이묘산(二妙散)과 구성약재(構成藥材)의 효능(效能) 비교(급성(急性) 췌장염(膵臟炎)을 중심으로) (The Comparison Between Single And Yimyosan in Acute Pancreatitis : Atractylodes chinensis(AC) and Phellodendron amurense Ruprecht(PAR))

  • Objectives : Atractylodes chinensis(AC) and Phellodendron amurense Ruprecht(PAR) have been used as anti-inflammatory medicines. However, the effects of AC, PAR and Yimyosan on AP were not examined. To prove this, We decocted the dried prescription of AC and PAR with boiling distilled water and freeze-dried to be powdered. AC, PAR and Yimyosan was administrated intraperitoneally. Methods : 1h after administration, cerulein was injected hourly six times. 6hrs after last cerulein injection, mice were sacrificed, then the pancreas and blood were harvested. Serum amylase and lipase, neutrophil infiltration, pancreatic cytokines were used as the parameter of severity of AP. Results : As a result of assessment the parameters of AP, AC alone treatment did not inhibit the severity of AP, however PAR treatment inhibited the severity of AP significantly. Yimyosan also showed the protective effects against AP at lower doses, however AC alone plus PAR alone extract did not show the protective effects significantly. Conclusions : In conclusion, PAR extract has a protective effects on AP, and the effects could be increased by co-treatment with AC.

친화막의 L-tryptophan 흡착특성 (Adsorption Characteristic of L-tryptophan of Affinity Membrane)

  • 전기방사를 이용하여 polyethersulfone (PES)-bovin albumin serum (BSA) 단백질 친화막을 제조하였으며, 이때 전기방사에 의한 공정상의 문제점을 용해도가 높고, 끓는점이 높은 2,2,3,4,4,4-Hexafluoro-1-butanol (HFB)를 공용매로 사용함으로써 해결하였다. 또한 공용매는 최적의 온도, 습도 범위를 확대시켜줌으로써 친화막의 대량 생산이 용이하리라 생각한다. 제조된 친화막은 biuret test를 통하여 친화막의 색깔이 무색에서 보라색으로 변함으로써 PES 섬유 내 BSA가 존재함을 확인할 수 있었다. 완충용액의 조성 성분 중 dimethyl sulfoxide (DMSO)는 세척과정에서 BSA와 L-tryptophan사이의 해리를 제어함으로써 용출과정에서 비교실험보다 약 5배의 높은 용출량을 보여주었다.

Lymphoscintigraphy에 이용되는 방사성의약품의 사용제한으로 인한 대체 방사성의약품의 성능평가 (Performance Evaluation of Substitution Radiopharmaceutical according to Restriction of the Radiocolloids in Lymphoscintigraphy)

  • Purpose: Several radioactive agents were used for lymphoscintigraphy in breast cancer. But recently, due to the change of local radiopharmaceutical licensing policy and discontinuation of supplies from manufacturer, we could not use many radiocolloids such as $^{99m}Tc$-Antimony Trisulfide Colloid, $^{99m}Tc$-Tin Colloid, $^{99m}Tc$-Human Serum Albumin. So it is necessary to use the substitution radiopharmaceutical. Therefore, this study aims to evaluate the performance of substitution radiopharmaceutical ($^{99m}Tc$-Phytate) including the existing radiocolloids and to set up of the appropriate protocol in lymphoscintigraphy. Materials and Methods: For each radiocolloids ($^{99m}Tc$-Antimony Trisulfide Colloid (ASC), $^{99m}Tc$-Tin Colloid (TC), $^{99m}Tc$-Human Serum Albumin (HSA), $^{99m}Tc$-Phytate) were performed the particle size by Zeta Sizer (Nano-ZS) and we compared the radiolabeling time, procedure and acquisition time for each of the radiocolloids (total 200 patients). For the last time, we made an analysis of image for each of the radiocolloids with our previous report (SJ Jang et al, Korean Nucl Med Mol imaging Vol.41, No.6, Dec 2007). Results: The particle size of each radiocolloids showed A.S.C (50 nm), T.C (310 nm), H.S.A (10.8 nm), $^{99m}Tc$-Phytate (499 nm). The labeling and acquisition time for each of the radiocolloids showed no substantial difference. But there is difference of the labeling time for the A.S.C. because the ASC procedure need to boiling process. There were no significant differences among those radiocolloids (p>0.005) in the identification rate (IR), false negative rate (FNR), and negative predictive value (NPV). Conclusions: $^{99m}Tc$-labeled radiocolloids showed equivalent results in lymphoscintigraphy. Therefore, in this exam of each radiocolloids could be applied appropriate protocol in lymphoscintigraphy.

