Molecular Subtyping of Methicillin-Resistant Staphylococcus aureus Isolated from Patients' Nasal Cavity (환자의 비강으로부터 분리된 메티실린 내성 황색 포도알균의 분자 아형 분석)

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium that causes infections in different parts of the body and causes skin and soft tissue infections (SSTI). The present study examined the antimicrobial resistance patterns and molecular epidemiological characteristics of MRSA isolated from nasal swabs in clinical patients. SCCmec type of MRSA isolates from clinical patients were analyzed: 24 cases were SCCmec type-II; two cases were type-II/IVa; one case was type-II/V; one case was type-IVa; 11 cases were not-typeable. The mec complex type of MRSA isolates from clinical patients were analyzed: 29 cases were mec complex type A, and 10 cases were not-typeable, but type B was not found in the present study. In conclusion, SCCmec type-II and mec complex type A were the most dominant MRSA subtypes among the MRSA isolates from a nasal swab of patients, and the results were similar to other studies on hospital-acquired MRSA (HA-MRSA). These results can not only provide basic data for hospital infection management but also be a good guideline for MRSA infections in the Republic of Korea.

Improvement of Durability and Change of Pore Structure for Concrete Surface by the Penetrative Surface Protection Agent (함침계 표면보호제에 의한 콘크리트 표면의 세공구조 변화 및 내구성 향상)

  • Recently, surface finishing and protection materials were developed to restore performance of the deteriorated concrete and inhibiting corrosion of the reinforcing-bar. For this purpose, surface protection agent as well as coatings are used. Coatings have the advantage of low Permeability of $CO_2,\;SO_2$ and water. However, for coatings such as epoxy, urethane and acryl, long-term adhesive strength is reduced and the formed membrane of those is blistered by various causes. Also when organic coatings are applied to the wet surface of concrete, those have a problem with adhesion. On the other hand, surface protection agent penetrates into pore structure in concrete through capillary and cm make a dense micro structure in concrete as a result of filling effect. Furthermore, the chemical reaction between silicate from surface protection agent and cement hydrates can also make a additional hydration product which is ideally compatible with concrete body. The aim of this study is to examine the effect of penetrative surface protection agent(SPA) by evaluating several concrete durability characteristics. The results show that the concrete penetrated surface protection agent exhibited higher durability characteristics for instance, carbonation velocity coefficient, resistance to chemical attack and chloride ion penetration than the plain concrete. These results due to formation of a discontinuous macro-pore system which inhibits deterioration factors of concrete by changed the pore structure(porosity and pore size distributions) of the concrete penetrated surface protection agent.

Subacute Toxicities of All-trans-Retinoic Acid Encapsulated in the Poly(D,L-Lactide) Microspheres

  • All-trans-retinoic acid (RA) plays essential roles in the regulation of differentiation and proliferation. It has been proved that RA is effective in the treatments of epithelial and hematologic malignancies. However, in spite of its pronounced effects, the clinical applications of RA are limited due to the retinoid acute resistance. Although RA induces complete remission in a high proportion of the patients of acute promyelocytic leukemia (APL), the cancer was relapsed in many patients after a brief remission in spite of a continued RA treatment. Patients who relapsed from remission that was initially induced by RA had clinically resistant to further RA treatment. That is, specific cytochrome P450 enzymes in the liver were induced by the continuous oral administration of RA, thereby accelerating the metabolism of RA. To overcome this problem, biodegradable microspheres were proposed by us, previously. And, several microsphere formulations for RA delivery have been prepared and studied on their effectiveness. Recently, poly(D,L-lactide) (PDLLA) microsphere formulation was optimized, And, from the animal studies by using a mouse and a rat, it have appeared to be effective on both the inhibition of tumor growth and chemoprevention of a carcinogenesis. In this study, subacute toxicities of the PDLLA microsphere formulation have been investigated as a preclinical test. For the test, the microspheres was injected subcutaneously into the back site of rats, and body weight change, clinical signs, hematological changes, blood biochemistry were evaluated. As a result, severe toxicities such as mortality were observed at the dose of 100mg RA/kg, and toxicities were not observed at the dose of 50mg RA/kg, which is the effective dose against carcinogenesis. Bone fracture, observed at several rats, might be inhibited by treating them with anti-inflammatory drugs.

