and/or brain herniation. Herein, we present the case of a 91-year-old male who had sustained PTCI following a low-energy penetrating craniocerebral injury due to a nail without evidence of hematoma. The patient survived after a decompressive craniectomy, but permanent neurological damage occurred. This is the first case of profound PTCI following a low-energy penetrating craniocerebral nail injury and reminds clinicians of possibility this rare dreadful complication for care of head-injured patients.

석곡(石斛)이 흰쥐의 뇌조직출혈에 미치는 영향 (Effects of Dendrobii herba against Intracerebral Hemorrhage in Rats)

  • Objects : This study was performed in order to observe the effects of water extract of Dendrobii herba on intracerebral hemorrhage(ICH), Method : ICH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. After the water extracts of Dendrobii herba were administrated orally once a day for 3 days, hematoma volume, percentage of brain edema, expression of iNOS and MPO were observed using immunohistochemistry. Results : Rats fed with water extracts of Dendrobii herba showed reduction of hematoma volume and percentage of brain edema compared with controls. In addition, Infiltration of myeloperoxidase (MPO) expressing neutrophil and expression of inducible nitric oxide synthatase(iNOS) were significantly reduced in rats fed with water extracts of Dendrobii herba. Conclusion : These results demonstrated that water extracts of Dendrobii herba reduced brain damage of intracerebral hemorrhage(ICH) and subsequent ICH-induced cerebral edema, and inhibited neutrophil infiltration.

  • Neuroendoscopic surgery is performed because it causes minimal damage to normal structures, carries a lower rate of complications, and achieves excellent outcomes. Surgeons using an endoscope and related instruments can perform complex operations through very small incisions, which is especially useful for minimally invasive procedures for the brain and spine. Neuroendoscopic surgery is now performed in cases of obstructive hydrocephalus, various intraventricular lesions, hypothalamic hamartomas, craniosynostosis, skull base tumors, and spinal lesions. This review discusses the brief history of neuroendoscopy and the current state and future perspectives of endoscopic surgery.

An Immunohistochemical Study of Viral Antigen in Aborted Fetuses Naturally Infected by Bovine Viral Diarrhea Virus

  • The tissue distribution and cellular localization of viral antigen in the brain of aborted fetus with bovine viral diarrhea virus(BVDV) infection was studied; BVDV antigens was detected in spleen, kidney, lung, eyelid as well as brain. In the brain, the virus was recognized in neurons and non-neuronal cells in the cerebellum and cerebrum. Many cells in the superficial layer and occasional Purkinje cells had BVDV antigens. As well, BVDV was also found in the perivascular cells, vascular endothelial cells and smooth muscle cells in the vessels and neuroglial cells in the white matter. This finding suggests that BVD virus favors infect progenitor cells in the brain, notably in the superficial layer of cerebellum, and damage normal development of cerebellum, which leads to cerebellar hypoplasia.

  • The proton magnetic resonance spectroscopy ($^1H-MRS$) is a tool used to detect concentrations of brain metabolites such as N-acetyl aspartate, choline, creatine, glutamate, and gamma-amino butyric acid (GABA). It has been widely used because it does not require additional devices other than the conventional magnetic resonance scanner and coils. Demyelination, or the neuronal damage due to loss of myelin sheath, is one of the common pathologic processes in many diseases including multiple sclerosis, leukodystrophy, encephalomyelitis, and other forms of autoimmune diseases. Rodent models mimicking human demyelinating diseases have been induced by using virus (e.g., Theiler's murine encephalomyelitis virus) or toxins (e.g., cuprizon or lysophosphatidyl choline). This review is an overview of the MRS findings on brain metabolites in demyelination with a specific focus on rodent models.

알코올 선별 검사법(Alcohol Use Disorders Identification Test)과 뇌 백질 영역의 확산텐서 비등방도 계측 값의 관련성 (Relationship between Alcohol Use Disorders Identification Test Fractional Anisotropy Value of Diffusion Tensor Image in Brain White Matter Region)

  • AUDIT(Alcohol Use Disorders Identification Test)에서 정상 음주자, 위험 음주자 및 알코올 사용 장애자로 분류하여 뇌 백질 영역의 ROI(Region Of Interest)에 대한 FA 측정값을 분석하였다. TBSS(Tract-Based Spatial Statics) 도구를 사용하여 ROI의 FA 값을 추출하였다. TBSS라는 도구는 뇌 회백질보다는 백질에 대한 FA값과 MD 값의 민감도가 더 높고 뇌 신경섬유의 비등방도를 정량적으로 도출해 낼 수 있는 장점이 있고 백질에 더 특화된 프로그램이라 할 수 있다. 특히 양쪽 맥락총 평균 차이가 높았고 정상 음주자에서는 FA의 평균값이 0.2831과 0.2872로 나타났으며, 알코올 사용 장애자의 경우 0.1972와 0.1936로 나타났다. 즉, AUDIT 척도에서 점수가 높을수록 뇌 백질의 계측한 모든 ROI 영역에서 FA 값이 더 낮게 측정되는 것을 알 수 있었으며 뇌 백질에 신경 섬유로의 손상에 대한 미세구조 변화를 확인할 수 있었다. AUDIT 척도를 사용하여 DTI의 FA 값에 대한 지침을 제시할 수 있으며 혈액학적 인자의 가이드인 처럼 FA 값을 산정한다면 알코올 사용 장애자의 선별 및 진단에 유용한 검사법이라 사료 된다.