Conditioned Media of ASC-17D Sertoli Cells Induce G1-Growth Arrest of DU145 Human Prostate Cancer Cells

  • We studied the effects of ASC-17D rat Sertoli cell-conditioned media (rSCCM) on the proliferation of the DU145 prostate cancer cells. rSCCM was prepared from ASC-17D cells cultured in DMEM/F-12 serum-free media at a nonpermissive temperature of $40^{\circ}C$, which is the condition for the high expression of c1usterin. We found that rSCCM could inhibit the proliferation of DU145 cells by arresting the cell cycle in the G1 phase in a dose-dependent manner. This growth arresting activity was abolished by boiling rSCCM for 5 min. The G1 growth-inhibiting activity of rSCCM was also detected in other prostate-originated cancer cells examined (i.e., LNCaP and PC-3) but not in other cells (ASC-17D, HepG2, SK-N-SH, and NIH3T3). Western blot analysis of partially purified growth inhibiting fractions with the clusterin antibody showed that the cytostatic factor in rSCCM was not c1usterin. This cytostatic factor was semi purified by DEAE-Sepharose, ammonium sulfate precipitation, and Phenyl-Sepharose column chromatography, and was estimated to have a molecular weight of 88 kDa by Sephacryl S-300 gel filtration.

인삼주정추출액이 스트레스에 폭로된 동물의 생체반응에 미치는 영향 (Influence of Panax Ginseng on the Responce of Btressful Stimuli in the Experimental Animal exposed to Various Stress)

  • Three hundred gram of Korean ginseng root was extracted with 95% ethanol on a boiling water bath for about 300 hr. Evaporation of alcohol yieled 50.2g of dark brown residue which was used by dissolving 4 mg of the residue in 1 ml of physiological saline. The ginseng group and the saline group received each day 0.5 ml per 100 g body weight of ginseng extract and physiological saline, respectively. Both the ginseng and saline group with stress were exposed to positive radial acceleration (1∼29g), cold (5$^{\circ}C$, 0$^{\circ}C$ &-10$^{\circ}C$) and heat (35$^{\circ}C$) environment, and surgical stress. After termination of the last stress, the tolerance, body weight, visceral organ weight, basal metabolism rate, rectal temperature, the number of erythrocyte and leucocyte, hemoglobin level, hematocrit ratio, total serum protein content and it's fraction and the content of adrenal ascorbic acid in the experimental animal exposed to stress were measured and at the corresponding periods, the same measurements were also carried out with the ginseng and the saline groups without stress exposure (serving as control). Results obtained were as follows. 1. Administration of ginseng does depressed the decrease of the tolerance, body weight, visceral organ weight, basal metabolism rate, the number of erythrocyte, hemoglobin value, hematocrit ratio and the A/G ratio in the mice and rats exposed to various stress. 2. The change of the rectal temperature, eosinophile counts, total serum protein content and the content of adrenal ascorbic acid of ginseng group that exposured to various stress facilitates the reaction to, and accelerates the recovery from the stress. 3. Even after hypophysectomy which served the link between the central and the peripheral portion of the stress mechanism, the adrenal ascorbic acid content of ginseng group decreased significantly more than that of the saline group 30 min. after administration of ACTH, while the value approached the normal level significantly closer in the ginseng group than in the saline group 1 and 2 hr after ACTH administration. Judging from the above results, it is concluded that administration of ginseng extract tolerated the experimental animals under the environment of stressfu1 stmuli, although the ginseng has no significant influence upon the stress mechanism in the absence of stressful stimuli. The site of action of the ginseng appears to be in the peripheral portion of the stress mechanism.

