Genetic Homogeneity in the Domestic Silkworm, Bombyx, and Phylogenetic Relationship Between B. mori and the Wild Silkworm, B. mandarina Using Mitochondrial COI Gene Sequences

  • Genetic variation in the domestic silkworm strains (Bombyx mori) and phylogenetic relationships between domestic silkworms and wild silkworms (B. mandarina) were investigated by using a portion of mitochondrial CGI gene sequences. Ten geographic strains of B. mori we sequenced were identical in the 410 bp-section of mitochondrial COI gene. This sequence was also identical to the homologous sequence of the four Gen-Bank-registered strains, but one strain of B. mori differed a single nucleotide (0.2%) from others. MtDNA homogeneity in the B. mori strains appears to be resulted from fixation into the mast frequent mtDNA type during the course of breeding for new strains, in which an extensive indoor rearing and removal of unwanted individuals were accompanied. In the comparisons between domestic and wild silkworms, some wild silkworms were closely related to domestic silkworms (0.2%-1.2% of divergence), but the others were not (2.7%-3.7% of sequence divergence). This result was also reflected in the phylogenetic analyses, showing two independent phylogenetic groups: one including all B. mandarina sequences and the other including both B. mandarina and B. mori sequences. Thus, domestic silkworms may have been derived from the ancestor of B. mandarina, which belongs to this group, alto-ough more extensive study will provide better understanding on this issue.

Characterization of Expressed Sequence Tags (ESTs) Generated from the Bombyx mandarina Whole Larvae and Molecular Cloning of Serine Protease Homologue Gene

  • We constructed an oligo-d(T) primed directional cDNA library from the Bombyx mandarina whole larvae. In an effort to isolate genes expressed in the B. mandarina, 227 expressed sequence tags (ESTs) were generated by single-pass sequencing from the cDNA library. Sequence analysis showed that 107 clones (47.1%) were classified into known genes and 120 clones (52.9%) were novel transcripts, which are unknown for their function. Of the 107 known genes, the most abundant gene was found to be actin and followed by serine protease in the expression profile. Among these clones, a serine protease homolog (BmSP) which is a class of proteolytic enzymes isolated. Full-length sequence of the BmSP cDNA clone was 922 bp in length and has an open reading frame of 276 amino acids. The conserved histidine, aspatic acid and serine residues forming the catalytic center as well as cysteine residues contributing to three disulphide bonds also were found in Bmsp gene. mRNA expression analysis revealed a high and specific expression of the gene only in midgut tissue, suggesting that BmSP gene is closely associated with the expression of digestive enzyme.

  • 천잠의 알에서 난황 주단백질인 vitellin을 분리·정제하고 가잠 및 상잠의 vitellin과 첨잠 nitellin과의 면역학적 특성을 비교하였으며 천잠의 배자발생에 따른 난내 vitellin의 변동을 조사하였다. 1. 전잠의 체액 vitellogenin은 유충기의 토사말기부터 검출되었다. 2. 점잠의 vitellinm은 가잠 및 상잠의 vitellin과 저기영동상의 이동도는 같았으나 면역학적인 특성에 있어서는 비동질성이었다. 3. 천잠 vitellin은 함량은 전체 가용성 난단백질의 46.0%였으며, 배자발생에 따른 vitellin의 변동은 산난후 8일까지는 같은 수준이었나, 10일 이후부터는 감소되는 경향을 보이다가 5개월된 난과 17개월된 난에서는 상당히 미량만이 검출되었다.

