반응표면분석법을 이용한 이부프로펜의 분쇄공정변수의 최적화 (Optimization of Process Variables for Grinding of Ibuprofen using Response Surface Methodology)

  • 이부프로펜은 비스테로이드성 소염진통제(Non-Steroidal Anti-Inflammatory Drugs; NSAIDs)의 대표적인 성분이다. 이부프로펜은 결정성이 크기 때문에 난용성이며, 따라서 생체이용률(bioavailability)도 낮다. 이와 같은 난용성을 개선하기 위해서는 이부프로펜의 입도를 감소시킬 필요가 있다. 본 연구의 목적은 이부프로펜의 분쇄조건을 최적화하는데 있다. 이부프로펜을 분쇄하기 위하여 유성밀을 사용하였으며, Box-Behnken 방법을 이용하여 분쇄변수들의 최적조건을 구하였다. 이부프로펜 분쇄생성물의 물성을 조사하기 위하여 입도, 결정크기 및 인장강도 측정에는 각각 입도분석기, XRD, tensile/compression tester를 사용하였다. 분쇄 최적조건은 밀회전수는 290 rpm, 시료장입량은 24.6 g, 분쇄시간은 10분이었으며, 이 조건에서 이부프로펜 분쇄생성물의 입도는 $13.5{\mu}m$이었다. 이부프로펜은 분쇄 후 결정크기가 감소하였다. 이부프로펜 분쇄생성물의 정제의 상대밀도가 0.85~0.90인 범위에서 그 정제의 인장강도는 $12{\sim}14Kg_f/cm^2$ 이었다.

Box-Behnken 및 반응표면 분석법을 이용한 음식물류 폐수 부상 스컴의 혐기성 소화를 위한 열-알칼리 전처리 최적화 (Optimization of Thermal-alkaline Pre-treatment for Anaerobic Digestion of Flotation Scum in Food Waste Leachate Using Box-Behnken Design and Response Surface Methodology)

  • Response surface methodology (RSM) based on a Box-Behnken Design (BBD) was applied to optimize the thermal-alkaline pre-treatment operating conditions for anaerobic digestion of flotation scum in food waste leachate. Three independent variables such as thermal temperature, NaOH concentration and reaction time were evaluated. The maximum methane production of 369.2 mL $CH_4/g$ VS was estimated under the optimum conditions at $62.0^{\circ}C$, 10.1% NaOH and 35.4 min reaction time. A confirmation test of the predicted optimum conditions verified the validity of the BBD with RSM. The analysis of variance indicated that methane production was more sensitive to both NaOH concentration and thermal temperature than reaction time. Thermal-alkaline pretreatment enhanced the improvement of 40% in methane production compared to the control experiment due to the effective hydrolysis and/or solubilization of organic matters. The fractions with molecular weight cut-off of scum in food waste leachate were conducted before and after pre-treatment to estimate the behaviors of organic matters. The experiment results found that thermal-alkaline pre-treatment could reduce the organic matters more than 10kD with increase the organic matters less than 1kD.

Simultaneous degradation of nitrogenous heterocyclic compounds by catalytic wet-peroxidation process using box-behnken design

  • The present study investigates the feasibility of nitrogenous heterocyclic compounds (NHCs) (Pyridine-Quinoline) degradation by catalytic wet peroxidation (CWPO) in the presence of nanoscale zerovalent iron supported on granular activated carbon (nFe0/GAC) using statistical optimization technique. Response surface methodology (RSM) in combination with Box-Behnken design (BBD) was used to optimize the process parameters of CWPO process such as initial pH, catalyst dose, hydrogen peroxide dose, initial concentration of pyridine (Py) and quinolone (Qn) were chosen as the main variables, and total organic carbon (TOC) removal and total Fe leaching were selected as the investigated response. The optimization of process parameters by desirability function showed the ~85% of TOC removal with process condition of initial solution pH 3.5, catalyst dose of 0.55 g/L, hydrogen peroxide concentration of 0.34 mmol, initial concentration of Py 200 mg/L and initial concentration of Qn 200 mg/L. Further, for TOC removal the analysis of variance results of the RSM revealed that all parameter i.e. initial pH, catalyst dose, hydrogen peroxide dose, initial concentration of Py and initial concentration of Qn were highly significant according to the p values (p < 0.05). The quadratic model was found to be the best fit for experimental data. The present study revealed that BBD was reliable and effective for the determination of the optimum conditions for CWPO of NHCs (Py-Qn).

