Development SCAR marker for the rapid authenticaton of Batryticatus Bombyx based on COI Sequences (COI 염기서열 기반 백강잠 신속 감별용 SCAR marker 개발 - 백강잠 유전자 감별 -)

  • Objectives : To ensure the safety, quality and pharmacological efficacy of Batryticatus Bombyx, it is important to discriminate with adulterants. In Korean Herbal Pharmacopoeias (KHP), the authentic species of Batryticatus Bombyx is defined only Bombyx mori. Therefore, the aim of this study is establishment of PCR assay method using the sequence characterized amplified region (SCAR) marker based on COI DNA barcode for discriminating six species related to Batryticatus Bombyx. Methods : Seventeen samples of six species (Bombyx mori, Bombyx mandarina, Rhodinia fugax, Oberthueria caeca, Actias artemis, and Caligula japponica) were collected from different habitate and nucleotide sequences of cytochrome c oxidase subunit I(COI) barcode regions were analyzed by Sanger sequencing methods. To develop SCAR-based PCR assay method, we designed species-specific primers based on COI sequence variabilities and verified those specificities using 17 samples of six species as well as commercial herbal medicines. Results : In comparative multiple analysis of COI sequences, six species were distinguished by species-specific nucleotides at the species level. To develop rapid and reliable PCR assay method for genetic authentication of Batryticatus Bombyx, therefore, we designed species-specific SCAR primers based on these nucleotide sequences and confirmed those specificities. Using these SCAR primers, We also established simple conventional PCR assay method using these SCAR primers at the species level. Conclusions : The comparative analysis of COI sequences and SCAR-based PCR assay methods represented equal results for distinguishing authentic Batryticatus Bombyx and adulterations at the species level. Therefore, our results are expected protecting adulteration of herbal medicine Batryticatus Bombyx.

Effects of a Two-Week-Oral-Toxicity Study of Bombyx batryticatus L. Extracts on C57BL/6 Mice (백강잠 추출물의 C57BL/6 마우스를 이용한 2주경구투여 독성시험)

  • Objectives : Bombyx batryticatus L. is the dried larval form of the silkworm (Bombyx mori L.) infected by Beauveria bassiania (Bals.) Vuill. It is used as a food and medicinal resource to treat asthma, headaches, epilepsy, and convulsions in traditional Korean and Chinese medicines. However, the research of the toxicity about B. batryticatus is not enough yet. Here, we investigate the effects of potential subacute toxicity following the repeated oral administration of B. batryticatus water extract to C57BL/6 mice, at various doses of 0, 50, 150, and 450 mg/kg/day during a two-week period. Methods : The following parameters were examined during the study period: body weight, gross findings, clinical signs, organ weight, hematology, serum biochemistry, histopathology, and mortality. At the end of the treatment period, all the mice were euthanized. Results : No changes were observed in the body weights, gross findings, clinical signs, organ weights, and mortality after two weeks of administration of the B. batryticatus extract. In addition, compared with the normal control group, no noticeable treatment-related changes were observed in the hematological, serum biochemical, and histopathological parameters in the treated group following treatment with doses of up to 450 mg/kg/day. Conclusion : Based on these findings, we conclude that the treatment of mice with the water extract of B. batryticatus did not cause considerable C57BL/6 toxicity, and therefore, it could be considered safe for further pharmacological studies.

Effects of Fructus Amomi Amari, Eucommiae Cortex, Bombyx Batryticatus Extract on Improving Symptoms of Late-onset Hypogonadism (익지인(益智仁), 두충(杜沖), 백강잠(白殭蠶) 혼합추출물이 남성갱년기 증상 개선에 미치는 영향)

