The mixotrophic dinoflagellate Cochlodinium polykrikoides was reported to be linked to major fish kills in Korea and Japan since the 1990s. Rapid and sensitive detection of microalgae has been problematic because morphological identification of dinoflagellates requires light microscopic and scanning electron microscopic observations that are time consuming and laborious compared to real-time PCR. To address this issue, a real-time PCR probe targeting the ITS2 rRNA gene was used for rapid detection and quantification of C. polykrikoides. PCR inhibitors in water column samples were removed by dilution of template DNA for elimination of false-negative reactions. A strong association between cell quantification using real-time PCR and microscopic counts suggests that the real-time PCR assay is an alternative method for cell estimation of C. polykrikoides in environment samples.

The research on Harmful Algal Blooms is generally in progress through field work, such as the naked eye and sampling. It was difficult to forecast exactly the course, from appearance of red tide to disappearance, with the established ways of investigation and analysis. Accordingly it is need to analyze environmental factors in time and space, the appearance of red tide and the path of its migration by more objective and scientific methods. In this study the remote sensing was used to diminish damage from the occurrence of red tide. Such as a temperature change of sea water and a change of tidal currents, the major cause for red tide. The probed data were utilized. The technique for forecast of red tide phenomenon on the south coast was researched by analyzing the cause of red tide, pollutant flowed from landand the possibility of application of the technique was showed.
Since algae had been issued an environmental problem, water blooms, deepened due to increase of retention water basin in Korea as well as a biomass resource for producing biofuel, this study conducted a series of experiments for Spirulina platensis using the flotation process with micro-bubble. To elevate utilization of collected-algae, this study focused on omitting or minimizing coagulant's doses as changing a cultivation period and condition affected on physical property change of algae. Two coagulants, PAC and Chitosan, were used to test the collecting rate of algae and the result found no difference between two rates. For flotation experiments without adding the coagulant, dried algae weight (passing 14 days after cultivation for 20 days) detected high separation efficiency 98.2 % and it (passing 7 days after long-term cultivation for 28 days) presented good separation efficiency 91.9 %. Chlorophyll's separation efficiency showed a similar tendency with the case of the dried algae weight. In endogeny conditions, a light source and a carbon source were not considerably affected on the flotation separation efficiency. Thus, this study confirms that algae biomass may be collected without the coagulant during the endogeny condition period after enough cultivation time, 3 weeks.
For the test organism of algal growth potential (AGP), the diatom in the genus Stephanodiscus which cause blooms in the Nakdong River was used instead of generally used strains of Selenastrum, Microcystis, or Anabaena. AGP results indicated that all the samples in the Nakdong River except for that from the Nakdan Bridge site were eutrophic state. Furthermore, the sample from Kumho River site was hypertrophic state. In the main stream Nakdong River, the value of AGP was lowest at the upstream Nakdan Bridge site and was highest at Koryoung Bridge site which is just downstream of Kumho River confluent point indicating the seriousness of pollution contributed by the Kumho River to the Nakdong River. Changes in the concentration of nutrients before and after the AGP tests and inter-relationship among the nutrients indicated that the growth of the Stephanodiscus in the AGP tests were mostly affected by the nitrate, silicate and phosphate. The limiting nutrient was identified by the nutrient addition experiments and the results showed that phosphate was the limiting nutrient for the growth of Stephanodiscus in the tested samples.
This paper demonstrates the possible nondestructive analysis of iron artifacts' metallurgical characteristics using neutron imaging. Ancient kingdoms of the Korean Peninsula used a direct smelting process for ore smelting and iron bloom production; however, the use of iron blooms was difficult because of their low strength and purity. For reinforcement, iron ingots were produced through refining and forge welding, which then underwent various processes to create different iron goods. To demonstrate the potential analysis using neutron imaging, while ensuring artifacts' safety, a sand iron ingot (SI-I) produced using ancient traditional iron making techniques and a sand iron knife (SI-K) made of SI-I were selected. SI-I was cut into 9 cm2, whereas the entirety of SI-K was preserved for analysis. SI-I was found to have an average grain size of 3 ㎛, with observed α-Fe (ferrite) and pearlite with a body-centered cubic (BCC) lattice structure. SI-K had a grain size of 1-3 ㎛, α-Ferrite on its backside, and martensite with a body-centered tetragonal (BCT) structure on its blade. Results show that the sample's metallurgical characteristics can be identified through neutron imaging only, without losing any part of the valuable artifacts, indicating applicability to cultural artifacts requiring complete preservation.
