Collagen Type VII (COL7A1) as a Longevity Mediator in Caenorhabditis elegans: Anti-Aging Effects on Healthspan Extension and Skin Collagen Synthesis

  • Longevity genes and senescence-related signaling proteins are crucial targets in aging research, which aims to enhance the healthy period and quality of life. Identifying these target proteins remains challenging because of the need for precise categorization and validation methods. Our multifaceted approach combined bioinformatics with transcriptomic data to identify collagen as a key element associated with the lifespan of the model organism, Caenorhabditis elegans. By analyzing transcriptomic data from long-lived mutants that involved mechanisms such as antioxidation, dietary restriction, and genetic background, we identified collagen as a common longevity-associated gene. We validated these findings by confirming that collagen peptides positively affect lifespan, thereby strengthening the validity of the target. Further verification through healthspan factors in C. elegans and functional assays in skin fibroblasts provided additional evidence of the role of collagen in organismal aging. Specifically, our study revealed that collagen type VII is a significant target protein for mitigating age-related decline. By validating these findings across different aging models and cell-based studies, we present compelling evidence for the anti-aging effects of collagen type VII, highlighting its potential as a target for promoting healthy aging. This study proposes that collagen not only serves as an indicative marker of organismal longevity across various senescence-related signaling pathways, but also offers a mechanistic understanding of skin degeneration. Consequently, collagen is an effective target for interventions aimed at mitigating skin aging. This study underscores the potential of collagen type VII (bonding collagen T7) as a therapeutic target for enhancing skin health and overall longevity.

상아질 접착에 대한 matrix metalloproteinase (MMP)의 영향과 이를 극복하기 위한 전략 (Effects of matrix metallproteinases on dentin bonding and strategies to increase durability of dentin adhesion)

  • The limited durability of resin-dentin bonds severely compromises the longevity of composite resin restorations. Resin-dentin bond degradation might occur via degradation of water-rich and resin sparse collagen matrices by host-derived matrix metalloproteinases (MMPs). This review article provides overview of current knowledge of the role of MMPs in dentin matrix degradation and four experimental strategies for extending the longevity of resin-dentin bonds. They include: (1) the use of broadspectrum inhibitors of MMPs, (2) the use of cross-linking agents for silencing the activities of MMPs, (3) ethanol wet-bonding with hydrophobic resin, (4) biomimetic remineralization of water-filled collagen matrix. A combination of these strategies will be able to overcome the limitations in resin-dentin adhesion.

산 표면처리된 상아질 표층의 교원섬유 용해가 레진-상아질간 결합에 미치는 영향 (EFFECT OF COLLAGEN DISSOLUTION IN ACID CONDITIONED DENTIN LAYER ON RESIN-DENTIN ADHESION)

  • The effect of collagen dissolution in acid conditioned dentin layer on resin - dentin adhesion was investigated. 160 freshly extracted human molars were divided into 4 groups randomly and dentin surfaces were exposed. 40 exposed dentin surfaces were not acid conditioned and each 10 of them were applied with bonding agents within dentin bonding systems of All Bond 2, Scotchbond Multipurpose, Clearfil Photobond and Superbond D - Liner respectively. Each 10 of another 40 exposed dentin surfaces were acid conditioned by the acid within the above four bonding systems respectively and applied with corresponding bonding systems. After acid conditioning of the other 40 exposed dentin surfaces as above, they were treated with 5% NaOCl for 2 minutes, and each 10 of them were applied with the above four dentin bonding systems respectively. The remaining 40 dentin surfaces were acid conditioned and treated with 10% NaOCl for 2 minutes, and each 10 of them were applied with corresponding bonding agents as the above. After the procedures were finished, composite resin (Z -100, 3M Dent. Prod., USA) were applied on the dentin surfaces and light cured. Shear bond strength values were measured. Surface changes of fractured dentin specimens were observed using SEM (Hitachi S-2350, Japan). The following results were obtained. 1. In all of dentin bonding systems, shear bond strengths of non - conditioned specimens were significantly lower than those of acid conditioned specimens (P<0.05). 2. A statistically significant difference of bond strengths did not exist between acid conditioned specimens and 5% NaGCI retreated specimens applied with All Bond 2, Scotchbond Multipurpose and Clearfil Photobond (P>0.05). However, strength values of 5% NaOCl retreated specimens applied with Superbond D - Liner were lower than those of acid conditioned specimens (P<0.05). 3. In all the applied dentin bonding systems except Clearfil Photobond, bond strengths of 10% NaOCl retreated specimens were lower than those of acid conditioned and 5% NaOCl retreated specimens (P<0.05). 4. The resin - dentin hybrid layer of 4 - $5{\mu}m$ thickness was formed in the acid conditioned specimens applied with All Bond 2, Scotchbond Multipurpose and Superbond D-Liner. 5. The resin - dentin hybrid layer of 3 - $4{\mu}m$ thickness was still formed in the 5% NaOCl retreated specimens applied with All Bond 2 and Scotchbond Multipurpose. In addition, this layer was not completely removed after the retreatment with 10% NaOCl. Above results indicate that the dissolution of collagen in acid conditioned dentin layer by NaOCl solution can not be achieved completely and the collagens contribute to the resin - dentin adhesion considerably.

