Controllable Biogenic Synthesis of Intracellular Silver/Silver Chloride Nanoparticles by Meyerozyma guilliermondii KX008616

  • Intracellular synthesis of silver/silver chloride nanoparticles (Ag/AgCl-NPs) using Meyerozyma guilliermondii KX008616 is reported under aerobic and anaerobic conditions for the first time. The biogenic synthesis of Ag-NP types has been proposed as an easy and cost-effective alternative for various biomedical applications. The interaction of nanoparticles with ethanol production was mentioned. The purified biogenic Ag/AgCl-nanoparticles were characterized by different spectroscopic and microscopic approaches. The purified nanoparticles exhibited a surface plasmon resonance band at 419 and 415 nm, confirming the formation of Ag/AgCl-NPs under aerobic and anaerobic conditions, respectively. The planes of the cubic crystalline phase of the Ag/AgCl-NPs were confirmed by X-ray diffraction. Fourier-transform infrared spectra showed the interactions between the yeast cell constituents and silver ions to form the biogenic Ag/AgCl-NPs. The intracellular Ag/AgCl-NPs synthesized under aerobic condition were homogenous and spherical in shape, with an approximate particle size of 2.5-30nm as denoted by the transmission electron microscopy (TEM). The reaction mixture was optimized by varying reaction parameters, including temperature and pH. Analysis of ultrathin sections of yeast cells by TEM indicated that the biogenic nanoparticles were formed as clusters, known as nanoaggregates, in the cytoplasm or in the inner and outer regions of the cell wall. The study recommends using the biomass of yeast that is used in industrial or fermentation purposes to produce Ag/AgCl-NPs as associated by-products to maximize benefit and to reduce the production cost.

A Study on the Map Accuracy Assessment of Positioning Data Using Statistical Approach Analysis (오차분석을 이용한 지도 위치정확도 평가기법에 관한 연구)

  • This paper suggests a Map Accuracy Standards by analyzing U.S. National Map Accuracy Standards, by considering korean terrain feature and statistical error theory for paper and digital maps on the scale of 1:50,000. Map accuracy standards require horizontal accuracy to be reported as a circular error with 90% confidence level through Linear Error Probable(LEP) theory and Circular Error Probable(CEP) theory. In order to verify the proposed methodology for positioning accuracy testing, several kinds of test point were selected and tested. These test points were extracted at the centers of roads and bridges, the comers of the independent building, the edges of geographical botany, and the tops of mountains. The positioning accuracy assessment was peformed by comparing the positions of test points in digital maps generated three different sources with those acquired by high accurate GPS surveying. The digital maps were produced from aerial photographs and SPOT satellite image using analytical plotter and 1:50,000 paper map.

Synthesis and Antimicrobial Activity of Some New 1-Alkyl-2-alkylthio-1,2,4-triazolobenzimidazole Derivatives

  • Some new derivatives of 1,2,4-triazolo[2,3-a]benzimidazoles were synthesized through the reaction of 1,2-diaminobenzimidazole with carbon disulfide. The resulting 1,2,4-triazolo-[2,3a]benzimidazole-2-thione intermediate was reacted with one equivalent of alkyl halides to give the corresponding 2-alkylthio derivatives, which were further alkylated through the reaction with another one equivalent of different alkyl halides to afford the target compounds; 1-alkyl-2alkylthio-1,2,4-traizolo[2,3-a]benzimidazoles. On the other hand, the 1,2-disubstituted derivatives with two identical alkyl substituents were prepared by the reaction of 1,2,4-triazolo[2,3-a]benzimidazole-2-thione with two equivalents of the alkyl halides. The structures of the new compounds were assigned by spectral and elemental methods of analyses. The synthesized compounds were tested for their antibacterial and antifungal activities. Most of the tested compounds proved comparable results with those of ampicillin and fluconazole reference drugs. The study indicated that, the antibacterial as well as the antifungal activities of the test compounds were improved with increase in the bulkiness of the introduced alkyl groups. Also, some active antibacterial compounds were tested for their antimycobacterial activity. All the test compounds showed equipotent antitubercular activity as that of INH as a reference drug.

