Calcium Carbonate Precipitation by Bacillus and Sporosarcina Strains Isolated from Concrete and Analysis of the Bacterial Community of Concrete

  • Microbially induced calcium carbonate precipitation (CCP) is a long-standing but re-emerging environmental engineering process for production of self-healing concrete, bioremediation, and long-term storage of CO2. CCP-capable bacteria, two Bacillus strains (JH3 and JH7) and one Sporosarcina strain (HYO08), were isolated from two samples of concrete and characterized phylogenetically. Calcium carbonate crystals precipitated by the three strains were morphologically distinct according to field emission scanning electron microscopy. Energy dispersive X-ray spectrometry mapping confirmed biomineralization via extracellular calcium carbonate production. The three strains differed in their physiological characteristics: growth at alkali pH and high NaCl concentrations, and urease activity. Sporosarcina sp. HYO08 and Bacillus sp. JH7 were more alkali- and halotolerant, respectively. Analysis of the community from the same concrete samples using barcoded pyrosequencing revealed that the relative abundance of Bacillus and Sporosarcina species was low, which indicated low culturability of other dominant bacteria. This study suggests that calcium carbonate crystals with different properties can be produced by various CCP-capable strains, and other novel isolates await discovery.

Root Exudation by Aphid Leaf Infestation Recruits Root-Associated Paenibacillus spp. to Lead Plant Insect Susceptibility

  • Aphids are a large group of hemipteran pests that affect the physiology, growth, and development of plants by using piercing mouthparts to consume fluids from the host. Based an recent data, aphids modulate the microbiomes of plants and thereby affect the overall outcome of the biological interaction. However, in a few reports, aboveground aphids manipulate the metabolism of the host and facilitate infestations by rhizosphere bacteria (rhizobacteria). In this study, we evaluated whether aphids alter the plant resistance that is mediated by the bacterial community of the root system. The rhizobacteria were affected by aphid infestation of pepper, and a large population of gram-positive bacteria was detected. Notably, Paenibacillus spp. were the unique gram-positive bacteria to respond to changes induced by the aphids. Paenibacillus polymyxa E681 was used as a rhizobacterium model to assess the recruitment of bacteria to the rhizosphere by the phloem-sucking of aphids and to test the effect of P. polymyxa on the susceptibility of plants to aphids. The root exudates secreted from peppers infested with aphids increased the growth rate of P. polymyxa E681. The application of P. polymyxa E681 to pepper roots promoted the colonization of aphids within 2 days of inoculation. Collectively, our results suggest that aphid infestation modulated the root exudation, which led to the recruitment of rhizobacteria that manipulated the resistance of peppers to aphids. In this study, new information is provided on how the infestation of insects is facilitated through insect-derived modulation of plant resistance with the attraction of gram-positive rhizobacteria.

Chronic Toxicity of the Triazole Fungicide Tebuconazole on a Heterocystous, Nitrogen-Fixing Rice Paddy Field Cyanobacterium, Westiellopsis prolifica Janet

  • This study explored the chronic effects of different doses of the triazole fungicide tebuconazole on the growth, and metabolic and enzymatic functions of the filamentous paddy field cyanobacterium, Westiellopsis prolifica Janet. The growth of the cyanobacterium was determined by an estimation of the change in pigment contents. Chlorophyll-a, carotenoids, and accessory pigments such as phycocyanin, allophycocyanin, and phycoerythrin were shown to decline over a 16-day period by a factor of 92%, 93%, 83%, 95%, and 100%, respectively, with increasing doses of the fungicide. Metabolic and enzymatic activities were also adversely affected. Over the 16 days, a gradual rise in total phenol content was recorded when Westiellopsis prolifica Janet was treated with 60 ppm of the fungicide, despite the reduction in carbohydrates, proteins, and amino acids by 96%, 92%, and 90%, respectively. Moreover, the enzymes nitrate reductase (NR), glutamine synthetase (GS), and succinate dehydrogenase (SDH) also registered reductions of 93%, 90%, and 98%, respectively. This study indicates that tebuconazole, although an important fungicide used extensively in rice fields, exhibits an inhibitory effect on the growth and metabolic activities of Westiellopsis prolifica Janet and hence possibly on other varieties as well.