Box-Behnken법을 이용한 Geobacter lovleyi의 TCE 탈염소화 공정 최적화 연구 (The Statistical Optimization of TCE Dechlorination by Geobacter lovleyi Using Box-Behnken Design)

  • 본 연구는 TBOS를 기질로 적용한 Geobacter lovleyi의 TCE 탈염소화 공정에 관하여 조사하였다. Geobacter lovleiy를 이용한 TCE 탈염소화 반응은 TCE 초기농도와 Geobacter lovleyi 주입량, 초기 TBOS 농도의 독립변수를 수학적으로 표시하였고, 반응표면법(RSM)을 활용하여 모델화하였다. 이 실험들은 Box-Behnken Design(BBD)을 통해 계획된 15개의 실험을 통해 이루어졌다. RSM을 통하여 TCE 제거효율과 독립변수들의 모델식이 도출되었다 : $Y_1$= -11.50(initial concentration of TCE) + 4.25(단백질 양, 주입된 Geobacter lovleyi의 양) - 4.75(initial concentration of TBOS) - ${6.58X_1}^2$ - ${8.583X_3}^2$ + 93.67, $Y_2=-10.92X_1+5.06X_2-4.89X_3-{4.93X_3}^2-2.19X_1X_2+2.54X_1X_3-2.19X_2X_3+16.71$. 도출된 반응모델은 수정결정계수는 각 0.975, 0.934로 1에 가깝게 나타났으며, 모델의 기여율이 높은 것으로 나타났다. 또한, 통계적 분석결과 TCE 탈염소화 효율에 미치는 영향은 TCE 초기농도 > TBOS 초기농도 > 단백질 양의 순으로 나타났으며, 상호항의 영향은 나타나지 않았다.

박스-벤켄 설계법을 이용한 폐감귤박 활성탄에 의한 수용액 중의 항생제 Trimethoprim의 흡착 연구 (Study on the Adsorption of Antibiotics Trimethoprim in Aqueous Solution by Activated Carbon Prepared from Waste Citrus Peel Using Box-Behnken Design)

  • 폐감귤박으로 제조한 활성탄(WCAC, waste citrus peel based activated carbon)에 의한 항생제 trimethoprim (TMP)의 흡착 특성을 조사하기 위해 반응표면법(RSM, response surface methodology)을 사용하여 TMP 흡착에 대한 운전인자들의 영향을 조사하였다. 농도($X_1$: 50-150 mg/L), pH ($X_2$: 4-10), 온도($X_3$: 293-323 K), 흡착제 투여량($X_4$: 0.05-0.15 g)의 4가지 입력 파라미터를 가진 4-요인 Box-Behnken 실험 설계에 따라 회분식 실험을 수행하고, 얻어진 실험 결과를 다중 회귀 분석으로 2차 다항식에 맞추고 통계적 방법을 사용하여 검토하였다. 독립 변수 및 변수들 간의 교호 작용의 유의성은 ANOVA 및 t-검정 통계기법으로 평가하였으며, 통계적 결과는 TMP 농도가 다른 요인들에 비하여 가장 많은 영향을 미치는 운전인자라는 것을 보여 주었다. 흡착공정은 유사 2차 속도식에 잘 부합하였으며, 등온흡착평형관계는 Langmuir 식이 Freundlich 식 보다 잘 부합하였다. Langmuir 등온식으로 부터 계산한 WCAC에 의한 TMP의 최대 흡착량은 293 K에서 144.9 mg/g이었다.