여주 추출물이 급성 납 중독된 흰쥐의 간과 콩팥에 미치는 효과 (Effects of Momordica Charantia L. Water Extracts on the Rat Liver and Kidney with Acute Toxicated by Lead)

  • 본 연구는 급성 납 독성에 대한 여주 추출물의 효과를 알아보기 위하여 시도되었다. 250 g 내외의 흰쥐를 대상으로 대조군, 납(1,000 ppm) 투여군, 납 투여 후 필리핀산과 일본산 여주 추출물을 5%와 10%로 희석하여 식수로 공급한 후 6주 후에 간과 콩팥 손상 억제 효과를 알아보기 위하여 생화학적 및 조직학적 실험을 실시하였다. 혈액중 AST, ALT와 BUN의 수치는 여주 추출물 투여군이 납 단독투여군보다 감소되었다. 간 조직의 광학현미경적 관찰에서 납 단독투여군은 간세포의 괴사와 염증세포의 침윤이 관찰되었으나 여주 추출물 투여군에서는 중심정맥 주위에 약간의 단핵세포의 조직 침윤만이 관찰되었다. 콩팥 조직의 광학현미경적 관찰에서 납 단독투여군은 콩팥토리 모세혈관의 위축과 괴사 현상이 관찰되었고 콩팥세관 주위 간질에 염증세포의 침윤이 관찰되었으나 여주 추출물 투여군에서는 일부 콩팥세관 주위에서 염증 세포의 침윤만이 관찰되었다. 이상의 연구 결과로 보아 여주가 납으로 유발된 간과 콩팥 손상을 보호하는 효과가 있는 것으로 사료되며 여주에 대한 다양한 연구가 필요할 것으로 사료된다.

Monitoring the Reproductive Status of Dairy Cows by Urinary Pregnanediol Glucuronide

  • This study was undertaken with the aim to establish a reliable radioimmunoassay (RIA) system for urinary pregnanediol glucuronide (PdG) and to employ it for monitoring the reproductive status of dairy cows. Urine and blood samples were collected from the Holstein cows both pregnant and non-pregnant. The samples were then investigated for evaluating the relationship between progesterone ($P_{4}$) in blood and PdG in urine adjusted with or without urinary creatinine basis. Biweekly urine collection was employed for three cows in estrous and those artificially inseminated, while urine from pregnant cows was collected on a monthly basis. P_{4}$ and PdG levels were measured by enzymeimmunoassay (EIA) and RIA techniques, respectively. Our results indicated the sensitivity of PdG for RIA being 35 pg/tube and the recovery rate of 100%. Urinary creatinine concentrations also fluctuated within a day, but change at midday was not noteworthy. Regardless of the time of urination the change in concentrations of PdG was relatively smaller and did not vary significantly. The urinary PdG concentration showed periodic changes as that with serum P_{4}$ levels during the cow's estrus cycle. The correlation coefficient rose when creatinine level in urine was adjusted but the change was also not significant. The concentrations of PdG during the luteal phase were detected between 8.2 and 17.4 ng/ml, three to five times higher than that in the follicular phase. The concentration of PdG from pregnant cows (21 days after conception) was three to four times higher than in the nonpregnant cows. Our finding suggests that the determination of urinary PdG could be reliably employed for early pregnancy detection. The urinary PdG level continued to raise until 30 days pre-partum while the concentration reached its peak at 30 ng/ml, after which it started to fall 18 to 30 days before parturition and finally fell to its nadir value one week after parturition. As the correlation coefficient between the urinary PdG and serum P_{4}$ was higher than that corrected by urinary creatinine it can be suggested that the adjustment is not needed. The concentrations of urinary PdG could be maintained stably for 2 days in urine samples stored at room temperature and extended to 8 days when the samples were pretreated by boiling for 30 minutes. In conclusion urinary PdG concentration even without the need for creatinine basis adjustment can be used directly for monitoring the reproductive status of dairy cows.