  • In this study, we studied strategic directions of urban management through evaluation the value of the investment with the consideration of the development plan, the condition of the location, the potential of the site, green and disaster prevention infrastructure, focused on Gugal-dong(Gugal Station area) and the region of Bora, Jung-dong, in Yongin city. In terms of the methodology, we tried to decide using Analytic Network Process which can consider the relation between the evaluation items. In conclusion, for the development plan, the order of evaluation items is development purpose, key tenant, fund raising and marketing, and for the condition of the location, the order of evaluation items is land use, surrounding environment, and traffic environment, and for the development potential, the order is marketability, identity, and historic character, and for the green and disaster prevention infrastructure, the order is prevention of human, natural, social disaster. The significance of the conclusion of this study is that it can be utilized in pre-evaluation in planning the urban development.

  • Recently, with rapid development of digital broadcasting technology, high-definition video service increased interest and demand. supplied mobile and image device support that improve 4~16 time existing Full HD. Such as high-definition contents supply, proposed compression for high-efficiency video codec (HEVC). Therefore, watermarking technology is necessary applying HEVC for protecting ownership and intellectual property. In this paper, analysis of prediction mode in intra frame and study feasibility of watermarking in re-encoding based HEVC. Proposed detect un-changed blocks in intra frame, using the result of analysis prediction mode.

Biodegradable Screws Containing Bone Morphogenetic Protein-2 in an Osteoporotic Rat Model

  • Objective : The aim of this study was to evaluate the effect for biodegradable screws containing bone morphogenetic protein-2 (BMP-2) in an osteoporotic rat model. Methods : Twenty-four female Wistar rat (250-300 g, 12 weeks of age) were randomized into four groups. Three groups underwent bilateral ovariectomy (OVX). Biodegradable screws with or without BMP-2 were inserted in the proximal tibia in two implantation groups. The extracted proximal metaphysis of the tibiae were scanned by exo-vivo micro-computed tomography. Evaluated parameters included bone mineral density (BMD), trabecular bone volume (BV/TV), trabecular number, trabecular thickness, and trabecular separation (Tb.Sp). The tibia samples were pathologically evaluated by staining with by Hematoxylin and Eosin, and trichrome. Results : Trabecular formation near screw insertion site was evident only in rats receiving BMP-2 screws. BMD and BV/TV significantly differed between controls and the OVX and OVX with screw groups. However, there were no significant differences between control and OVX with screw BMP groups. Tb.Sp significantly differed between control and OVX and OVX with screw groups (p<0.05), and between the OVX and OVX with screw BMP group (p<0.05), with no statistically significant difference between control and OVX with screw BMP groups. Over the 12 weeks after surgery, bone lamellae in direct contact with the screw developed more extensive and thicker trabecular bone around the implant in the OVX with screw BMP group compared to the OVX with screw group. Conclusion : Biodegradable screws containing BMP-2 improve nearby bone conditions and enhance ostoeintegration between the implant and the osteoporotic bone.

First Report of Six Trichoderma Species Isolated from Freshwater Environment in Korea

  • Trichoderma (Hypocreaceae) is one of the most ubiquitous genera worldwide. This genus has an excellent ability to adapt to diverse environments, even under poor nutritional conditions, such as in freshwater. However, little is known about the diversity of Trichoderma species in freshwater environments. In this study, we isolated diverse fungal strains from algae, plant litter, and soil sediment in streams in Korea. The strains were identified based on molecular phylogenetic analyses of internal transcribed spacer (ITS) rDNA and translation elongation factor 1 ($TEF1{\alpha}$) sequences. We also investigated their morphological characteristics by microscopic observation and determination of cultural features on different media. As a result, six Trichoderma species were found in Korea: T. afroharzianum, T. capillare, T. guizhouense, T. paraviridescens, T. reesei, and T. saturnisporum. Interestingly, T. paraviridescens showed both cellulose activity and hypoxia stress tolerance phenotypes, indicating its role as a decomposer in freshwater ecosystems. Our study revealed that freshwater environment could be a good candidate for investigating the species diversity of Trichoderma.

