The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

This study was carried out to determine the action mechanism of sulfur-grain larvae extract (SGE) on anti-obesity and the reduction of blood lipid level in high-fat diet induced obese model animals. Animals were classified into a normal diet group (NC, normal control), HFD (high-fat diet without SGE), HFD 15 (high-fat diet + oral administration of 15 mg of SGE extract per 100 g body weight) and HFD 30 (high-fat diet + oral administration of 30 mg of SGE extract per 100 g body weight). The body weight gain declined in HFD 15 and HFD 30 groups compared with the HFD group, even though the diet intake increased significantly. The weight of liver and adipose tissue increased significantly in HFD group compared with in the HFD 15 and HFD 30 groups. Triglyceride, total cholesterol, LDL-C and AI decreased in HFD 15 and HFD 30 groups compared with in the HFD group, but the contents of HDL-C increased significantly. Expression of SREPB-$1{\alpha}$, SREPB-2 mRNA in the liver was lower in the high-fat diet group compared with the HFD group, but the expression of LPL mRNA in adipose tissue and $PPAR{\alpha}$ increased significantly. Fat accumulation in the liver tissues and liver damage were greatly reduced in HFD 15 and HFD 30 groups compared with in the HFD group. The size of adipocytes became smaller in the HFD 15 and HFD 30 groups compared with HFD group. In conclusion, this research discovered for the first time that grain maggot has anti-obesity effects, by reducing the abdominal fat of obese model animals and lowering blood lipid level through the down-regulation of PPAR-$1{\alpha}$ and SREPB-2 mRNA and the up-regulation of PPAR-${\alpha}$ mRNA.
This study aims to investigate changes in plasma lipid concentrations and appetite-regulating hormone levels after a 4% body fat reduction using a 9-week intervention involving aerobic exercise, a fat-oxidizing agent, and diet limitation. After the 9-week intervention, the aerobic exercise plus hydroxycitric acid (EX+HCA), exercise (EX), and diet limitation (DIET) groups achieved the target 4% body fat reduction from the baseline value. None of the plasma lipid indicators showed significant intergroup differences, indicating that plasma lipid levels are not influenced by body weight regulation. With regard to appetite-regulating hormones, no significant intergroup differences were observed in glucose, insulin, or glucagon-like peptide-1 levels, unlike ghrelin and leptin. Ghrelin levels in particular tended to decrease in the DIET group and increase in the HCA+EX and EX groups. Leptin levels significantly decreased in the HCA+EX and EX groups, whereas no differences were observed in the DIET group. Such results indicate that exercise alone without the administration of obesity diet supplements induces elevation in ghrelin levels and reduction in leptin levels, but that diet restriction alone does not influence changes in leptin levels. Taken together, we could not confirm any synergic effects arising from the use of a fat-oxidizing agent during an exercise program to control body weight. Furthermore, diet limitation unsupported by exercise had no effect on muscle mass reduction or appetite-regulating hormone levels; thus, it is not recommended as an effective body weight control method.
The influence of (-0-Hydroxycitrate(HCA), shown to be a competitive inhibitor of adenosine 5-triphosphate(ATP) citrate lyase, on food intake and body weight, serum triglyceride and cholosterol level, in vivo rates of fatty acid and cholesterol synthesis, and fat cell number and size was investigated. 3 groups of female, 5 weeks old Sprague Dawley rats, 8 animals each, were ad libitum meal-fed or pair-fed(3 hours from 10 : 00 to 13 : 00) AIN based high glucose diet for a total period of 8 weeks. Providing normolipidemic rats orally with 400mg of HCA formula containing approximately 20mg of HCA 1 hour prior to daily feeding schedule significantly depressed in vivo hepatic rates of fatty acid and cholesterol synthesis in the liver and adipose tissue. Serum triglyceride and cholesterol levels were significantly reduced by HCA. At the end of treatment period, the rats administered with HCA resulted in a significantly reduction in body weight gain. The reduction in weights was attributable to a significant decrease in fat cell size with a smaller extent, but not significant, reduction in fat cell number. Rats receiving HCA demonstrated less food intake than the controls ; however, this decreased caloric intake was not fully responsible for the HCA induced depression of hepatic and adipocytic lipogenesis, since experiment using pair-fed cojntrol rats showed, less magnitude but similar results. Both a anorectic and an antilipogenic properties of HCA seem to be responsible for this weight reduction activity of HCA. The outcome of this study suggest that metabolic regulation may be a feasible approach to the control of obesity and hyperlipidemia.