Potential impact of climate change on plant invasion in the Republic of Korea

  • Background: Invasive plant species are considered a major threat to biodiversity, ecosystem functioning, and human wellbeing worldwide. Climatically suitable ranges for invasive plant species are expected to expand due to future climate change. The identification of current invasions and potential range expansion of invasive plant species is required to plan for the management of these species. Here, we predicted climatically suitable habitats for 11 invasive plant species and calculated the potential species richness and their range expansions in different provinces of the Republic of Korea (ROK) under current and future climate change scenarios (RCP 4.5 and RCP 8.5) using the maximum entropy (MaxEnt) modeling approach. Results: Based on the model predictions, areas of climatically suitable habitats for 90.9% of the invasive plant species are expected to retain current ecological niches and expand to include additional climatically suitable areas under future climate change scenarios. Species richness is predicted to be relatively high in the provinces of the western and southern regions (e.g., Jeollanam, Jeollabuk, and Chungcheongnam) under current climatic conditions. However, under future climates, richness in the provinces of the northern, eastern, and southeastern regions (e.g., Seoul, Incheon, Gyeonggi, Gyeongsangnam, Degue, Busan, and Ulsan) is estimated to increase up to 292%, 390.75%, and 468.06% by 2030, 2050, and 2080, respectively, compared with the current richness. Conclusions: Our study revealed that the rates of introduction and dispersion of invasive plant species from the western and southern coasts are relatively high and are expanding across the ROK through different modes of dispersion. The negative impacts on biodiversity, ecosystem dynamics, and economy caused by invasive plant species will be high if preventive and eradication measures are not employed immediately. Thus, this study will be helpful to policymakers for the management of invasive plant species and the conservation of biodiversity.

Nutrient dynamics in decomposing litter from four selected tree species in Makurdi, Benue State, Nigeria

  • Background: Nutrient release during litter decomposition was investigated in Vitex doniana, Terminalia avecinioides, Sarcocephallus latifolius, and Parinari curatellifolius in Makurdi, Benue State Nigeria (January 10 to March 10 and from June 10 to August 10, 2016). Leaf decomposition was measured as loss in mass of litter over time using the decay model Wt/W0 = e-kd t, while $Kd=-{\frac{1}{t}}In({\frac{Wt}{W0}})$ was used to evaluate decomposition rate. Time taken for half of litter to decompose was measured using T50 = ln 2/k; while nutrient accumulation index was evaluated as $NAI=(\frac{{\omega}t\;Xt}{{\omega}oXo})$. Results: Average mass of litter remaining after exposure ranged from 96.15 g, (V. doniana) to 78.11 g, (S. lafolius) in dry (November to March) and wet (April to October) seasons. Decomposition rate was averagely faster in the wet season (0.0030) than in the dry season (0.0022) with P. curatellifolius (0.0028) and T. avecinioides (0.0039) having the fastest decomposition rates in dry and wet seasons. Mean residence time (days) ranged from 929 to 356, while the time (days) for half the original mass to decompose ranged from 622 to 201 (dry and wet seasons). ANOVA revealed highly significant differences (p < 0.01) in decomposition rates and exposure time (days) and a significant interaction (p < 0.05) between species and exposure time in both seasons. Conclusion: Slow decomposition in the plant leaves implied carbon retention in the ecosystem and slow release of CO2 back to the atmosphere, while nitrogen was mineralized in both seasons. The plants therefore showed effectiveness in nutrient cycling and support productivity in the ecosystem.

Studies on Plant Succession of Sand Bars at the Nagdong River Estuary I. Vegetation and Soil Environment (낙동강 하구 사주식생의 천이에 관한 연구)

  • Plant succession and subsequent changes of soil properties were studied in sand bars at the Nagdong River estuary in Korea. By old maps, ages of sand bars such as Namusitdeung, Galmaegideung, Baeghapdeung and Ogryudeung were estimated about 10, 15, 25 and 60 years old, respectively. The dominance-diversity curves and plant species diversity indices among the sand bars showed increasing trend of stability from Galmaegideung toward Ogryudeung. The soil acidity, contents of organic matter and total nitrogen of soil increased significantly from Galmaegideung toward Ogryudeung. The order of successional degree among the sand bars on the basis of the number of plant species, the results of vegetation analysis and changes of soil properties almost coincide with the order of age among the sand bars. However, the order of successional degree between Namusitdeung and Galmaegideung was more or less obscured because of cyclic succession driven by allogenic processes.