  • The effect of collagen dissolution in acid conditioned dentin was morphologically examined by both scanning and transmission electron microscopy. 18 freshly extracted human molars and dentin bonding systems of All Bond 2, Scotchbond Multipurpose, Superbond D-Liner were used in this study. For SEM preparation, each 3 of ~ exposed dentin surfaces were acid conditioned by using various acids within the above three bonding systems respectively. After acid conditioning of the other 3 exposed dentin surfaces as above, they were treated with 1.7% NaOCl for 2 minutes. The remaining 3 dentin surfaces were acid conditioned and treated with 3.3 % NaOCl for 2 minutes. All of the specimens were then fixed in 4 % glutaraldehyde for 12 h at $4^{\circ}C$ and dehydrated in ethanols grades from 50 % to 100 %, then surface changes of the specimens were observed by using SEM. For TEM preparation, exposed dentin surfaces were acid conditioned with the same acid as SEM specimens and treated with 1.7%, 3.3 % NaOCl respectively, then applied with corresponding bonding agents. After the procedures were finished, composite resin were applied on the dentin surfaces and light cured. Small, rectangular sticks with end dimensions of approximately 1 by 1 mm were sectioned and further sample preparative techniques for transmission electron microscopy were performed in accordance with the procedures used for ultrastructural TEM observations of calcified tissues. The results were as follows : 1. In the 1.7 % NaOCl retreated specimens after acid conditioning, the porous dentin surface of intertubular dentin and wide opening of dentinal tubules were appeared. And there were fine irregularities on the intertubular dentin, indicating a clear difference as compared with the acid conditioned specimens. 2. In the 3.3% NaOCl retreated specimens after acid conditioning, the intertubular dentin was further eroded causing a more porous and wider opening of dentinal tubules. Moreover, sharp irregularities on the intertubular dentin were more evident than those of acid conditioned and 1.7% NaOCl retreated specimens. 3. In all of the acid conditioned specimens, the resin-dentin hybrid layer of approximately 3.5mm thickness was formed and the collapsed collagen layer was observed on the uppermost part of hybrid layer in the specimens applied with All Bond 2. The collgen fibrils of intertubular dentin in specimens applied with Scotchbond Multipurpose were running perpendicular to the interface, and electron dense black layer demarcated from the deep unaltered dentin was more evident in the specimen applied with Superbond D-Liner than any other specimens. 4. In the 1.7 % NaOCl retreated specimens after acid conditioning, the resin-dentin hybrid layer of approximately 2.5-3.0mm thickness was formed and the collapsed collagen layer and longitudinally running collagen fibrils as shown in the acid conditioned specimens were observed in the specimens applied with All Bond 2 and Superbond D-Liner. 5. In all of the 3.3% NaOCl retreated specimens after acid conditioning, the evidence of resin-dentin hybrid layer was not identified ; nevertheless, the longitudinally running collagen fibrils remained slightly in the specimens applied with All Bond 2.

In vivo에서 습윤접착과 상아질 교원섬유용해가 변연누출에 미치는 영향 (IN VIVO EFFECT OF WET BONDING AND COLLAGEN DISSOLUTION IN ACID CONDITIONED DENTIN ON MARGINAL LEAKAGE)

  • The purpose of this study was to evaluate the degree of marginal leakage that is made by wet bonding to dentin and dissolution of dentinal collagen layer in vivo and in vitro. Class V cavities were prepared to 32 cat's canine teeth and they were randomly assigned into the following 4 groups : Group 1 (not acid conditioned) ; Group 2 (acid-conditioned and blot-dried) ; Group 3 (acid-conditioned and air-dried for 30 seconds) ; Group 4 (acid-conditioned and retreated with 10% NaOCl solution). The acetone-based primer and bonding agent of All Bond 2(Bisco Inc., U.S.A.) and composite resin(Z-100, 3M. Dent. Prod., U.S.A.) were then applied. The resin collar and cap attached to each specimen and 50% silver nitrate solution was placed inside the resin cap for 3 hours. After the canine tooth was resected in level of cervical third, specimens were highly polished and observed the degree of the dye penetration on incisal and gingival margins. The results were as follows ; 1. There was no significant difference of marginal leakage in all the groups between in vivo and in vitro. 2. In vivo, the degree of leakage on the gingival margin showed the lowerest in group 4 and followed by group 2, 3 and group 1 with ascending order. However, there was no statistical difference among all the groups(p>0.05). 3. In vitro, the degree of leakage on the gingival margin showed the lowerest in group 2 and followed by group 4, 3 and group 1 with ascending order. However, there was no significant difference among the experimental groups(p>0.05). 4. The marginal leakage of the incisal margin in vitro was significantly higher in group 1 than the remaining groups(p<0.05).