A Study of Social Media User Response about Firms' Crisis Response Strategies (기업의 위기대응전략에 대한 소셜 미디어 이용자의 반응 연구)

  • The importance of online communication is getting increased by the rapid growth of smartphone supply and Social Network Service (SNS) use. Catching up with the trend, firms are actively use SNS to improve brand image, promote products, and communicate with customer. On the one hand, SNS is the channel for firms' marketing activities, but on the other, it is also the channel where the events related to the firms propagate in real time. Firms are led to unexpected state of crisis, when events are quickly spread out on SNS. Then firms are assessed their image by the way they deal with the state of crisis. This paper proposes to figure out user response on SNS according to each crisis response strategies by analyzing event-related twitter data when crisis situations of firms arise. We classify crisis response strategies into response attitude, defensive and accommodative response, and response speed, fast and slow response. This paper suggests optimal crisis response strategy to firms regarding state of crisis propagated on SNS.

Risk Factors for Suicidal Ideation among Middle Class Korean: Focusing on Psychosocial Comparison - An Analysis of a Nationwide Survey of the 8th Korea Health Panel Data (중산층 한국인의 생애주기별 자살 충동경험과 위험요인: 비교의식과 관련된 사회심리적 특성을 중심으로 - 8차 한국의료패널 자료분석)

  • Purpose: The purpose of this study was to examine the prevalence and risk factors of suicidal ideation among middle class Korean. Methods: Cross sectional study was designed for secondary data analysis. From the 8th Korea Health Panel survey (2008~2013), a total of 6,037 data was drawn and analyzed by developmental stage using descriptive statistics including frequency, percentage, $x^2$ test, and logistic regression analysis. Results: Across all age groups, high physical-mental stress, frustration, anxiety about the future and low self-perceived health status or social class were found to be the risk factors of suicidal ideation. Peer-compared subjective health status and frustration significantly influenced the adolescents. The young adults'suicidal ideation was mainly influenced by physical and mental stress, frustration and absence of economic activity. For the middle-aged, physical and mental stress, frustration, future anxiety, low peer-compared subjective health status were found to be the major influencing factors. The predominant risk factors for the elderly were frustration and low peer-compared subjective health status. Conclusion: Making comparisons to others significantly influence suicidal ideation throughout all life cycles. Improvement of mental health and suicide prevention can be enhanced by avoiding negative comparison to others.

The Korea National Suicide Survey (KNSS) : Rationale and Design

  • Suicide, the fourth leading cause of death in Korea, is a serious national problem. The Korea National Suicide Survey (KNSS) is the result of the first legislation to address this issue, "Article 11 of THE ACT FOR THE PREVENTION OF SUICIDE AND THE CREATION OF A CULTURE OF RESPECT FOR LIFE" (the "Act for the Prevention of Suicide"). To overcome the limitations of previous studies, the KNSS was designed by collaborators from a variety of fields : psychology, epidemiology, social welfare, and psychiatry. The KNSS was composed of four substudies that addressed the multifaceted process involved in suicide-related behavior over time, exploring general attitudes toward suicide and suicidal ideation, suicide planning, suicide attempts, and completed suicides. Study 1 examined the risk factors for suicide based on data regarding completed suicides ; Study 2 adopted the approach of a psychological autopsy ; Study 3 focused on suicide attempters ; and Study 4 explored attitudes toward suicide in the general population. The KNSS was designed to provide a comprehensive understanding of suicide from a longitudinal, multifaceted perspective to serve as a basis for policies aimed at suicide prevention.
