Behaviors of Arsenic in Paddy Soils and Effects of Absorbed Arsenic on Physiological and Ecological Characteristic of Rice Plant;IV.   • A water culture experiment was conducted to investigate the effect of As content in a culture solution on the water status and growth of rice plants. Rice (Oryza sativa L. Line. Iri 316) seeds were germinated in bentonite and cultivated there for 30 days. Rice seedlings were transplanted into 3.5l pots containing the culture solution on May 1, 1985 and allowed to grow without As treatment for one month. Afterwards, they were grown in a culture solution maintaining the final concentration of As, 0, 1, 5, 10 and 15mg/1 renewing in the solution dissolved sodium arsenate at intervals of 3 to 7 days. Plants were cultivated in the green house during the growing period and harvested 60 days after As treatment. The results obtained were as follows: Transpiration of rice plants was decreased with the increase of the As level in the culture solution. Stomatal diffusive resistance and leaf temperature increased with increase of As levels though the humidity and the air flow rate in leaf decreased. Air flow rate, transpiration and stomatal diffusive resistance showed a highly significant correlation with As contents in shoots and roots of rice plants: Espally The air flow rate and transpiration revealed a significantly higher correlation with As contents in the root than that in the shoot, but diffusive resistance showed adverse tendency. High levels of As in the culture solution depressed plant height, no. of tillers, leaf width and dry weight of plant remarkably. Typical symptoms of As toxicity were root discoloration, and necrosis of leaf tips and margins, and leaf rolling during the sunny daytime were also another symptoms.

A Study on Vascular Plants, Distribution Status and Management Plans of the Cactus Habitat (No. 429 Natural Monument) in Wolryung-ri, Jeju Island (제주 월령리 선인장군락지(천연기념물 제429호)의 관속식물상, 분포실태, 관리방안에 관한 연구)

  • The cactus habitat in Jeju Island has a phytogeographically specific distribution in the East Asian region, and forms a unique landscape as the only native cactus (Opuntia ficus-indica (L.) Mill.) habitat in Korea. However, there has been no detailed investigation on the distribution of cacti in the habitat and no investigation on the diversity of the mixed composition of plants in the habitats and their correlation with the distribution of cactus populations. This study attempted to investigate the diversity of vascular plants in the Wollyeong-ri cactus habitat and record the actual distribution and trends of cactus distribution. In addition to the distribution characteristics of cacti, we also discuss the characteristics of species reflecting the characteristics of the habitat among the mixed population of plant species, as well as the biological and environmental factors that threaten the maintenance of cactus populations or require management for preservation of cacti. Considering the phenological character, we conducted the field surveys for flora identification six times between June 2015 and September 2017. The Engler classification system was used for the arrangement sequence and names of plants, and the Korean Plant Names Index was adopted for the Korean name of the species. The study results showed that the Wolryung-ri cactus habitat in Jeju Island has the characteristic physiognomy of an area dominated by cactus. For the vascular plants, a total of 125 taxa were identified, including 53 families, 104 genera, 109 species, 15 varieties and 1 forma. Endangered plants specified by the Ministry of Environment were not found. Two species, Cyrtomium falcatum and Asplenium incisum, were identified as the ferns, and no gymnosperms were found. In addition, 123 taxa of angiosperms, 91 taxa of dicotyledones and 32 taxa of monocotyledons were identified. The distributions of cacti were confirmed in 289 meshes corresponding to 59.3% of the total 487 meshes in the cactus protected area, which showed various coverage distributions ranging from 5% to 95%. Most of the meshes where no cacti were found are coastal areas with exposed basalt rocks where the soil depth has not developed or extremely restricted due to repeated waves, or areas where artificial facilities, grasslands, and observation paths have been constructed. On the other hand, there were 71 lattice points in 14.5% of the total area where the cactus showed 70% or higher dominance. Cacti are randomly distributed in these areas. They have adapted to the microhabitat environment and are found to be opportunistically distributed along the growable locations. Considering that the reproduction of cacti in the habitat is mostly dependent on parthenogenesis, the present distribution seems to reflect the potentially distributable regions of cacti in the habitat. Based on the results of field surveys, a management plan for conservation and protection of the protected areas has been proposed.

