Streptococcus uberis의 락토페린 결합단백질 추출을 위한 두 가지 방법의 비교 (A Comparison of Two Methods for the Extraction of Lactoferrin-binding Proteins from Streptococcus uberis)

  • 락토페린 결합단백질(Lactoferrin-binding proteins, LBP)은 젖소유방염 원인균인 Streptococcus uberis의 막단백질로서 그 특성에 관해서는 잘 규명되어 있지 않지만, 특히 최근에는 스트렙토코커스성 유방염의 독성인자로서 중요시되고 있다. 본 연구에서는 S. uberis 네 가지 균주를 대상으로 LBP를 보다 효율적으로 추출하기 위하여 mutanolysin 및 sodium dodecyl sulfate(SDS)를 이용한 두 가지 다른 추출 방법을 사용하였다. 추출된 세균단백질을 SDS-polyacrylamide gel electrophoreis(SDS-PAGE)로 전기영동을 하였고, 겔을 니트로셀룰로스 막으로 이동시켰다. Rabbit anti-bovine lactoferrin 항체와 HRP-conjugated donkey anti-rabbit IgG 항체를 사용하여 LBP를 검출하였다. 이러한 웨스턴 블롯팅 분석을 통해 SDS 추출법이 mutanolysin 추출법에 비해 보다 효율적으로 110 kDa 및 112 kDa의 LBP를 추출할 수 있음을 증명하였다.

Cytokine expression pattern in milk somatic cells of subclinical mastitis-affected cattle analyzed by real time PCR

  • The expression profiles of inflammatory cytokines viz. interleukins (IL)-6, IL-8, IL-12, granulocyte macrophage-colony stimulating factor, interferon-${\gamma}$ and tumor necrosis factor-${\alpha}$ in response to subclinical mastitis in indigenous cattle breed Kankrej (n = 6), Gir (Bos indicus) (n = 12) and crossbred (Bos taurus${\times}$Bos indicus) (n = 7) were investigated using quantitative real time PCR. Significant correlation (p < 0.05) was observed between total bacterial load and somatic cell count (SCC) in all three breeds of cattle. All the cytokines were observed to be up-regulated compared to cows with healthy quarters, however, level of their expression varied among three breeds of cattle. In Kankrej most cytokines were found to be transcribed to higher levels than in other two breeds; the milk had higher load of bacteria but not so high SCC, implying that Kankrej has a higher inherent resistance against mastitis. The results of present study indicated that mammary glands of crossbred cattle are more sensitive to bacterial infection than indigenous breed of cattle as they elicit immune response at lower bacterial load and result into higher SCC. Research on identification of factors responsible for differentially expressed cytokines profiles and use of cytokines as immunomodulatory tools can pave way for formulating control strategies against bovine mastitis.

준임상형(準臨床型) 유방염(乳房炎) 및 집합유(集合乳)의 Staphylococcus 속균(屬菌)에 대하여 (Studies on Staphylococci from Subclinical Mastitic and Bulk Milk Samples)

  • This paper deals with the incidence of bovine mastitis for 743 quarters and distribution of Staphylococci for the quarter and 70 bulk milk samples in the northern area of Gyeongbuk during the period from January to December 1984. Isolated Staphylococci were examined for species, subgroups, antibiotic resistance and penicillinase production. The results obtained were summarized as follows : A total of 25(73.5%) of 34 herds, 102(54.3%) of 188 cows and 208(30.3%) of 743 quarters were found to be infected with subclinical mastitis. A total of 83(83.1%) of 102 cows, 94(45.2%) of 208 mastitic quarters and 55(78.6%) of 70 bulk milk samples were isolated Staphylococci. Three hundred and eighteen strains of Staphylococci were classified into 11 species. Of these speoies, S. aureus from mastitis and S. sciuri from bulk milk were found most frequently, followed by S. epidermidis, S. simulans, S. cohnii, S. haemolyticus, S. xylosus, S. hyicus subsp. chromogenes, S. saprophyticus, S. warneri, S hyicus subsp. hyicus. Subgroups of catalase-positive and negative cocci were belonged most frequently to subgroup I, and subgroups III and III b, respectively. The method of Pelzer of al(97.8%) was more classified than that of Baird-Parker (68.5%). One hundred and sixty one strains(50.6%) of 318 Staphylococci isolates were resistance to one or more antibiotics such as ampicillin, chloramphenicol, gentamicin, kanamycin, streptomycin, and tetracycline. Isolates from subclinical mastitis were more resistant to antibiotics than its from bulk milk. Of the 318 Staphylococci Isolates, 128(40.3%) gave positive reaction for the penicillinase test, all of ampicillin resistance strains produced this emzyme.