치과용복합레진과의 결합에 있어, 상아질 내 교원섬유의 역할에 관한 연구 (TITLE : THE ROLE OF COLLAGEN FIBER IN DENTIN BONDING)

  • The purpose of this study was to investigate the effects of moistening mechods of dentin on the morphologic states of hybrid layers and on the interfacial bond strength between dentin and composite. Specimens were divided into 6 groups based on the surface moistening methods and materials used. After the dentin surface was conditioned with 10 % phoporic acid and irrigated: 1. The dentin surface was dried. Then bonding agent and composite were applied. 2. The dentin surface was blot-dryed. Then primer, bonding atent and composite were applied. 3. The dentin surface was dryed first. Within 20 seconds, the surface was rewetted, then primer, bonding agent and composite were applied. 4. The dentin surface was dryed. Then primer, bonding agnent and composite were applied. 5. The dentin surface was dryed first. Atter 24hrs, the surface was rewetted, and then primer, bonding agent and composite were applied. 6. The surface was conditioned with NaOCl for 5min. Then primer, bonding agent and composite were applied. To reveal the hybrid layer, scanning electron microscopy was used after the samples were ion beam etched. The shear bond strength of each group was tom pared by ANOVA. In groups 2, 3 and 4, the hybrid layer was clearly visible, but the width was more limited in group 4. In group 1 and 5, the hybrid layer was not found, and a gap was formed between dentin and composite. In group 6, the hybrid layer was not found, but the interface between the dentin and composite was intimate. The shear bond strength of each group was as follows: Group 1 : 4MPa, Group 2 : 14MPa, Group 3 : 12MPa, Group 4 : 14MPa, Group 5 : 5MPa, Group 6 : 9MPa.

Is dentin biomodification with collagen cross-linking agents effective for improving dentin adhesion? A systematic review and meta-analysis

  • Objectives: The aim of this investigation was to evaluate the effectiveness of collagen cross-linking agents (CCLAs) used in combination with the adhesive technique in restorative procedures. Materials and Methods: In this systematic review, the authors followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses checklist. An electronic search was performed using PubMed, Scopus, Web of Science, Cochrane Library, LILACS, and DOSS, up to October 2020. The gray literature was also researched. Only randomized clinical trials were selected. Results: The selection process yielded 3 studies from the 838 retrieved. The addition of CCLAs in the retention of restorations increased the number of events. The postoperative sensitivity scores and marginal adaptation scores showed no significant difference between the CCLA and control groups, and the marginal pigmentation scores showed a significant increase in the CCLA group. There were no caries events in any group throughout the evaluation period. Conclusions: This systematic review showed that there is no clinical efficacy to justify the use of CCLAs in the protocols performed.

Cyanate methacrylate가 상아질 결합강도에 미치는 영향 (THE EFFECT OF CYANATE METHACRYLATE ON THE SHEAR BOND STRENGTHS TO DENTIN)

  • 접착제의 술식을 간편하게 하고 효과적이며 안정적인 접착강도를 지닌 상아질 접착제를 개발하고자 isocyanate methacrylate의 농도를 달리해 상아질에 적용하여 전단결합강도를 측정하고 응집파괴양상을 분석, 평가하였다. Isocyanate methacrylate의 농도에 따라 0% (대조군), 2%, 4%, 6%, 8%, 10%, 및 12%의 7개 실험군으로 분류하였으며 , Instron (No.4466.USA)를 이용하여 복합레진의 전단 결합강도를 측정하고 Resin tag 및 응집파괴를 주사전자현미경으로 관찰해 다음 결론을 얻었다. 1. Cyanate methacrylate 8%군이 가장 높은 전단 결합강도는 나타내었으나 (33.62 KPa), 통계학적 유의성이 없었다 (P > 0.05). 2. Cyanate methacrylate 2% ,4% 및 6%군은 응집성파괴빈도가 대조군 (0%군)과 비교하여 높게 나타났다. 3. Cyanate menthacrylate군에서 레진tag의 길이는 짧게 나타났다. 이상의 연구의 결과로 cyanate methacrylate가 collagen과 반응하여 상아질 내로 침투를 방해하며 레진 tag의 길이를 짧게 하며 상아질의 유기성분과의 반응에 의하여 결합강도는 높아질 수 있으나, 상아질 자체의 강도가 낮아질 수 있어 상아질에 대한 결합강도의 실질적인 증가는 확인할 수 없었다.