The Localization of the Excretory, Purified and Infected Antigenic Protein in the Tissue of Trichinella spiralis Larval Worm (선모충(Trichinella spiralis) 유충의 조직 내 배설, 분리 및 감염항원 단백의 분포)

  • In order to observe the localization of excretory, purified and infected antigenic protein in the tissue of Trichinella spiralis larvae, immunogoldlabeling methodology using IgG and protein A-gold complex was implemented. T. spiralis larvae obtained from rat muscle were initially cultured in medium, and secreted excretory antigen was collected for 1 or 3 days. Purified antigenic protein was obtained from homogenized T. spiralis larvae. Rabbits were then immunized with 1 or 3 days secreted excretory protein and purified 45 kDa protein, and IgG was purified from collected serum. Serum, against infected antigen, collected from rat on 1 and 4 weeks after infection with T. spiralis larvae, and IgG was purified from collected serum. T. spiralis larvae were embedded in Lowicryl HM20 medium. Then they were finally treated with immunized IgG and protein A-gold complex (particle size; 15 nm) and observed under electron microscope. In T. spiralis larvae tissue, the tissue antigen reacted with rabbit IgC antigen Day 1 secreted excretory protein, infected antigenic protein and purified 45 kDa protein. But different distribution pattern of labeled gold particles were observed. When Day 1 secreted excretoy protein was used, gold particle labeling was observed specifically on the cuticle, basal layer, esophagus interstitial matrix (EIM) and ${\alpha}_0,\;{\alpha}_1$ granules of stichocyte of the worm. In a separate group of tissue, the antigen reacted with rabbit IgG against Day 3 secreted excretory protein. Labeled gold particles were specifically distributed on the surface layer of cuticle, EIM and ${\alpha}_0$ granules of stichocyte of the worm. In case of using infected antigenic protein, gold particle labeling was specifically distributed on the cuticle and EIM of the worm. When purifed 45 kDa protein was used gold particle labeling was specifically distributed on the cuticle, basal layer, EIM and ${\alpha}_0,\;{\alpha}_1$ granules of stichocyte of the worm. Therefore, excretory antigens appeared to originate from the cuticle and ${\alpha}_0,\;{\alpha}_1$ granules of stichocyte for the first day but the cuticle layer associated with globular proteins and ${\alpha}_0$ granules of stichocyte after 3 days and infected antigens appeared to originate from the cuticle for 1 and 4 weeks after infection. These results suggest that excretory and infection specific antigens are secreted into the cuticle, basal layer, EIM and ${\alpha}_0,\;{\alpha}_1$ granules of stichocyte and 45 kDa protein may be contained these specific antigens.

Dietary Fiber and β-Glucan Contents of Sparassis crispa Fruit Fermented with Lactobacillus brevis and Monascus pilosus (유산균 및 홍국균 발효 꽃송이버섯 추출물과 잔사의 식이섬유와 베타 글루칸의 함량)

  • Sparassis (S.) crispa is an edible mushroom abundant in dietary fiber and ${\beta}$-glucan. The aim of this study was to prepare extracts and residues of the fruit bodies of S. crispa fermented with Lactobacillus (L.) brevis and Monascus (M.) pilosus and to measure the remaining dietary fiber and ${\beta}$-glucan. Dried powder of S. crispa containing 64.4 g/100 g total dietary fiber (2.6 g/100 g soluble and 61.8 g/100 g insoluble dietary fibers) and 24.0 g/100 g ${\beta}$-glucan was used as the starting material for the extraction. Raw and fermented S. crispa were extracted with hot water and three kinds of aqueous ethanol (50, 70, and 90%, v/v), respectively. A hot water extract from S. crispa fermented with M. pilosus had greater soluble dietary fiber content (19.3 g/100 g) than that from raw S. crispa with 14.6 g/100 g soluble dietary fiber or that from L. brevis-fermented S. crispa with 8.2 g/100 g soluble dietary fiber. The yield of the extract was 16.6% of intial weight of dried S. crispa. After hot water extraction of S. crispa fermented with M. pilosus, residues containing 90.5 g/100 g total dietary fiber (1.3 g/100 g soluble and 89.2 g/100 g insoluble dietary fibers) were obtained, and the yield was 69.6% of intial weight of dried S. crispa. The residue (31.0 g/100 g) contained more ${\beta}$-glucan than raw S. crispa or M. pilosus-fermented S. crispa (24.4 g/100 g). The resulting hot water extract and residue from S. crispa fermented with M. pilosus would be suitable for use in preparing liquid and powdered health functional foods, respectively.