Proliferation of Tricholoma matsutake Mycelial Mats in Pine Forest Using Mass Liquid Inoculum

  • Two isolates of Tricholoma matsutake T-008 and T-034, preserved in Entomopathogenic Fungal Culture Collection (EFCC) of Korea, were used in the present study. The isolates had 100% Bootstrap homology with Tricholoma matsutake U62964 and T. matsutake AB188557 and AF309538 preserved in Gene Bank of NCBI. Mycelial growth of T. matsutake was highest in TMM and MYA at $25^{\circ}C$. The highest dry wt. of mycelium was obtained after 65 days of culture, when 6 mycelial discs were inoculated in 100 ml of broth in 250 ml shaking flask. Mycelial mats were observed in clumped condition at the inoculation sites of pine forest after two weeks of inoculation. After 5 months of inoculation, mycelia mats were observed growing inside soil and walls of a few inoculation sites, while mycelial mats growth up to $5{\sim}8$ cm were observed in the roots of pine tree after 6 months. The survival rate of the inoculum was about 40% of the total inoculation sites. The survival rate was found below 20% when the mycelium was inoculated in the summer. The reasons for low survival rates of the mycelium were mainly due to dry season and the soil-borne small animals such as earthworm and mole. After one year of inoculation, no external difference was observed between the artificially inoculated mycelia and the naturally existing mycelia of T. matsutake. The present study showed that fruiting bodies of T. matsutake could be produced by artificial inoculation under the appropriate environmental conditions.

Characteristics of Fusion Products between Pleurotus ostreatus and Pleurotus florida Following Interspecific Protoplast Fusion (느타리버섯과 사철느타리버섯의 종간 원형질체융합 균주의 특성)

  • Interspecific heterokaryons were obtained between auxotrophic mutants of Pleurotus ostreatus and Pleurotus florida by polyethylene $glycol-Ca^{++}$ induced fusion of somatic protoplasts. The fusion products produced colonies of dense growing mycelium after 10-14 days culture on hypertonic Mushroom Minimal Medium. When they were transferred to Mushroom Minimal Medium plates, the sectors showed normal vegetative morphology but the colonies of irregular shape varied in growth rate. All of the colonies produced fruit body of normal pilei. Some heterokaryon colonies bearing none or only a small amounts of basidiospores were isolated. Fusion products, generally, gave higher basidiocarp yields than the parents.

  • This study was carried out to isolate and characterize $A{\alpha}$ mating locus controlling fruiting body formation directly in the Basidiomycete Schzophyllum commune growing in the North America. Total numbers of genomic library of S. commune UVM1-34 was about $2{\times}10^4$ cells. About 90% library was appeared to have about 35 kb inserted genome DNA in cosmid pTC20 vector. 6 clones were proved to have positive signal to probes within Z and Y region in colony and southern hybridization. In the mating activity test, all the 6 positive clones were appeared to have $A{\alpha}3$ mating activity although they had two different restriction patterns. pSC13 containing 5.7 Kb PstI-fragment of UVM 1-34 $A{\alpha}3$ allele showed about 50% clamp cell formation indicating mating activity when cotransformation was done together with cosmid pTC20.

Ecological Studies on the Montane Grassland of Mt. Soback in Korea (소백산 산지초원의 생태학적 연구 II. 물질생산과 염류순환)

  • Comparative study of the biomass productioin and the cyclings of nitrogen, phosphorus and potassium was carried out on the east and the northwest facing slope in the montane grassland of Mt. Soback. The maximum productin during the growing season in the east and the northwest slope were 1, 150g/m2.yr, and 755g/m2.yr. in the August, respectively. The positive correlation was appearent between biomass production and A-layer depth of the soil and the correlation coefficient (r=.964) was very significant at 1% level. In the above ground materials, the nutrients contents were high at the early of the growing season but decreased gradually. In roots, however, there are no significant trend throughout the growing season. Total amounts of minerals uptaken by plants during the growing season in the east and the northwest site were 18.20 and 10.66g/m2.yr for N, 0.06 and 0.03g/m2.yr for P, 0.23 and 0.11g/m2.yr for K, respectively. the minerals returned to the soil by litter decomposition and roots decay in the east and the northwest site were 16.19 and 9.62g/m2.yr for N, 0.046 and 0.021g/m2.yr for P, 0.211 and 0.099g/m2.yr for K, respectively. The turnover the rate (absorbed/returned) of the nutrients in the east and the northwest site were 1.13 and 1.14 for N, 1.30 and 1.43 for P, 1.09 and 1.11 for K, and the absorption rates of minerals were 0.39 and 0.29% for N, 3.16 and 1.88% for P, 0.91 and 0.57% for K, respectively.