Effects of Walking and Band Exercise on C-reactive Protein and Cardiovascular Disease Risk Factor in Overweight and Obese Children (걷기와 밴드운동이 과체중 및 비만아동의 C-반응성단백질 및 심혈관질환 위험인자의 변화에 미치는 영향)

  • The purpose of this study was to demonstrate the effectiveness of walking and band exercise for 12 weeks on c-reactive protein and cardiovascular disease risk factor in overweight and obese children. Body composition, blood lipids, insulin sensitivity, c-reactive protein (CRP) were assessed before, after 4 weeks and after 12 weeks of combined exercise. Sixteen participants (BMI${\geq}$21.3) were randomly allocated to exercise group (n=8) and control group (n=8). The exercise group participated in 50 minutes of walking exercise and band exercises as resistance training two days a week for 12 weeks. There were significant different on weight (p<0.001), BMI (p<0.001), fat mass (p<0.001), fat percentage (p<0.001), LBM percentage (p<0.001), TG (p<0.05), HDL-C (p<0.01), insulin (p<0.05), HOMA-IR (p<0.05) in exercise group after intervention. And the change of weight (p<0.001), BMI (p<0.001), fat mass (p<0.001), fat percentage (p<0.001), LBM mass (p<0.05), LBM percentage (p<0.001), insulin (p<0.05), HOMA-IR (p<0.05) were significant difference between groups after intervention. These findings suggest that 12 weeks of walking and band exercise can be useful intervention in the improvement of cardiovascular disease risk factor in overweight and obese children. But c-reactive protein was no change.

Preparation and Properties of Silicon Nitride Ceramics by Nitrided Pressureless Sintering (NPS) Process (Nitrided Pressureless Sintering 공정을 이용한 질화규소 세라믹스의 제조 및 특성)

  • The mechanical properties and microstructure and thermal properties of Nitrided Pressureless Sintering(NPS) silicon nitride ceramics, containing three type of $Al_{2}O_3,\;Y_{2}O_3$ sintering additives, were investigated. Also, we have investigated the effect of silicon metal content changing with 0, 5, 10, 15, and $20wt\%$ Si in each composition. In $5wt\%\;Al_{2}O_3,\;5wt\%\;Y_{2}O_3,\;and\;5wt\%$ Si composition, silicon nitride sintered body was successfully densified to a high density. The average 4-point flexural strength and relative density of these specimens were 500 MPa and 98% respectively. Also, Thermal expansion coefficient and thermal conductivity of specimens at room temperature were $2.89{\times}10^{-6}/^{\circ}C\;and\;28W/m^{\circ}C$, respectively. The flexural strength of sintered specimens after thermal shock test of 20,000 cycles was maintained as-received value of 500 MPa.

A Study on the Stability Analysis of Reinforced Embankment on the Soft Ground (연약지반상의 보강성토의 안정해석에 관한 연구)

  • Preloading method is used to prevent the settling of a foundation and to increase the strength of ground by consolidation settlement in advance. But, the embankment used in preloading method brings large deformation and sliding failure in the soft ground. Recently, reinforcement method is often used in embankment in order to prevent sliding failure. But, until now, the research on the stability analysis considering both the rate of strength increase of clay by embankment load and increase of resistance force by the geosynthetics in the embankment body is not found. In this study, the stability analysis program(REAP) for embankment including these two points is developed. By this program(REAP), the stability analysis can be done about during the gradual increase of embankment and the stability counterplan can be established when the safety factor is lower than allowable safety factor of design. After calculating the position of sliding failure surface, the force of geosynthetics which is selected by either the effective tensile strength or tensile force caused by the displacement of soil mass in this position is applied to stability analysis. And the increase of resisting moment can be calculated by this force. Also, the construction period can be estimated and the time for the appropriate counterplan can be decided in order to maintain the stability of embankment. And then, safe and economical embankment design can be performed.

Effect of Microbe Control and Water Temperature on Early Growth and Yield of Soybean Sprouts (콩 종실의 미생물 제어방법과 수주온도에 따른 콩나물의 초기 생육 및 수율)

  • High temperature sterilization method (30min. at $40^{\circ}C$longrightarrow30 min. at $70^{\circ}C$longrightarrow30 min. cooling at $20^{\circ}C$) was evaluated better than that of chloride, ion water, $O_3$ water, and salt water for the controling of micro-organisms at the early stage of sprout culture. Due to the improved germination rate and effective control of micro-organisms at early stage, the method resulted in much lower in the number of micro-organisms, higher in sprout yield, and 2 days longer in quality keeping at market than sprouts prepared by conventional method. There are two methods related with water temperature; constant temperature method (18.5$\pm$ $0.5^{\circ}C$) which keeps the same water temperature during the culture and 3-stage temperature method which changes the water temperature depending on the days after culture. Three-stage temperature method set the temperature at 21$\pm$ $1^{\circ}C$) in the second and third days after the initial acceleration of germination (about 4 hours), at 18.5$\pm$0.5$^{\circ}C$ in the fourth and fifth days, and at 17.0$\pm$ $0.5^{\circ}C$) in the sixth and seventh days. Three-stage temperature method could enhance the resistance power to diseases at the early growth stage, control the growth to standard goods (8-9cm in length and 2.15-2.30mm in width), and keep good body color at the middle or final stage. This method also increased the sprout yield by 6% compared with the conventional method, constant temperature method.