Analysis of Efficiency of Bacillus subtilis To Treat Bagasse Based Paper and Pulp Industry Wastewater-A Novel Approach

  • In this present study, bagasse based pulp and paper industry wastewater was treated under different operating conditions such as initial pH (6-8), temperature ($25-35^{\circ}C$) and contact time (3-7 days) by using Bacillus subtilis. Response surface methodology (RSM) coupled with Box-Behnken response surface design (BBD) was employed to investigate the effect of process variables on the responses such as turbidity, biological oxygen demand (BOD) and chemical oxygen demand (COD) removal. The experimental data were analyzed by Pareto analysis of variance (ANOVA) and the second order polynomial models were developed. Interactive effects of the process variables on the responses were studied using plotting 3D response surface contour graph and the optimum process conditions were found to be: initial pH of 7, temperature of $30^{\circ}C$ and contact time of 5 days. Under these conditions, removal efficiencies of turbidity, BOD and COD were found to be 85%, 93% and 80% respectively which are close agreement with real experiments. These results indicate that the treatment of bagasse based pulp and paper industry wastewater using Bacillus subtilis is an effective and novel technique.

Optimization of Transesterification Process of Biodiesel from Nyamplung (Calophyllum inophyllum Linn) using Microwave with CaO Catalyst

  • Nyamplung (Calophyllum inophyllum Linn) is one of the most widely grown plants in Indonesia. In addition, nyamplung oil has a future competitive advantage in that it can be processed into biodiesel. However, conventional methods for transesterification of nyamplung oil have been less effective. Therefore, in this study biodiesel was produced using microwaves as one of the alternative methods that can improve the shortcomings of conventional methods. In addition, optimization of parameters such as microwave power, catalyst concentration and transesterification time was done using Box-Behnken design. The combination of microwave with CaO catalyst and treated with Box-Behnken design are considered as a new and modern method for production of biodiesel from nyamplung oil and optimizing the factors that affected the transesterification process. The results showed that factors such as microwave power of 449.29 W, concentration of catalyst of 4.86% and transesterification time of 10.07 min can produce optimal yield of biodiesel of 92.73% with reliability of 93.22%.

Optimization of operating parameters to remove and recover crude oil from contaminated soil using subcritical water extraction process

  • Box-Behnken Design (BBD) under response surface methodology (RSM) was implemented to optimization the operating parameters and assess the removal and recovery efficiencies of crude oil from contaminated soil using subcritical water extraction. The effects of temperature, extraction time and water flow rate were explored, and the results indicate that temperature has a great impact on crude oil removal and recovery. The correlation coefficients for oil removal ($R^2=0.74$) and recovery ($R^2=0.98$) suggest that the proposed quadratic model is useful. When setting the target removal and recovery (>99%), BBD-RSM determined the optimum condition to be a temperature of $250^{\circ}C$, extraction time of 120 min, and water flow rate of 1 mL/min. An experiment was carried out to confirm the results, with removal and recovery efficiencies of 99.69% and 87.33%, respectively. This result indicates that BBD is a suitable method to optimize the process variables for crude oil removal and recovery from contaminated soil.

Medium optimization for keratinase production by a local Streptomyces sp. NRC 13S under solid state fermentation

  • Thirteen different Streptomyces isolates were evaluated for their ability to produce keratinase using chicken feather as a sole carbon and nitrogen sources under solid state fermentation (SSF). Streptomyces sp. NRC 13S produced the highest keratinase activity [1,792 U/g fermented substrate (fs)]. The phenotypic characterization and analysis of 16S rDNA sequencing of the isolate were studied. Optimization of SSF medium for keratinase production by the local isolate, Streptomyces sp. NRC13S, was carried out using the one-variable-at-a-time and the statistical approaches. In the first optimization step, the effect of incubation period, initial moisture content, initial pH value of the fermentation medium, and supplementation of some agro-industrial by-products on keratinase production were evaluated. The strain produced about 2,310 U/gfs when it grew on chicken feather with moisture content of 75% (w/w), feather: fodder yeast ratio of 70:30 (w/w), and initial pH 7 using phosphate buffer after 8 days. Based on these results, the Box-Behnken design and response surface methodology were applied to find out the optimal conditions for the enzyme production. The corresponding maximal production of keratinase was about 2,569.38 U/gfs.