  • In recent times, the number of men with late-onset hypogonadism has increased, and interest on this topic has also increased. This study was conducted to investigate effects of the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus on improve late-onset hypogonadism. The experimental subjects consisted of three groups: a control group consisting of 8-week-old male ICR mice that had undergone no treatment, an aging-elicited group (AE group) consisting of 50-week-old ICR male mice that had undergone no treatment, and a Mixed herbal extract treatment group (MT group) consisting of 50-week-old ICR male mice that had undergone the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus treatment (0.1 g/kg/day) for 6 months. After the experiment, the mice from all the experimental groups were dissected, and they were analyzed through histochemical and immunohistochemical methods. The mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus reduces aging-induced cell damage and oxidative stress and increases the secretion of serotonin and B-endorphin in aged mice, and promotes spermatogenesis in seminiferous tubules and reduces apoptosis and oxidative stress, and increases androgen receptor, $17{\beta}-HSD$ and GnRH, increases the ratio of smooth muscle to collagen fibers in the corpus cavernosum, increases eNOS, decreases PDE-5 and oxidative stress in aged mice, so it improves depression, reproductive, sexual problems caused by Late-onset hypogonadism. the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus inhibits the induction of osteoporosis by increasing decreased bone matrix distribution due to aging, increasing the activities of OPC and OPN, which are produced in osteoblasts, and decreasing RANKL, MMP-3 activity, increasing OPG activity. It also reduces muscle damage, oxidative stress, inflammation and apoptosis of muscle tissue, and increases Myo-D in the sartorius muscle of aged mice for improving muscle atrophy caused by by Late-onset hypogonadism.

Effect of Saryuktangkamibang on Streptozotocin-induced Diabetic Rat (사육탕가미방(四六湯加味方)이 streptozotocin으로 유발(誘發)된 백서(白鼠)의 실험적(實驗的) 탕뇨(糖尿)에 미치는 영향)

  • Objectives : The objective of this study is to observe the effect of Saryuktangkamibang, Saryuktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, on the experimental diabetes. Methods : In order to induce diabetes experimentally, injected streptozotocin to the vein in the tail of rats and then treated oral administration of Saryuktangkamibang water extracts. In the measurement of the variation levels of glucose, ALP, creatinine, and BUN concentration for each concentration levels for serum (0.2g/100g, 0.4g/l00g, 0.6g/100g), concentration level of glucose significantly decreased in 0.4g/l00g concentration level of Saryuktangkamibang. With this 0.4g/l00g concentration level of Saryuktangkamibang, the following conclusion was derived from the measurement of the serum levels of glucose. ALP, AST, ALT, creatinine, and BUN concentration for the injection of each 4th, 11th, and 18th day. Results : 1. In the measurement for each concentrations, the glucose concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group. 2. The creatinine and BUN concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 3. In the measurement for each dosage dates, the glucose concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 4. The AST concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group. Conclusion : Saryuktangkamibang, Saruktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, has an effect of hypoglycemia, with no hepato-renal dysfunction. For the experimental results above, in that nephrotoxicity significantly decreased on the 0.6g/100g concentration, another research such as study for effect in other ranges and study for effect of dosage term except the concentration range of this study is necessary. Also dosage of appropriate concentration of Saryuktangkamibang is thought to provide valuable aid to cure on diabetes.

Studies on Antioxidant, Anti-inflammatory and Whitening Effects of Oriental Herbal Extracts (Mix) including Eucommiae cortex (두충을 포함하는 한방추출물(Mix)의 항노화, 항염, 미백 효능 활성에 관한 연구)

  • Recently, due to the increase in skin diseases caused by particulate matter, endocrine disruptor and environmental changes, the trend of development of cosmetic materials has been shifting to the more safe and effective ingredients based on natural materials rather than existing synthetic compounds like steroids and antihistamines. This study aimed to develop a new natural cosmetic materials using oriental herbs such as Eucommiae cortex, Alpinia oxyphylla Miquel and Bombyx batryticatus. First, DPPH assay was performed to examine the antioxidative activity of the herbal extract (Mix) and 98.8% DPPH radical scavenging activity was confirmed at $400{\mu}g/mL$ concentration of it. In order to confirm the whitening efficacy of oriental herbal extracts(mix), the amount of melanin synthesized after stimulation of ${\alpha}-MSH$ with B16F10 cells was measured. Results showed that it was decreased to 27.1% comparing with the only ${\alpha}-MSH$ treated group, which confirmed the whitening efficacy. Also, both nitric oxide(NO) production and iNOS and COX-2 expression were significantly reduced in RAW264.7 macrophages activated by LPS in the presence of the extracts(Mix). The mRNA expression of the inflammatory cytokines such as $IL-1{\alpha}$, $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ was also analyzed to confirm the inhibition effect of the extracts on inflammation. Finally, to confirm the enhancement of skin barrier function, the expression of claudin 1 gene, a tight junction protein, was observed using human keratinocyte HaCaT cells and increased as concentration dependent manner. From these results, it is concluded that the oriental herbal extracts(Mix) containing Eucommiae cortex, Alpinia oxyphylla Miquel and Bombyx batryticatus is effective for antioxidant, anti-inflammation, skin whitening, and skin barrier and thus could be applied as a new natural cosmetic material.