Infrared Thermography in the Assessment of Temporomandibular Joint Dysorder (측두하악장애에서의 적외선 체열 촬영 검사의 유용성)

  • Background: Temporomandibular joint disorder (TMD) is a group of musculoskeletal conditions characterized by pain in the pre-auricular area, limitation of jaw movement and palpable muscle tenderness. Thermography is a nonionizing, noninvasive diagnostic alternative for the evaluation of TMD. This study was conducted to evaluate the usefulness of thermography in the assessment of TMD. Methods: Thermography was conducted on the 61 patients who had been diagnosed with TMD, and on the 34 normal symptom-free volunteers. The temperature differences between opposite sides of the temporomandibular joint (${\Delta}T_{TMJ}$) and the masseter muscle (${\Delta}T_{MST}$) were calculated. The sensitivity and specificity of thermography was calculated at the cut off values of 0.2, 0.3, and $0.4^{\circ}C$. Results: In the patient group, the ${\Delta}T_{TMJ}$ was $0.42{\pm}0.38^{\circ}C$ and the ${\Delta}T_{MST}$ was $0.38{\pm}0.33^{\circ}C$, whereas in the control group the ${\Delta}T_{TMJ}$ was $0.10{\pm}0.07^{\circ}C$ and the ${\Delta}T_{MST}\;0.15{\pm}0.10^{\circ}C$. In addition, the patient group demonstrated a significantly lower level of thermal symmetry than the control group (P < 0.001) in both the temporomandibular joints and the masseter muscles. The sensitivity of thermography at the cut off values of 0.2, 0.3 and $0.4^{\circ}C$ was 67.2, 49.2, and 42.6% in the temporomandibular joint (TMJ) and 60.7, 49.2 and 37.7% in the masseter muscle, respectively. The specificity of thermography at the cut off values of 0.2, 0.3 and $0.4^{\circ}C$ was 88.2, 100, and 100% in the TMJ and 61.8, 91.2 and 100% in the masseter muscles, respectively. The accuracy of thermography at the cut off values of 0.2, 0.3 and $0.4^{\circ}C$ was 74.7, 67.4, and 63.2% in TMJ and 61.1, 64.2 and 60.0% in the masseter muscles, respectively. Conclusions: Temperature differences exist between the opposite sides of the TMD and masseter muscles in patients with TMD. Although the sensitivity of thermography in the diagnosis of TMD is low, it has high specificity in the evaluation of TMD, and is therefore applicable to patients with TMD.

A Research on Service and Awareness of Dental Coordinators by Manpower at Dental Care Service Institutions - Centering on Manpower Other than Dentists (치과코디네이터 업무 및 인식에 관한 조사연구 - 치과의사를 제외한 기타 인력을 중심으로)