Differential Toxicity of the Water and Ethanol Extracts of Chung-Sang, an Experimental Herbal Formula

  • Chung-Sang (CS) is an experimental herbal remedy that is formulated to treat respiratory diseases implicated by inflammation. The herbs comprised of CS are frequently prescribed for treating various inflammatory symptoms: Menthae haplocalycis Herba, Magnoliae Flosis, Xanthii Fructus, Herba Asari, and Caryphylli Flos. Here, we prepared the extract of CS with boiling water (wCS) or with 50 % ethanol (eCS) and examined whether the two different extracts of CS exhibit a toxicity to cultured cells and mice. RAW 264.7 cells were treated with wCS or eCS, and the cytotoxicity of these extracts to RAW 264.7 cells was determined by an MTT assay. Although the production of intracellular reactive oxygen species that are detrimental to the cells was not increased by the extracts, the cytotoxicity to the cells was evident from 10 mg/ml of wCS and 100 mg/ml of eCS, suggesting that eCS is less cytotoxic. When mice (n = 10/group) received a single intratracheal wCS or eCS daily for 14 days, wCS yielded 40 % mortality, whereas eCS showed none. Both wCS and eCS did not significantly affect the weight of the body and of vital organs, except the lung. Biochemical analyses of mice blood indicated no damage to liver or kidney. However, unlike eCS, wCS significantly increased the level of IgE in serum. Collectively, our results show that eCS was less toxic than wCS, suggesting that CS prepared with 50 % ethanol is preferential over the conventional way of preparing CS.

시너(thinner)의 물리화학적 특성과 랫트의 생식기에 미치는 영향 연구 (A Study on thinner's Physicochemical property and its effect on genital organ of rat)

  • The aim of this study is to confirm the physicochemical property and hazard of thinner (012), which is a diluent of enamel paint used for floor coating for waterproofing and oil painting for the outer wall. The literatures of physicochemical property and hazard of thinner were surveyed and its physicochemical property were evaluated. And then, the inhalation toxicity of thinner affecting the central nervous system and reproductive organs in rats were examined by subchronic (6 h./day. 5 days/ week for 13 weeks) inhalation test. 1) According to the 13-week subchronic inhalation test, there were no significant changes in clinical test and body weight. However, a significant evidence of toxicity was observed in the hematological test and organ weight such as heart, kidney, liver and brain (p<0.01) in the 200 ppm and 1,000 ppm exposure groups in a dose response manner. In the histopathology analysis, there were no significant evidence of toxicity. Therefore, thinner was not classified as an organ targeted toxic agent. In case of Harmfulness, it could be classified as a chronic toxic agent 3($500 ppm/4hr, rat). 2) The reproductive toxicity such as extension of the period of estrous cycle, reduction of serum estradiol concentration and increase of frequency of the abnormal sperm was observed in the 1,000 ppm exposed animals. 3) The result of the physicochemical property of the test material showed that the specific gravity was 0.793, boiling point $155.8^{\circ}C$, steam pressure 2.1 kPa, ignition point $34.5^{\circ}C$, and spontaneous ignition point $280^{\circ}C$. The endothermic and exothermic values were 371.4 J/g and 159.1 J/g. respectively. The explosion limit was 214 mg/l. These data showed that thinner could be classified as an explosion agent level 1.2 and ignitive liquid agent 3 ($23-60^{\circ}C$) according to the notification No. 2008-1 of the Labor Ministry, "Classifying Standard of Chemical Materials."

다양(多樣)한 황금약침제제(黃芩藥鍼製劑)의 안전성(安定性) 및 효능(效能)에 관한 연구(硏究) (Studies on effect and Safety of various Aqua-acupunctures from Scutellariae Radix)

