Adenosine derived from Staphylococcus aureus-engulfed macrophages functions as a potent stimulant for the induction of inflammatory cytokines in mast cells

  • In this study, we attempted to isolate novel mast cell-stimulating molecules from Staphylococcus aureus. Water-soluble extract of S. aureus cell lysate strongly induced human interleukin-8 in human mast cell line-1 and mouse interleukin-6 in mouse bone marrow-derived mast cells. The active molecule was purified to homogeneity through a $C_{18}$ reverse phase HPLC column. By determination of its structure by MALDITOF and $^1H$- and $^{13}C$-NMR, adenosine was revealed to be responsible for the observed cytokine induction activities. Further studies using 8-sulfophenyl theophylline, a selective adenosine receptor blocker, verified that purified adenosine can induce interleukin-8 production via adenosine receptors on mast cells. Moreover, adenosine was purified from S. aureus-engulfed RAW264.7 cells, a murine macrophage cell line, used to induce phagocytosis of S. aureus. These results show a novel view of the source of exogenous adenosine in vivo and provide a mechanistic link between inflammatory disease and bacterial infection.

A FINITE ELEMENT STRESS ANALYSIS OF TOOTH AND IMPLANT SUPPORTED FIXED PARTIAL DENTURE ACCORDING TO THE LOCATION OF NON-RIGID CONNECTOR (치아 및 임플랜트 지지 고정성 국소의치의 비고정성 연결부의 위치에 따른 유한요소법적 응력분석)

  • The purpose of this study was to analyze the stress distribution and the displacement happened to the abutment, the prosthesis, and the surrounding structure according to the location of the nonrigid connector, that is, the keyway in the distal of canine and the mesial of the implant in the three unit fixed partial denture. Two-dimensional finite element model ws constructed and analyzed for the stress distribution and the displacement using software ABAQUS(Ver 5.2 Hibbitt, Karisson & Sorenson, Inc., 1992). After finishing the finite element model, the distribution load of 15kg was applied simultaneously to the all cusp tips of the prosthesis and the concentration load of 10㎏ was applied respectively at the each cusp tip of the prosthesis. The following results were obtained : 1. The amount of displacement of the implant was greater in case of the non-rigid connection than the rigid connection, and the more favorable displacement was shown in case of the IKb than the IKa. 2. Without regard to the connection method, the stress represented at the surrounding bone was similar, and the more favorabel stress distribution was shown in case of IKb. 3. The maximum stress was concentrated at the fastening screw and the neck of implant in all experimental groups, and their stress magnitudes were in the order of IKb, IR, and IKa.

  • In this study, new platinum complexes were synthesized utilizing the ligand of a 2,2'-bispyrimidine (bpim), 2,2'-bipyridine (bpy), 5,5'-dimethyl-2,2'-bipyridine (5,5-mebpy), 5'-bromo-2,2'-bipyridine (5-brbpy), 5,5'-dibromo-2,2'-bipyridine (5,5-brbpy), 4,4'-dimethyl-2,2'-bipyridine (4,4-mebpy), 4,4'-dihexyl-2,2'-Bipyridine (4,4-hebpy), 1,10'-Phenanthroline (phen), 3,4,7,8'-tetramethyl-1,10'-Phenanthroline (3,4,7,8-phen). In order to determine chemical structure of Synthesized platinum complexes, $^1H(^{13}C)$-NMR, UV-vis and FT-IR were used and optical physics and chemical properties were measured PL. In the case of platinum complexes, wavelength has been identified 356~421 nm. Quantum efficiency in DMSO solution was appeared 0.05~0.46.

  • The differential electromagnetic transducer for IME(implantable middle ear) system, which have two small permanent magnets glued with the same pole facing each other in the coil, has high vibration efficiency and does not influenced by external magnetic field. In this paper, using finite element method, highly efficient structure of the transducer was proposed and vibration force of the transducer was calculated by electromagnetic theory. And the necessary vibration force of transducer to transmit the sound signal to inner ear when the transducer is attached at stapes was calculated and the design parameters of the transducer were investigated. Using the parameters, the differential electromagnetic transducer was manufactured in small size to implant in confined human middle ear. And it was examined by unloaded and loaded vibration experiment using temporal bone sampled from cadaver.

  • Severely absorbed edentulous ridge cannot bear mechanical stress, causes undesired transformation of oral environment and makes patients difficult to adapt to dentures. Nowadays implant overdenture can be a treatment of choice in order to relieve patients' discomfort and improve stability and retention of the denture. Placement of implant on maxilla is difficult because of its bone quality and anatomic structure. It also has wide supportive tissue and convenience of border sealing, which provides sufficient support and stabilization with conventional complete denture. Mandible, on the other hand, is difficult to obtain sufficient support, retention and stabilization with conventional complete denture. Therefore, implant overdenture is recommended on mandible. Locator attachment has been improved for convenience of use and male parts of various retention enabled it to replace ball type attachment clinically. In this study, we restored maxillary arch with conventional denture, and mandibular arch with implant and tissue-supported overdenture and Locator attachment system.

