Effects of Silkworm Extract on Streptozotocin-induced Diabetic Rats

  • This study was undertaken to find effects of ethanol extracts from Bombyx mori on the hyperglycemia induced by streptozotocin in rats. Blood glucose level of rats increased depending on experimental days after streptozotocin treatment. But there was no difference in blood glucose level between the rats injected with ethanol extracts of B. mori and normal rats. There was no significant difference in the kidney fibrosis between the rats injected with ethanol extrscts of B. mori and normal rats at all ages examined. The transforming growth factor-$\beta$l ($TGF-{\beta}1$) protein was detected in the glomerulay endothelial cells and tubular epithelial cells of streptozotocin rats at 14 days of age. In the rats injected with ethanol extracts of B. mori to the kidneys, the $TGF-{\beta}1$ protein was detected very faintly. The $TGF-{\beta}1$ mRNA of streptozotocin rats increased at 14 days of age, and this was higher than the rats injected with ethanol extrscts of B. mori. Taken these together, the results suggest that the ethanol extracts of B. mori ameliorate hyperglycemia of the rats induced by streptozotocin.

  • Soluble proteins of silkworm hemolymph were investigated by means of isoelectric focusing (IEF). The protein spectra during ontogenesis of races and inter-races hybrids kept in Bulgaria was studied. A total of 51 protein bands in the hemolymph from fourth larval instar to imago were ascertained. Stage specific expression was established. The specific expression of some protein bands in the individual spectra manifest phenotype of gene determinate polymorphism (HP F, HP J, HP K, HP L, HP Q - in the zone with pH gradient 3.5-6.2 and HP K, HP L, HP N, HP P, HP T - in the zone with pH gradient 9.5 - 6.2). Breed specific expression was observed. On the basis of the obtained results, it was established that the investigated breeds are heterogeneous and the isoelectric focusing method is successful when specifying the inner-race and inter-race polymorphism in silkworm.

  • In order to study the feasibility of utilizing bivoltine foundation cross (FC2) males of the silkworm as male components with Pure Mysore (PM), six foundation crosses viz,CSR2 x CSR27, CSR27 x CSR2, CSR27 x CSR50, CSR50 x CSR27, CSR2 x CSR50 and CSR50 x CSR2 along with CSR2 pure breed were assessed for performance of parental crop,grainage, rearing and reeling performance. The foundation crosses exhibited better performance than CSR2 as a parental crop and male component which resulted in higher egg recovery. Though there was not much difference between PM crossed with FC2 and CSR2 males pertaining to important bioassay parameters, PM x CSR2 exhibited superiority in reeling traits particularly with reelability and raw silk %. This study reveals that FC2 seed cocoons can be utilized during exigency when there is a dearth for CSR2 seed cocoons but not regularly.

Studies on the Hereditary Properties of SSR Marker in Silkworm (Bombyx mori L.)

  • Two BC1 group, c108 $\times$ (p50 $\times$ c108) and p50 $\times$ (p50 $\times$ c108), one group of F$_{2}$ progeny, (p50 $\times$ c108) F$_{2}$ ,and 3 SSR markers, F10539, FlO626 and FlO618 were used to test the hereditary properties of SSR markers in silkworm. FI0539, FlO626 were proved to be linkage, and FlO618 was proved to be independent to those two markers. According to Mendel's law, the recombinant value between F10539, FlO626 was calculated in all of these groups, and they were 8.55$\%$ (c108BC1), 8.02$\%$ (p50BC1) and 7.81 $\%$ (F$_{2}$) respectively. There was dominant difference among the crossing-over value using paired-samples tests by SPSS 10.0 software. This research proved that SSR markers were co-dominant in B. mori too, and F 2 progeny could be used to construct SSR linkage map although B. mori lacked of crossing over in females.

Preliminary Study on a Microsporidian Isolate Occurring in the Lamerin Breed of the Silkworm Bombyx mori L. in India

  • The silkworm, Bombyx mori L. is prone to infection of various pathogenic organisms. Pebrine, one of the deadliest disease of silkworm caused by highly virulent parasitic microsporidian, Nosema bombycis has been understood since long. Infections of the disease range from chronic to highly virulent and can result in complete lose to the sericulture industry. Several strains and species of microsporidians have since been isolated from the infected silkworms. A new microsporidian spore was isolated from Lamerin breed of the silkworm B. mori have been studied under scanning electron microscope, found to be different in spore size (length 4.36$\pm$0.06 ${\mu}{\textrm}{m}$, width 2.14$\pm$0.01${\mu}{\textrm}{m}$) and shape (ova cylindrical with slight depression) from standard strain N. bombycis (length 3.08$\pm$0.21 ${\mu}{\textrm}{m}$, width 2.01$\pm$0.05 ${\mu}{\textrm}{m}$ and ovidal respectively). In immunological test, the silkworm breed Lamerin isolated micrisporidian spore does not react to different antibody (N. bombycis, M$_{11}$ and M$_{12}$) sensitized latex particle and thus appeared to be a different strain of microsporidian parasitic to the Lamerin breed of the silkworm B. mori.i.i.

