Two ${\beta}$-1,4-glucanases (DI and DIII fractions) were purified to homogeneity from the culture filtrate of a cellulolytic bacteria, Cellulomonas sp. CS 1-1, which was classified as a novel species belonging to Cellulomonas uda based on chemotaxanomic and phylogenetic analyses. The molecular mass was estimated as 50,000 Da and 52,000 Da for DI and DIII, respectively. Moreover, DIII was identified as a glycoprotein with a pI of 3.8, and DI was identified as a non-glycoprotein with a pI of 5.3. When comparing the ratio of the CMC-saccharifying activity and CMC-liquefying activity, DI exhibited a steep slope, characteristic of an endoglucanase, whereas DIII exhibited a low slope, characteristic of an exoglucanase. The substrate specificity of the purified enzymes revealed that DI efficiently hydrolyzed CMC as well as xylan, whereas DIII exhibited a high activity on microcrystalline celluloses, such as Sigmacells. A comparison of the hydrolysis patterns for pNP-glucosides (DP 2-5) using an HPLC analysis demonstrated that the halosidic bond 3 from the nonreducing end was the preferential cleavage site for DI, whereas bond 2, from which the cellobiose unit is split off, was the preferential cleavage site for DIII. The partial N-terminal amino acid sequences for the purified enzymes were $^1Ala-Gly-Ser-Thr-Leu-Gln-Ala-Ala-Ala-Ser-Glu-Ser-Gly-Arg-Tyr^{15}$-for DI and $^1Ala-Asp-Ser-Asp-Phe-Asn-Leu-Tyr-Val-Ala-Glu-Asn-Ala-Met-Lys^{15}$-for DIII. The apparent sequences exhibited high sequence similarities with other bacterial ${\beta}$-1,4-glucanases as well as ${\beta}$-1,4-xylanases.
Polyurethane dispersion (PUD) polymers were synthesized by using polyether and polyester polyol. The effect of ionomeric centers, r(NCO / OH) values, chain extender process, and chain extender types on the adhesion properties was investigated. In the case of polyether-based PUD, the ionic center, r value, chain extension process and chain extender types were not adjusted even after adjustment. In the case of polyester-based PUD, when the ionic center content was more than 2.5%, the state of adhesive strength was $2.0kgf/cm^2$ or more. On the other hand, the initial adhesive strength was excellent at about $1kgf/cm^2$ when the ionic center content was over 3.5%. When the r value was 1.3 or more, it was found that the initial bonding strength and the state of bonding strength were excellent at about $1kgf/cm^2$ and $2.1kgf/cm^2$ or higher, respectively. An IR spectrum analysis of the synthesized PUD confirmed that PUD was composed of urethane based on the N-H characteristic peak at $3340cm^{-1}$ and the urethane characteristic peak at $1730cm^{-1}$. Moreover, the characteristic peaks of the isocyanate ($2260cm^{-1}$) used in the preparation of the prepolymer were not observed. As a result, the residual -NCO was not observed, and urethane was completely synthesized.
This study presents the microstructure-electrical property relationship of reactive-sputtered Ta$_2$O$_{5}$ MIM capacitor structure processed by annealing in a vacuum and $O_2$ ambience. A microstructural investigation showed the existence of amorphous phase in as-deposited condition and the formation of preferentially oriented-Ta$_2$O$_{5}$ in $700^{\circ}C$ annealing. On annealing under the $O_2$ atmosphere, the Ta$_2$O$_{5}$ film exhibited the trend of its composition\`s approaching to stoichiometry from off-stoichiometry, analyzed by EPMA, the leakage current decrease and the enhanced temperature-capacitance characteristic stability. In the case of low temperature vacuum-annealing treatment, the leakage current behavior was stable irrespective of applied electric field. In the high temperature-annealed film at a vacuum condition, the electrical properties was observed to deteriorate. The results state that in Ta$_2$O$_{5}$ film annealed at $O_2$ atmosphere, gives rise to improvement of electrical characteristics in the capacitor were improved by reducing oxygen-vacancy and dandling Ta-O bond.-O bond.