  • To analyze dental hygienists and other manpower at dental care service institutions where a dental coordinator was working among about 200 dental care service institutions in Seoul, Gyeonggi Province, and Incheon as of June 2005 for contents of training for dental coordinators, opinions of qualification of dental coordinators, present and future services provided by dental coordinators, and awareness of dental coordinators and to provide basic data about future services, roles, and cultivation of dental coordinators, a survey was conducted and 216 copies returned were analyzed, obtaining the following results. 1. 83.8 percent needed an educational program for dental coordinators as an educational content; 41.7% had awareness of the educational content; and 83.8 percent insisted that over the intermediate level of curricula should be taken. Dental coordinator cultivation institutions identified included the institution under the control of the Korean Dental Hygienists Association and the education center for the department of dental hygiene; 76.9% insisted that an appropriate qualifying examination should be necessary. They suggested the central government department and the local government as a certification institution; 39.4% insisted that financial support for the education should be provided by financing education alone. Only 28.7% experienced dental coordinator education and 73.1% hoped to serve as a dental coordinator. They were found to expect a rise in payment(64.4%) and in the title(46.8%) after completion of the educational program. 2. 66.2% saw a dental hygienist as the most appropriate for a dental coordinator; clinical career (39.4%) and practical capacity(29.2%) were suggested as requirements for a dental coordinator; and a period of over three years(47.2%) was suggested for appropriate dental career. 3. Dental coordinators' present services included 'reservation management' for customer management, 'staff service training' for organization management, 'understanding of customer reception attitudes and actions' for self-management, 'hospital information management' for hospital marketing, 'acceptance' for hospital affairs management, and 'hospital environment management' for hospital facilities management; their future services included 'acquisition of ability to use a foreign language' for self-management, followed by 'staff service training' for organization management, 'training and counseling' for customer management, 'acquisition of counseling capacity' for self-management, 'complaining customer reception' for customer management, and 'marketing strategy implementation' for hospital marketing. 4. After comparing dental hygienists and other manpower in terms of dental coordinators' future services, dental hygienists showed interest in 'acquisition of ability to use a foreign language,' 'staff service training,' 'complaining customer reception,' and 'acquisition of counseling capacity' while other manpower showed interest in 'acquisition of ability to use a foreign language,' 'document data management,' 'acquisition of basic service manner,' 'acquisition of counseling capacity,' 'manpower management,' 'establishment and evaluation of a marketing strategy,' and 'education and counseling.' 5. As for awareness of dental coordinators, they were thought of as helpful in improving image of a dental clinic; it was found that continuous training should be necessary to develop dental coordinators' capacity; dental coordinators' services should be important and contribute to patients' qualitative satisfaction.

Assessment of the Heavy Metal Contamination in Paddy Soils Below Part of the Closed Metalliferous Mine (폐금속광산 하류 논토양의 중금속 오염도 평가)

  • BACKGROUND: Most of the tailings have been left without any management in abandoned metalliferous mines and have become the main source of heavy metal contamination for agricultural soils and crops in the these areas. METHODS AND RESULTS: This experiment was carried out to investigate the assessment of the heavy metal contamination in paddy soils located on downstream of the closed metalliferous mine. The average total concentrations of cadmium (Cd), copper (Cu), lead (Pb), zinc (Zn), and arsenic (As) in paddy soils were 8.88, 56.7, 809, 754, and 37.9 mg/kg, respectively. Specially, the average concentrations of Cd, Pb and Zn were higher than those of warning criteria for soil contamination(4 mg/kg for Cd, 200 mg/kg for Pb, and 300 mg/kg for Zn) in agricultural soil established by Soil Environmental Conservation Act in Korea. The proportions of 0.1 M HCl extractable Cd, Cu, Pb, Zn, and As concentration to total concentration of these heavy metals in paddy soils were 27.7, 21.3, 35.1, 13.8 and 10.5%, respectively. The pollution index of these five metals in paddy soils ranged from 0.42 to 11.92. Also, the enrichment factor (EFc) of heavy metals in paddy soils were in the order as Cd>Pb>Zn>Cu>As, and the enrichment factor in paddy soil varied considerably among the sampling sites. The geoaccumulation index (Igeo) of heavy metals in soils were in the order as Cd>Pb>Zn>Cu>As, specially, the average geoaccumulation index of Cd, Pb, and Zn (Igeo 2.49~3.10) were relatively higher than that of other metals in paddy soils. CONCLUSION: Based on the pollution index, enrichment factor, and geoaccumulation index for heavy metal in paddy soils located on downstream of closed metalliferous mine, the main contaminants are mine waste materials and mine drainage including mine activity.

Assessment of Human Bioavailability Quotient for the Heavy Metal in Paddy Soils Below Part of the Closed Metalliferous Mine (폐금속광산 하류 논토양의 중금속에 대한 인체흡수도 평가)