소 유방염 유래 Staphylococcus aureus의 AFLP 지문분석 (Amplified fragment length polymorphism fingerprinting analysis of Staphylococcus aureus isolated from bovine mastitis milk)

  • Amplified fragment length polymorphism(AFLP) technique is based on the polymorphism detection through selective PCR amplification of restriction fragments from digested genomic DNA and thus includes the procedures of the total DNA digestion by endonucleases, ligation of adapters to the ends of the fragments, and following the selective amplification of the restricted DNA fragments. This study were aimed to : (1) determine the genetic variability of S aureus strains, (2) estimate genetic diversity within and among these strains, (3) compare phylogenetic relationships among these strains as genetic markers using AFLP techniques. Genomic DNA was digested with a particular combination of three restriction enzymes with specific recognition sites and the DNA fragments were ligated to restriction specific adapters and amplified using the selective primer combinations. In the S aureus strain, the number of scorable AFLP bands detected per each primer combination varied from 29 to 102, with an average of 61.59 using 27 primer combinations. A total of 1,663 markers were generated, 904 bands of which were polymorphic, showing a 33.48% level of polymorphism with these primer combinations. Among the primer combinations, E02/T02, E02/T03, E04/H02, E02/T01 and E04/H03 primer combinations showed a high level of polymorphism with 0.78, 0.76, 0.74, 0.71 and 0.70, respectively. But T03/H01, E01/T02 and E01/T03 primer combinations showed a low level of polymorphism with 0.38, 0.37 and 0.15, respectively, Therefore, the former primer combinations will be the most effective for AFLP analysis of S aureus. In SA1 sub-types the level of polymorphism of S aureus KCTC 1927 was similar to that of S aureus CU 01(0.825) and higher than those of other strains such as S aureus CU 02 (0.715), S aureus KCTC 2199(0.625), S aureus KCTC 1916(0.607) and S aureus KCTC 1621 (0.553). In SA2 sub-types the level of polymorphism of S aureus CU 07 was similar to that of S aureus CU 08(0.935) and higher than those of both S aureus CU 04(0.883) and S aureus CU 05(0.883) and lower than those of S aureus CU 03(0.583). In SA3 subtypes the level of polymorphism of S aureus CU 11 was similar to that of S aureus CU 12(0.913) and lower than that of S aureus CU 15(0.623). The results proved that AFLP marker analysis of S aureus strain could be used to study the epidemiology of mastitis and in addition, common genotype in geographic region could be useful for the development of an effective vaccine or DNA marker for easy diagnosis of mastitis caused by S aureus infection.

Prototheca에 의한 소 유방염 발생예 (Association of Prototheca with Bovine Mastitis)

  • 7세의 홀스타인 암소에서 엽록소가 없는 Prototheca에 의한 만성적인 진행성 유방 염의 발생에 대해 보고한다. 유방염 우유로 만든 습윤표본에서 명확한 세포벽과 내생포자를 갖고 있는 구형 내지 난원형의 병원체가 직접 관찰되었고, Sabouraud배지에서의 순수배양물 에 대해 Wright염색 및 PHOL염색을 했을 때 algae가 증명되었으며, nutrient배지에서 세균 이 중식하지 않아서 Prototheca가 유방염의 원인으로 제시되었다. 이 병원체는 자연상태에서 는 사물기생성이지만 감염원은 확실하지 않았다. algae와 진균의 형태학적인 연구를 하는데 있어서 PHOL염색액을 광범위하게 응용할 수 있음을 제시하였다. Prototheca의 ecology와 병원성을 규명하기 위해 더 많은 연구가 이루어져야 될 것으로 생각된다.