  • 황금약침제제(黃芩藥鍼製劑)의 안정성(安定性) 및 효능(效能)에 관한 연구(硏究)하기 위해, 약침제제(藥鍼製劑)들은 황금(黃芩)을 원료(原料)로 하여 전탕(煎蕩) 여과(濾過) 희석(稀釋)의 방법으로 제조(製造)하였다. 황금약침제제(黃芩藥鍼製劑)들은 랫트의 족삼리 근육에 반복투여(反復投與)하여 아급성(亞急性) 독성(毒性) 및 효능(效能)을 조사(調査)하였고, 대조군(對照群) 주사용(注射用) 생리식염수(生理食鹽水)와 연유수(燕溜水)의 매체대조군(媒體對照郡) 침자재군(鍼刺栽郡)을 설정하여 비교(比較)하였다. 1. 랫트에 황금약침제제(黃芩藥鍼製劑)들을 0.2cc 근육투여(筋肉投與)한 결과(結果) 대조군(對照群)과 시험군(試驗群)에서 1마리도 사망하지않았고, 일반증상(一般症狀) 및 부검소견(剖檢所見)에서도 특별한 이상이 발견(發見)되지 않았다. 또한 검액(檢液) 황금약침제제(黃芩藥鍼製劑)들 각각을 랫트에 반복 약침(藥鍼) 시술(施術)하여 체중변화(體重變化) 장기중량(臟器重量) 및 혈액생화학적(血液生化學的) 검사등(檢査等)을 실험(實驗)한 결과(結果), 검액(檢液)에 기인한 독성증상(毒性症狀)은 나타나지 않았다. 따라서 시험군(試驗群)에 사용한 황금약침제제(黃芩藥鍼製劑)은 안전성(安全性)에는 문제가 없는 것으로 사료되었다. 2. 경시적(輕視的) 체중변화(體重變化)의 결과(結果)에서 침자재군(鍼刺栽郡)(acupuncture)은 시술(施術) 3일후(日後)부터 p-value가 <0.05, 7일(日)부터는 p-value가 <0.001로 유의성(有意性)이 있는 체중감소(體重減少)가 나타났고, 9일후(日後)에는 모든 시험군(試驗群)이 체중(體重) 증가(增加)가 억제되었다. 3. 장기중량(臟器重量)에 대한 결과(結果)에서 D-2군(群)과 생리식염수군군(生理食鹽水群)은 간장중량(肝臟重量)이 통계적(統計的) 유의성(有意性)을 보이고 있는데 침자재(鍼刺栽)의 경우 간장(肝臟)과 비장(脾臟)에서 유의(有意)한 결과(結果)를 얻었다. 이러한 결과(結果) 효능면(效能面)에서 약침(藥鍼)의 침(鍼)과 약(藥)이 효과(效果)에서 침(鍼)의 효과(效果)가 우선하고 있다고 추측되어진다. 4. 약침(藥鍼) 시술군(施術群)에 대한 혈액생화학적(血液生化學的) 검사(檢査)는 GLU, T.G., CHOL에 대해 조사하였는데, glucose에 대한 영향(影響)은 황금약침제제(黃芩藥鍼製劑) ${\times}$10군(群)에서 대조군(對照群)에 비해 유의성(有意性) 있는 감소를 확인할 수 있었고, 황금약침제제(黃芩藥鍼製劑) ${\times}$100군(群), Distillation-2군(群), 생리식염수군군(生理食鹽水群)에서는 대조군(對照群)에 비해 유의성(有意性) 있는 증가(增加)를 확인할 수 있었고, 모든 시술군(施術群)에서 CHOL의 유의성(有意性)있는 감소(減少)를 보였고, T.G는 황금약침제제(黃芩藥鍼製劑)와 복합주사제군(複合注射劑群)를 제외한 모든 시술군(施術群)에서 통계학적(統計學的) 유의성(有意性)이 있었는데, 황금약침제제(黃芩藥鍼製劑)를 희석(稀釋)한 황금수액희석제제군(黃芩水液稀釋製劑群)에서 유의성(有意性)있는 감소가 일어나는 것을 확인(確認)할 수 있었다. 농도와는 관계없이 황금약침제제(黃芩藥鍼製劑)들이 함유하고 있는 성분상호간(成分相互間)의 작용에 의해서 T.G.가 감소(減少)된다고 생각되어진다. 또한 침자재군(鍼刺栽群)에서도 T.G.를 감소시키는 유의성있는 흥미로운 결과(結果)를 얻을수 있었다. 5. 혈액학적(血液學的) 검사결과(檢査結果)는 과립구(顆粒球)에서 황금수액제제(黃芩水液製劑)만을 제외한 모든 시술군(施術群)에서 유의성(有意性)있는 증가(增加)를 보였으며, 황금수액희석제제군(黃芩水液稀釋製劑群)들에서의 과립구(顆粒球)는 매우 유의적(有意的)인 증가(增加)(p-value가 <0.001)를 보였다. 침자재군(鍼刺栽群)은 과립구(顆粒球)와 평균혈구용적(平均血球容積)에서 통계적(統計的) 유의성(有意性)이 인정되었다.