  • BACKGROUND: For the heavy metal cotamination sites, it is very important to estimate the human bioavailability quotients for heavy metals in paddy soils released from mine tailings, which is a major source of contamination in Korea, and to assess the human health risks of heavy metals. METHODS AND RESULTS: This experiment was carried out to investigate the human bioavailability quotient of the heavy metals in paddy soils below part of the closed metalliferous mine. For estimating the human bioavailability quotients for heavy metals, 30 paddy soils below part of the closed mine were collected, and analyzed for Cd, Cu, Pb, Zn, and As using simple bioavailability extraction test(SBET). The quantities of Cd, Cu, Pb, Zn and As extracted from paddy soils below part of the mine by using the SBET analysis were 28.1, 17.3, 34.1, 14.6 and 2.3% respectively. Specially, the maximum values of Cd, Pb and Zn were 73.3, 81.5 and 58.1% of human bioavailability quotient, respectively, and varied considerably among the sampling sites. The human bioavailability quotient of Cd, Cu, Pb and Zn in soils near the closed mine showed significant positive correlation among soil pH value, O.M. and Ex. Ca. contents, while it correlated negatively between soil Ex. K and Ex. Mg contents in paddy soils. Also, its of Cd, Cu, Pb and Zn in paddy soils showed significant positive correlation with 0.1M HCl extractable and total contents, while in soils, it correlated negatively with As content in soil near the closed mine. CONCLUSION: The results of the simple bioavailability extraction test (SBET) indicate that regular ingestion of soils by the local population could be closed a potential health threat due to long-term heavy metals exposure in these mine areas.

Effects of Molasses Addition on the Feeding Value of Socheongryongtang(Herbal Medicine) Meal Silage (당밀 첨가가 소청룡탕박 사일리지의 품질에 미치는 영향)

  • Two experiments were conducted to investigate effects of molasses addition to silage materials in ensiling Socheongryongtang meal on the nutritive quality of silage, palatability and various ruminal parameters in Korean native goats. In Experiment 1, Socheongryongtang meal silage was produced by the addition of 0, 0.5, or 1.0% molasses and stored for 40 days at room temperature. There were three replicates per treatment. Lactic acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were significantly(p<0.05) higher than that of the control(0%). However, the pH and butyric acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were lower than those of the control(0%). In addition, molasses(0.5 and 1.0%) increased the number of lactobacillus, but decreased the number of fungi in Socheongryongtang meal silage. In vitro dry matter disappearance tended to increase by molasses addition. In Experiment 2, three 1.5-yr-old Korean native female goats were employed in cross-over design to measure the palatability and various ruminal parameters of Socheongryongtang meal silage. Feed intake of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) for 30 min significantly higher than that of control(0%). Silmilar trend was found when the feed intake was measured for 6 hr. In various ruminal parameters, molasses addition to silage materials increased propionic acid(P) and total volatile fatty acid contents, but decreased pH, acetic acid (A), and butyric acid. The A/P ratio was the lowest in 1.0% molasses treatment group. It is concluded that molasses addition in ensiling Socheongryongtang meal could enhance its nutritional quality and lengthen storage period. Also the palatability of silage was improved by the addition of molasses.

Analysis of Environmental Design Data for Growing Pleurotus ervngii (큰 느타리버섯 재배사의 환경설계용 자료 분석)

  • This study was carried out to file up using effect and requirement of energy for environmental design data of Pleurotus eryngii growing houses. Heating and cooling Degree-Hour (D-H) were calculated and compared for. some Pleurotus eryngii growing houses of sandwich-panel (permanent) o. arch-roofed(simple) type structures modified and suggested through field survey and analysis. Also thermal resistance (R-value) was calculated for the heat insulating and covering materials of the permanent and simple-type, which were made of polyurethane or polystyrene panel and $7\~8$ layers heat conservation cover wall. The variations of heating and cooling D-H simulated for Jinju area was nearly linearly proportional to the setting inside temperatures. The variations of cooling D-H was much more sensitive than those of heating D-H. Therefore, it was expected that the variations of required energy in accordance with setting temperature or actual temperature maintained inside of the cultivation house could be estimated and also the estimated results of heating and cooling D-H could be effectively used far the verification of environmental simulation as well as for the calculation of required energy amounts. When the cultivation floor areas are all equal, panel type houses to be constructed by various combinations of materials were found to by far more effective than simple type pipe house in the aspect of energy conservation maintenance except some additional cost invested initially. And also the energy effectiveness of multi-span house compared to single span together with the prediction of energy requirement depending on the level insulated for the wall and roof area could be estimated. Additionally, structural as well as environmental optimizations are expected to be possible by calculating periodical and/or seasonal energy requirements for those various combinations of insulation level and different climate conditions, etc.