The Clinical Implications of Hepatic Enzymes in Metabolically Healthy Obese Men (대사적으로 건강한 비만남성에서 간 효소의 임상적 의의)

  • Increased hepatic enzymes are associated with insulin resistance, metabolic complications, and type 2 diabetes mellitus. Metabolically healthy obese (MHO) phenotype is not accompanied by metabolic complications and maintains insulin sensitivity, despite excessive body fat. The purpose of this study was to evaluate the clinical implications of hepatic enzymes in MHO men. The diagnostic criteria for MHO were based on NCEP-ATP III and obesity in adults was defined using WHO Asian-Pacific criteria. We used the data from 9,683 obese men aged between 20 and 70 years. The subjects were divided into three groups according to the diagnostic criteria: The metabolically healthy non-obese (MHNO, N=2,878), metabolically healthy obese (MHO, N=5,427), and metabolically abnormal obese (MAO, N=1,378). Obesity criteria were classified according to the standards set forth by WHO Asia-Pacific Criteria. AST, ALT, and GGT were significantly lower in the MHO group than in the MAO group (p<0.001, respectively). However, the hepatic enzyme levels were higher in the MHO group than in the MHNO group (p<0.001). Liver enzymes were associated with metabolic syndrome risk factors. Waist circumference, fasting glucose, total cholesterol, triglyceride, and HDL-C were risk factors for metabolic syndrome affecting liver enzymes. In conclusion, hepatic enzymes were found to predict metabolic abnormalities in metabolically healthy obese men.

Changes in Neuropeptide Y-Immunoreactive Cells in the Hypothalamus and Cajal Interstitial Cells in the Small Intestine of Rats with High-Fat Diet (고지방식이에 의한 흰쥐의 시상하부 Neuropeptide Y-면역반응 신경세포와 장내 Cajal 세포의 변화)

  • Purpose: The aim of this study was to assess changes in neuropeptide Y (NPY) immunoreactivity in the hypothalamus and interstitial cells of Cajal (ICC) in the small intestine of rats fed high-fat diets (HFD). Methods: Male Sprague-Dawley rats (200~250 g body weight) were randomly divided into two groups, which were the control group (normal chow diet for 6 weeks), and the HFD group (rodent diet with 60% kcal fat for 6 weeks). The immunoreactivity of NPY in the hypothalamus and ICC in the small intestine was evaluated after every feed for 6 weeks. Results: NPY immunoreactivity was observed strongly in the hypothalamic nuclei in the HFD group compared to the control group. The numbers of NPY-immunoreactive (IR) cells were significantly higher in the paraventricular hypothalamic nucleus in the HFD group than in the control group. In the region of Auerbach's plexus (AP) of small intestine, the staining intensity of the ICC-IR cells was reduced in the HFD group compared to the control group. The numbers of ICC in the small intestine with HFD, including ICC in the inner circular and outer longitudinal muscle were significantly lower than in the control group. Conclusion: This study suggested that increasing NPY-IR cells in the hypothalamus may reflect resistance of NPY action after a HFD, and decreasing ICC-IR cells in the small intestine after a HFD is functionally significant in gastrointestinal motility.

A Study on the Effect of Sangbaegpitang & Supungsungiwhan on the Glucose Metabolism of db/db Mice (상백피탕(桑白皮湯)과 수풍순기환(搜風順氣丸)이 db/db Mice의 당대사(糖代謝)에 미치는 영향(影響))

  • In this study, body weight levels of glucose, insulin and triglyceride in blood and glucosidase activity of the small intestine were investigated to determine the effect of Sangbaegpitang and Supungsungiwhan on the glucose metabolism of db/db mice. The GLUT4 mRNA of muscle tissue and the Acetyl CoA Carboxylase and the activation rate of GLUT2 mRNA of liver tissue were measured by the reverse transcription-polymerase chain reaction method and by the vitro transcription. The results were obtained as follows: 1. In the Sangbaegpitang administration group, (1) The level of triglyceride was decreased significantly and the glucosidase activity of the small intestine was inhibited remarkably, (2) The amounts of the GLUT4 mRNA in muscle tissue and Acetyl CoA Carboxylase mRNA in liver tissue were increased significantly. (3) Though glucose level in both fasting and non-fasting, were decreased and the insulin level in blood was increased, the results showed no statistical significance. 2. In the Supungsungiwhan administration group, (1) The levels of glucose and triglyceride were decreased significantly in the blood of non-fasting animals. (2) The glucosidase activity of small intestine was inhibited markedly and the amounts of GLUT4 mRNA of muscle tissue and GLUT2 mRNA of liver tissue were increased significantly. (3) The glucose levels in the fasting group were reduced, while insulin level was increased but showed no statistical significance, Based on the above results, our conclusions are as follows: Sangbaegpitang & Supungsungiwhan are thought to be capable of inhibiting the activity glucosidase, the enzyme which influences carbohydrate metabolism in the small intestine of db/db mice(the experimental diabetic model) and delaying the absorption of carbohydrate, thus proving effective on inhibiting the increase of non-fasting glucose level effectively. Futhermore Sangbaegpitang and Supungsungiwhan are though: to be capable of preventing the composition of free fatty acids by restoring the production of GLUT4 mRNA of muscle tissues and GLUT2 mRNA of liver tissues. Those results suggests that above prescriptions can be applied to non-insulin dependent diabetes mellitus in order to improve insulin resistance.