비교원성 단백질이 상아질 접착제의 결합강도와 교원질의 형태에 미치는 영향 (THE EFFECT OF THE REMOVAL OF CHONDROITIN SULFATE ON BOND STRENGTH OF DENTIN ADHESIVES AND COLLAGEN ARCHITECTURE)

  • 본 연구는 상아질의 비교원성 단백질을 chondroitinase ABC (C-ABC)를 이용하여 제거함으로써 비교원성 단백질의 제거가 상아질 접착제의 미세인장결합강도와 교원질망의 형태에 미치는 영향을 상아질의 다양한 습윤상태에 따라 평가하고자 시행하였다. 비교원성 단백질의 상아질접착제의 미세인장강도에 대한 영향을 평가하기 위해 제 3대구치의 상아질을 노출시키고, 두 군으로 나누고 한 군은 C-ABC, 다른 군은 증류수를 $37^{\circ}C$에서 48시간 동안 적용한 후, 상아질의 습윤상태(wet, dry 및 re-wet)와 상아질 접착제(Single Bond 2, One Step Plus)를 다르게 이용하여 복합레진을 수복하였다. 24시간 후 가로 1 mm, 세로 1mm의 시편을 제작하고 미세인장강도를 측정하였다. 상아질 교원질의 형태변화를 관찰하기 위하여 상아질 시편에 산부식을 시행하고 C-ABC 적용 후, 시편을 제작하였고 미세인장강도 측정후 파괴된 접착면의 파괴양상과 각 접착제의 접착계면 관찰을 위하여 FE-SEM 관찰하였다. C-ABC 처리여부와 관계없이 습윤한 상아질면에 접착한 군은 모든 접착제에서 통계학적으로 유의성있는 미세인장결합강도의 차이를 나타나지 않았다(p > 0.05). C-ABC를 적용하였을 경우, Single Bond 2에서는 재수화한 상아질면에 접착한 군이 습윤한 상아질면에 접착한 군에 비해 미세인장결합강도가 감소하였다(p < 0.05). FE-SEM 관찰결과, C-ABC를 적용후에는 접착성 파괴가 주로 일어났으며, 교원질 섬유간 거리가 증가하였으며 부분적으로 교원질 섬유들간에 응집된 양상이 관찰되었다.

단일용기 상아질 접착제 처리 후 레진-상아질 경계면에 대한 투과전자현미경적 연구 (A TEM STUDY OF THE RESIN-DENTIN INTERDIFFUSION ZONE FORMED BY ONE-BOTTLE DENTIN ADHESIVE SYSTEMS)

  • One bottle system was recently developed in order to simplify the clinical skills and save chair time after continuous improvements on dentin bonding agents. There has been many studies to measure the bond strength of one bottle systems but no actual work has been done on micromorphologic study of resin-dentin interdiffusion zone after one bottle system application. To evaluate the bonding patterns of various commercially available one bottle systems to dentin, observation of resin-dentin interdiffusion zone under TEM was performed. Caries-free human third molars within one month of extractions were chosen for the experiments. The molars were sectioned 1mm above the cementoenamel junction and got rid of the root portions. Crown portions of the teeth were sectioned parallel to occlusal surface so that dentin discs of 1mm in thickness were remained. 7 one bottle systems and 1 two bottle system were applied according to manufacturer's instructions and followings were the results. 1. In every experimental groups, cross bandings of collagen fiber were distinguishable and tight bon dings between the bonding agents and dentin were observed. 2. Hybrid layer was clearly observed in ONE-STEP$^{(R)}$, Prime & Bond$^{(R)}$ 2.1, Syntac$^{(R)}$ SC, MAC-BOND II groups but it was not clear in Single Bond, D-Liner Dual PLUS, ONE COAT BOND groups. 3. Electron-density of hybrid layer was uniform in pattern in MAC-BOND II, Prime & Bond$^{(R)}$ 2.1 groups but not so uniform in ONE-STEP$^{(R)}$ group. 4. Electron-dense amorphous phase in most superior layer of the resin-dentin interdiffusion zone was characteristically observed in Single Bond, Syntac$^{(R)}$ SC, ONE COAT BOND groups. It can be concluded that bondings between the dentin bonding agents and dentin can be various in pattern according to their chemical compositions and the condition during applications.
