RNA-RNA interactions and the dynamic RNA conformations are important regulators in virus replication in several RNA virus systems and may also involved in the regulation of many important virus life cycle phases, including translation, replication, assembly, and switches in these important stages. The 5' non-translated region of Potato virus X(PVX) contains multiple cis-acting elements that facilitate various viral processes. It has previously been proposed that RNA-RNA interactions between various RNA elements present in PVX RNA genome are required for PVX RNA accumulation(Hu et al., 2007; Kim and Hemenway, 1999). This model was based on the potential base-pairing between conserved sequence elements at the upstream of subgenomic RNAs(sgRNAs) and at the 5' and 3' end of RNA genome. We now provide more evidence that RNA-RNA base-pairing between elements present at the 5' end and upstream of each sgRNA is required for efficient replication of genomic and subgenomic plus-strand RNA accumulation. Site-directed mutations introduced at the 5' end of plus-strand RNA replication defective mutant(${\Delta}12$) increasing base-pairing possibility with conserved sequence elements located upstream of each sgRNAs restored genomic and subgenomic plus-strand RNA accumulation and caused symptom development in inoculated Nicotiana benthamiana plants. Serial passage of a deletion mutant(${\Delta}8$) caused more severe symptoms and restored wild type sequences and thus retained possible RNA-RNA base-pairing. Altogether, these results indicate that the RNA element located at the 5' end of PVX genome involved in RNA-RNA interactions and play a key role in high-level accumulation of plus-strand RNA in vivo.
In this study, the anti-osteoarthritis effects of Cynanchum wilfordii, Phlomis umbrosa, and Angelica gigas extract (CPAE), observed and confirmed in previously clinical studies were further investigated by in vitro and in vivo studies. Anabolic biomarkers related to healthy cartilage maintenance, such as aggrecan, type II collagen ${\alpha}$-1 (Col2a1), sex determining region Y-box-9 (Sox-9), and catabolic biomarkers related to osteoarthritis, such as cyclooxygenase-2 (Cox-2), matrix metalloproteinase-13 (Mmp13), and nuclear factor kappa-light-chain-enhancer of activated B cells ($Nf{\kappa}b$), were evaluated by quantitative reverse transcriptase polymerase chain reaction and reporter gene assay. In vitro study results showed significant changes in both anabolic and catabolic biomarkers. For anabolic factors, significant changes in the level of aggrecan (P<0.05), Col2a1 (P<0.05), and Sox-9 (P<0.01) activation were shown after treatment of cartilage cells with CPAE (50 ng/mL) with similar efficacy compared to insulin growth factor, the positive control (100 ng/mL). For catabolic factors, significant changes in the inhibition activity of Cox-2 (P<0.05), Mmp13 (P<0.01), and $Nf{\kappa}b$ (P<0.05) were shown for CPAE (50 ng/mL) with similar efficacy compared to Celecoxib, the positive control ($10{\mu}M$). In the in vivo carrageenan-induced paw edema model study results showed that CPAE-treated groups (100 mg/kg) and Celecoxib-treated groups (60 mg/kg) showed comparably significant efficacy of inhibition by 37.1% and 52.1%, respectively. Furthermore, CPAE (200 mg/kg) showed similar effect to Celecoxib (60 mg/kg) with an inhibition rate of 54.3%. This result confirms that CPAE effectively inhibited the inflammation-induced osteoarthritis symptoms.
Rates of the sedimentation and particle mixing have been estimated by applying uranium-series disequilibrium techniques to three sediment cores collected from the korea Deep Ocean Study (KODOS) site between the clarion and Clipperton Fracture Zones (CCFZ) of the Equatorial Pacific. Sedimentation rates based on the profiles of excess /SUP 230/Th activity and /SUP 230/ Th/SUB xs//SUP 232/ Th activity ratios at the southeastern part of the study area were estimated to be in the order of a few millimeters per thousand year, while at the northwestern part a factor of ten lower. Excess activities of /SUP 230/Th and /SUP 230/Th ratios showed intervals of constant values in the upper part of the sediment cores, probably generated by biological particle mixing. A "two-box" advection-diffusion steady state mixing model was employed in order to estimate particle mixing rates in the upper and the lower layers, based on the distribution profiles of excess /SUP 210/Pb activities. Particle mixing coefficients were estimated to be in the order of 10$^1$ cm$^2$/y in the upper layer and 10/SUP -1/-10/SUP 0/ cm$^2$/y in the lower layer.