A Survey of Aflatoxin Contamination in Medicinal Herbs for Food and Medicine (식약공용한약재의 아플라톡신 오염실태 조사)

  • The aim of this study was to determine the incidence and contamination levels of aflatoxin in Medicinal Herbs for Food and Medicine at Yakyeang market in seoul. 191 Samples 11 items medicinal herbs for food and medicine were evaluated for the aflatoxin contamination. in result 41 samples 10 items (21.5%) were detected in the alfatoxin, a high incidence of aflatoxins items are cassiae semen (50.0%), testudinis plastrum (43.8%) and Batryticatus Bombyx (40.0%), Polygalae Radix (31.2%), Zizyphi Semen (23.5%), Dolichoris Semen, Myristicae Semen (20.0%), Nelumbinis Semen (15.8%), Glycyrrhizae Radix et Rhizoma (7.4%), Hoveniae Semen Cum Fructus (4.3%). AFB1 were detected 27 cases (14.1%), AFB2, AFG1 and AFG2 were detected 18cases (9.4%), 16cases (8.4%) and 5cases (2.6%). The excess cancer risk estimated using the cancer potency of aflatoxin B1 (7(mg/kg/day)-1 for HBsAg-and 230(mg/kg/day)-1 HBsAg+) was N.D ~ $3.79{\times}10^{-6}$ for hepatits B surface antigen negative (HBsAg-) and N.D ~ $9.68{\times}10^{-5}$ hepatits B surface antigen positive (HBsAg+) respectively.

Anti-diabetic effect of medicinal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats (Streptozotocin으로 유도된 당뇨 흰쥐에서 하소에 사용되는 한약의 항당뇨 효과 비교 연구)

  • Objectives : To prove the channel-tropism theory in herbology, we investigated the anti-diabetic effect of six herbal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats. Methods : Diabetes was induced in male Sprague-Dawley rats by consecutive injection of streptozotocin (30 mg/kg i.p.) for 5 days. The rats were divided into normal control, diabetic control, and diabetic treatment with Lycii Radicis Cortex (LRC, 300 mg/kg); Corni Fructus (CF, 300 mg/kg); Bombyx Batryticatus (BB, 50 mg/kg); Lycii Fructus (LF, 300 mg/kg); Phellodendri Cortex (PC, 300 mg/kg); Epimedii Herba (EH, 300 mg/kg); and glibenclimide (10 mg/kg) as a reference drug. Herbal extracts or reference drug were administered orally for 28 days. The changes of body weight, food intake and water intake, and serological markers such as blood glucose, serum total cholesterol, triglyceride (TG), blood urea nitrogen (BUN), and creatinine (Cr) were measured. Results : The decrease of body weight and the increase of food and water intake in STZ-induced diabetic rats was improved by the administration of CF and LF. Also, the enhancement of blood glucose and serum total cholesterol, TG, BUN and Cr in STZ-induced diabetic rats was significantly inhibited by the administration of CF, BB, LF and glibenclimide. On the other hand, EH strongly inhibited the increase of BUN and Cr in the sera of STZ-induced diabetic rats. Conclusions : These results suggest that among six herbal medicines used lower emaciation of emaciation-thirst disease, CF, BB, LF and EH show a characteristics including the channel-tropism theory.

