Purpose: The goal of this research is to find the role of collagen membrane, which can reduce physical damage, as a scaffold for possible alternative to the corticotomy which causes Regional Acceleratory Phenomenon (RAP). Materials and Methods: The experiments were carried out on 12 New Zealand white rabbits, approximately 3.5 kg in bodyweight. We made an incision on the skin of the mandibular border and applied 37% phosphoric acid and collagen membrane to the mandibular bone surface of the first group (experimental group), and only phosphoric acid to the second group (control group). After 3 days, 1 week, and 2 weeks, 4 rabbits each were sacrificed and specimens were obtained. Each specimen was stained by H&E and Tartrate-resistant acid phosphatase (TRAP), and histological changes were observed by light microscope. Results: The demineralization of the experimental group was weak compared to the control group. It also showed a gradual increase of demineralization (after 3 days, 1 week, and 2 weeks) and the control group showed more extensive demineralization than the experimental group. Conclusion: This study demonstrates the amount of demineralization as a result of using phosphoric acid, and as time went by, demineralization increased. The absorbable collagen membrane was used as a scaffold to increase bone demineralization effect and prevent dispersion to adjacent tissues, but rather the amount of bone demineralization decreased. Therefore, the role of collagen membrane as a scaffold for RAP was weak.

Autogenous tooth bone graft material contains organic and inorganic components for osteoinductive and osteoconductive healing. The clinical availability and safety of this material have been confirmed by various experimental and clinical studies. In the future, allogenic and xenogenic tooth bone graft materials, ideal scaffold using teeth for stem cells and bone growth factors, and endodontic and tooth restorative material will be developed.

A highly porous Biphasic Calcium Phosphate (BCP) scaffold was fabricated by the sponge replica method with a microwave sintering technique. The BCP scaffold had interconnected pores ranging from $80\;{\mu}m$ to $1000\;{\mu}m$, which were similar to natural cancellous bone. To enhance the mechanical properties of the porous scaffold, infiltration of polycaprolactone (PCL) was employed. The microstructure of the BCP scaffold was optimized using various volume percentages of polymethylmethacrylate (PMMA) for the infiltration process. PCL successfully infiltrated into the hollow space of the strut formed after the removal of the polymer sponge throughout the degassing and high pressure steps. The microstructure and material properties of the BCP scaffold (i.e., pore size, morphology of infiltrated and coated PCL, compressive strength, and porosity) were evaluated. When a 30 vol% of PMMA was used, the PCL-BCP scaffold showed the highest compressive strength. The compressive strength values of the BCP and PCL-BCP scaffolds were approximately 1.3 and 2MPa, respectively. After the PCL infiltration process, the porosity of the PCL-BCP scaffold decreased slightly to 86%, whereas that of the BCP scaffold was 86%. The number of pores in the $10\;{\mu}m$ to $20\;{\mu}m$ rage, which represent the pore channel inside of the strut, significantly decreased. The in-vitro study confirmed that the PCL-infiltrated BCP scaffold showed comparable cell viability without any cytotoxic behavior.

Silk sericin, a natural protein from silkworm cocoons, is emerging as a multifunctional biomaterial in biomedicine, particularly in tissue engineering and wound healing. Recent studies have highlighted its biocompatibility, biodegradability, and potential for chemical modification, which allows it to be incorporated into various scaffold architectures. This review article synthesizes current research, including the development of sericin-based hydrogel scaffolds for tissue engineering and sericin's role in enhancing wound healing. Key findings demonstrate sericin's ability to refine scaffold porosity and mechanical strength, expedite tissue healing, and reduce bacterial load in wounds. The integration of sericin into novel bioactive dressings and its use in peripheral nerve injury repair are also discussed, showcasing its adaptability and efficacy. The convergence of these studies illustrates the broad applications of sericin, from scaffold design to clinical interventions, making it a promising material in regenerative medicine and tissue engineering, with the potential to improve patient outcomes significantly.
최근 조직 공학 분야에서는 폴리카프로락톤(PCL), 수산화인회석, 삼인산칼슘, 이상인산칼슘(BCP), 지르코니아(Zirconia, $ZrO_2$) 와 같은 합성 생체폴리머와 생체세라믹 등은 다양한 생체 조직 또는 장기를 재생하는데 필요한 대체재로 사용되고 있다. 따라서, 본 연구의 목적은 골 조직 재생을 위한 혼합된 $ZrO_2$/BCP/PCL(ZBP) 인공지지체의 특성을 관찰하기 위함이다. 단선 패터닝 실험의 결과를 토대로 내부연결성 있는 공극을 가지고 $45^{\circ}+135^{\circ}$ 타입과 격자타입의 새로운 복합 공극 패턴을 가지는 혼합된 ZBP 인공지지체는 폴리머 적층시스템에 의해 성공적으로 제작되었다. 뿐만 아니라 기계적 특성에 대한 ZBP 인공지지체의 효과를 분석하였다. 게다가 MG63 세포에 대한 ZBP 인공지지체의 세포 상호작용은 CCK-8 분석을 이용함으로써 평가되었다.