젖소 유방감염증(乳房感染症)으로부터 분리(分離)한 Gram 음성간균(陰性桿菌) (Gram-Negative Bacilli Isolated from Bovine Udder Infections)

  • In order to know the main causative agent and distribution of the agents in Gram-negative bacillus infections of bovine udders, one hundred and forty-eight cultures of Gram-negative bacilli isolated from clinical cases of mastitis during lactation over a period of 2 years in 6 herds of 180 Holstein cows were studied. Escherichia coli occupied 54.7% of the isolates was the most frequently isolated Gram-negative bacillus. 30 of the isolates (20.3%) were Enterobacter aerogenes, but Enterobacter cloacae was not isolated. Remaining isolates were Klebsiella pneumoniae, Proteus spp., Preudomonas aeruginosa and Alcaligenes faecalis. Most of the isolates (75.0%) were isolated from rear quarters.

젖소 유방유래(乳房由來) 난포구원(蘭葡球園)에 관한 연구(硏究) : III. 분리균주(分離菌株)에 대한 Penicillins 및 Gentamicin의 항균효과(抗菌效果) (Studies on Staphylococci Isolated from Bovine Mastitis: III. Activity of Penicillins and Gentamicin to Isolates)

  • The in vitro activity of penicillins(penicillin G, ampicillin, carbenicillin, methicillin and cloxacillin) and gentamicin to Staphylococcus aureus and coagulase-negative staphylococci isolated from bovine mastitic milk samples was determined. The growth of all isolates of staphylococci tested was inhibited by cloxacillin and methicillin at a concentration of $0.78{\mu}g/ml$ and by gentamicin at a minimum inhibitory concentration (MIC) of $1.56{\mu}g/ml$. Of the 140 strains of Staphylococcus aureus isolated 99(70.7%) gave positive reactions for penicillinase on the starch-iodine test. Of 121 isolates of coagulase-negative staphylococci, 58(47.9%) showed penicillinase production, but all the Staph. xylosus lacked the ability to produce penicillinase. MIC of penicillin G of the penicillinase-positive strains was $0.2{\mu}g/ml$ or more. The combination of cloxacillin with gentamicin showed a synergistic effect by inhibiting regrowth of the tested organism.

Antimicrobial resistance studies in staphylococci and streptococci isolated from cows with mastitis in Argentina

  • Background: Staphylococcus aureus and Streptococcus agalactiae are the main cause of clinical mastitis in dairy cattle in Argentina, whereas coagulase-negative staphylococci (CNS) and environmental streptococci are the main cause of subclinical mastitis. Bacteria isolated from infected animals show increasing antimicrobial resistance. Objectives: This study aims to determine the antimicrobial resistance of staphylococci and streptococci isolated from milk with mastitis, and to genotypically characterize the methicillin-resistant (MR) staphylococci. Methods: Isolation was performed on blood agar and identification was based on biochemical reactions. Antimicrobial susceptibility was according to the Clinical and Laboratory Standards Institute guidelines. The antimicrobial resistance genes, SCCmec type and spa type were detected by the polymerase chain reaction method. Results: We isolated a total of 185 staphylococci and 28 streptococci from 148 milk samples. Among the staphylococcal isolates, 154 were identified as CNS and 31 as S. aureus. Among the 154 CNS, 24.6% (n = 38) were resistant to penicillin, 14.9% (n = 23) to erythromycin, 17.5% (n = 27) to clindamycin, 6.5% (n = 10) to cefoxitin and oxacillin. Among the S. aureus isolates, 16.1% (n = 5) were resistant to penicillin, 3.2% (n = 1) to cefoxitin and oxacillin (MRSA). Six MR isolates (5 CNS and 1 MRSA) were positive to the mecA gene, and presented the SCCmec IVa. The MRSA strain presented the sequence type 83 and the spa type 002. Among the 28 streptococcal isolates, 14.3% (n = 4) were resistant to penicillin, 10.7% (n = 3) to erythromycin and 14.3% (n = 4) to clindamycin. Conclusions: The present findings of this study indicate a development of antimicrobial resistance in main bacteria isolated from cows with mastitis in Argentina.