Analysis of trends in patents on insect-derived medicinal materials for skin diseases (곤충 유래 한약재를 활용한 피부질환 개선 기능성 소재에 대한 특허 동향 분석)

  • Objective: In this study, we analyzed the trend of patents registered prior to October 4, 2019, to understand the industrial trends in insect-derived medicinal materials used for the treatment of skin diseases. Methods: Using the WIPSON database, we collected information regarding the patents related to insect-derived materials for treating various skin diseases. Results: The patents registered prior to October 4, 2019, from Korea, Japan, the USA, China, and the EU, along with those registered under PCT were selected. There were 195 patents related to the use of insect-derived medicinal materials in treating various skin diseases such as psoriasis, inflammatory skin diseases, eczema, pruritus, and atopic dermatitis. China is mostly superior in total number of registered patents compared with the other countries. Korea was the major patent technology-holder for atopic dermatitis, but China dominated in the remaining categories of skin diseases. Upon first patent registration in 1992, there had been a continual increase in the number of patents. Especially, patents related to eczema, psoriasis, inflammatory skin disease were markedly increased. Most frequently used insect-derived medicinal materials was Scolopendra, Cicadidae Periostracum, Scorpions, Cantharides, and Batryticatus Bombyx. The insect-derived medicinal materials were generally used as a combined preparation with other medicinal materials in patents. Conclusion: This study could help to establish the basis for future research and development related treating skin diseases using insect-derived medicinal materials. In order to provide sufficient data, further study including analysis of rejected patents is needed.

Studies on the Residual Contents of Sulfur Dioxide in Herbal Medicines Distributed at Domestic (국내 유통 한약재에서 이산화황 잔류량에 대한 연구)

  • We investigated the residual contents of sulfur dioxide ($SO_2$) on the 280 kinds of herbal medicines distributed at 8 cities including in Seoul, Busan, Daegu, Gwangju, Jecheon, Yeongju, Geumsan and Jeonju in Korea. The residual contents of $SO_2$ were determinated by modified Monier-Williams method. The residual contents of $SO_2$ were not detected at 206 products in total 280 products. However, it was detected below 100 ppm in 39 products, between 101 and 1000 ppm in 30 products and exceeded 1000 ppm in 5 products. $SO_2$ contents ranged 11 ${\sim}$ 2339 mg/kg (mean 293 mg/kg) at domestic samples distributed. Regardless of region, $SO_2$ contents were not found at Notopterygii Rhizoma, Ligustici Scinensis Rhizoma et Radix, Bombyx Batryticatus, Coicis Semen, Cnidii Rhizoma, Anemarrhenae Rhizoma, Gardeniae Fructus, Alismatis Rhizoma. But it's found at Batatatis Rhizoma, Paeoniae Radix and Codonopsitis Radix of every region collected the samples. $SO_2$ contents were not detected at 58 products which collected cultural fields of dometic. After treated with water wash and hot water extraction, the reduction rates of sulfur dioxide were appeared with 14.3% ${\sim}$ 40.4% and 55.2% ${\sim}$ 100.0%, respectively. These data will be used to establish a criteron of residual sulfur dioxide in herbal medicines.

  • Humans have been breeding the mulberry silkworm for the long period of time to obtain silk fabric and nutrient-rich pupae. Currently, silkworm larvae, pupae, and silk-Fibroin hydrolysates are registered as food raw materials, while silkworm feces and Bombyx batryticatus are registered as Korean traditional medicines. Among sericulture products, individually recognized health functional food ingredients include silk-protein acid-hydrolysates for immunity enhancement, Fibroin-hydrolysates for memory improvement, and freeze-dried 5th instar and 3rd-day-silkworm powder for lowering-blood sugar. Recently, HongJam produced by steaming and freeze-drying mature silkworms were reported to have various health-promoting effects such as preventing the onset of Alzheimer's disease and Parkinson's disease, enhancing gastro-intestinal functions, improving skin-whitening and hair growth, and extending healthspan. By consuming silkworm products with various health-promoting effects, it is possible to increase the healthspan of human beings, thereby reducing personal and national medical expenses, resulting in increasing the individual's happiness.