The United States adopted DRG based prospective payment system (PPS) in order to control the inflation of health care costs. No study used statistical test while many studies reported the cost containing effect of the PPS. To study impacts of the PPS on the Medicare expenditure, this study set the following three hypotheses (1) The PPS decelerated the increase in the hospital expenditure (Part A), (2) the PPS accelerated the increase in the expenditure of outpatients and physicians (Part B), (3) the increase in total expenditure was decelerated inspite of the spill over (substitution) effect because saving in the Part A expenditure were greater than losses in the Part B expenditure. The dependent variables are per capita hospital expenditure, per capita Part B expenditure, and per capita total expenditure for the Medicare beneficiaries. An intervention analysis, which added intervention effect to the time series variation on the Box-Jenkins model, was used. The observations included 120 months from 1978 to 1987. The results are as follows : (1) The annual increase in the per capita Part A expenditure was $5.11 after the implementation of DRG where as that before the PPS had been $11.1. The effect of the reduction ($5.99) was statistically significient (t=-3.9). (2) The spill over (substitution) effect existed because the annual increase in the per capita Part B expenditure was accelerated by $1.73 (t=1.91) after the implementation of the PPS. (3) The increase in the total Medicare expenditure per capita was reduced by $4.26 (t=-2.19) because the spill over effect was less than cost savings in the Part A expenditure.
Increasingly stringent emission regulations of EU and CARB (California Air resource Board) require mandatory OBD (On Board Diagnostics) far the catalytic converters of a vehicle. It demands that MIL(Malfunction Indication Light) should be tuned on to inform the driver of catalytic converter failures. Currently dual oxygen sensor method Is widely used for the converter OBD. However, since it works only alter converter light-off, it has a serious limitation when applied to TLEV or more stringent emission regulations where more than 85% of total emission is coming out before converter light-off. In addition, a recent development in catalyst material. coating technology and additive catalysts leads to a much improved OSC (Oxygen Storage Capacity) after converter light-off, current methods are very difficult to determine levels of converter aging. Therefore, it is desired to develop an OSC detecting method before converter light-off to diagnose converter failures with higher reliability. In this study, OSCs of converters are measured by an absolute measuring method and a dynamic measuring method, and some of fundamental ideas are suggested about converter OBD before converter light-off. The converters are aged with two different aging methods; those are a furnace aging and an engine bench aging: to represent aging conditions in actual field applications. Dual oxygen sensor method at the lower temperature than light-off is also studied at a model gas bench with the converters. It is fecund that there is a certain point in temperature lower than light-off where difference due to aging level becomes maximum, thus a proper dynamic method to effectively monitor catalytic converters could be implemented fur the range lower than light-off temperatures. With this result, the aging level of converters is examined at an engine bench.
Automobile black boxes store and provide accident and driving information. The accident and driving information can be utilized to build scientific traffic-event database and can be applied in various industries. The objective of this study is to develop a recognition system of dangerous driving through analyzing the driving characteristic patterns. In this paper, possible dangerous driving models are classified into four models on the basis of vehicle behaviors(acceleration, deceleration, rotation) and accident types from existing statistical data. Dangerous driving data have been acquired through vehicle tests using automobile black boxes. Characteristics of driving patterns have been analyzed in order to classify dangerous driving models. For the recognition of dangerous driving, this study selected critical value of each dangerous driving model and developed the recognition algorithm of dangerous driving. The study has been verified by the application of recognition algorithm of dangerous driving and vehicle tests using automobile black boxes. The presented recognition methods of dangerous driving can be used for on-line/off-line management of drivers and vehicles.