The purpose of this study is to analyze the characteristics and correlations of the Yeongcheon Lake in order to reduce the occurrence of harmful cyanobacteria. In this study, we investigated the water quality and phytoplankton of the lake from May to November in 2017. Correlation and data mining analyses were performed to analyze the relationship between the two factors. The water temperature was lowest at the point where the Yeongcheon Lake inflow occurs at Imha Lake. It was highest at the point where the outflow occurs to Angye Lake. The pH was also highest at the outflow point, but in the case of DO, it was highest at the midpoint between the inflow and outflow. The main cyanobacteria that emerged during the study period were Oscillatorialimosa, Microcysti saeruginosa and Aphanizomenon flos-aquae. As a result of correlation analysis, the water temperature, inflow, COD loading, TOC loading at the inflow point of the Yeongcheon Lake were the items that were related to the harmful cyanobacteria. The data mining analysis indicated that the TP loading and harmful cyanobacteria in the inflow point of the Yeongcheon Lake were influential on the detrimental cyanobacteria in the Yeongcheon Lake outflow point. When the TP loading was less than 39.0 kg/day at the inflow site, it was expected that the amount of harmful cyanobacteria could be maintained below 10,000 cells/mL.
The present study assessed the efficient removal of nutrients and Chlorophyll-a (Chl-a) by using methyl esterified sericite (MES) and pine needle extracts (PNE), a low cost and abundant green hybrid material from nature. For this purpose, the optimal conditions were investigated, such as the pH, temperature, MES and PNE ratio, and MES-PNE dose. In addition, a Microcystis aeruginosa control using MES-PNE was also analyzed with various inhibition models. The removal of the nutrient and Chl-a onto MES-PNE was optimized for over 95% removal as follows: 2-2.5 for the MES-PNE ratio, 7-8 pH and a $22-25^{\circ}C$ temperature. In this respect, approximately 1.52-2.20 g/L of MES-PNE was required to remove each 1 g of dry weight/L of Chl-a. Total phosphorus (TP) has a greater influence on the increase in Chl-a than total nitrogen (TN) according to the correlation between TN, TP and Chl-a. Moreover, the Luong model was the best model for fitting the biodegradation kinetics data from Chl-a on MES-PNE from lake water. The novel hybrid material MES-PNE was very effective at removing TN, TP and Chl-a from the lake and can be applied in the field.
In this study, we designed a data-driven model to predict chlorophyll-a using M5P model tree and extreme learning machine (ELM). The Juksan weir in the Youngsan River has high chlorophyll-a, which is the primary indicator of algal bloom every year. Short-term algal bloom prediction is important for environmental management and ecological assessment. Two models were developed and evaluated for short-term algal bloom prediction. M5P is a classification and regression-analysis-based method, and ELM is a feed-forward neural network with fast learning using the least square estimate for regression. The dataset used in this study includes water temperature, rainfall, solar radiation, total nitrogen, total phosphorus, N/P ratio, and chlorophyll-a, which were collected on a daily basis from January 2013 to December 2016. The M5P model showed that the prediction model after one day had the highest performance power and dropped off rapidly starting with predictions after three days. Comparing the performance power of the ELM model with the M5P model, it was found that the performance power of the 1-7 d chlorophyll-a prediction model was higher. Moreover, in a period of rapidly increasing algal blooms, the ELM model showed higher accuracy than the M5P model.
The aim of this review is to introduce the progress in trauma surgery made during war. In the 16th century, Paré reintroduced ligature of arteries, which had been introduced by Celsus and Galen, instead of cauterization during amputation. Larrey, a surgeon in Napoleon's military, adapted the "flying artillery" to serve as "flying ambulances" for rapid transport of the wounded. He established rules for the triage of war casualties, treating wounded soldiers according to the seriousness of their injuries and the urgency of medical care. To treat fractures and tuberculosis, Thomas created the "Thomas splint", which was used to stabilize fractured femurs and prevent infection; in World War I (WWI), use of this splint reduced the mortality of compound femur fractures from 87% to less than 8%. During WWI, Cushing systematized the treatment of head injuries, reducing mortality among head injury patients. Gillies repaired facial injuries, and his experiences became the basis of craniofacial and aesthetic surgery. In WWII, McIndoe discovered that immersion in saline promoted burn healing and improved survival rates, and thus began saline baths and early grafting instead of using tannic acid. Harmful algae blooms have become a serious environmental problem in major river basins in Korea. They are known to produce various algal organic matters (AOMs) including intracellular organic matters (IOMs) and extracellular organic matters (EOMs). Generally AOMs cannot be easily removed by coagulation/flocculation process in conventional drinking water plants. AOMs produced by blue-green algae also include various toxins such as Microcystins, Anatoxin-a, and Saxitoxin known to have harmful effects on living organisms in aquatic environment. In this study, toxic effects of EOMs produced by three different algae species (Microcystis sp., Anabaena sp., and Oscillatoria sp.) on zebrafish were investigated using electroencephalography (EEG) recording method, a technology for recording brain activity. Electroencephalographic changes in zebrafish revealed that a low EOM had a negative effect on zebrafish compared to both Anabaena sp. and Oscillatoria sp. at 30 ppm EOM exposures. This result might be due to Microcystins present in EOMs produced by Microcystis sp. As a result of power spectrum density anallysis, exposure to EOMs produced by Microcystis sp. caused a state of vigilance in zebrafish. This EEG based toxicity test can be used to examine effects of harmful materials at low levels on living organisms in an aquatic system.