Hepatoprotective Effect of Bacillus subtilis-fermented Silkworm (Bombyx mori L.) Extract on Non-alcoholic Fatty Liver in Rats (고초균 발효누에 추출물이 비알코올성 지방간 유발 흰쥐에 미치는 간 기능 개선 효과)

  • The aim of this study was to investigate the potential effects of extracts from silkworm Bombyx mori L. fermented with Bacillus subtilis KACC 91157 at levels of 5%(v/w) and 10%(v/w) in Sprague-Dawley rats intoxicated with 1%(w/w) orotic acid (OA) for 10 days. The rats were divided into a normal group (N), a control group (C: OA), and treatment groups (SP10: OA + 10% extracts from B. mori L.; BSP5: OA + 5% extracts from B. mori L. fermented with B. subtilis KACC 91157; BSP10: OA + 10% extracts from B. mori L. fermented with B. subtilis KACC 91157). Serum activities of aspartate aminotransferase (AST), alanine transferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) increased following OA feeding, but the rise was slightly reduced by administration of BSP10. The total lipid, free fatty acid, phospholipid, total cholesterol, and triglyceride contents in serum were significantly lower in the OA treatment groups than in the N group. However, the contents slightly increased following the administration of BSP10. Glutathione concentrations in liver and serum were reduced in the OA-induced fatty liver, but they increased following the administration of BSP10. Hepatocytes in the OA-induced fatty liver contained numerous large droplets. However, SP10, BSP5, and BSP10 feeding prevented OA-induced lipid droplet accumulation in hepatocytes. Accordingly, extracts from silkworm powder fermented with B. subtilis could be an ideal material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

Egg Diapause Induction in Multivoltine Silkworm Bombyx mori for Long-term Germplasm Preservation

  • At present, multivoltine silkworm races reared five times per year involving huge manpower and rearing expenditure. Egg diapausing behavior is facultative in multivoltine and egg diapause was induced in selected multivoltine races by regulating temperature at $18^{\circ}C$, relative humidity 80% and photoperiod (6L:18D) in the late stage silkworm rearing. The maximum percentage of egg diapause induction was recorded in Rong Diazo, Diazo and MW13 showed 94%, 93% and 92% respectively, whereas the races A14DY and OS-616 showed minimum diapause induction 15% and 18% respectively. The diapause induced multivoltine eggs were preserved up to six months by cold preservation schedule normally adopted for bivoltine. After three and six months egg preservation, the diapause induced layings were released and observed for hatching percentage, all races showed above 82 % of hatching except the race AP12, which showed only 78 % of hatching. This methods reduce the crop cycle, gives strong safety backup and preventing the genetic erosion. This study helps formulating a new conservation method for multivoltine silkworm germplasm.

  • To evolve silkworm hybrids with higher survival and better cocoon characters, evaluation of semen hybrids in the laboratory (short-listed out of 143 hybrids) resulted in the identification of two promising hybrids, A3${\tines}$935 E (HSP1) and A3${\tines}$916 B (HSP2). The hybrids were evaluated at three different Regional Sericultural Research Stations of Central Silk Board during 1997-1998 and also tested with the farmers along with two control hybrids, KAxNB4D2 and PMxNB4D2. Evaluation of the hybrids indicated that these hybrids can be reared in all the seasons, especially during summer season. These hybrids show shorter larval duration (22 days 17 hrs against 23 days in control KAxNB4D2 and better cocoon characters as compared to the crossed breeds PM ${\tines}$NB4D2).

Evaluation and Identification of Promising Bivoltine Hybrids of Silk-worm, Bombyx mori L., for Tropics

  • Ten newly evolved bivoltine hybrids of silkworm (Bombyx mori L) were evaluated with control hybrid KA${\pm}$NB4D2 during three seasons of a year for their seasonal performance. Analysis of variance and other statistical methods were employed and the performance was observed in respect of 10 quantitative traits. The results showed significant genotype${\pm}$environment interaction with respect to four quantitative characters viz. fecundity, yield/10,000 larvae, filament length and raw silk (%). Environmental effects were significant for nine characters out of ten characters evaluated. A 105${\pm}$J2 and B${\pm}$NB4D2 were considered as highly adaptable hybrids to local conditions with high mean for maximum characters studied and found suitable to rear in all seasons.

  • Systemic fungicides viz., Bavistin and Beyleton are reported to have curative effect against the infection of Beauveria bassiana in silkworm Bombyx mori L. To understand the influence of the systemic fungicides on the disease suppression/development, hematological studies were carried out. There was an increase in the percent total hemocyte count, granulocyte and plasmatocyte in silkworm treated with the systemic fungicides. It possibly indicates the influence of systemic fungicide on the hemocyte mediated defense system leading to the higher resistance and suppression of disease development.