Objectives: This study is to investigate life style and weight control based on the body mass index, and tongue diagnosis in adolescent girls. Help controling unnecessary weight and establishing proper understanding of desirable body images and healthy life for adolescents are expected. Methods: The questionnaire surveys and physical examinations were conducted to examine the life style, weight-control behaviors, and the health status of 173 adolescent girls in Jeonju. Among the students surveyed, 162 answered seriously and were accepted as subjects for the study. Results: 1. The mean height was 160.4 cm and weight was 54.21 kg. The mean BMI of the girls was 21.1 kg/m2, which is normal. The percentages of the normal weight group (71.6%), overweight group (16.7%), obesity group (9.9%), underweight group (1.9%) were shown respectively. 2. The oriental medicine doctor examined the tongue diagnosis. The thin and white tongue fur was the most common (20.7%). This diagnosis showed no statistical significance to BMI. 3. The ideal height for the girls was 165 cm, and the ideal weight was 49 kg. 4. A considerable number of girls (50.6%) thought that a thin body figure was the most beautiful body shape for the women. 5. Even though the percentages of an actual overweight or obese group were 26.6%, 42.0% of the girls thought that they were plump. 6. 94.83% of the normal weight group concerned about the weight regulation. 7. Many girls were dissatisfied with their own body shape, and 86.9% of girls regarded being in a shape was important. 70.69% of the normal weight group answered that they were under stress of their body shapes. Conclusions: The findings of this study demonstrate that there are serious misconceptions on body shapes of the adolescent girls, and mostly, unnecessary and inadequate weight control behaviors were done. Therefore, establishing adequately education on their desirable body images and healthy life is needed for the adolescent girls. In this study, there is no significant relationship between BMI and tongue diagnosis. For the number of subjects is not enough, further study on the relationship between the two is expected.

Acupuncture has fairly good weight-reducing effect in treating simple obesity due to the neuroendocrine regulation. In this study, the antiobesity effects of herbal acupuncture(HA) with Sang-hwang(phellinus linteus) at Fuai(SP16) were investigated in the rat fed on high-fat(HF) diet. Sang-hwang mushroom has been proven to have anti-carcinogenic effects and Sang-hwang extracts are highly effective in treatment and preventive treatment of AIDS, diabetes and high blood-pressure. To determine whether the Sang-hwang herbal acupuncture may have the anti-obesity effect, male Sprague-Dawley(4-wk-old) rats were fed a HF diet for 5 wk, which produced significant weight gain compared to rats were fed a normal diet, and then herbal acupuncture were treated for 3 wk in HF diet group. The body weight, food consumption, food effeciency ratio(FER), body fat mass, plasma nitric oxide(NO) were investigated in rats fed on normal diet, HF diet, and HF diet with HA(HF-diet-HA) groups. NO has been proposed to be involved in the regulation of food intake. In addition, the expression of appetite peptides such as orexigenic peptide neuropeptide Y(NPY) and the anorectic peptide cholecystokinin(CCK) were observed in the hypothalamus. HF-HA group reduced body weight gain, FER, body fat contents and NO concentration compared to HF diet group. The expression of NPY was reduced in arcuate nucleus(ARC), and CCK was increased in the paraventricular nucleus(PVN) after treatment of HA. In conclusion, Sang-hwang HA reduced adipocity, plasma NO and hypothalamic NPY, but increased CCK expression in the HF dietinduced obesity rat, therefore HA may have anti-obesity action through regulating body weight and appetite peptide of the central nervous system.