The cracking behavior of prestressed concrete members is important for the rational design of prestressed concrete structures. However, the test data on the cracking behavior of prestressed concrete structures are very limited. The purpose of the present study is to investigate the crack spacing and crack width in transversely post-tensioned decks of concrete box girder bridges under applied loading. For this purpose, large scale test members of concrete box girder segments were fabricated and tested. The crack widths, crack spacings and crack patterns were investigated for various load levels. The crack widths and steel strains were continuously monitored during the loading process. To derive a rational predicton equation for crack width, the bond characteristics of post-tensioned steel and nonprestressed rebar in the PSC members were explored first. This was done by measuring the strains of prestressing steel and nonprestressed rebar in the test members under loading. A simple equation for the prediction of maximum crack width in transversely post-tensioned concrete one-way slabs is proposed by considering bond characteristic of prestressing steel and nonprestressed reinforcement. The comparison of proposed equation with experimental data shows good correlation. The present study indicates that ACI and CEB-FIP code equations exhibit rather large deviation from test data on prestressed concrete members.
Si direct bonding(SDB) technology is very attractive for both Si-on-insulator(SOI) electric devices and MEMS applications because of its stress free structure and stability. This paper presents on pre-bonding according to HF pre-treatment conditions in Si wafer direct bonding. The characteristics of bonded sample were measured under different bonding conditions of HF concentration, and applied pressure. The bonding strength was evaluated by tensile strength method. The bonded interface and the void were analyzed by using SEM and IR camera respectively. A bond characteristic on the interface was analyzed by using IT- IR. Si-F bonds on Si surface after HF pre-treatment are replaced by Si-OH during a DI water rinse. Consequently, hydrophobic wafer was bonded by hydrogen bonding of Si $OH{\cdots}(HOH{\cdots}HOH{\cdots}HOH){\cdots}OH-Si$. The bond strength depends on the HF pre-treatment condition before pre- bonding (Min:$2.4kgf/crn^2{\sim}Max:14.9kgf/crn^2$)
Structures of unprotonated [(NH3)n+(n = 1-6)] and protonated [NH4+(NH3)n-1(n = 1-6)] ammonia cluster cations have been optimized with ab initio Hartree-Fock (HF) and second-order MФller-Plesset (MP2)/6-31+G ** levels and the harmonic vibrational frequencies have also been evaluated. In unprotonated cluster cations, NH3+ forms as a central core of the first ammonia solvation shell. In protonated cluster cations, NH4+ forms as a central core. In unprotonated dimer and trimer cations, there are two types of isomers (hydrogen-bonded and head-to-head interactions). In both cluster cations, the hydrogen-bonded isomers are more stable. In the hydrogen-bonded dimer cation, the proton transfer reaction takes place from (NH3-HN+H2) to (NH4+-NH2). But in the other unprotonated cluster cations, the proton transfer does not take place. In unprotonated pentamer and hexamer, a NH3+ core has both interactions in a complex. On the other hand, in unprotonated tetramer a core has only the hydrogen-bonded type combined with neutral ammonia molecules. With increasing cluster cation size, the bond lengths [R(NN)] between two nitrogen atoms and the distances [R(N ...H)] of the hydrogen-bond increase reg-ularly. In the calculated infra-red absorption bands for ammonia cluster cations, the characteristic peaks of the bridged NH vibration of the hydrogen-bonded clusters appear near 2500 cm-1 . With increasing size, the peaks shift from 2306 cm-1 to 2780 cm-1 .
Concrete is the well-used material in many architectural and civil structures. The behavior of concrete does exhibit a different characteristic in compression and tension, and it also shows an inelastic-nonlinear behavior. In addition, the concrete properties vary slightly depending on the environmental factor and manufacturer. These properties of concrete make the modeling or simulation of concrete material difficult. In reinforced concrete, particularly, there is a difficulty in bond-slip relationship between concrete and steel. However, in this paper, reserving remainder of these limits the finite element analysis for reinforced concrete beams through ABAQUS simulation has been carried out with some assumptions. Assumptions include the perfect bond of steel and concrete as well as the concrete damaged plasticity (CDP) in concrete property. There is a reasonable agreement between the experimental and numerical results, although the analytical strength and external rod deformation are slightly overestimated. The average and standard deviation between two results are 1.05 and 0.05, respectively. And the models and the computations lead to the evolution of fracture in bending beam.
