Effect of BODUOPAESAN(BOS) on Gastrointestinal Tract (오패산(烏貝散)과 보두(寶豆)의 병용투여(倂用投與)가 위장관(胃腸管)에 미치는 영향(影響))

  • In order to investigate the effect of BODUOPAE-SAN(BOS), experiments were performed on analgesic effect induced by acetic acid, duration of hypnosis induced by pentobarbital-Na in mice and gastric ucler in Shay rats and indomethacin induced gastric ulcer, HCI-ethanol induced gastrci lesion in rats and gastric juice secretion in Shay rats. The results are as followings. 1. BOS showed significantly an analgesic effect induced by acetic acid. 2. BOS prolonged the duration of hypnosis induced by pentobarbital-Na in rats 3. BOS rised the spontaneous motility of isolatied ileum of mice temporarily. 4. BOS depressed the gastric motility of rats significantly. 5. BOS showed an anti-ulcer effect in Shay rats and indometacine-induced ulcer rats significantly. 6. BOS reduced the ulcer index of the HCl-ethanol induced gastric lesion in rats. 7. BOS reduced the gastric juice secretion in Shay rats. From above results think BOS is more effective in comparison to the OPAESAN alone on gastrointestinal disorder.

Allele Frequency of the Bovine Y-chromosomal Microsatellite Locus in the Cattle Breeds (소 Y 염색체 특이 Microsatellite를 이용한 품종별 대립유전자 빈도 분석)

  • The INRA124 is a bovine Y-chromosomal specific microsatellite locus that has been revealed a polymorphism. This locus has two alleles. The 132 bp allele is specific to cattle (humpless) of taurine origin and the 130 bp allele is specific to cattle (humped) of indicine origin. A total 822 males of 20 breeds or populations; North Eastern Asian breeds (Hanwoo, Korean Black cattle, Chik-so, CBK, Japanese Black cattle, Japanese Brown cattle, Yanbian cattle), Chinese yellow cattle (Luxi cattle, Nanyang cattle), European origin (Angus, Hereford, Charolais, Simmental, Brown swiss, Holstein, Limousin), African origin (Kavirondo zebu, White Fulani, crossbreed of N'Dama and Boran), Indian origin (Sahiwal) were characterized the distribution of alleles using INRA124 locus. Any individuals of European, Japanese origins and Hanwoo were not detected 130 bp allele, Bos indicus specific allele. Bos indicus breeds of Indian and African origins were not detected 132 bp allele, Bos taurus specific allele. CBK population that the crossbreed of Hanwoo, Brahman and Charolais showed the frequency of 0.19 in indicine specific allele. The breeds of Chinese mainland, Luxi and Nanyang cattle were detected 0.46 and 0.29 frequencies in indicine specific allele, respectively. These results suggest that Korean cattle, Hanwoo, had not been originated from a crossbred between Bos primigenius in Europe and Bos indicus in India.

A Comparison on Polymorphism of Beta-lactoglobulin Gene in Bos indicus, Bos taurus and Indicine×Taurine Crossbred Cattle

  • The study was carried out on eight Bos indicus cattle breeds namely, Sahiwal, Tharparkar, Nimari, Khilari, Deoni, Amritmahal, Hariana and Hilly cattle; two Bos taurus cattle breeds namely, Jersey and Holstein Friesian and Indicine${\times}$Taurine crossbred cattle to find out the polymorphic pattern of beta-lactoglobulin gene. The polymorphism at beta-lactoglobulin gene was detected by conducting PCR-RFLP studies on 398 bp fragment spanning over 104 bases of exon IV and 294 bases of intron IV. Two alleles A and B and three genotypes AA, AB and BB were observed in all the cattle breeds. The frequency of B allele was comparatively higher than that of A allele. The allelic frequency of A varied from 0.20 to 0.30 in Bos indicus cattle breeds and 0.19 to 0.34 in Bos taurus breeds while in crossbred cattle the frequency was estimated as 0.21. The weighted frequency of A allele was highest in Indian cattle and lowest in crossbred cattle while the frequency in taurine cattle was found to be in between indicus and crossbred cattle. The non-significant differences of allelic frequency amongst Bos indicus, Bos taurus and crossbred cattle was observed. The effect of genotype on fat percentage was also found to be non-significant in cattle.

  • Objectives : The purpose of this study was to investigate the effect of Bo-du-san (BOS) on apoptosis in human gastric cancer cells, SNU-l cells. BOS, a drug preparation consisting of two herbs, that is, Crotonis Fructus (Strychni ignatii Semen, bodu in Korean) and Glycyrrhizae Radix (Glycyrrhizae uralensis FISCH, Gamcho in Korean). Methodss : In this study, methanol extract of BOS was examined for cytotoxic activity on human gastric cancer cells, SNU-1 cells, using XTT assay, with an IC50 value was 0.7 mg/ml and 0.3 mg/ml at 24 hrs and 48 hrs, respectively. Apoptosis induction by BDS in SNU-l cells was verified by the induction of DNA fragmentation, cleavage of poly ADP-ribose polymerase (PARP), and activation of caspase-3, -8 and -9. Inhibitors of caspase-3, -8 and -9 (Ac-DEVD-CHO, Z-IETD-FMK and Z-LEHD-FMK) efficiently blocked BOS-induced cell death of SNU-l. Resultss : BOS-induced cell death was via caspase dependent apoptosis. Moreover, treatment of BOS result in the decrease the G1/S cycle regulation proteins (cyclin D1 and E) expression and increase CDK inhibitor proteins (p21 and p27) expression, and increase apoptotic protein, p53 expression. Thus, BOS induces apoptosis in SNU-1 cells via cell cycle arrested in G1 phase. Conclusions : These results indicated that BOS has some potential for use as an anti-cancer agent.

