  • The early tissue reactions in the periodontal tissues of the tension zones following the application of force (30gm) to the maxillary first molar teeth of the albino rats were studied by the light microscopy and electron microscopy The increase of periodontal fibroblasts was evident, particularly in 1 day survival period. Osteoblast differentiation and new bone formation on the alveolar bone surface were occurred from 1 day survival period. Mononuclear phagocytes occurred consistently and in relatively high number adjacent to and at some distance from blood vessel Extensive breakdown of collagen fibers was observed. The increase of the phagocytosis of collagen by the active fibroblasts was evident Also, collagen fibrils were sparse or lost near the macrophage.

단치소요산가미방이 Dexamethasone 처리한 랫드의 두개골 세포에 미치는 영향 (Effects of DSG on Osteoblastic Cell from Rat Calvariae in the Presence of Dexamethasone)

  • It is well known that glucocorticoid may induce osteoporosis as its side effect in long-term therapy. The inhibition of osteoblast by glucocorticoid is also recognized as its action mechanism of decreased bone formation. In this study, the effect of DSG, Danchisoyosangamibang, on the differentiation and function of osteoblastic cells was investigated. The osteoblastic cells were isolated from rat calvariae using collagenase treatment. The cell counting, enzyme activity assay, MTT assay, collagen content assay were done to determine the cell proliferation, intracellular alkaline phosphatase (ALP) activity, bone martrix production, and cell apoptosis. DSG enhanced the cell proliferation after the culture for 10 days. ALP activity and total protein synthesis, and intracelluar collagen synthesis were increased time dependently when the cells were treated with DSG in the presence of dexamethasone. And, DSG restored calvarial cell function decreased by dexamethasone.

  • The inhibition of osteoblast by glucocorticoid is recognized as its action mechanism of decreased bone formation. In this study, the effect of JY, Jahyulyangkeuntang, on the differentiation and mineralization of osteoblastic cells was investigated in the presence of dexamethasone. The cell counting, enzyme activity assay, MTT assay, collagen content assay were done to determine the cell proliferation, alkaline phosphatase(ALP) activity, bone martrix production, and cell apoptosis. Protective Action of Cartilage and Bone Destruction by Deer Antler Herbal-acupuncture Solution, the Pilose Antler of Cervus Korean TEMMINCK Var. Mantchuricus Swinhoe, on Type II Collagen-induced Arthritis in Mice

  • Objectives : The effects of water extract of deer antler herbal-acupunture solution(DHS), prepared from the pilose antler of Cervus korea TEMMINCK var. mantchuricus Swinhoe (Nokyong), a traditional immunosuppressive and immune-activating Korean herbal- acupuncture, on collagen-induced arthritis(CIA:RA model) in mice was studied. Destruction of cartilage and bone are hallmarks of human rheumatoid arthritis, and controlling these erosive processes is the most challenging objective in the treatment of RA. Methods : We investigated the tissue protective effects of deer antler treatment using established murine collagen-induced arthritis(CIA) as a model. Potential synergy of low dosages of anti-inflammatory glucocorticosteroids and deer antler was also evaluated. Results : Treatment of established murine CIA with deer antler herbal-acupunture solution(DHS) $(10-50{\mu}g/day)$ suppressed disease activity and protected against cartilage and bone destruction. Although $10-50{\mu}g/day$ DHS had only a moderate effect on the inflammatory component of the disease activity, it strongly reduced cartilage pathology, as determined by histological examination. Serum cartilage oligomeric matrix protein(COMP) levels were significantly reduced, confirming decreased cartilage involvement. Histological analysis showed that bone destruction was prevented. DHS administration increased serum IL-1Ra levels and reduced anticollagen type II antibody levels. Treatment with low-dose $DHS(1{\mu}g/day)$ was ineffective in suppressing disease score, serum COMP or joint destruction. Synergistic suppression of both arthritis oseverity and COMP levels was noted when low-dose DHS was combined with prednisolone(0.05mg/kg/day), however, which in itself was not effective. Conclusion : DHS was shown to have the inhibiting effects against $IL-1{\alpha}-$ and $IL-1{\beta}-stimulated$ bone resorption. These results indicated that the DAS is not only highly stable and applicable to clinical uses in bone resorption, but also it will be served as a potent anti-inflammatory and anti-arthritic agents for treatment of human RA.