Treatment of polymicrobial infected musculoskeletal defects continues to be a challenge in orthopaedics. This research investigated single and dual-delivery of two antibiotics, vancomycin and amikacin, targeting different classes of microorganism from a biodegradable calcium sulfate-chitosan-nHA microsphere composite scaffold. The addition of chitosan-nHA was included to provide additional structure for cellular attachment and as a secondary drug-loading device. All scaffolds exhibited an initial burst of antibiotics, but groups containing chitosan reduced the burst for amikacin at 1hr by 50%, and vancomycin by 14-25% over the first 2 days. Extended elution was present in groups containing chitosan; amikacin was above MIC ($2-4{\mu}g/mL$, Pseudomonas aeruginosa) for 7-42 days and vancomycin was above MIC ($0.5-1{\mu}g/mL$ Staphylococcus aureus) for 42 days. The antibiotic activity of the eluates was tested against S. aureus and P. aeruginosa. The elution from the dual-loaded scaffold was most effective against S. aureus (bacteriostatic 34 days and bactericidal 27 days), compared to vancomycin-loaded scaffolds (bacteriostatic and bactericidal 14 days). The dual- and amikacin-loaded scaffolds were effective against P. aeruginosa, but eluates exhibited very short antibacterial properties; only 24 hours bacteriostatic and 1-5 hours bactericidal activity. For all groups, vancomycin recovery was near 100% whereas the amikacin recovery was 41%. In conclusion, in the presence of chitosan-nHA microspheres, the dual-antibiotic loaded scaffold was able to sustain an extended vancomycin elution longer than individually loaded scaffolds. The composite scaffold shows promise as a dual-drug delivery system for infected orthopaedic wounds and overcomes some deficits of other dual-delivery systems by extending the antibiotic release.
배경: 혈관질환의 수술에 사용되는 인공 도관의 막힘과 문합부위의 좁아짐 등을 개선하기 위한 방법으로 조직공학적인 방법과 자가 세포를 이용한 인공혈관의 제작이 대안으로 대두되고 있다. 저자들은, 생흡수성이 있는 고분자 폴리머 지지체와 자가 골수세포를 이용한 인공혈관으로 생체실험을 시행하였다. 대상 및 방법: 생분해성 고분자 재료인 poly (lactide-co-${\varepsilon}$-caprolactone) (PLCL)과 poly(glycolic acid) (PGA) fiber로 혈관용 지지체를 제작한 후, 피실험 동물의 골수를 채취하여 혈관 내피 세포와 평활근 세포로 분열시켜 배양한 후 혈관 지지체위에 이식하였다. 만들어진 인공 혈관을 잡견의 복부대동맥에 이식한 후 3주 후에 혈관 조영술을 시행하고, 안락사 후에 혈관을 제거하여 조직학적 검사를 시행하였다. 결과: 6마리의 잡견 중 2마리에서 수술 후 10일에 혈관 지지체의 균열에 의한 대량 출혈로 사망하였다. 나머지 4마리의 잡견은 수술 후 3주까지 생존하였으며, 혈관 조영술상 혈관의 막힘이나 좁아짐은 발견되지 않았다. 인공 혈관의 내면은 작은 혈전들이 붙어 있었으며, 조직학 검사에서 정상 혈관과 유사한 3층의 구조를 나타내었다. 또한 면역화학 검사에서 혈관 내피세포와 혈관 평활근 세포가 재생된 것을 확인하였다. 결론: 고분자 폴리머와 자가 골수세포를 이용한 인공혈관은 생체 내에서 정상혈관과 유사한 모양으로 재생이 가능함을 보여주었다. 그러나, 동맥압력에 견디기 위해 혈관 지지체의 물성에 대한 개량과 충분한 양의 혈관 세포를 얻기 위한 연구가 더 필요할 것으로 생각된다.
direct bone apposition during bone remodelling. To address these problem, we developed a new ceramic, calcium metaphosphate(CMP), and report herein the biologic response to CMP in subcutaneous tissue, muscle and bone. Porous CMP blocks were prepared by condensation of anhydrous $Ca(H_2PO_4)_2$ to form non-crystalline $Ca(PO_3)_2$. Macroporous scaffolds were made using a polyurethane sponge method. CMP block possesses a macroporous structure with approximate pore size range of 0.3-1mm. CMP blocks were implanted in 8mm sized calvarial defect, subcutaneous tissue and muscle of 6 Newzealand White rabbits and histologic observation were performed at 4 and 6 weeks later. CMP blocks in subcutaneous tissue and muscle were well adapted without any adverse tissue reaction and resorbed slowly and spontaneously. Histologic observation of calvarial defect at 4 and 6 weeks revealed that CMP matrix were mingled with and directly apposed to new bone without any intervention of fibrous connective tissue. CMP blocks didn't show any adverse tissue reaction and resorbed spontaneously also in calvarial defect. This result revealed that CMP had a high affinity for bone and was very biocompatible. From this preliminary result, it was suggested that CMP was a promising ceramic as a bone substitute and tissue engineering scaffold for bone formation.
Scaffolds play a crucial role in the tissue engineering. Biodegradable polymers with great processing flexibility and biocompatability are predominant scaffolding materials. New developments in biodegradable polymers and their nanocomposites for the tissue engineering are discussed. Recent development in the scaffold designs that mimic nano and micro features of the extracellular matrix (ECM) of bones, cartilages, and vascular vessels are presented as well.