Motion Analysis and EMG Analysis of the Pelvis and Lower Extremity according to the Width Variation of the Base of Support

  • The purpose of this study is to identify which width of the base of support(BOS) is safer and more effective in lifting by comparing muscle activations and body sways when lifting objects under the width variation of the BOS. A total of fifteen healthy adults participated in this study. For the width variation of the BOS, the participants changed the width between their feet into three different types(10cm, 32cm, 45cm) and lifted a 10kg four times in each type after going up on a force plate. In order to measure body sways according to the width variation of the BOS, a motion analysis system was used. In addition, in order to measure the muscle activations of lower extremities, including the erector spinae, gluteus maximus, rectus femoris, and tibialis anterior, an electromyogram(EMG) analysis was employed. In addition, the Borg's scale was drawn by quantifying the subjective discomfort levels felt from each width of the BOS. In conclusion, no statistically significant differences according to the width variation of the BOS were observed(p=.295, .308)(p>.05). However, a statistically significant difference was exhibited between the Borg's scale, which indicates the discomfort levels from lifting performances, and the width variation of the BOS (p=$.000^*$).

  • This paper describes the effect of DME, biodiesel blended fuels on the macroscopic spray characteristics in a high pressure diesel injection system using Background Oriented Schlieren (BOS) method. The BOS method for visualization of impingement evaporation sprays to analyze macroscopic spray properties and evolutionary processes. In this work, the blending ratio of DME in the blended fuel are 0, 50, 100% by weight ratio. In order to investigate the macroscopic impinged spray characteristics under the various injection parameters and blending ratio. In this work, a mini-sac type single-hole nozzle injector with nozzle hole was length 0.7 mm and diameter of 0.3 mm was used. According to the result, the spray area of the collision wall increased as the DME mixing ratio increased, and the evolutionary pattern showed a stepwise increase due to the collision effect of the wall. Also, results of impinged spray area were increased according to increasing injection pressure.

  • 본 논문에서는 소형 PEM(Proton Exchange Membrane) 연료전지 스택의 BOS(Balance of Stack) 제어에 관하여 논의한다. 별도의 가습 장치가 필요치 않고 BOS의 구성이 비교적 간단한 소형 PEM 연료전지 시스템에서는 팬과 퍼지밸브만의 제어를 통해 스택 내부 수분의 조절과 열 관리가 수행된다. 따라서 본 논문에서는 부하에 따른 최적의 공기유량을 계산하고 요구되는 유량의 공급을 위해 팬을 제어하는 알고리즘을 통해 소형 연료전지 시스템의 과도응답 특성과 안정성을 향상시키는 방법에 관하여 제안한다. 150W급 소형 연료전지 시스템을 꾸미고, 마이크로컨트롤러를 이용한 제어회로를 구현하여 실험함으로써 제안된 알고리즘의 유용함을 검증하였다.

Genetic Relationships of Cattle Breeds Assessed by PCR-RFLP of the Bovine Mitochondrial DNA D-loop Region

  • To investigate the genetic relationships among various cattle breeds, bovine mtDNA D-loop region was used in 411 animals of 18 cattle breeds, including 8 Asian Bos taurus, 7 European Bos taurus, 1 Asian Bos indicus, and 2 African Bos indicus. The size of amplified PCR products from mtDNA D-loop region was 964 bp and the products were digested by 15 different restriction enzymes. Two different band patterns were identified in eight restriction enzymes (BstXI, Hae III, Msp I, Apa I, Taq I, Alu I, BamH I, EcoN I) and the rest of restriction enzymes showed more than 3 different band patterns among which Apo I and MspR9 resulted in 7 different restriction patterns. The genotypes, number of haplotype, effective number of haplotype, and degree of heterozygosity were analyzed. Based on all the PCR-RFLP data, different haplotypes were constructed and analyzed for calculating genetic distances between these breeds using Nei's unbiased method and constructing a phylogenetic tree.

Molecular Phylogeny of the Gayal in Yunnan China Inferred from the Analysis of Cytochrome b Gene Entire Sequences

  • The gayal (Bos frontalis) in China is a very rare semi-wild and semi-domestic bovine species. There still exist remarkable divergences on the gayal's origin and taxonomic status. In the present study, the cytochrome b (Cyt b) gene entire sequences (1,140 bp) of 11 gayals in Yunnan China were analyzed. Combined with other bovine Cyt b sequences cited in GenBank, the phylogenetic trees of genus Bos were reconstructed by neighbor-joining (NJ) and maximum parsimony (MP) methods with Bubalus bubalis as outgroup. Sequence analysis showed that, among 1,140 sites compared for 11 gayals, 95 variable sites (8.33% of all sites) and 6 different haplotypes were observed, showing abundant mitochondrial genetic diversity in gayals. Both NJ and MP trees demonstrated that gayals in this study were markedly divided into three embranchments: one embranchment clustering with Bos gaurus, another clustering with Bos taurus, and the third clustering with Bos indicus. The result of phylogenetic analysis suggested that the gayal might be the domesticated form of the gaur, and a great proportion of the gayal bloodline in China was invaded by other bovine species.