Magnetic Properties of Multiferroic h-HoMnO3 (Multiferroic h-HoMnO3의 자기적 성질 연구)

  • Multiferroic $HoMnO_3$ single crystal was prepared using 4-point focused floating zone furnace, and polycrystalline $HoMn_{1-x}\;^57Fe_xO_3$ (x=0.00, 0.01, 0.02, 0.05) powders have been prepared by solid state reaction. Their magnetic and crystallographic properties are studied using MPMS, PPMS, and $M\ddot{o}ssbauer$ spectroscopy. The crystal structure found to be a hexagonal and a magnetic easy-axis is (110) direction. As the external applied magnetic field increases, temperature of the dielectric constant anomaly is decreased. $HoMn_{0.95}\;^{57}Fe_{0.05}O_3$ shows huge quadrupole splitting value from the $M\ddot{o}ssbauer$ spectra.

  • Computed tomography(CT) has become more widely available in recent years and is recognized as an important diagnostic tool for complex disease and condition of the orofacial structure in adults. In pediatric patients, especially in the mixed dentition period, the number of superimposing dental structures in the jaw bone is much greater than in the permanant dentition period. Therefore, in spite of radiation risk, computed tomography should sometimes be used in pediatric patients in order to obtain a precise and valid diagnosis. This case study presents clinical application of computed tomography on patients visiting department of pediatric dentistry in Seoul National University Dental Hospital. The results obtained through these cases were summarized as follow : 1. Localization of impacted teeth using CT allows for efficient treatment planning leading to reduced postoperative complications. 2. In orthodontic traction of impacted permanent teeth, utilization of CT made exact localization of window site and determination of prognosis possible. 3. Use of CT made possible definite diagnosis of lesions in the jaw that were only suspected on plain film radiographs. 4. Careful selection is necessary when applying CT on children due to increased radiation dose and high cost.

  • Today, there is considerable evidence to support a cause-effect relationship between microbial colonization and the pathogenesis of implant failures. The presence of bacteria on implant surfaces may result in an inflammation of the peri-implant mucosa, and, if left untreated, it may lead to a progressive destruction of alveolar bone supporting the implant, which has been named as peri-impantitis. Several maintenance regimens and treatment strategies for failing implants have been suggested. Recently, in addition to these conventional tools, the use of different laser systems has also been proposed for treatment of peri-implant infections. As lasers can perform excellent tissue ablation with high bactericidal and detoxification effects, they are expected to be one of the most promising new technical modalities for treatment of failing implants. It is introduced that Er,Cr:YSGG laser, operating at 2780nm, ablates tissue by a hydrokinetic process that prevents temperature rise. We studied the change of the titanium implant surface under scanning electron microscopy after using Er,Cr:YSGG laser at various energies, irradiation time. In this study, Er,Cr:YSGG laser irradiation of implant fixture showed different effects according to implant surface. Er,Cr:YSGG laser in TPS surface with RBM not alter the implant surface under power setting of 4 Watt(W) and irradiation time of 30sec. But in TPS surface with $Ca_3P$ coating alter above power setting of 2W and irradiation time of 10sec. TPS surface with RBM showed microfracture in 4W, 30sec and TPS surface with $Ca_3P$ coating showed destruction of fine crystalline structure, melting in excess of 2W, 10sec. We concluded that proper power setting, air, water of each implant surface must be investigated and implant surface must be irradiated under the damaged extent.

  • Currently, the lack of equipments for the medical practice and education made it impossible for the people in medical institution to carry out suitable experiments for observing human bodies. In this paper, the authors embodied three dimensional images and moving pictures for the human skeletal structure, digestive organs, cardiovascular system and their processes over the internet framework. The three dimensional images and moving picture made it possible for the general people as well as the specialists to observe and obtain informations with regard to the human body. specially, the authors realized a framework for visualizing the human bodies in three dimensional images, via which a detailed and realistic architecture for the human body and its organs tan be obtained. The system developed in this paper can be used in the practice and education of the people engaged in medical fields.

Synthesis and bioactivity evaluation of metal ion-substitution biphasic calcium phosphate for bone defect reconstruction (골결손부 재건을 위한 금속 이온 치환 이상인산칼슘 합성 및 생체 활성 평가)

  • The co-precipitation technique has been applied to synthesize Biphasic Calcium Phosphate (BCP), Mg-BCP and Si-BCP. X-ray diffraction (XRD) and Fourier transformed infrared (FTIR) spectroscopy were used to characterize the structure of synthesized BCP, Mg-BCP and Si-BCP powders. The results have shown that BCP and substitution of magnesium and silicon in the calcium deficient apatites revealed the formation of biphasic mixtures of Hydroxyapatite (HAp)/${\beta}$-Tricalcium phosphate (${\beta}$-TCP) ratios after heating at $1000^{\circ}C$. Ionic substituted BCP is able to develop a new apatite phase on the surface in contact with physiological fluids faster than BCP does. An MTT assay indicated that BCP, Mg-BCP, and Si-BCP powders had no cytotoxic effects on MG-63 cells, and that they have good biocompatibility.