  • Purpose : The aim of this study was to investigate healed bovine bone particles ($Bio-Oss^{(R)}$) and absorbable collagen sponge ($CollaPlug^{(R)}$) applied extraction socket site at 4-6 months' post-extraction. Material and methods : From August, 2004 to October, 2005, 17 sockets in 5 adult patients were selected out of the patients whose received ridge preservation using bovine bone particles and absorbable collagen sponges at Dept. of oral and maxillofacial surgery in Samsung Medical Center. There were 5 male patients, ages 30 to 58 years. Immediate postoperation and 4-6 months after operation study models were compared to evaluate the ridge dimension by measuring vertical height and horizontal width of alveolar ridge. Results : The measurements at 4-6 months revealed, in the ridge dimension, a loss of vertical height of 0.91${\pm}$0.40mm and horizontal width of 1.25${\pm}$0.58mm. There was no adverse reaction. Conclusion : This study suggests that treatment of extraction sockets with graft materials and collagen sponges is valuable in preserving alveolar bone in extraction sockets and preventing alveolar ridges defects.

  • The role of cultured bone marrow stromal cells (BMSCs) in peripheral nerve regeneration was examined using an established rabbit peroneal nerve regeneration model. A 15-mm peroneal nerve defect was bridged with a vein filled with BMSCs $(1{\times}10^6)$, which had been embedded in collagen gel. On the contralateral side, the defect was bridged with a vein filled with collagen gel alone. When the regenerated tissue was examined 4, 8 and 12 weeks after grafting, the number and diameter of the myelinated fibers in the side with the BMSCs were significantly higher than in the control side without the BMSCs. This demonstrates the potential of using cultured BMSCs in peripheral nerve regeneration.

Bone regeneration and graft material resorption in extraction sockets grafted with bioactive silica-calcium phosphate composite (SCPC) versus non-grafted sockets: clinical, radiographic, and histological findings

  • Purpose: The purpose of the present study was to evaluate the effect of silica-calcium phosphate composite (SCPC) granules on bone regeneration in extraction sockets. Methods: Ten patients were selected for a split-model study. In each patient, bone healing in SCPC-grafted and control ungrafted sockets was analyzed through clinical, radiographic, histomorphometric, and immunohistochemical assessments 6 months postoperatively. Results: A radiographic assessment using cone-beam computed tomography showed minimal ridge dimension changes in SCPC-grafted sockets, with 0.39 mm and 1.79 mm decreases in height and width, respectively. Core bone biopsy samples were obtained 6 months post-extraction during implant placement and analyzed. The average percent areas occupied by mature bone, woven bone, and remnant particles in the SCPC-grafted sockets were 41.3%±12%, 20.1%±9.5%, and 5.3%±4.4%, respectively. The percent areas of mature bone and woven bone formed in the control ungrafted sockets at the same time point were 31%±14% and 24.1%±9.4%, respectively. Histochemical and immunohistochemical analyses showed dense mineralized bundles of type I collagen with high osteopontin expression intensity in the grafted sockets. The newly formed bone was well vascularized, with numerous active osteoblasts, Haversian systems, and osteocytes indicating maturation. In contrast, the new bone in the control ungrafted sockets was immature, rich in type III collagen, and had a low osteocyte density. Conclusions: The resorption of SCPC granules in 6 months was coordinated with better new bone formation than was observed in untreated sockets. SCPC is a resorbable bone graft material that enhances bone formation and maturation through its stimulatory effect on bone cell function.