Paecilomyces japonica is widely cultured to produce mycelium for medicinal and health food use. Illumination is an important factor in the growth and production of mycelium in submerged culture. The effects of different light-emitting diode (LED) combinations on the growth and cordycepin content as bioactive substances of mycelium were investigated. The results showed that the mycelium dry weights were lower under dark condition and red LED treatments. Dark condition, fluorescent light, and ultraviolet-A failed to increase the cordycepin content. Blue light was necessary to increase the cordycepin content, and a red-to-blue ratio of 3:7 induced the highest cordycepin content. The cordycepin contents of mycelium in submerged culture were significantly higher in a 12 h/day illumination time under red and blue (red-to-blue ratio of 3:7) LED treatments, showing an increase of up to 38% compared with those under the fluorescent-light control condition. The results demonstrated the roles of light with different wavelengths on the biosynthesis of cordycepin as bioactive substances. The low-heat release and replacement of traditional fluorescent lights with low-energy-consuming LEDs could increase the contents of bioactive substances. After optimization of the cordycepin production using response surface methodology (Box-Behnken design) to its canonical form, the optimum combination was found to be as follows: illumination time = 17.7 h/day, sugar content in the medium = 9.7 g/50 mL, and incubation time = 61.2 h. The model predicted a maximum response of 3779.2 ㎍/mL cordycepin yield.
This study was conducted to develop a functional feed additive for pig with spent $Lycium$$chinense$ Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent $Lycium$$chinense$ Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression ($p$ = 0.001), linear ($p$ = 0.002), square ($p$ = 0.017) and interaction ($p$ = 0.047) was significant and adjusted determination coefficient ($R^2$) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.

A series of Quasi-Elastic Neutron Scattering (QENS) experiments helps us to understand the single-particle (hydrogen atom) dynamics of a globular protein and its hydration water and strong coupling between them. We also performed Molecular Dynamics (MD) simulations on a realistic model of the hydrated hen-egg Lysozyme powder having two proteins in the periodic box. We found the existence of a Fragile-to-Strong dynamic Crossover (FSC) phenomenon in hydration water around a protein occurring at TL=$225{\pm}5K$ by analyzing Intermediate Scattering Function (ISF). On lowering of the temperature toward FSC, the structure of hydration water makes a transition from predominantly the High Density Liquid (HDL) form, a more fluid state, to predominantly the Low Density Liquid (LDL) form, a less fluid state, derived from the existence of a liquid?liquid critical point at an elevated pressure. We showed experimentally and confirmed theoretically that this sudden switch in the mobility of the hydration water around a protein triggers the dynamic transition (so-called glass transition) of the protein, at a temperature TD=220 K. Mean Square Displacement (MSD) is the important factor to show that the FSC is the key to the strong coupling between a protein and its hydration water by suggesting TL${\fallingdotseq}$TD. MD simulations with TIP4P force field for water were performed to understand hydration level dependency of the FSC temperature. We added water molecules to increase hydration level of the protein hydration water, from 0.30, 0.45, 0.60 and 1.00 (1.00 is the bulk water). These confirm the existence of the FSC and the hydration level dependence of the FSC temperature: FSC temperature is decreased upon increasing hydration level. We compared the hydration water around Lysozyme, B-DNA and RNA. Similarity among those suggests that the FSC and this coupling be universal for globular proteins, biopolymers.
Lab-scale experiments were conducted to evaluate the effect of surface tension reduction on the extension of the influence zone and the VOC removal efficiency of ground water sparging. A glass column packed with coarse sand was used for VOC removal test at two different surface tensions. A glass column without porous media was also used fer control purpose prior to sand-packed column test. A quasi-two-dimensional glass box model, packed with a sand, was used fer sparging zone tests at different water surface tensions. Surface tension of the aquoues solution used in this study was controlled using sodium dodecyl sulfate (SDS). For the glass, sand column experiments, total amount of air filled in the media increased as surface tension decreased. Toluene (used as VOC in this study) removal rate increased slightly with decreased surface tension f3r both free water column and sand-packed column. Air sparging zone extended up to 500% as the surface tension decreased. Combining the results from two different experiments, VOC removal efficiency is expected to increase significantly with surface tension reduction.