In a previous study, we developed a new food additive as an egg yolk antibody (IgY) against carbohydrate digestion enzymer for the regulation of blood glucose level and weight control. The IgY delayed and decreased the increment if blood glucose level after administration of sucrose in human being by 30% in 20∼30 min. We also developed a lipase inhibitor as a water extract of two kinds of herb, Platycodon grandiflorum and Solanum Melongena, Twenty three volunteers were subjected to the intake of the egg yolk IgY Plus the herbal extracts for 50 days. In average, the treated subjects appeared to lose 1.96 kg of body weight and 3.4 kg of body fat mass during the treated period. Furthermore, Panniculus adiposus and breech size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of egg yolk IgY and natural herbal extract improve the obesity by the decrement of body weight and body fat mass.
During the 5 years period(93-197) of Seoul racecourse Thoroughbred racing, 2i963 holies took an active part in Seoul Racecourse and 61,181 horses entered the racing.400 horses(305 : 1 time, 76 :2 times, 19 :3 times, total : 514 cases) had bled(EIPH) from their nostils after their races. Incidence of bleeding horses from the 21963 racehorses was 13.5%, and incidence of bleeding cases from the 61,181 horses was 0.84%. The average racing frequence (1.05 times/1 month) of bleeders was more than that(0.8 times/1 month) of total race-horses. There was tendency to higher incidence of bleeding at older in age, female in sex, England/ Ireland horses in origin, higher grade in performances heavier in body weight increased, larger in weight to be carried, mid or longer in racing distance, handicap race, spring in season. Relationship did not exist between bleeding and respiratory disease. These results suggested that frequent entering into the race should be controlled. And body weight regulation with conditioning is recommended far reducing incidence of EIPH. It is supposed to prevent bleeding that decrescent performance horses should not be entering long distance or handicap race.
Modification of thyroid hormone levels has a profound effect on skeletal muscle differentiation, predominantly through direct regulation involving thyroid hormone receptors. Nevertheless, little is known about the regulation of myostatin gene expression in skeletal muscle due to altered concentrations of thyroid hormone. Thus, the goal of our study was to find out whether altered thyroid states could change the gene expression of myostatin, the most powerful inhibitor of skeletal muscle development. A hyperthyroid state was induced in rats by daily injections of L-thyroxine 20 mg/100 g body weight for 14 days, while a hypothyroid state was induced in another group of rats by administering methimazole (0.04%) in drinking water for 14 days. After a period of 14 days of L-thyroxine treatment we observed a significant increase of myostatin expression both in mRNA and protein level. However, decreased expression of myostatin mRNA and protein were observed in hypothyroid rats. Furthermore, our studies demonstrated that the upregulation of myostatin gene expression might be responsible for the loss of body weight induced by altered thyroid hormone levels. We concluded that myostatin played a role in a metabolic process in muscle that was regulated by thyroid hormone.

Objective : To suggest a scientific evidence of Aconitum carmichaeli Debx. (Aconiti Lateralis Radix Preparata: ALRP) as one of cooling and heating medicines on the regulation of body temperature, we investigated the effects of ALRP water extract on hypothyroidism. Methods : Hypothyroidism was induced by intradermal injection with PTU for 4 weeks in SD rats. ALRP extract or L-thyroxine as a control drug was orally administrated for 2 weeks with PTU injection in rats. The physiological and serological parameters were measured in rats. The histological change of thyroid tissues was observed by H&E staining, and also the expression of thermo-regulating proteins was determined by Western blot in dorsal root ganglia and brain tissues of rats. Results : The administration of ALRP extract in PTU-induced hypothyroidism rats was significantly increased body temperature, but did not changes on body weight, food and water intake. ALRP extract did not effect on the levels of TSH and T4 in the hypothyroidism rats. ALRP extract significantly decreased the levels of GPT, glucose, HDL-cholesterol, LDL-cholesterol, and total cholesterol in the hypothyroidism rats. In histological observation, the enlarged epithelium and atrophic follicles with higher concentration of follicular cells on hypothyroidism were improved by ALRP extract. In addition, ALRP extract increased the expression of TRPV1 and TRPM8 ion channel proteins in hypothyroidism rats. Conclusion : These results indicate that ALRP extract can improve PTU-induced hypothyroidism through regulation of body temperature and lipid accumulation. The action mechanism of ALRP extract is related with body temperature control by thermoregulation with TRP ion channels.