강성(剛性) 고정(固定)이 성장중(成長中)인 가토(家兎)의 전두비골봉합(前頭鼻骨封合)에 미치는 영향(影響)에 관(關)한 연구(硏究) (THE EFFECT OF RIGID FIXATION ON THE FRONTONASAL SUTURE IN GROWING RABBITS)

  • This study was aimed to clarify the histopathological changes in the experimental animal model subjcted to rigid fixation performed across the frontonasal sutrue in growing rabbits. Sixteen rabbits aged 6 weeks used. In experimental group(n=12), rigid fixation with miniplates and screws was performed across the frontonasal suture. Control group(n=4) was those with periosteal elevation only. Experimental animals were sacrificed on the 2nd, 4th, 8th, and 12th week after operation, and frontonasal suture area was excised for light microscopic and scanning electron microscopic examination. The results obtained were as follows : 1. In control groups, collagen fiber bundles ran in the midportion of bone sutrue and cambial layers were seen at bone surface. Sutural surfaces are beveled and external and internal bony projected portions were observed. 2. In experimental groups, distance of bone suture was decreased by new bone formation on the 2nd week, while increased by bone resorption at the miniplate applied area and bone formation in the adjacent bone on the 4th week. 3. In experimental groups, the original bone surface was almost resorbed and new bone formation was found on the 8th week. Regulary-run collagen fibers, smooth and dense bone surfaces were similar to the bone patterns of control groups on the 12th week. Above results suggest that bone formation is restricted where the miniplate is applied, while compensatory growth is appeared in the adjacent bont. It is considered that rigid fixation with miniplates and acrews results in a little disturbance of sutural growth of the craniofacial bone in infancy and children when applied for short duration.

Changes in Serum Biochemical Markers of Bone Cell Activity in Growing Thoroughbred Horses

  • We studied the changes in biochemical markers of bone metabolism in growing Thoroughbred horses. Serum osteocalcin (OC), as a marker for bone formation, and carboxy-terminal propeptide of type-I collagen (PICP), as a marker for bone formation, carboxy-terminal telopeptide of type-I collagen (ICTP), as a marker for bone resorption, were determined in nine clinically healthy horses from 3 d to 17 mo of age. The BW and withers height (WH) increased during the study. On the other hand, a rapid reduction in body weight gain (BWG) was observed between 1 mo and 9 mo of age and a rapid reduction in withers height gain was observed between 1 mo and 5 mo of age. The serum markers decreased significantly with increasing age. In particular, dramatic changes in serum markers occurred between 3 d to 1 wk and 5 to 7 mo of age in these horses, which suggests that bone turnover rapidly decreased after birth. On the other hand, the ratio of PICP to ICTP decreased through the experiment. This result suggests that the reduction in bone formation exceeded that of bone resorption. There was a significant correlation between markers and growth parameters, except for the correlation between PICP and BWG on single linear regression analysis. Serum OC and ICTP were affected by the WH in multiple linear regression analysis. These results indicated that the age-related variation in serum biochemical markers of bone metabolism reflected bone growth, but neither BW nor BWG. Therefore, we consider that changes in bone modeling are the major factor affecting the levels of serum biochemical markers by 17 mo of age in horses.

Benefits of mineralized bone cortical allograft for immediate implant placement in extraction sites: an in vivo study in dogs

  • Purpose: The aim of the present study was to evaluate the effectiveness of using a mineralized bone cortical allograft (MBCA), with or without a resorbable collagenous membrane derived from bovine pericardium, on alveolar bone remodeling after immediate implant placement in a dog model. Methods: Six mongrel dogs were included. The test and control sites were randomly selected. Four biradicular premolars were extracted from the mandible. In control sites, implants without an allograft or membrane were placed immediately in the fresh extraction sockets. In the test sites, an MBCA was placed to fill the gap between the bone socket wall and implant, with or without a resorbable collagenous membrane. Specimens were collected after 1 and 3 months. The amount of residual particles and new bone quality were evaluated by histomorphometry. Results: Few residual graft particles were observed to be closely embedded in the new bone without any contact with the implant surface. The allograft combined with a resorbable collagen membrane limited the resorption of the buccal wall in height and width. The histological quality of the new bone was equivalent to that of the original bone. The MBCA improved the quality of new bone formation, with few residual particles observed at 3 months. Conclusions: The preliminary results of this animal study indicate a real benefit in obtaining new bone as well as in enhancing osseointegration due to the high resorbability of cortical allograft particles, in comparison to the results of xenografts or other biomaterials (mineralized or demineralized cancellous allografts) that have been presented in the literature. Furthermore, the use of an MBCA combined with a collagen membrane in extraction and immediate implant placement limited the extent of post-extraction resorption.