  • 누에 난황단백질의 주성분인 난황소(vitellin)를 분리하고 그 면역반응적 특성과 난소 이식과의 관계 및 배발생중의 양적 변화를 조사한 결과를 요약하면 다음과 같다. 1. 누에의 용혈액 vitellogenin과 난황소는 원심분리에 이은 DEAE-cellulose column chromatography에 의해 거의 순수하게 분리되었으며 두 단백질의 polyacrylamide gel 전기영동상의 이동도는 같았다. 2. 누에의 난황소와 이의 전구물질인 용혈림프 vitellogenin은 면역반응적으로 서로 동질임이 확인되었고, 가잠의 근록곤충인 상잠(Bombyx mandarina)의 난황단백질에는 가잠의 난황소와 면역학적으로 같은 반응을 나타낸 항원이 있는 반면, 천잠(Antheraea yamamai)의 난황단백질에는 가잠과의 공통항원이 없었다. 3. 수누에에 이식한 난소에서 생성된 난황단백질에는 난황소가 결여됨이 확인되어 난황소의 합성에는 난소이외의 다른 암컷 기관에서 생성된 전구물질이 필요함을 나타내었다. 4. 배발생중의 난황소는 배자발육 후기에 주로 기관의 형성에 이용되어 배자발육 초기부터 꾸준하게 이용되는 난특이단백질(ESP)과는 이용상 다른 특성을 보였다.

견사곤충에서 Mariner Transposase-like Element의 분자적 동정 (Molecular identification of Mariner Transposase-like Element from Four silkmoths)

  • 누에, 멧누에, 천잠 및 작잠에서의 형질전환용 vector 개발을 위한 첫단계로 이미 인간을 포함한 여러 곤충에서 그 존재가 확인된 mariner transposable element가 이들 견사 곤충에 존재하는지의 여부를 탐색하기 위해서 이미 보고된Drosophila mauritiana와 Hyalophora ceropia의 mariner transposase 유전자의 고도로 보존성이 높은 부위에 대한 degenerctive primer pair를 제작하여 이들의 존재 여부를 탐색하였다. 결과적으로 예상되는 약 500bp의 PCR product가 관찰됨에 따라서 모든 견사곤충의 genome에는 maminer-like element(MLE)가 존재한다는 것을 간접적으로 확인할 수 있었다. 이미 cloning된 pBmoMAR를 probe DNA로하여 southern hybridization을 수행한 결과에서도 확인 할 수 있었으며, 각 견사층의 genome내에 high copy로 존재한다는 것을 추정할 수 있었다. 따라서 이와 같은 결과는 견사곤충에서 MLE가 vector system으로써 개발될 수 있는 기초자료로 제공될 수 있을 것이다.

한국산 멧누에의 염색제수(예보) (The Number of Chromosomes of Wild Mulberry Silkworm, Bombyx mori, in Korea)

  • The number of chromosomes of the wild mulberry silkworm, Bombyx mandarina, collected in Suwon area in the middle part of Korean peninsula was investigated. The chromosomes were examined after smearing and squashing the testis of the last instar larvae which were at spining stage and the microscopic observation showed that the number of chromosomes of the wild mulberry silkworm was 27.

Comparison of gloverin gene expression patterns between domesticated and wild silkworms

  • Bombyx mandarina is widely accepted as ancestor of B. mori. Silkworms are served as well-characterized models for understanding the mechanism for the genetic regulation of development. In this study, we performed RNA-Seq analysis to examine tissue-expression of gloverin isoforms of the silk-gland, mid-gut, and fat body in B. mandarina. BLAST analysis revealed that four gloverin isoform gene sequences of B. mandarina were highly similar to B. mori. To identify the difference between two species, the expression profile of gloverin was measured by semi- RT-PCR analysis. The specific expression of gloverin isoform genes was observed mainly in the fat body from B. mori but not B. mandarina. However, all of tissues in the wild-type silkworm could induce the upregulation of compared with the B. mori. To validate the sudden increase in gloverin gene expression in the mid-gut tissue of B. mandarina, we were using qRT-PCR. Relative mRNA expression rate of gloverin at the wild-type silkworm was much higher than domestic silkworm. Comparative genomics between domesticated and wild silkworms showed different tissue-expression levels in some of immune related genes. These results are suggesting a trend toward decreasing immunity related genes expression during domestication. Further studies are needed to elucidate the silkworm domestication and an invaluable resource for wild silkworm genomics research.
